Assessment of the Therapeutic Effects of Okn-007 in Rodent Glioma Models Using Advanced Mri Techniques
Glioblastoma is a malignant grade IV glioma with a poor prognosis in humans. New therapeutics are desperately required. The nitrone OKN-007 (2,4-disulfophenyl-PBN) has demonstrated effective anti-glioma properties in several rodent models and is currently being used as a clinical investigational drug for recurrent gliomas. In this study, we assessed the efficacy of OKN-007 in two adult (rat F98 and mouse GL261) and one novel pediatric (IC-3752GBM in nude mice) high grade glioma model. OKN-007 significantly reduced the percent necrosis (p<0.01) and tumor cell proliferation (p<0.05) in treated F98-tumor bearing rats compared to the untreated group. Based on molecular magnetic resonance imaging (MRI) results, OKN-007 was also found to significantly decrease (p<0.05) free radicals levels in treated F98 rats compared to the untreated group. We also assessed VEGFR-2 (vascular endothelial growth factor receptor 2) levels in untreated and OKN-007-treated GL261 mouse gliomas using molecular MRI. OKN-007 decreased significantly the levels of VEGFR-2 (p<0.05) in GL261 gliomas compared to the untreated group. Furthermore, OKN-007 was also able to decrease significantly the tumor volumes (p<0.01) and increase survival (p< 0.001) in GL261 treated mice. For the pediatric glioma study, MRI results showed that OKN-007 significantly decreased tumor volumes (p<0.05), increased diffusion (p<0.01) and perfusion rates (p<0.05) in a responsive cohort of IC-3752GBM treated animals. In addition, OKN-007 significantly increased animal survival (p<0.05), decreased tumor cell proliferation (p<0.05) and microvessel density (p<0.05) in responsive IC-3752GBM treated mice. Furthermore, the nitrone was also able to significantly decrease the immunoexpression of SULF2 (p<0.05) and PDGFRα (platelet-derived growth factor receptor-α) (p<0.05), and significantly increase the expression of decorin (p<0.05) in responsive IC-3752GBM pGBM tumor bearing mice. Taken together, our findings showed that OKN-007 mediated multiple effects in different rodent glioma models, indicating that the efficacy associated with OKN-007 is a result of anti-proliferative, free radical scavenging, and anti-angiogenic properties. Furthermore, OKN-007 might be also an effective anti-cancer agent for some patients with pediatric gliomas, possibly by inhibiting the PDGFRα pathway, and could be considered as an additional therapy for pediatric brain tumor patients.Veterinary Biomedical Science
Frequency and Burden of Neurological Manifestations Upon Hospital Presentation in COVID-19 Patients: Findings From a Large Brazilian Cohort
BACKGROUND: Scientific data regarding the prevalence of COVID-19 neurological manifestations and prognosis in Latin America countries is still lacking. Therefore, the study aims to understand neurological manifestations of SARS-CoV 2 infection and outcomes in the Brazilian population.
METHODS: This study is part of the Brazilian COVID-19 Registry, a multicentric cohort, including data from 37 hospitals. For the present analysis, patients were grouped according to the presence of reported symptoms (i.e., headache; anosmia and ageusia; syncope and dizziness) vs. clinically-diagnosed neurological manifestations (clinically-defined neurological syndrome: neurological signs or diagnoses captured by clinical evaluation) and matched with patients without neurological manifestations by age, sex, number of comorbidities, hospital of admission, and whether or not patients had underlying neurological disease.
RESULTS: From 6,635 hospitalized patients with COVID-19, 30.8% presented reported neurological manifestations, 10.3% were diagnosed with a neurological syndrome and 60.1% did not show any neurological manifestations. In patients with reported symptoms, the most common ones were headache (20.7%), ageusia (11.1%) and anosmia (8.0%). In patients with neurological syndromes, acute encephalopathy was the most common diagnosis (9.7%). In the matched analysis, patients with neurological syndromes presented more cases of septic shock (17.0 vs. 13.0%, p = 0.045), intensive care unit admission (45.3 vs. 38.9%, p = 0.023), and mortality (38.7 vs. 32.6%, p = 0.026; and 39.2 vs. 30.3%, p \u3c 0.001) when compared to controls.
CONCLUSION: COVID-19 in-hospital patients with clinically defined neurological syndromes presented a higher incidence of septic shock, ICU admission and death when compared to controls
Associação entre a ooforectomia bilateral precoce e o desenvolvimento do parkinsonismo e Doença de Parkinson em mulheres na pré-menopausa
O parkinsonismo é um distúrbio do sistema nervoso de maior incidência masculina do que feminina, visto que, por mecanismos fisiológicos, o estrogênio possui efeitos neuroprotetores, com funções como aumento da dopamina, um neurotransmissor essencial para o controle das funções motoras. Além disso, previne a formação dos corpúsculos de Lewy e da agregação da α-sinucleína, responsáveis pela progressão da Doença de Parkinson. Por isso, a doença se apresenta diferentemente nas mulheres. A remoção cirúrgica de ambos os ovários em mulheres na pré-menopausa para a prevenção do câncer de ovário parece favorecer o surgimento da doença, tendo em vista a perda da produção do hormônio protetor. Assim, o objetivo do estudo é analisar a associação entre a ooforectomia bilateral precoce e o desenvolvimento de parkinsonismo e Doença de Parkinson em mulheres na pré-menopausa. Trata-se de uma revisão bibliográfica sistemática, do tipo quantitativa, que utilizou as plataformas do PubMed, SciELO e Cochrane Library como bases de dados para seleção dos artigos, todos na língua inglesa. Foram utilizadas literaturas publicadas com recorte temporal de 2017 a 2022. De acordo com as literaturas analisadas, a ooforectomia bilateral precoce em mulheres na pré-menopausa aumenta o risco do desenvolvimento de parkinsonismo. Desse modo, a diminuição dos procedimentos cirúrgicos profiláticos para câncer de ovário nas pacientes com risco médio de malignidade reduziria o risco dessa condição

A social and ecological assessment of tropical land uses at multiple scales: the Sustainable Amazon Network
Science has a critical role to play in guiding more sustainable development trajectories. Here, we present the Sustainable Amazon Network (Rede Amazonia Sustentavel, RAS): a multidisciplinary research initiative involving more than 30 partner organizations working to assess both social and ecological dimensions of land-use sustainability in eastern Brazilian Amazonia. The research approach adopted by RAS offers three advantages for addressing land-use sustainability problems: (i) the collection of synchronized and co-located ecological and socioeconomic data across broad gradients of past and present human use; (ii) a nested sampling design to aid comparison of ecological and socioeconomic conditions associated with different land uses across local, landscape and regional scales; and (iii) a strong engagement with a wide variety of actors and non-research institutions. Here, we elaborate on these key features, and identify the ways in which RAS can help in highlighting those problems in most urgent need of attention, and in guiding improvements in land-use sustainability in Amazonia and elsewhere in the tropics. Inhibition of Pediatric Glioblastoma Tumor Growth by the Anti-Cancer Agent OKN-007 in Orthotopic Mouse Xenografts

This work was also supported by Oklahoma State University, Center of Veterinary Health Science (Support Grant AE-1-50060 to P.C.S.), the Musella Foundation (R.A.T.), and the Childhood Brain Tumor Foundation (R.A.T.).Pediatric glioblastomas (pGBM), although rare, are one of the leading causes of cancer-related deaths in children, with tumors essentially refractory to existing treatments. Here, we describe the use of conventional and advanced in vivo magnetic resonance imaging (MRI) techniques to assess a novel orthotopic xenograft pGBM mouse (IC-3752GBM patient-derived culture) model, and to monitor the effects of the anti-cancer agent OKN-007 as an inhibitor of pGBM tumor growth. Immunohistochemistry support data is also presented for cell proliferation and tumor growth signaling. OKN-007 was found to significantly decrease tumor volumes (p<0.05) and increase animal survival (p<0.05) in all OKN-007-treated mice compared to untreated animals. In a responsive cohort of treated animals, OKN-007 was able to significantly decrease tumor volumes (p<0.0001), increase survival (p<0.001), and increase diffusion (p<0.01) and perfusion rates (p<0.05). OKN-007 also significantly reduced lipid tumor metabolism in responsive animals (Lip1.3 and Lip0.9)-to-creatine ratio (p<0.05), as well as significantly decrease tumor cell proliferation (p<0.05) and microvessel density (p<0.05). Furthermore, in relationship to the PDGFRα pathway, OKN-007 was able to significantly decrease SULF2 (p<0.05) and PDGFR-α (platelet-derived growth factor receptor-α) (p<0.05) immunoexpression, and significantly increase decorin expression (p<0.05) in responsive mice. This study indicates that OKN-007 may be an effective anti-cancer agent for some patients with pGBMs by inhibiting cell proliferation and angiogenesis, possibly via the PDGFRα pathway, and could be considered as an additional therapy for pediatric brain tumor patients.Yeshttp://www.plosone.org/static/editorial#pee
Taking the pulse of Earth's tropical forests using networks of highly distributed plots
Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science. Pulmonary and splenic immunoexpression of IL-12, TGF-β and TNF-α cytokines and S-100 and Lysozyme proteins in Pontoporia blainvillei (Gervais e d\'Orbigny, 1844) (Mammalia, Cetacea)
Compreender a função das diferentes proteínas e citocinas durante a vigência de alterações inflamatórias pode contribuir para um melhor conhecimento do sistema imunológico dos mamíferos marinhos e sua relação com os processos patológicos. A toninha, Pontoporia blainvillei, é um cetáceo costeiro e o único golfinho brasileiro considerado ameaçado de extinção devido às atividades antrópicas. O objetivo desse trabalho foi avaliar as imunoexpressões das citocinas TGF-β, TNF-α e IL-12 e proteínas S-100 e lisozima em amostras pulmonares e esplênicas de 34 espécimes de P. blainvillei. Com base nas alterações histopatológicas, os animais foram divididos em dois grupos: G1 (n = 8) - animais sem doença inflamatória pulmonar; G2 (n = 26) - animais com doenças inflamatórias pulmonares. Os fragmentos de pulmão e baço foram processados de acordo com os protocolos de rotina. A padronização das reações de imuno-histoquímica e análise morfométrica foram realizadas conforme procedimentos descritos na literatura. Os nossos dados demonstram que as P. blainvillei pertencentes ao G2 apresentam imunoexpressão pulmonar das citocinas TGF-β, TNF-α e IL-12 e proteínas S-100 e lisozima significativamente maior que os animais do G1. Tais resultados corroboram os dados disponíveis na literatura, indicando que a P. blainvillei apresenta citocinas com funções semelhantes às encontradas em outros mamíferos.Evidenciamos também, que as P. blainvillei pertencentes ao G2 não apresentam imunoexpressão esplênica das citocinas TGF-β, TNF-α e IL-12 e da proteína lisozima significativamente maior que os animais do G1. Estudos mais detalhados, incluindo aqueles relacionados com os métodos utilizados, são necessários para a adequada compreensão destes resultados. E finalmente, apesar de tratar-se de dados ainda preliminares, observou-se uma possível associação entre elevadas concentrações de organoclorados no tecido adiposo subcutâneo de parcela dos exemplares de P. blainvillei estudados na presente investigação e a ocorrência de depleção linfóide esplênica nestes animais. Tais dados ratificam a importância do desenvolvimento de análise mais detalhada sobre estes tópicos em futuras investigações científicas.The understanding of the role of different proteins and cytokines during the inflammatory diseases could lead to better knowledge of the marine mammal immune system and its relation to pathological process. Franciscana dolphin, Pontoporia blainvillei, is a coastal cetacean and the only Brazilian dolphin considered threatened with extinction, due to the human activities. The aim of the current study was to investigate the immunoexpression of TGF-β, TNF-α and IL-12 cytokines, and S-100 and Lysozyme proteins in pulmonary and splenic samples of 34 Pontoporia blainvillei. Based on histopathological examination, the animals were divided in two groups: G1 (n = 8) animals without pulmonary inflammatory disease; G2 (n = 26) animals with pulmonary inflammatory diseases. Fragments of lung and spleen were processed according to routine protocols. The immunohistochemistry and morphometric procedures were tested and optimized according to proceedings described in literature. Our results showed that P. blainvillei from G2 presented significant increase in the pulmonary immunoexpression of TGF-β, TNF-α and IL-12 cytokines, and S-100 and Lysozyme proteins in comparison with animals from G1. These results are according to the literature and suggest that P. blainvillei presents same cytokines observed in other mammals. We evidenced also that the P. blainvillei belonging to the G2 did not show splenic immunoexpression to TGF-β, TNF-α and IL- 12 cytokines and Lysozyme protein significantly higher than animals from G1. Further studies, including those related to the methods used, are necessary for the proper understanding of these results. And finally, although these are still preliminary data, we observed a possible association between high concentrations of organochlorines in subcutaneous adipose tissue of a portion of P. blainvillei studied in this research and the occurrence of splenic lymphoid depletion in these animals. These data confirm the importance of developing more detailed analysis on these topics in future scientific investigations
Uso de diferentes fontes de carboidratos em meio MS na germinação de Cattleya labiata (ORCHIDACEAE).
A família Orchidaceae é uma das mais numerosas entre as fanerógamas, sendo encontrada em quase todas as regiões do planeta, com exemplares até nas regiões boreais. Atualmente, muitas orquídeas em estado silvestre encontram-se em extinção, tendo-se, portanto, a importância de seu cultivo (Moura, 1979).
O gênero Cattleya destaca-se no Brasil na maioria das espécies, tanto pela coloração belíssima, como pelo grande tamanho das flores (Silva, 1986).
As plantas propagadas in vitro necessitam de uma fonte de energia externa pois, nesta fase são praticamente heterotróficas não encontrado as condições favoráveis para realizar fotossíntese. Assim, fontes de carboidratos são adicionadas ao meio nutritivo fornecendo energia metabólica e esqueletos carbônicos para a síntese de compostos orgânicos necessários para o crescimento das células (Caldas; Haridasam; Ferreira, 1998).
A sacarose é o carboidrato mais utilizado em meio de cultura, visando a propagação de plantas ornamentais, sendo que no meio Murashige & Skoog (1962) sua concentração é de 30g.L-1. Na propagação in vitro de orquídeas, outras fontes também têm sido utilizadas com menos freqüência como a glicose, frutose, maltose, entre outros (Tombolato; Costa, 1998).
O tipo e a concentração dos açúcares são importantes para promover a germinação e o crescimento das plântulas in vitro, assim como a própria manutenção de crescimento radicular (Arditti, 1967; Ernest, 1967; Kraus; Kerbauy, 1992; Collins; Dixon, 1992; Kerbauy 1993).
