    Saguenay Youth Study : a multi-generational approach to studying virtual trajectories of the brain and cardio-metabolic health

    This paper provides an overview of the Saguenay Youth Study (SYS) and its parental arm. The overarching goal of this effort is to develop trans-generational models of developmental cascades contributing to the emergence of common chronic disorders, such as depression, addictions, dementia and cardio-metabolic diseases. Over the past 10 years, we have acquired detailed brain and cardio-metabolic phenotypes, and genome-wide genotypes, in 1029 adolescents recruited in a population with a known genetic founder effect. At present, we are extending this dataset to acquire comparable phenotypes and genotypes in the biological parents of these individuals. After providing conceptual background for this work (transactions across time, systems and organs), we describe briefly the tools employed in the adolescent arm of this cohort and highlight some of the initial accomplishments. We then outline in detail the phenotyping protocol used to acquire comparable data in the parents

    A note on the distribution and abundance of blue whales (<i>Balaenoptera musculus</i>) in the Central and Northeast North Atlantic

    The distribution and abundance of blue whales (Balaenoptera musculus) was assessed from ship surveys conducted in the Central and Northeast Atlantic in 1987, 1989, 1995 and 2001. Blue whales were most commonly sighted off western Iceland, and to a lesser extent northeast of Iceland. They were very rare or absent in the Northeast Atlantic. Sightings were combined over all surveys to estimate the detection function using standard line transect methodology, with the addition of a covariate to account for differences between surveys. Total abundance was highest in 1995 (979, 95% CI 137-2,542) and lowest in 1987 (222, 95% CI 115-440). Uncertainty in species identity had little effect on estimates of abundance. There was a significant positive trend in abundance northeast of Iceland and in the total survey area

    Minke whale abundance estimation from the NASS 1987 and 2001 aerial cue–counting surveys taking appropriate account of distance estimation errors

    We estimate the abundance of minke whales (Balaenoptera acutorostrata) from the Icelandic coastal shelf aerial surveys carried out as part of the 1987 and 2001 North Atlantic Sightings Surveys (NASS). In the case of the 1987 survey, the probability of detecting animals at distance zero (g(0)) is very close to 1 but there is substantial random measurement error in estimating distances. To estimate abundance from these data, we use methods which assume g(0)=1 but which includea distance measurement error model. In the case of the 2001 survey, measurement errors were sufficiently small to be negligible, and we use double platform methods which estimate g(0) and assume no measurement error to estimate abundance. From the 1987 survey, we estimate abundance to be 24,532 animals, with 95% CI (13,399; 44,916). From the 2001 NASS survey data, minke whale abundance is estimated to be 43,633 animals, with 95% CI (30,148; 63,149).Publisher PDFPeer reviewe

    Development of functional connectivity during adolescence:A longitudinal study using an action-observation paradigm

    Successful interpersonal interactions rely on an ability to read the emotional states of others and to modulate one's own behavior in response. The actions of others serve as valuable social stimuli in this respect, offering the observer an insight into the actor's emotional state. Social cognition continues to mature throughout adolescence. Here we assess longitudinally the development of functional connectivity during early adolescence within two neural networks implicated in social cognition: one network of brain regions consistently engaged during action observation and another one associated with mentalizing. Using fMRI, we reveal a greater recruitment of the social-emotional network during the observation of angry hand actions in male relative to female adolescents. These findings are discussed in terms of known sex differences in adolescent social behavior

    Fundamental stellar parameters of zeta Pup and gamma^2 Vel from HIPPARCOS data

    We report parallax measurements by the HIPPARCOS satellite of zeta Puppis and gamma^2 Velorum. The distance of zeta Pup is d=429 (+120/ -77) pc, in agreement with the commonly adopted value to Vela OB2. However, a significantly smaller distance is found for the gamma^2 Vel system: d=258 (+41/-31) pc. The total mass of gamma^2 Vel derived from its parallax, the angular size of the semi-major axis as measured with intensity interferometry, and the period is M(WR+O)=29.5 (+/-15.9) Msun. This result favors the orbital solution of Pike et al. (1983) over that of Moffat et al. (1986). The stellar parameters for the O star companion derived from line blanketed non-LTE atmosphere models are: Teff=34000 (+/-1500) K, log L/Lsun=5.3 (+/-0.15) from which an evolutionary mass of M=29 (+/-4) Msun and an age of 4.0 (+0.8/-0.5) Myr is obtained from single star evolutionary models. With non-LTE model calculations including He and C we derive a luminosity log L/Lsun~4.7 (+/-0.2) for the WR star. The mass-luminosity relation of hydrogen-free WR stars implies a mass of M(WR)~5 (+/-1.5) Msun. From our data we favor an age of ~10 Myr for the bulk of the Vela OB2 stars.     Pulmonary Abnormalities and Carotid Atherosclerosis in Ex-Smokers without Airflow Limitation.

    Abstract It is well-established that COPD patients have a burden of vascular disease that cannot be fully-explained by smoking history but the mechanistic links between atherosclerosis and pulmonary disease in COPD patients are not well-understood. Moreover, in ex-smokers without symptoms or other evidence of COPD, subclinical pulmonary and vascular disease, although potentially present, has not been described or evaluated. Hence our aim was to use sensitive three-dimensional (3D) pulmonary and carotid imaging to quantify pulmonary airway/parenchyma abnormalities and atherosclerosis in ex-smokers without airflow limitation or symptoms consistent with COPD. We evaluated 61 subjects without airflow limitation including 34 never- (72 ± 6 years) and 27 ex-smokers (73 ± 9 years), who provided written informed consent to spirometry, plethysmography, (3)He magnetic resonance imaging (MRI) and carotid ultrasound (US) and, for ex-smokers alone, thoracic X-ray computed tomography (CT). Ex-smokers had significantly greater (3)He ventilation defect percent (VDP = 7%, p = 0.001) and carotid total plaque volume (TPV = 250 mm(3), p = 0.002) than never-smokers, although there were no significant differences for spirometry or plethysmography, and CT airway and emphysema measurements were normal. There were univariate relationships for (3)He VDP with carotid intima media thickness (IMT, r = 0.42, p = 0.004), TPV (r = 0.41, p = 0.006) and vessel wall volume (VWV, r = 0.40, p = 0.007). Multivariate models that included age, BMI, FEV1, DLCO and VDP showed that only VDP significantly predicted IMT (ÎČ = 0.41, p = 0.001), VWV (ÎČ = 0.45, p = 0.003) and TPV (ÎČ = 0.38, p = 0.005). In summary, there was imaging evidence of mild airways disease and carotid plaque burden that were related and significantly greater in ex-smokers without airflow limitation than in never-smokers

    Visceral adiposity is associated with metabolic profiles predictive of type 2 diabetes and myocardial infarction

    Background: Visceral fat (VF) increases risk for cardiometabolic disease (CMD), the leading cause of morbidity and mortality. Variations in the circulating metabolome predict the risk for CMD but whether or not this is related to VF is unknown. Further, CMD is now also present in adolescents, and the relationships between VF, circulating metabolome, and CMD may vary between adolescents and adults. Methods: With an aim to add understanding to the metabolic variations in visceral obesity, we tested associations between VF, measured directly with magnetic resonance imaging, and 228 fasting serum metabolomic measures, quantified with nuclear magnetic resonance spectroscopy, in 507 adults (36-65 years) and 938 adolescents (12-18 years). We further utilized data from published studies to estimate similarities between VF and CMD-associated metabolic profiles. Results: Here we show that VF, independently of body mass index (BMI) or subcutaneous fat, is associated with triglyceride-rich lipoproteins, fatty acids, and inflammation in both adults and adolescents, whereas the associations with amino acids, glucose, and intermediary metabolites are significant in adults only. BMI-adjusted metabolomic profile of VF resembles those predicting type 2 diabetes in adults (R 2 = 0.88) and adolescents (R 2 = 0.70), and myocardial infarction in adults (R 2 = 0.59) and adolescents (R 2 = 0.40); this is not the case for ischemic stroke (adults: R 2 = 0.05, adolescents: R 2 = 0.08). Conclusions: Visceral adiposity is associated with metabolomic profiles predictive of type 2 diabetes and myocardial infarction even in normal-weight individuals and already in adolescence. Targeting factors contributing to the emergence and maintenance of these profiles might ameliorate their cumulative effects on cardiometabolic health

    LSST: from Science Drivers to Reference Design and Anticipated Data Products

    (Abridged) We describe here the most ambitious survey currently planned in the optical, the Large Synoptic Survey Telescope (LSST). A vast array of science will be enabled by a single wide-deep-fast sky survey, and LSST will have unique survey capability in the faint time domain. The LSST design is driven by four main science themes: probing dark energy and dark matter, taking an inventory of the Solar System, exploring the transient optical sky, and mapping the Milky Way. LSST will be a wide-field ground-based system sited at Cerro Pach\'{o}n in northern Chile. The telescope will have an 8.4 m (6.5 m effective) primary mirror, a 9.6 deg2^2 field of view, and a 3.2 Gigapixel camera. The standard observing sequence will consist of pairs of 15-second exposures in a given field, with two such visits in each pointing in a given night. With these repeats, the LSST system is capable of imaging about 10,000 square degrees of sky in a single filter in three nights. The typical 5σ\sigma point-source depth in a single visit in rr will be ∌24.5\sim 24.5 (AB). The project is in the construction phase and will begin regular survey operations by 2022. The survey area will be contained within 30,000 deg2^2 with ÎŽ<+34.5∘\delta<+34.5^\circ, and will be imaged multiple times in six bands, ugrizyugrizy, covering the wavelength range 320--1050 nm. About 90\% of the observing time will be devoted to a deep-wide-fast survey mode which will uniformly observe a 18,000 deg2^2 region about 800 times (summed over all six bands) during the anticipated 10 years of operations, and yield a coadded map to r∌27.5r\sim27.5. The remaining 10\% of the observing time will be allocated to projects such as a Very Deep and Fast time domain survey.     A Simple Approach to Assign Disulfide Connectivity Using Extracted Ion Chromatograms of Electron Transfer Dissociation Spectra

    Increasing interest in production of protein-based pharmaceuticals (biotherapeutics) is accompanied by an increased need for verification of protein folding and correct disulfide bonding. Recombinant protein expression may produce aberrant disulfide bonds and could result in safety concerns or decreased efficacy. Thus, the thorough analysis of disulfide bonding is a necessity for protein therapeutics. The use of ETD facilitates this analysis because disulfide bonds are preferentially cleaved when subjected to ETD. Here, we make use of this well-characterized reaction to assign disulfide bonding networks by coupling the use of extracted ion chromatograms (XICs) of cysteine-containing peptides with ETD analysis to produce an efficient assignment approach for disulfide bonding. This method can be used to assign a disulfide pattern in a de novo fashion, to detect disulfide shuffling, and to provide information on heterogeneity, when more than one disulfide bonding pattern is present. The method was applied for assigning the disulfide-bonding network of a recombinant monomer of the HIV envelope protein gp120. It was found that one region of the protein, the V1/V2 loops, had significant heterogeneity in the disulfide bonds
