    Evidence-based vector control? Improving the quality of vector control trials.

    Vector-borne diseases (VBDs) such as malaria, dengue, and leishmaniasis cause a high level of morbidity and mortality. Although vector control tools can play a major role in controlling and eliminating these diseases, in many cases the evidence base for assessing the efficacy of vector control interventions is limited or not available. Studies assessing the efficacy of vector control interventions are often poorly conducted, which limits the return on investment of research funding. Here we outline the principal design features of Phase III vector control field studies, highlight major failings and strengths of published studies, and provide guidance on improving the design and conduct of vector control studies. We hope that this critical assessment will increase the impetus for more carefully considered and rigorous design of vector control studies

    Recognising the socio-technical opportunity of workplace: an analysis of early responses to COVID-19

    COVID-19 has disrupted the ways in which we work, offering an opportunity to rethink our workplaces. Organisations have had to adapt and respond in unprecedented ways to enable continued organisational performance that have come to see many working from home. Early responses to ‘return-to-work’ have sought to repurpose existing workspace arrangements, but they miss the unique opportunity to reconceive ‘workplace’ more comprehensively, as well as the role the property community have in enabling work. This paper aims to highlight the opportunity of viewing workplace holistically through the lens of socio-technical systems. An examination of the early responses to the pandemic identified a focus on the technical aspects of reoccupying workspaces, but taking from socio-technical systems, this should not be at the detriment to other factors. A more nuanced debate regarding who should return to work and how this will occur is presented, which highlights further a need to move beyond the physical workspace and to reflect on how we can enable ways of working

    Wrappings of power: a woman’s burial in cattle hide at Langwell Farm, Strath Oykel

    A well-preserved burial, discovered during peat clearing on Langwell Farm in Strath Oykel, Easter Ross, consisted of a stone cist that held the skeleton of a woman who had died in 2200–1960 cal BC. Although the cist contents were disturbed and partly removed before archaeological investigation took place, the burial rite can be interpreted to some extent. The woman, who died in her late 20s, had been wrapped in brown cattle hide, and wooden and woven objects were placed with her body. Periodic waterlogging created conditions that allowed the rare, partial preservation of the organic materials. Analysis of bone histology indicated that decay of the human remains had been arrested, either by deliberate mummification or waterlogging. The cist had been set into a low knoll on the valley floor and it may have been covered with a low cairn or barrow. This spot had been the site of a fire several hundred years earlier, and it may have been a node on a cross-country route linking east and west coasts in the Early Bronze Age. The use of animal hide suggests the creation and use of particular identities, linking the dead to ancestors and to powerful spiritual properties attributed to the natural world.     A mechanism for the suppression of homologous recombination in G1 cells

    DNA repair by homologous recombination (HR)(1) is highly suppressed in G1 cells(2,3) to ensure that mitotic recombination occurs solely between sister chromatids(4). Although many HR factors are cell cycle-regulated, the identity of the events that are both necessary and sufficient to suppress recombination in G1 cells is unknown. Here we report that the cell cycle controls the interaction of BRCA1 with PALB2-BRCA2 in order to constrain BRCA2 function to the S/G2 phases. We found that the BRCA1-interaction site on PALB2 is targeted by an E3 ubiquitin ligase composed of KEAP1, a PALB2-interacting protein(5), in complex with CUL3-RBX1(6). PALB2 ubiquitylation suppresses its interaction with BRCA1 and is counteracted by the deubiquitylase USP11, which is itself under cell cycle control. Restoration of the BRCA1-PALB2 interaction combined with the activation of DNA end resection is sufficient to induce HR in G1, as measured by RAD51 recruitment, unscheduled DNA synthesis and a CRISPR/Cas9-based gene targeting assay. We conclude that the mechanism prohibiting HR in G1 minimally consists of the suppression of DNA end resection coupled to a multi-step block to BRCA2 recruitment to DNA damage sites that involves the inhibition of BRCA1-PALB2-BRCA2 complex assembly. We speculate that the ability to induce HR in G1 cells with defined factors could spur the development of gene targeting applications in non-dividing cells

    Designing Durable Vapor-Deposited Surfaces for Reduced Hydrate Adhesion

    The formation and accumulation of clathrate hydrates inside oil and gas pipelines cause severe problems in deep-sea oil/gas operations. In the present work, durable and mechanically robust bilayer poly-divinyl benzene/poly(perfluorodecylacrylate) coatings are developed using initiated chemical vapor deposition (iCVD) to reduce the adhesion strength of hydrates to underlying substrates (silicon and steel). Tetrahydrofuran (THF) dissolved in water with a wt% concentration of 0–70 is used to study the formation of hydrates and their adhesion strength. Goniometric measurements of the THF–water droplets on the substrates exhibit a reduction in advancing and receding contact angles with an increase in the THF concentration. The strength of hydrate adhesion experiences a tenfold reduction when substrates are coated with these iCVD polymers: from 1050 ± 250 kPa on bare silicon to 128 ± 100 kPa on coated silicon and from 1130 ± 185 kPa on bare steel to 153 ± 86 kPa on coated steel. The impact of subcooling temperature and time on the adhesion strength of hydrate on substrates is also studied. The results of this work suggest that the THF–water mixture repellency of a given substrate can be utilized to assess its hydrate-phobic behavior; hence, it opens a pathway for studying hydrate-phobicity.Chevron Corporation (MIT-Chevron University Partnership Program

    Early preparation during turn-taking:Listeners use content predictions to determine <i>what</i> to say but not <i>when</i> to say it

    During conversation, there is often little gap between interlocutors' utterances. In two pairs of experiments, we manipulated the content predictability of yes/no questions to investigate whether listeners achieve such coordination by (i) preparing a response as early as possible or (ii) predicting the end of the speaker's turn. To assess these two mechanisms, we varied the participants' task: They either pressed a button when they thought the question was about to end (Experiments 1a and 2a), or verbally answered the questions with either yes or no (Experiments 1b and 2b). Predictability effects were present when participants had to prepare a verbal response, but not when they had to predict the turn-end. These findings suggest content prediction facilitates turn-taking because it allows listeners to prepare their own response early, rather than because it helps them predict when the speaker will reach the end of their turn
