    Fingolimod reduces hemorrhagic transformation associated with delayed tissue plasminogen activator treatment in a mouse thromboembolic model

    BACKGROUND AND PURPOSE: The sphingosine 1-phosphate receptor agonist fingolimod reduces infarct size in rodent models of stroke and enhances blood-brain barrier integrity. Based on these observations, we hypothesized that combination of fingolimod with tissue plasminogen activator (tPA) would reduce the risk of hemorrhagic transformation associated with delayed administration of tPA. METHODS: We evaluated the effects of fingolimod in a mouse model of thromboembolic stroke, in which both the beneficial effect of reperfusion associated with early tPA treatment and hemorrhagic transformation associated with delayed administration mimic clinical observations in humans. RESULTS: Our results demonstrate that fingolimod treatment attenuates the neurological deficit and reduces infarct volume after in situ thromboembolic occlusion of the middle cerebral artery. Combination of fingolimod and tPA improves the neurological outcome of the thrombolytic therapy and reduces the risk of hemorrhagic transformation associated with delayed administration of tPA. CONCLUSIONS: This study confirms the protective efficacy of fingolimod as a treatment against ischemic stroke in another rodent model of stroke (thromboembolic occlusion), and suggests that fingolimod could potentially be used in combination with tPA to reduce the risk of brain hemorrhage

    Reliability and validity of the geriatric depression scale in a sample of Portuguese older adults with mild-to-moderate cognitive impairment

    Although the Geriatric Depression Scale (GDS) is a well-established instrument for the assessment of depressive symptoms in older adults, this has not been validated specifically for Portuguese older adults with cognitive impairment. The objective of this study was to analyze the psychometric properties of two Portuguese versions of the GDS (GDS-27 and GDS-15) in a sample of Portuguese older adults with mild-to-moderate cognitive impairment. Clinicians assessed for major depressive disorder and cognitive functioning in 117 participants with mild-to-moderate cognitive decline (76.9% female, Mage = 83.66 years). The internal consistency of GDS-27 and GDS-15 were 0.874 and 0.812, respectively. There was a significant correlation between GDS-27 and GDS-15 with the Beck Depression Inventory-II (GDS-27: rho = 0.738, p < 0.001; GDS-15: rho = 0.760, p < 0.001 ), suggesting good validity. A cutoff point of 15/16 in GDS-27 and 8/9 in GDS-15 resulted in the identification of persons with depression (GDS-27: sensitivity 100%, specificity 63%; GDS-15: sensitivity 90%, specificity 62%). Overall, the GDS-27 and GDS-15 are reliable and valid instruments for the assessment of depression in Portuguese-speaking older adults with cognitive impairment.info:eu-repo/semantics/publishedVersio

    Examination of the Feynman-Hibbs Approach in the Study of NeN_N-Coronene Clusters at Low Temperatures

    Feynman-Hibbs (FH) effective potentials constitute an appealing approach for investigations of many-body systems at thermal equilibrium since they allow us to easily include quantum corrections within standard classical simulations. In this work we apply the FH formulation to the study of NeN_N-coronene clusters (N=N= 1-4, 14) in the 2-14 K temperature range. Quadratic (FH2) and quartic (FH4) contributions to the effective potentials are built upon Ne-Ne and Ne-coronene analytical potentials. In particular, a new corrected expression for the FH4 effective potential is reported. FH2 and FH4 cluster energies and structures -obtained from energy optimization through a basin-hoping algorithm as well as classical Monte Carlo simulations- are reported and compared with reference path integral Monte Carlo calculations. For temperatures T>4T> 4 K, both FH2 and FH4 potentials are able to correct the purely classical calculations in a consistent way. However, the FH approach fails at lower temperatures, especially the quartic correction. It is thus crucial to assess the range of applicability of this formulation and, in particular, to apply the FH4 potentials with great caution. A simple model of NN isotropic harmonic oscillators allows us to propose a means of estimating the cut-off temperature for the validity of the method, which is found to increase with the number of atoms adsorbed on the coronene molecule

    UHMWPE/SBA-15 nanocomposites synthesized by in situ polymerization

    Different nanocomposites have been attained by in situ polymerization based on ultra-high molecular weight polyethylene (UHMWPE) and mesoporous SBA-15, this silica being used for immobilization of the FI catalyst bis [N-(3-tert-butylsalicylidene)-2,3,4,5,6-pentafluoroanilinato] titanium (IV) dichloride and as filler as well. Two distinct approaches have been selected for supporting the FI catalyst on the SBA-15 prior polymerization. A study on polymerization activity of this catalyst has been performed under homogenous conditions and upon heterogenization. A study of the effect of presence of mesoporous particles and of the immobilization method is also carried out. Moreover, the thermal characterization, phase transitions and mechanical response of some pristine UHMWPEs and UHMWPE/SBA-15 materials have been carried out.     Association between the histopathologic findings of a zero-time biopsy and the donor kidney function 24 hours and a year after nephrectomy

    Background: The living-donor transplantation (LDT) is essential to provide patients with chronic kidney disease (CKD) a superior quality of life and improve their lifespan. Recent investigations prove that the living donors (LD) have a risk of developing CKD, without there being a way to anticipate it. The zero-time biopsies provide valuable information that may contribute to this objective since they report histopathologic findings of subclinical chronic damage. Methods: Retrospective, observational and analytical study. The information from the medical files and pathology department of LD attended at “Dr. Miguel Silva” general hospital from January 2006 to January 2018 was analyzed. The glomerular filtrate rate was obtained 24 hours and a year after nephrectomy and was estimated based on CDK-EPI. The comparison among groups was made through Mann-Whitney testing for continuous numeric. A value of p36 years, the dropping of the GFR >43%, the GFR and the creatinine levels after 24 hours of nephrectomy were associated with a reduction of the GFR the year after nephrectomy

    Dataset from analytical pyrolysis assays for converting waste tires into valuable chemicals in the presence of noble-metal catalysts

    About 25.7 million tons of waste tires (WT) are discarded each year worldwide causing important environmental, and health problems. This waste is difficult to manage and dispose due to its huge rate of generation and its extremely slow biodegradation. Therefore, many efforts are being made to valorise WTs into a series of marketable products under a circular economy framework. In the attempt to convert WT into higher-value products, thermochemical decomposition by pyrolysis has emerged as a promising process. The pyrolysis is a thermochemical transformation (under an oxygen-depleted atmosphere) of the tire´s polymeric constituents: natural rubber (NR), styrene-butadiene rubber (SBR), and butadiene rubber (BR) into three major fractions. These fractions are a gas (10–35%, TPG) which is usually used as a heat source (50 MJ kg⁻¹), a solid consisting mainly of recovered carbon black (12–45%, rCB), and a liquid fraction (35-65%, TPO) containing a complex mixture of organic compounds. Among the high-value compounds that can be found in the TPO are D,L-limonene, isoprene, benzene, toluene, mixed-xylene, ethylbenzene, styrene, p-cymene, and some polycyclic aromatic hydrocarbons. This mixture is commonly used as a diesel substitute and owing to its complex composition it rarely is seen as a source for more valuable products. To overcome such a complexity, and selectively produce specific chemical identities, different types of catalysts have been used. Herein, we provide a dataset from a systematic study about catalytic pyrolysis of WT for selectively producing benzene, toluene, and xylenes (BTX) and p-cymene on noble metals (Pd, Pt, Au) supported on titanate nanotubes (NT-Ti). The comprehensive analysis of this data was recently published, thus, the analytical techniques, experimental conditions and dataset are given in the present paper as a complement to that publication. The reaction was evaluated in an analytical pyrolysis unit consisting in a micropyrolizer coupled to a mass spectrometer (Py-GC/MS) operating at temperatures between 400 and 450 °C in a fast pyrolysis regime (12 s). The effectivity of catalysts was measured in terms of selectivity to monoaromatics as BTX and p-cymene, under non-catalytic and for catalytic pyrolysis conditions. Moreover, the reaction was conducted on individual rubbers (Polyisoprene, Polybutadiene, and Styrene-Butadiene) and DL-limonene, to get deep insights into the transformation behaviour and reaction pathways. Therefore, the reader will find a data-in-brief paper containing some characterizations of the WTs used for the investigation, along with a complete dataset of Py-GC/MS results.     Height-based equations as screening tools for high blood pressure in pediatric practice, the GENOBOX study

    Due to the absence of easily applicable cut-off points to determine high blood pressure or hypertension in children, as in the adult population, blood pressure is rarely measured in the pediatrician''s clinical routine. This has led to an underdiagnosis of high blood pressure or hypertension in children. For this reason, the present study evaluate the utility of five equations for the screening of high blood pressure in children: blood pressure to height ratio, modified blood pressure to height ratio, new modified blood pressure to height ratio, new simple formula and height-based equations. The authors evaluated 1599 children between 5 and 18 years. The performance of the five equations was analyzed using the receiver-operating characteristics curves for identifying blood pressure above P90th according to the American Academy of Pediatrics Clinical Practice Guideline 2017. All equations showed an area under the curve above 0.882. The new modified blood pressure to height ratio revealed a high sensitivity whereas the height-based equations showed the best performance, with a positive predictive value above 88.2%. Finally, all equations showed higher positive predictive values in children with overweight or obesity. The height-based equation obtained the highest PPV values above 71.1% in children with normal weight and above 90.2% in children with overweight or obesity. In conclusions, the authors recommend the use of the height-based equations equation because it showed the best positive predictive values to identify children with elevated blood pressure, independently of their sex, pubertal and weight status. © 2022 The Authors.
