    Assessing the genetic diversity of farmed and wild Rufiji tilapia (Oreochromis urolepis urolepis) populations using ddRAD sequencing

    Rufiji tilapia (Oreochromis urolepis urolepis) is an endemic cichlid in Tanzania. In addition to its importance for biodiversity conservation, Rufiji tilapia is also attractive for farming due to its high growth rate, salinity tolerance, and the production of all-male hybrids when crossed with Nile tilapia (Oreochromis niloticus). The aim of the current study was to assess the genetic diversity and population structure of both wild and farmed Rufiji tilapia populations in order to inform conservation and aquaculture practices. Double-digest restriction-site-associated DNA (ddRAD) libraries were constructed from 195 animals originating from eight wild (Nyamisati, Utete, Mansi, Mindu, Wami, Ruaha, Kibasira, and Kilola) and two farmed (Bwawani and Chemchem) populations. The identified single nucleotide polymorphisms (SNPs;n = 2,182) were used to investigate the genetic variation within and among the studied populations. Genetic distance estimates (F-st) were low among populations from neighboring locations, with the exception of Utete and Chemchem populations (F-st = 0.34). Isolation-by-distance (IBD) analysis among the wild populations did not detect any significant correlation signal (r = .05;p-value = .4) between the genetic distance and the sampling (Euclidean distance) locations. Population structure and putative ancestry were further investigated using both Bayesian (Structure) and multivariate approaches (discriminant analysis of principal components). Both analysis indicated the existence of three distinct genetic clusters. Two cross-validation scenarios were conducted in order to test the efficiency of the SNP dataset for discriminating between farmed and wild animals or predicting the population of origin. Approximately 95% of the test dataset was correctly classified in the first scenario, while in the case of predicting for the population of origin 68% of the test dataset was correctly classified. Overall, our results provide novel insights regarding the population structure of Rufiji tilapia and a new database of informative SNP markers for both conservation management and aquaculture activities

    International audience(Market)places are spatial entities which individuals and groups might experience as meaningful. By highlighting the role of place in ethnic consumer research, this article argues that increased mobility and changing places render relatively stable notions of ethnicity outdated. We identify three main trajectories to revitalize future research on ethnicity. First, we demonstrate the need for research on ethnic identity to be underpinned by a better understanding of the role of place in identity processes. Second, we contend that the established migration/acculturation paradigm should be replaced by the mobility/adaptiveness paradigm. Third, we consider the profound effects of interethnic contact among mobile and immobile populations within shared places on individual and societal well-being

    CHIMPS2: Survey description and <sup>12</sup>CO emission in the Galactic Centre

    Abstract The latest generation of Galactic-plane surveys is enhancing our ability to study the effects of galactic environment upon the process of star formation. We present the first data from CO Heterodyne Inner Milky Way Plane Survey 2 (CHIMPS2). CHIMPS2 is a survey that will observe the Inner Galaxy, the Central Molecular Zone (CMZ), and a section of the Outer Galaxy in 12CO, 13CO, and C18O (J = 3 → 2) emission with the Heterodyne Array Receiver Program on the James Clerk Maxwell Telescope (JCMT). The first CHIMPS2 data presented here are a first look towards the CMZ in 12CO J = 3→2 and cover −3○ ≤ ℓ ≤ 5○ and ∣b{b}∣ ≤ 0.{_{.}^{\circ}}5 with angular resolution of 15 arcsec, velocity resolution of 1 km s−1, and rms ΔTA=\Delta \, T_A ^\ast = 0.58 K at these resolutions. Such high-resolution observations of the CMZ will be a valuable data set for future studies, whilst complementing the existing Galactic Plane surveys, such as SEDIGISM, the Herschel{Herschel} infrared Galactic Plane Survey, and ATLASGAL. In this paper, we discuss the survey plan, the current observations and data, as well as presenting position-position maps of the region.     Characterization of CorR, a transcriptional activator which is required for biosynthesis of the phytotoxin coronatine

    Coronatine (COR) is a plasmid-encoded phytotoxin synthesized by several pathovars of phytopathogenic Pseudomonas syringae. The COR biosynthetic gene cluster in P. syringae pv. glycinea PG4180 is encoded by a 32-kb region which contains both the structural and regulatory genes needed for COR synthesis. The regulatory region contains three genes: corP, corS, and corR. corS is thought to function as a histidine protein kinase, whereas corP and corR show relatedness to response regulators of the two-component regulatory paradigm. In the present study, we investigated whether CorR is a positive activator of COR gene expression. We also studied whether CorR specifically binds the DNA region located upstream of cfl, a gene located at the 5* end of the gene cluster encoding coronafacic acid, the polyketide portion of COR. Complementation analysis with a corR mutant, PG4180.P2, and transcriptional fusions to a promoterless glucuronidase gene (uidA) indicated that CorR functions as a positive regulator of COR gene expression. Deletion analysis of the 5* end of the cfl upstream region was used to define the minimal region required for COR gene expression. A 360-bp DNA fragment located over 500 bp upstream from the cfl transcriptional start site was used in DNase I protection assays to define the specific bases bound by CorR. An area extending from 2704 to 2650 with respect to the cfl transcriptional start site was protected by DNase I footprinting, indicating a rather large area of protection. This area was also conserved in the promoter region for cmaA, which encodes a transcript containing genes for coronamic acid synthesis, another intermediate in the COR biosynthetic pathway. The results obtained in the current study suggest that both the coronafacic acid and the coronamic acid structural genes are controlled by CorR, a positive activator of COR gene expression.Peer reviewedEntomology and Plant Patholog

    Attitudes and burden in relatives of patients with schizophrenia in a middle income country

    BACKGROUND: Most studies of family attitudes and burden have been conducted in developed countries. Thus it is important to test the generalizability of this research in other contexts where social conditions and extended family involvement may be different. The aim of this study was to assess the relationship between the attitudes of caregivers and the burden they experience in such a context, namely Arica, a town located in the northernmost region of Chile, close to the border with Peru and Bolivia. METHODS: We assessed attitudes towards schizophrenia (including affective, cognitive and behavioural components) and burden (including subjective distress, rejection and competence) in 41 main caregivers of patients with schizophrenia, all of whom were users of Public Mental Health Services in Arica. RESULTS: Attitude measures differed significantly according to socio-demographic variables, with parents (mainly mothers) exhibiting a more negative attitude towards the environment than the rest of the family (t = 4.04; p = 0.000).This was also the case for caregivers with a low educational level (t = 3.27; p < 0.003), for the oldest caregivers (r = 0.546; p = 0.000) and for those who had spent more time with the patient (r = 0.377; p = 0.015). Although attitudes had significant association with burden, their explanatory power was modest (R2 = .104, F = 4,55; p = .039). CONCLUSIONS: Similar to finding developed countries, the current study revealed a positive and significant relationship between the attitudes of caregivers and their burden. These findings emphasize the need to support the families of patients with schizophrenia in this social context
