    Haemoatopoietic alterations induced by carbaryl in Clarias batrachus (LINN)

    The aim of the study was to assess the effect induced by Sevin a carbamate pesticide on hematological indices of Clarias batrachus. The effect was studied based on the results of acute toxicity tests and comparison of control with experimental group. The fish was exposed to varying levels of the toxicant and sub lethal exposure to varying concentrations was used to study the effect on blood cells. LC50 of Sevin for Clarias batrachus after 96hr exposure was found to be 15.3mg/l. Examination of hematological indices was performed on 6 control and 9 experimental specimens after 96hr exposure to Sevin in concentration of 12.6mg .1-1 and 14.6mg .l -1. Reduction in the number of red blood cells, erythrocytes, packed cell volume, Mean corpuscular hemoglobin, mean corpuscular volume decreased indicating Anemia.     A six year old Bengal non-descript pregnant cow weighing 200kg with a hemorrhagic tumour mass in right nasal cavity attached to nasal septum compressing the left passage was presented with stretor, unilateral epistaxis and difficulty in breathing. Administration of Lithium Antimony thiomalate at deep intramuscular route was not effective. Vincrystine @0.025 mg/kg I/V was without response. The growth was removed by dorsal rhinotomy. Diathermy and diluted phenol were used for separation and hemostasis, respectively. In histopathology, the mass was diagnosed as chondro-adenofibroma

    JAK3 Inhibition Regulates Stemness and Thereby Controls Glioblastoma Pathogenesis

    Glioblastoma multiforme (GBM) is the most deadly brain tumor, effective treatment options for which still remain elusive. The current treatment procedure of maximal resection followed by chemotherapy has proved to be grossly insufficient to prevent disease progression and death. Despite best efforts, the maximum survival post-diagnosis is a mere 1.5 years. Therefore, there is a huge unmet clinical need to find effective therapeutic procedures to prevent the pathogenesis and relapse of GBM. Small-molecule inhibitors of signaling pathways are an attractive option to prevent various types of tumors. However, no effective small-molecule inhibitors have been successful against GBM in clinical trials. Various signaling pathways are altered and an array of signaling molecules, transcription factors (TFs), and epigenetic modifying factors have been implicated in the pathogenesis of GBM. JAK-STAT pathway alteration is an important contributor to GBM pathogenesis and relapse. Many small-molecule inhibitors of JAKs, or STAT TFs, especially JAK2 and STAT3, have been assessed for their anti-tumor activity in GBM. However, no definitive success so far has been achieved. Herein, by using two small-molecule inhibitors of JAK3, we show that they are quite effective in inhibiting GBM cell proliferation and neurosphere formation, downregulating their stemness character, and inducing differentiation into neuronal origin cells. The effect of a single treatment with the drugs, both in a serum-containing differentiation medium and in a proliferation medium containing EGF and FGF, was really strong in limiting GBM cell growth, suggesting a potential therapeutic application for these JAK inhibitors in GBM therapy

    Tillage and nutrient management influence net global warming potential and greenhouse gas intensity in soybean-wheat cropping system

    207-214Conservation tillage has proven advantageous in improving soil health and productivity. However, the greenhouse gases (GHGs) emission and intensity from different conservation tillage and nutrient management systems under Indian conditions are less understood. Therefore, here, we compared the effect of tillage and nutrient management on GHGs emissions, net global warming potential (NGWP), and greenhouse gas intensity (GHGI) from a field experiment under five years in a soybean-wheat cropping system in the Vertisols. The tillage treatments comprised of reduced tillage (RT) and no tillage (NT). The three nutrient management treatments included application of 100% NPK (T1), 100% NPK + 1.0 Mg FYM-C ha-1 (T2), 100% NPK +2.0 Mg FYM-C ha-1 (T3). The results showed significantly higher SOC sequestration under NT (1388 kg ha-1 yr-1) followed byRT (1134 kg ha-1 yr-1) with application of FYM (2.0 Mg C ha-1) (T3) every year. Across tillage, integrated nutrient management(T2 and T3) lowered NGWP and GHGI compared to NPK (T1). The GHGI of NT system was less by 33% compared to RT. The results suggest that GHGs mitigation and sustained food production in the soybean-wheat system can be achieved in NT and RT with integrated use of organic and inorganic fertilizer as the major component of nutrient management

    The terrestrial carbon budget of South and Southeast Asia

Available from IOP Publishing via the DOI in this record.Accomplishing the objective of the current climate policies will require establishing carbon budget and flux estimates in each region and county of the globe by comparing and reconciling multiple estimates including the observations and the results of top-down atmospheric carbon dioxide (CO2) inversions and bottom-up dynamic global vegetation models. With this in view, this study synthesizes the carbon source/sink due to net ecosystem productivity (NEP), land cover land use change (E LUC), fires and fossil burning (E FIRE) for the South Asia (SA), Southeast Asia (SEA) and South and Southeast Asia (SSEA = SA + SEA) and each country in these regions using the multiple top-down and bottom-up modeling results. The terrestrial net biome productivity (NBP = NEP - E LUC - E FIRE) calculated based on bottom-up models in combination with E FIRE based on GFED4s data show net carbon sinks of 217 ±147, 10 ±55, and 227 ±279 TgC yr-1 for SA, SEA, and SSEA. The top-down models estimated NBP net carbon sinks were 20 ±170, 4 ±90 and 24 ±180 TgC yr-1. In comparison, regional emissions from the combustion of fossil fuels were 495, 275, and 770 TgC yr-1, which are many times higher than the NBP sink estimates, suggesting that the contribution of the fossil fuel emissions to the carbon budget of SSEA results in a significant net carbon source during the 2000s. When considering both NBP and fossil fuel emissions for the individual countries within the regions, Bhutan and Laos were net carbon sinks and rest of the countries were net carbon source during the 2000s. The relative contributions of each of the fluxes (NBP, NEP, E LUC, and E FIRE, fossil fuel emissions) to a nation's net carbon flux varied greatly from country to country, suggesting a heterogeneous dominant carbon fluxes on the country-level throughout SSEA.This research was partly supported by the NASA Land Cover and Land Use Change Program (NNX14AD94G) and the US National Science Foundation (No. NSF-AGS-12-43071)

    A threshold level of NFATc1 activity facilitates thymocyte differentiation and opposes notch-driven leukaemia development.

    International audienceNFATc1 plays a critical role in double-negative thymocyte survival and differentiation. However, the signals that regulate Nfatc1 expression are incompletely characterized. Here we show a developmental stage-specific differential expression pattern of Nfatc1 driven by the distal (P1) or proximal (P2) promoters in thymocytes. Whereas, preTCR-negative thymocytes exhibit only P2 promoter-derived Nfatc1beta expression, preTCR-positive thymocytes express both Nfatc1beta and P1 promoter-derived Nfatc1alpha transcripts. Inducing NFATc1alpha activity from P1 promoter in preTCR-negative thymocytes, in addition to the NFATc1beta from P2 promoter impairs thymocyte development resulting in severe T-cell lymphopenia. In addition, we show that NFATc1 activity suppresses the B-lineage potential of immature thymocytes, and consolidates their differentiation to T cells. Further, in the pTCR-positive DN3 cells, a threshold level of NFATc1 activity is vital in facilitating T-cell differentiation and to prevent Notch3-induced T-acute lymphoblastic leukaemia. Altogether, our results show NFATc1 activity is crucial in determining the T-cell fate of thymocytes

    Cancer-selective, single agent chemoradiosensitising gold nanoparticles

    Two nanometre gold nanoparticles (AuNPs), bearing sugar moieties and/or thiol-polyethylene glycol-amine (PEG-amine), were synthesised and evaluated for their in vitro toxicity and ability to radiosensitise cells with 220 kV and 6 MV X-rays, using four cell lines representing normal and cancerous skin and breast tissues. Acute 3 h exposure of cells to AuNPs, bearing PEG-amine only or a 50:50 ratio of alpha-galactose derivative and PEG-amine resulted in selective uptake and toxicity towards cancer cells at unprecedentedly low nanomolar concentrations. Chemotoxicity was prevented by co-administration of N-acetyl cysteine antioxidant, or partially prevented by the caspase inhibitor Z-VAD-FMK. In addition to their intrinsic cancer-selective chemotoxicity, these AuNPs acted as radiosensitisers in combination with 220 kV or 6 MV X-rays. The ability of AuNPs bearing simple ligands to act as cancer-selective chemoradiosensitisers at low concentrations is a novel discovery that holds great promise in developing low-cost cancer nanotherapeutics
