    Improving Evidence-Based Practice education in healthcare courses: A Participatory Action Research multiple-case study

    [EN] This paper synthesises the results of three participatory action research (PAR) studies undertaken to improve the integration of evidence-based practice (EBP) education in three undergraduate health courses at one Australian university: Bachelor of Nursing, Bachelor of Occupational Therapy, and Bachelor of Physiotherapy. The PAR process with interested academics uncovered a range of EBP education strengths and weaknesses in the three courses. Common themes were evident, which are likely to be applicable in other similar courses. Identified weaknesses included a lack of explicit teaching about the meaning, principles, steps, and importance of EBP, partly stemming from a lack of shared understanding. A relative lack of emphasis on certain EBP steps was also noted, particularly the first step of ‘asking’ questions. A lack of communication with workplace learning (WPL) supervisors about how to facilitate EBP was also noted, raising concerns about variable EBP-education quality across WPL settings. Opportunities for improvement were identified by academics in each course, across multiple subjects and year levels. In our experience, PAR has been a highly constructive approach to EBP curriculum improvement.     The nocturnal activity of a commonly housed rodent: How African pygmy dormice (Graphiurus murinus) respond to an enriched environment

African pygmy dormice (Graphiurus murinus) are typical in this respect. This research aimed to determine the effect of environmental enrichment on the behavior (including stereotypical scratching at the glass walls of the enclosure) and space use of a group of eight African pygmy dormice at Sparsholt College Hampshire, UK. An apple-wood climbing grid and three raised (at various heights above the substrate) woven-wicker nest boxes were provided. Instantaneous scan sampling was used to record 150 hours of nocturnal behavior (19:00 – 07:00 daily) over five experimental phases (Phase 1 baseline; Phase 2 climbing grid provided; Phase 3 lower nest box provided; Phase 4 middle nest box provided; Phase 5 higher nest box provided). Space use was determined using the modified Spread of Participation Index. Nest box use was recorded continually. The provision of the climbing grid significantly increased the groups’ time spent eating, digging, gnawing and climbing, and significantly decreased stereotypic scratching at glass. It also significantly changed the use of all enclosure zones, with mice utilizing the highest zones as soon as they were accessible. The addition of raised nesting opportunity saw the highest zones of the enclosure become those preferentially used. It also totally diminished stereotypic scratching at glass. The highest nest box was preferentially used and use of terrestrial nest boxes (those placed directly on top of the substrate) reduced significantly when raised alternatives were provided. This study suggests those working with African pygmy dormice should provide an enriched enclosure via ‘arboreal’ opportunity to increase active behaviors and reduce stereotypy

    Pebbles and avalanches

    This joint exhibition of ten artists from the University of Lincoln was curated by Clare Charnley and used artworks to draw attention to the complex dynamics of the two-way process of influence between teachers and students of art. This show included stills from one of my video pieces - ‘Domain of formlessness’, which was inspired by Steve Dutton whom I met whilst doing my Masters. The video relates (quite literally) to the notion of ‘avalanches’ in the title of this exhibition and the film stems from a conversation that started between Steve and myself (and continues to this day) about the catastrophes that result from trying to deal with landslides of material from abandoned artistic activities which often end up strewn chaotically across the studio. Dutton and Peacock made a film called ‘Plague-Orgy-Time’ in 1997 and I first saw it in their exhibition ‘Apocatropes’ at the Mappin Gallery in the same year. I was taken by their approach to a ‘series of accretions of things in a space punctuated by scattered evidence of artistic activity’ (Glover, I. 1997). My film deals with a similar issue but in this case the studio is reduced to a model and artistic activities are miniaturised in a series of ‘Gulliver-esque’ tableaux or enactments – each one punctuated by the close of a stage curtain. In a way my film consciously tends towards 'the reproduced', the constructed stage, film or theatre set in its depictions of a series of avalanches and visually references old slapstick humour films in a sequence of ‘vignettes’.     XLF-Cernunnos promotes DNA ligase IV–XRCC4 re-adenylation following ligation

    XLF-Cernunnos (XLF) is a component of the DNA ligase IV–XRCC4 (LX) complex, which functions during DNA non-homologous end joining (NHEJ). Here, we use biochemical and cellular approaches to probe the impact of XLF on LX activities. We show that XLF stimulates adenylation of LX complexes de-adenylated by pyrophosphate or following LX decharging during ligation. XLF enhances LX ligation activity in an ATP-independent and dependent manner. ATP-independent stimulation can be attributed to enhanced end-bridging. Whilst ATP alone fails to stimulate LX ligation activity, addition of XLF and ATP promotes ligation in a manner consistent with XLF-stimulated readenylation linked to ligation. We show that XLF is a weakly bound partner of the tightly associated LX complex and, unlike XRCC4, is dispensable for LX stability. 2BN cells, which have little, if any, residual XLF activity, show a 3-fold decreased ability to repair DNA double strand breaks covering a range of complexity. These findings strongly suggest that XLF is not essential for NHEJ but promotes LX adenylation and hence ligation. We propose a model in which XLF, by in situ recharging DNA ligase IV after the first ligation event, promotes double stranded ligation by a single LX complex

    Orbital effects of a monochromatic plane gravitational wave with ultra-low frequency incident on a gravitationally bound two-body system

    We analytically compute the long-term orbital variations of a test particle orbiting a central body acted upon by an incident monochromatic plane gravitational wave. We assume that the characteristic size of the perturbed two-body system is much smaller than the wavelength of the wave. Moreover, we also suppose that the wave's frequency is much smaller than the particle's orbital one. We make neither a priori assumptions about the direction of the wavevector nor on the orbital geometry of the planet. We find that, while the semi-major axis is left unaffected, the eccentricity, the inclination, the longitude of the ascending node, the longitude of pericenter and the mean anomaly undergo non-vanishing long-term changes. They are not secular trends because of the slow modulation introduced by the tidal matrix coefficients and by the orbital elements themselves. They could be useful to indepenedently constrain the ultra-low frequency waves which may have been indirectly detected in the BICEP2 experiment. Our calculation holds, in general, for any gravitationally bound two-body system whose characteristic frequency is much larger than the frequency of the external wave.     DNA non-homologous end-joining (NHEJ) is the major DNA double strand break (DSB) repair pathway inmammalian cells. Defects in NHEJ proteins confer marked radiosensitivity in cell lines and mice models,since radiation potently induces DSBs. The process of V(D)J recombination functions during the devel-opment of the immune response, and involves the introduction and rejoining of programmed DSBs togenerate an array of diverse T and B cells. NHEJ rejoins these programmed DSBs. Consequently, NHEJdeficiency confers (severe) combined immunodeficiency – (S)CID – due to a failure to carry out V(D)Jrecombination efficiently. NHEJ also functions in class switch recombination, another step enhancing Tand B cell diversity. Prompted by these findings, a search for radiosensitivity amongst (S)CID patientsrevealed a radiosensitive sub-class, defined as RS-SCID. Mutations in NHEJ genes, defining human syn-dromes deficient in DNA ligase IV (LIG4 Syndrome), XLF-Cernunnos, Artemis or DNA-PKcs, have beenidentified in such patients. Mutations in XRCC4 or Ku70,80 in patients have not been identified. RS-SCIDpatients frequently display additional characteristics including microcephaly, dysmorphic facial featuresand growth delay.     Multiple novel prostate cancer susceptibility signals identified by fine-mapping of known risk loci among Europeans

    Genome-wide association studies (GWAS) have identified numerous common prostate cancer (PrCa) susceptibility loci. We have fine-mapped 64 GWAS regions known at the conclusion of the iCOGS study using large-scale genotyping and imputation in 25 723 PrCa cases and 26 274 controls of European ancestry. We detected evidence for multiple independent signals at 16 regions, 12 of which contained additional newly identified significant associations. A single signal comprising a spectrum of correlated variation was observed at 39 regions; 35 of which are now described by a novel more significantly associated lead SNP, while the originally reported variant remained as the lead SNP only in 4 regions. We also confirmed two association signals in Europeans that had been previously reported only in East-Asian GWAS. Based on statistical evidence and linkage disequilibrium (LD) structure, we have curated and narrowed down the list of the most likely candidate causal variants for each region. Functional annotation using data from ENCODE filtered for PrCa cell lines and eQTL analysis demonstrated significant enrichment for overlap with bio-features within this set. By incorporating the novel risk variants identified here alongside the refined data for existing association signals, we estimate that these loci now explain ∼38.9% of the familial relative risk of PrCa, an 8.9% improvement over the previously reported GWAS tag SNPs. This suggests that a significant fraction of the heritability of PrCa may have been hidden during the discovery phase of GWAS, in particular due to the presence of multiple independent signals within the same regio

    Analyzing the economic benefits and costs of smart growth

    California’s Senate Bill 375, (Chapter 728, Statutes of 2008), aims to reduce transportationrelated greenhouse gas emissions through more efficient patterns of land development. Advocates claim these smart growth policies will reduce vehicle travel while benefitting residents, cities, and regions in the form of more attractive communities, more affordable housing, and healthier municipal finances. In this study, we analyzed the economic impacts of existing smart growth plans similar to those currently being considered and adopted throughout metropolitan California. Through five case studies of neighborhood-level plans already implemented in California, we examined the effects of smart growth interventions on residential development, commercial development, municipal budgets, and vehicle travel. We used a combination of quantitative and qualitative methods to estimate the net benefits and costs from the regional, municipal, and household perspectives. We found the plans, in most cases, produced net benefits for the stakeholders considered. The benefits emerged from plans that resulted in denser development in relatively central locations with good access to transit. However, in some cases the plans produced costs, and impacts were not evenly distributed. The research suggests smart growth policies can produce benefits, but planners must be aware of potential costs
