Assessment of anticancer properties of essential oils from aromatic plants of the sand dunes of peniche (portugal)
O cancro é um problema de saúde crescente no mundo e é a segunda causa de morte depois das doenças cardíacas. De acordo com a Agência Internacional de Investigação em Cancro (IARC) existem atualmente mais de 10 milhões de casos de cancro por ano no mundo.
Os produtos naturais oferecem oportunidades de inovação na descoberta de novos fármacos. Neste sentido, os compostos naturais isolados a partir de plantas medicinais, como potenciais fontes de novas drogas anticancerígenas, têm tido um interesse crescente.
Os Óleos Essenciais (OEs) são sintetizados pelas plantas e têm sido estudados pelas suas inúmeras atividades biológicas, incluindo anticancerígena, anti-inflamatória, antimicrobiana, antiviral, antioxidante e repelente de insetos.
Este estudo tem como objetivos determinar a eficácia de OEs de seis espécies de plantas das dunas de Peniche (Portugal), como potenciais agentes terapêuticos anticancerígenos em linhas celulares de cancro da mama (MCF7) e do colo-rectal (RKO), assim como perceber o mecanismo de ação destes OEs.
Neste estudo, partes aéreas de Artemisia campestris subsp. maritima, Crithmum maritimum, Eryngium maritimum, Juniperus turbinata subsp. turbinata, Otanthus maritimus e Seseli tortuosum foram colhidas na praia da Consolação, em Peniche (Portugal), e os seus OEs isolados através de hidrodestilação. A composição química dos OEs foi investigada por cromatografia gasosa (GC) e por cromatografia gasosa com espetrofotometria de massa (GC-MS) e os compostos maioritários foram descritos para cada óleo.
Para avaliar a atividade anticancerígena nas linhas celulares MCF7 e RKO, o método MTS (3- (4, 5-dimethyl- 2 -thiazolyl) - 2, 5-dyphenyl-2H-tetrazolium bromide) foi usado e a viabilidade celular avaliada, através de diluições sucessivas, a concentrações iniciais de 5 μL/mL e 1 μL/mL, com diluição de 1:2 e 1:10, respetivamente, comparando com o controlo (DMSO). De todos os OEs testados, a atividade anticancerígena foi descrita, em ambas as linhas celulares, como observado pela diminuição da viabilidade/proliferação celular – exceto o OE Eryngium maritimum a uma concentração inicial de 5 μL/mL.Com o objetivo de avaliar o mecanismo biológico de ação dos OEs, foi realizado um western blot para marcadores relativos ao bloqueio do ciclo celular e apoptose (p53, p21 e caspase 3 clivada), para Seseli tortuosum e Otanthus maritimus. Foi observado um aumento do nível proteína p53 nas células tratadas com estes OEs, sugerindo a indução de stress celular nas células cancerígenas testadas. No entanto, não foi observada caspase 3 clivada, sugerindo que a apoptose não terá sido a causa para a diminuição da viabilidade/proliferação celular observada. Foi ainda observado o aumento da expressão da p21 com os OEs selecionados, sugerindo que o tratamento com OE está associado ao bloqueio do ciclo celular. Para validar estas observações, a análise realizada por FACS, depois do tratamento indica um possível bloqueio do ciclo celular na fase G1.
Concluindo, a concentração inicial de 5 μL/mL revelou ser muito tóxica para as linhas celulares testadas. No entanto, a uma concentração final de 1 μL/mL foi demonstrada uma diminuição da viabilidade/proliferação celular para todos os OEs.
No estudo preliminar do mecanismo de ação dos OEs, foi demonstrado, face à presença da p21, que os óleos de Seseli tortuosum e Otanthus maritimus atuam bloqueando o ciclo celular. Para comprovar estes resultados, o FACS realizado (apenas no OE de Seseli tortuosum) revelou que este bloqueio pode ocorrer, pelo aumento da percentagem de células observadas, na fase G1.
Estes resultados demonstram o interesse destes OEs de Peniche na procura de novos agentes quimo preventivos contra a progressão do cancro da mama e colo-rectal
Óbitos Por Causas Externas e Acidentes de Trabalho no Município da Serra-es.
Estudo descritivo das mortes por causas externas, ocorridas entre 2000 e 2005,
entre homens e mulheres, de 10 anos ou mais de idade, residentes no município da
Serra, Espírito Santo, com objetivos de descrever o perfil da população ocupada;
descrever o padrão dos óbitos por causas externas segundo as variáveis: tipo de
causa, sexo, faixa etária e ocupação; e identificar as relações existentes entre os
óbitos por causas externas e os acidentes de trabalho. Foram utilizadas bases de
dados oficiais. Os resultados obtidos foram analisados em dois períodos agregados:
2000 a 2002 e 2003 a 2005. Observou-se que em cinco anos o número de
empregos formais no município aumentou 85,07% enquanto a população geral
aumentou 19,31%, destacando o setor da construção como o que mais contribuiu
com a expansão, sendo responsável por 25,55% do acréscimo. Observou-se ainda
que, no período de 2000 a 2002, as causas externas foram a principal causa de
morte respondendo por 30,1% das mortes e, para os anos de 2003 a 2005, a
segunda causa com 28,2% dos óbitos. Quanto ao sexo, encontrou-se que, em
média, morreram oito homens para cada mulher por causas externas. O impacto dos
homicídios permanece superior na população masculina - variando de 72,2% a
73,61% entre os dois períodos estudados - e na faixa etária de 15 a 19 anos - com
83,56% e 89,18%, respectivamente - diminuindo nas faixas de maior idade. Para a
maioria das ocupações, a causa externa representa a maior proporção de mortes,
sendo os homicídios responsáveis por 67,22% das mortes nos anos de 2000 a 2002
e 69,70% nos anos de 2003 a 2005. Os estudantes representaram a maior
proporção de óbitos por causas externas nos dois períodos, com 12,81% e 12,07%,
respectivamente. Entre 2000 e 2002, observaram-se percentuais de 10,4% para
trabalhadores braçais, 9,94% para refrataristas e 5,01% para tratoristas. Nos anos
de 2003 a 2005, houve um aumento de 14,36% nos óbitos por causas externas,
sendo os refrataristas com 11,74%, trabalhadores braçais com 9,93% e protocolistas
com 4,52%. Em média, 98% dos óbitos não possuem informação declarada sobre
acidente de trabalho. Os acidentes de trabalho declarados encontravam-se na
categoria outros acidentes (51,43%), acidentes de transportes (25,71%) e quedas
(22,86%); para 14,29% dos óbitos não havia informação sobre a ocupação.
Observou-se que os acidentes de trabalhos são identificados nas ocupações dos

setores industriais e da construção civil, não havendo visibilidade para os acidentes
que ocorrem no contexto da rua. O perfil de óbitos por causas externas encontrado
pode ser explicado pelas características do município marcado pela industrialização
e urbanização tardias; pelo desenvolvimento econômico recente, onde as indústrias
de transformação são motores para a ampliação dos empregos nos setores da
construção civil, do comércio e da prestação de serviços; pela precariedade de
vínculos e pela informalidade, que criam condições estruturais para as
desigualdades sociais. Neste cenário, os acidentes de trabalho constituem a outra
face a ser enfrentada, com a melhoria da produção de informações no nível local,
visando instrumentalizar ações efetivas para seu controle
Consumption: the elephant in the room: Understanding, changing, and reducing consumption for sustainability
The consumption of goods and services is an understudied key contributor to the climate and environmental crises. In this dissertation we investigated how to change and reduce consumption for sustainability, from a broad multi-disciplinary perspective. First, based on a ten-discipline review of what influences consumption, we present a framework emphasizing that consumption, much more than driven by “consumers”, is a complex phenomenon that arises from the (inter)actions of multiple agents and contexts. Governments, businesses, marketers, producers, and media are some of these actors. They influence and are influenced by social, cultural, geographic, infrastructural, technological, political, institutional, economic, and historical contexts. Historical research indicates that the shift to a consumer society was a deliberate process set with the intention of stimulating the growth-economy. As a next step we investigated what it takes to reduce consumption. We concluded that it requires abstaining from consumption, but also consuming differently (e.g., second-hand, good-quality, non-disposable), engaging in low-consumption practices (e.g., sharing, repairing, self-provisioning) and contexts that enable low-consumption living. The remaining chapters of the dissertation focused on how to change food systems for sustainability, by investigating in detail actors and contexts of Alternative Food Networks (AFNs). We interviewed organisers of AFNs in three countries (Portugal, Poland, and the Netherlands) to understand the factors that support or hinder the development of diverse types of AFNs. Finally, in a case-study of a non-profit food co-op supermarket we studied how food waste is managed (quantitatively and qualitatively) and showed how it differed from conventional retail
Existence and Complexity of Approximate Equilibria in Weighted Congestion Games
We study the existence of approximate pure Nash equilibria (-PNE) in
weighted atomic congestion games with polynomial cost functions of maximum
degree . Previously it was known that -approximate equilibria always
exist, while nonexistence was established only for small constants, namely for
-PNE. We improve significantly upon this gap, proving that such games in
general do not have -approximate PNE, which provides
the first super-constant lower bound.
Furthermore, we provide a black-box gap-introducing method of combining such
nonexistence results with a specific circuit gadget, in order to derive
NP-completeness of the decision version of the problem. In particular,
deploying this technique we are able to show that deciding whether a weighted
congestion game has an -PNE is NP-complete. Previous
hardness results were known only for the special case of exact equilibria and
arbitrary cost functions.
The circuit gadget is of independent interest and it allows us to also prove
hardness for a variety of problems related to the complexity of PNE in
congestion games. For example, we demonstrate that the question of existence of
-PNE in which a certain set of players plays a specific strategy
profile is NP-hard for any , even for unweighted congestion
games.
Finally, we study the existence of approximate equilibria in weighted
congestion games with general (nondecreasing) costs, as a function of the
number of players . We show that -PNE always exist, matched by an almost
tight nonexistence bound of which we can again transform into
an NP-completeness proof for the decision problem
The Different Shades of Infinite Session Types
Many type systems include infinite types. In session type
systems, infinite types are important because they specify communication protocols that are unbounded in time. Usually infinite session
types are introduced as simple finite-state expressions rec X.T or by nonparametric equational definitions X
.= T. Alternatively, some systems of
label- or value-dependent session types go beyond simple recursive types.
However, leaving dependent types aside, there is a much richer world of
infinite session types, ranging through various forms of parametric equational definitions, to arbitrary infinite types in a coinductively defined
space. We study infinite session types across a spectrum of shades of grey
on the way to the bright light of general infinite types. We identify four
points on the spectrum, characterised by different styles of equational
definitions, and show that they form a strict hierarchy by establishing
bidirectional correspondences with classes of automata: finite-state, 1-
counter, pushdown and 2-counter. This allows us to establish decidability
and undecidability results for type formation, type equivalence and duality in each class of types. We also consider previous work on context-free
session types (and extend it to higher-order) and nested session types,
and locate them on our spectrum of infinite types

Photoluminescence Study Of Interfaces Between Heavily Doped Al 0.48in 0.52as:si Layers And Inp (fe) Substrates
Properties of the interface between the epitaxial layer of heavily doped Al 0.48In 0.52As:Si and the InP(Fe) substrate are investigated by photoluminescence in AlInAs:Si/InP(Fe) heteroestructures grown by molecular beam epitaxy. The effect on heterostructure optical properties of including a thin Al 0.22Ga 0.26In 0.52As:Si layer at the interface is investigated as well. To explain the different interface emission energies observed, the results are analyzed by using the mixed-type I-II interface model, which considers in the type II interface a narrow InAs well, with variable width, between AlInAs and InP. The observation of the interface emission at energies as high as 1.36 eV, at low excitation intensity, is explained taking into account the high doping level of the samples. Este proyecto se presentó por demanda de un grupo de personas nucleadas en el Barrio San Carlos de la Plata (Las Quintas y El Triunfo) que a través del Programa “Barrio Adentro” dependiente del Organismo Provincial de Niñez y Adolescencia, perteneciente al ministerio de Desarrollo de la Comunidad de la Provincia de Buenos Aires, quienes detectaron la necesidad de implementar acciones sanitarias en niñas y niños jóvenes socialmente vulnerables y sus familias. Las acciones se abocaron en restituir piezas dentarias perdidas a través de elementos protésicos. Objetivos: Elevar los niveles de salud de la población realizando tratamientos que contemplen la eliminación de patologías orales y la posterior confección, prueba e instalación de prótesis completas y parciales de acrílico.Facultad de Odontologí
2015/16 seasonal vaccine effectiveness against hospitalisation with influenza a(H1N1)pdm09 and B among elderly people in Europe: Results from the I-MOVE+ project
We conducted a multicentre test-negative caseâ\u80\u93control study in 27 hospitals of 11 European countries to measure 2015/16 influenza vaccine effectiveness (IVE) against hospitalised influenza A(H1N1)pdm09 and B among people aged â\u89¥ 65 years. Patients swabbed within 7 days after onset of symptoms compatible with severe acute respiratory infection were included. Information on demographics, vaccination and underlying conditions was collected. Using logistic regression, we measured IVE adjusted for potential confounders. We included 355 influenza A(H1N1)pdm09 cases, 110 influenza B cases, and 1,274 controls. Adjusted IVE against influenza A(H1N1)pdm09 was 42% (95% confidence interval (CI): 22 to 57). It was 59% (95% CI: 23 to 78), 48% (95% CI: 5 to 71), 43% (95% CI: 8 to 65) and 39% (95% CI: 7 to 60) in patients with diabetes mellitus, cancer, lung and heart disease, respectively. Adjusted IVE against influenza B was 52% (95% CI: 24 to 70). It was 62% (95% CI: 5 to 85), 60% (95% CI: 18 to 80) and 36% (95% CI: -23 to 67) in patients with diabetes mellitus, lung and heart disease, respectively. 2015/16 IVE estimates against hospitalised influenza in elderly people was moderate against influenza A(H1N1)pdm09 and B, including among those with diabetes mellitus, cancer, lung or heart diseases
