    An atomic scale comparison of the reaction of Bioglass® in two types of simulated body fluid

    A class of melt quenched silicate glasses, containing calcium, phosphorus and alkali metals, and having the ability to promote bone regeneration and to fuse to living bone, is produced commercially as Bioglass. The changes in structure associated with reacting the bioglass with a body fluid simulant (a buffered Tris(hydroxymethyl)aminomethane growth medium solution or a blood plasma-like salt simulated body fluid) at 37°C have been studied using both high energy and grazing incidence x-ray diffraction. This has corroborated the generic conclusions of earlier studies based on the use of calcia–silica sol-gel glasses whilst highlighting the important differences associated with glass composition; the results also reveal the more subtle effects on reaction rates of the choice of body fluid simulant. The results also indicate the presence of tricalcium phosphate crystallites deposited onto the surface of the glass as a precursor to the growth of hydroxyapatite, and indicates that there is some preferred orientation to their growth

    The Mode of Death of Pig Kidney Cells Infected with Cowpox Virus Is Governed by the Expression of thecrmAGene

    AbstractPig kidney cells (LLC-PK1) were infected with one of three viruses: wild-type cowpox virus (Brighton red strain) expressing thecrmAgene; recombinant cowpox virus A602, lacking thecrmAgene; or cowpox virus A604, a revertant of virus A602, expressing thecrmAgene. The wild-type virus and virus A604 produced identical cytopathic effects consistent with death by necrosis. In these cells, the structural features of the plasma membrane, the nuclear membrane, and the chromatin were maintained until lysis of the cells. In contrast, cowpox virus A602 produced cytopathic effects consistent with death by apoptosis. These effects included loss of microvilli on the cell surface, margination and condensation of the chromatin, progressive convolution of the nuclear membrane, release of dense chromatin masses on disintegration of the nucleus, fragmentation of the DNA, and the generation of apoptotic bodies. These results suggest that thecrmAgene is necessary to inhibit processes of apoptosis induced in LLC-PK1cells by infection with cowpox virus. Thus in cells of certain types, thecrmAgene can act with other viral genes to control the mode of death of the virus-infected cell. This capability may be advantageous to virus replicationin vivo,potentially facilitating both virus trafficking and interference with antiviral immune defenses

    The Effect of Aquifer/Caprock Interface on Geological Storage of CO2

    AbstractThe migration of CO2 stored in deep saline aquifers depends on the morphology of the top of the aquifer. Topographical highs, such as anticlines, may trap CO2 and limit the distance migrated, or elevated ridges may provide pathways enabling CO2 to migrate further from the injector. For example, seismic data of the Utsira formation at the Sleipner storage site indicates that a branch of the CO2 plume is moving to the north [1]. It is therefore important to study the interface between the aquifer and the caprock when assessing risk as CO2 storage sites.Undulations in the top surface of an aquifer may either be caused by sedimentary structures [2], or by folding. In addition, irregularities may be generated by faulting [2]. Large-scale features are detected using seismic data (i.e. structures with amplitudes greater than 10 m), and such structures will generally be included in reservoir or aquifer models. However, smaller- scale features could also have an effect on a CO2 plume migration, and this is the topic of our study. We have conducted simulations in models with a range of top-surface morphology, and have examined the distance migrated and the amount of dissolution.The results from this study suggest that the effects of sub-seismic variations in the topography of the aquifer/caprock interface are unlikely to have a significant impact on the migration and dissolution of CO2 in a saline aquifer, compared with tilt or permeability anisotropy. The results were most sensitive to the kv/kh ratio during the injection period

    Emergency supply of prescription-only medicines to patients by community pharmacists: a mixed methods evaluation incorporating patient, pharmacist and GP perspectives

    Objective To evaluate and inform emergency supply of prescription-only medicines by community pharmacists (CPs), including how the service could form an integral component of established healthcare provision to maximise adherence. Design Mixed methods. 4 phases: prospective audit of emergency supply requests for prescribed medicines (October–November 2012 and April 2013); interviews with CPs (February–April 2013); follow-up interviews with patients (April–May 2013); interactive feedback sessions with general practice teams (October–November 2013). Setting 22 community pharmacies and 6 general practices in Northwest England. Participants 27 CPs with experience of dealing with requests for emergency supplies; 25 patients who received an emergency supply of a prescribed medicine; 58 staff at 6 general practices. Results Clinical audit in 22 pharmacies over two 4-week periods reported that 526 medicines were requested by 450 patients. Requests peaked over a bank holiday and around weekends. A significant number of supplies were made during practice opening hours. Most requests were for older patients and for medicines used in long-term conditions. Difficulty in renewing repeat medication (forgetting to order, or prescription delays) was the major reason for requests. The majority of medicines were ‘loaned’ in advance of a National Health Service (NHS) prescription. Interviews with CPs and patients indicated that continuous supply had a positive impact on medicines adherence, removing the need to access urgent care. General practice staff were surprised and concerned by the extent of emergency supply episodes. Conclusions CPs regularly provide emergency supplies to patients who run out of their repeat medication, including during practice opening hours. This may aid adherence. There is currently no feedback loop, however, to general practice. Patient care and interprofessional communication may be better served by the introduction of a formally structured and funded NHS emergency supply service from community pharmacies, with ongoing optimisation of repeat prescribing

    Free jet feasibility study of a thermal acoustic shield concept for AST/VCE application: Single stream nozzles

    A technology base for the thermal acoustic shield concept as a noise suppression device for single stream exhaust nozzles was developed. Acoustic data for 314 test points for 9 scale model nozzle configurations were obtained. Five of these configurations employed an unsuppressed annular plug core jet and the remaining four nozzles employed a 32 chute suppressor core nozzle. Influence of simulated flight and selected geometric and aerodynamic flow variables on the acoustic behavior of the thermal acoustic shield was determined. Laser velocimeter and aerodynamic measurements were employed to yield valuable diagnostic information regarding the flow field characteristics of these nozzles. An existing theoretical aeroacoustic prediction method was modified to predict the acoustic characteristics of partial thermal acoustic shields
