Gold Nanoplates as Cancer-Targeted Photothermal Actuators for Drug Delivery and Triggered Release
The selective exposure of cancerous tissue to systemically delivered chemotherapeutic agents remains a major challenge facing cancer therapy. To address this question, a near infrared responsive oligonucleotide-coated (AS1411, hairpin, or both) gold nanoplate loaded with doxorubicin is demonstrated to be nontoxic to cells without triggered release, while being acutely toxic to cells after 5 minutes of laser exposure to trigger DOX release. Conjugation of oligonucleotides to the nanoplates is confirmed by an average increase in hydrodynamic diameter of 30.6ânm, an average blue shift of the plasmon resonance peak by 36ânm, and an average â10âmV shift in zeta potential of the particles. DOX loading through intercalation into the hairpin DNA structure is confirmed through fluorescence measurements. For both GNP-Hairpin and GNP-Hairpin-AS1411, ~60% of loaded DOX is released after the first 5 minutes of laser exposure (λ=817ânm), with complete release after two more 5-minute exposures. Preliminary proof of concept is demonstrated in vitro using A549 and MDA-MB-231 cell lines as models for breast and lung cancer, respectively. Exposure of cells to untriggered DOX-loaded conjugate with no laser exposure results in little to no toxicity, while laser-triggered release of DOX causes significant cell death
Cancer-selective antiproliferative activity is a general property of some G-rich oligodeoxynucleotides
Oligodeoxynucleotide libraries containing randomly incorporated bases are used to generate DNA aptamers by systematic evolution of ligands by exponential enrichment (SELEX). We predicted that combinatorial libraries with alternative base compositions might have innate properties different from the standard library containing equimolar A + C + G + T bases. In particular, we hypothesized that G-rich libraries would contain a higher proportion of quadruplex-forming sequences, which may impart desirable qualities, such as increased nuclease resistance and enhanced cellular uptake. Here, we report on 11 synthetic oligodeoxynucleotide libraries of various base combinations and lengths, with regard to their circular dichroism, stability in serum-containing medium, cellular uptake, protein binding and antiproliferative activity. Unexpectedly, we found that some G-rich libraries (composed of G + T or G + C nucleotides) strongly inhibited cancer cell growth while sparing non-malignant cells. These libraries had spectral features consistent with G-quadruplex formation, were significantly more stable in serum than inactive libraries and showed enhanced cellular uptake. Active libraries generally had strong protein binding, while the pattern of protein binding suggested that G/T and G/C libraries have distinct mechanisms of action. In conclusion, cancer-selective antiproliferative activity may be a general feature of certain G-rich oligodeoxynucleotides and is associated with quadruplex formation, nuclease resistance, efficient cellular uptake and protein binding
Microenvironment Changes (in pH) Affect VEGF Alternative Splicing
Vascular endothelial growth factor-A (VEGF-A) has several isoforms, which differ in their capacity to bind extracellular matrix proteins and also in their affinity for VEGF receptors. Although the relative contribution of the VEGF isoforms has been studied in tumor angiogenesis, little is known about the mechanisms that regulate the alternative splicing process. Here, we tested microenvironment cues that might regulate VEGF alternative splicing. To test this, we used endometrial cancer cells that produce all VEGF isoforms as a model, and exposed them to varying pH levels, hormones, glucose and CoCl2 (to mimic hypoxia). Low pH had the most consistent effects in inducing variations in VEGF splicing pattern (VEGF121 increased significantly, pâ<â0.001, when compared to VEGF145, 165 or 189). This was accompanied by activation of the p38 stress pathway and SR proteins (splicing factors) expression and phosphorylation. SF2/ASF, SRp20 and SRp40 down-regulation by siRNA impaired the effects of pH stimulation, blocking the shift in VEGF isoforms production. Taken together, we show for the first time that acidosis (low pH) regulates VEGF-A alternative splicing, may be through p38 activation and suggest the possible SR proteins involved in this process
Search for CP violation in decays
A model-independent search for direct CP violation in the Cabibbo suppressed
decay in a sample of approximately 370,000 decays is
carried out. The data were collected by the LHCb experiment in 2010 and
correspond to an integrated luminosity of 35 pb. The normalized Dalitz
plot distributions for and are compared using four different
binning schemes that are sensitive to different manifestations of CP violation.

Observation of excited Lambda_b0 baryons
Using pp collision data corresponding to 1.0 fb-1 integrated luminosity
collected by the LHCb detector, two narrow states are observed in the
Lambda_b0pi+pi- spectrum with masses 5911.97 +- 0.12(stat) +- 0.02(syst) +-
0.66(Lambda_b0 mass) MeV/c^2 and 5919.77 +- 0.08(stat) +- 0.02(syst) +-
0.66(Lambda_b0 mass) MeV/c^2. The significances of the observations are 5.2 and
10.2 standard deviations, respectively. These states are interpreted as the
orbitally-excited Lambda_b0 baryons, Lambda_b*0(5912) and Lambda_b*0(5920).Comment: Replaced by version published in Phys. Rev. Lett, modified fit with

Measurement of the CP-violating phase phi_s in the decay Bs->J/psi phi
We present a measurement of the time-dependent CP-violating asymmetry in B_s
-> J/psi phi decays, using data collected with the LHCb detector at the LHC.
The decay time distribution of B_s -> J/psi phi is characterized by the decay
widths Gamma_H and Gamma_L of the heavy and light mass eigenstates of the
B_s-B_s-bar system and by a CP-violating phase phi_s. In a sample of about 8500
B_s -> J/psi phi events isolated from 0.37 fb^-1 of pp collisions at sqrt(s)=7
TeV we measure phi_s = 0.15 +/- 0.18 (stat) +/- 0.06 (syst) rad. We also find
an average B_s decay width Gamma_s == (Gamma_L + Gamma_H)/2 = 0.657 +/- 0.009
(stat) +/- 0.008 (syst) ps^-1 and a decay width difference Delta Gamma_s ==
Gamma_L - Gamma_H} = 0.123 +/- 0.029 (stat) +/- 0.011 (syst) ps^-1. Our
measurement is insensitive to the transformation (phi_s,DeltaGamma_s --> pi -

Strong constraints on the rare decays Bs -> mu+ mu- and B0 -> mu+ mu-
A search for Bs -> mu+ mu- and B0 -> mu+ mu- decays is performed using 1.0
fb^-1 of pp collision data collected at \sqrt{s}=7 TeV with the LHCb experiment
at the Large Hadron Collider. For both decays the number of observed events is
consistent with expectation from background and Standard Model signal
predictions. Upper limits on the branching fractions are determined to be BR(Bs
-> mu+ mu-) mu+ mu-) < 1.0 (0.81) x 10^-9 at

Measurements of the branching fractions of the decays B°s â Dâs K± and B°s â DÂŻsÏ+
The decay mode B°s â Dâs K± allows for one of the theoretically cleanest measurements of the CKM angle Îł through the study of time-dependent CP violation. This paper reports a measurement of its branching fraction relative to the Cabibbo-favoured mode B°s â DÂŻsÏ+ based on a data sample corresponding to 0.37 fbÂŻÂč of proton-proton collisions at âs = 7TeV collected in 2011 with the LHCb detector. In addition, the ratio of B meson production fractions fs/fd, determined from semileptonic decays, together with the known branching fraction of the control channel B°s â DÂŻsÏ+ is used to perform an absolute measurement of the branching fractions: B(B°s â DÂŻsÏ+) = (2.95 ± 0.05 ± 0.17 -0.22 +0.18) Ă 10ÂŻÂł ; B(B°s â Dâs K±) = (1.90 ± 0.12 ± 0.13 -0.14 +0.12) Ă 10ÂŻ4 ; where the first uncertainty is statistical, the second the experimental systematic uncertainty, and the third the uncertainty due to f s/f
First evidence of direct CP violation in charmless two-body decays of Bs mesons
