Caracterización preliminar de frutos de granada china (Passiflora ligularis Juss.) en Hueyapan y Teziutlán, Puebla
En la Sierra Nororiental de Puebla, México, los frutos de granada china son muy apreciados por la población local, pero existen pocos datos sobre su calidad hortofrutícola. Con el fin de iniciar la selección de individuos productores de frutos de buena calidad, se tomaron como muestra diez frutos de 20 plantas, cinco de cada localidad, Talzinta, Nexpan, Tanamacoyan y Mexcalcuautla, para analizarlos física (peso, diámetro, color, porcentaje de los componentes del fruto) y bioquímicamente (sólidos solubles totales, pH, acidez titulable, densidad del jugo). El valor medio para peso de frutos varió de 85 a 57 g; el porcentaje de pulpa de 81 a 51%; y el contenido de sólidos solubles totales (SST) fue de 17.5 a 12.2 ºB. El análisis de conglomerados, a una distancia euclidiana de 12.6, formó cinco grupos; en los grupos 2, 3 y 4 se observaron frutos de mayor tamaño (+ 75 g), con altos porcentajes de pulpa (60%) y valores aceptables de SST (14 ºB); éstos fueron los colectados en Talzinta y Nexpan. El análisis de componentes principales (CP) indicó que los primeros cinco CP explicaron el 75% de la variabilidad observada; siendo 25 y 18% la variabilidad que explican los CP 1 y CP 2, respectivamente. CP 1 estuvo altamente influenciado por el diámetro polar (0.900), luminosidad (-0.767) y el peso fresco del fruto (0.750); el CP 2 se relacionó con porcentaje de pulpa (0.866), porcentaje de cáscara (-0.866) y porcentaje de semillas (0.857). Accelerated directed evolution of dye-decolorizing peroxidase using a bacterial extracellular protein secretion system (BENNY)
Background
Dye-decolorizing peroxidases (DyPs) are haem-containing peroxidases that show great promises in industrial biocatalysis and lignocellulosic degradation. Through the use of Escherichia coli osmotically-inducible protein Y (OsmY) as a bacterial extracellular protein secretion system (BENNY), we successfully developed a streamlined directed evolution workflow to accelerate the protein engineering of DyP4 from Pleurotus ostreatus strain PC15.
Result
After 3 rounds of random mutagenesis with error-prone polymerase chain reaction (epPCR) and 1 round of saturation mutagenesis, we obtained 4D4 variant (I56V, K109R, N227S and N312S) that displays multiple desirable phenotypes, including higher protein yield and secretion, higher specific activity (2.7-fold improvement in kcat/Km) and higher H2O2 tolerance (sevenfold improvement based on IC50).
Conclusion
To our best knowledge, this is the first report of applying OsmY to simplify the directed evolution workflow and to direct the extracellular secretion of a haem protein such as DyP4
Assessment of the toll-like receptor 4 Asp299Gly, Thr399Ile and interleukin-8 -251 polymorphisms in the risk for the development of distal gastric cancer
<p>Abstract</p> <p>Background</p> <p>The intensity of the inflammation induced by <it>Helicobacter pylori </it>colonization is associated with the development of distal gastric cancer (GC). The host response to <it>H</it>. <it>pylori </it>has been related to genetic polymorphisms that influence both innate and adaptive immune responses.</p> <p>Our aim was to investigate whether the presence of the <it>TLR4 Asp299Gly</it>, <it>TLR4 Thr399Ile </it>and <it>IL-8-251 </it>A/T polymorphisms had any influence in the development of distal GC in a Mexican population.</p> <p>Methods</p> <p>We studied 337 patients that were divided in two groups: 78 patients with histologically confirmed distal GC and 259 non-cancer controls. The presence of <it>H. pylori </it>in the control population was defined by positive results of at least two of four diagnostic tests: serology, histology, rapid urease test and culture. Human DNA was purified and genotyped for <it>TLR4 Asp299Gly </it>polymorphism by pyrosequencing, for <it>TLR4 Thr399Ile </it>by PCR-RFLP and for <it>IL8-251 </it>by the amplification refractory mutation system (ARMS)-PCR.</p> <p>Results</p> <p>The non-cancer control group was found to be in Hardy-Weinberg equilibrium at the polymorphic loci studied (chi-square <sub>H-W </sub>= 0.58 for <it>IL8-251</it>, 0.42 for <it>TLR4 Asp299Gly </it>and 0.17 for <it>TLR4 Thr399Ile</it>). The frequencies of mutated alleles (homozygous plus heterozygous) were compared between cases and controls. We found no significant difference for <it>TLR4- Asp299Gly </it>[the 7.7% of distal GC patients and 7.7 % non-cancer controls (p = 0.82)] and for <it>TLR4 Thr399Ile </it>[the 1.3% of GC patients and the 5% of the control population (p = 0.2)]. In contrast, for <it>IL-8-251 </it>A/T, 80.77% of the GC patients and 66.4% in the control group age and gender matched had at least one copy of mutated allele (OR = 2.12, 95% CI = 1.1–4.2) (p = 0.023).</p> <p>Conclusion</p> <p>This study showed that the <it>IL8-251*A </it>allele could be related to the development of distal gastric cancer in this Mexican population.</p
Increased circulating ANG II and TNF-α represents important risk factors in obese Saudi adults with hypertension irrespective of diabetic status and BMI
Central adiposity is a significant determinant of obesity-related hypertension risk, which may arise due to the pathogenic inflammatory nature of the abdominal fat depot. However, the influence of pro-inflammatory adipokines on blood pressure in the obese hypertensive phenotype has not been well established in Saudi subjects. As such, our study investigated whether inflammatory factors may represent useful biomarkers to delineate hypertension risk in a Saudi cohort with and without hypertension and/or diabetes mellitus type 2 (DMT2). Subjects were subdivided into four groups: healthy lean controls (age: 47.9±5.1 yr; BMI: 22.9±2.1 Kg/m2), non-hypertensive obese (age: 46.1±5.0 yr; BMI: 33.7±4.2 Kg/m2), hypertensive obese (age: 48.6±6.1 yr; BMI: 36.5±7.7 Kg/m2) and hypertensive obese with DMT2 (age: 50.8±6.0 yr; BMI: 35.3±6.7 Kg/m2). Anthropometric data were collected from all subjects and fasting blood samples were utilized for biochemical analysis. Serum angiotensin II (ANG II) levels were elevated in hypertensive obese (p<0.05) and hypertensive obese with DMT2 (p<0.001) compared with normotensive controls. Systolic blood pressure was positively associated with BMI (p<0.001), glucose (p<0.001), insulin (p<0.05), HOMA-IR (p<0.001), leptin (p<0.01), TNF-α (p<0.001) and ANG II (p<0.05). Associations between ANG II and TNF-α with systolic blood pressure remained significant after controlling for BMI. Additionally CRP (p<0.05), leptin (p<0.001) and leptin/adiponectin ratio (p<0.001) were also significantly associated with the hypertension phenotype. In conclusion our data suggests that circulating pro-inflammatory adipokines, particularly ANG II and, TNF-α, represent important factors associated with a hypertension phenotype and may directly contribute to predicting and exacerbating hypertension risk
European propolis is highly active against trypanosomatids including Crithidia fasciculata.
Extracts of 35 samples of European propolis were tested against wild type and resistant strains of the protozoal pathogens Trypanosoma brucei, Trypanosoma congolense and Leishmania mexicana. The extracts were also tested against Crithidia fasciculata a close relative of Crithidia mellificae, a parasite of bees. Crithidia, Trypanosoma and Leishmania are all members of the order Kinetoplastida. High levels of activity were obtained for all the samples with the levels of activity varying across the sample set. The highest levels of activity were found against L. mexicana. The propolis samples were profiled by using liquid chromatography with high resolution mass spectrometry (LC-MS) and principal components analysis (PCA) of the data obtained indicated there was a wide variation in the composition of the propolis samples. Orthogonal partial least squares (OPLS) associated a butyrate ester of pinobanksin with high activity against T. brucei whereas in the case of T. congolense high activity was associated with methyl ethers of chrysin and pinobanksin. In the case of C. fasciculata highest activity was associated with methyl ethers of galangin and pinobanksin. OPLS modelling of the activities against L. mexicana using the mass spectrometry produced a less successful model suggesting a wider range of active components

We investigated the clinical and prognostic importance of whole blood global and site-specific DNA methylation in GC. tests. Burden and challenges of heart failure in patients with chronic kidney disease. A call to action
Patients with the dual burden of chronic kidney disease (CKD) and chronic congestive heart
failure (HF) experience unacceptably high rates of symptom load, hospitalization, and mortality. Currently, concerted efforts to identify, prevent and treat HF in CKD patients are
lacking at the institutional level, with emphasis still being placed on individual specialty
views on this topic. The authors of this review paper endorse the need for a dedicated
cardiorenal interdisciplinary team that includes nephrologists and renal nurses and jointly manages appropriate clinical interventions across the inpatient and outpatient settings.
There is a critical need for guidelines and best clinical practice models from major cardiology
and nephrology professional societies, as well as for research funding in both specialties to
focus on the needs of future therapies for HF in CKD patients. The implementation of crossspecialty educational programs across all levels in cardiology and nephrology will help train
future specialists and nurses who have the ability to diagnose, treat, and prevent HF in CKD
patients in a precise, clinically effective, and cost-favorable manner.Los pacientes con enfermedad renal crónica (ERC) que desarrollan insuficiencia cardíaca (IC)
congestiva crónica presentan cifras inaceptablemente altas de síntomas, hospitalización y
mortalidad. Actualmente, se echan en falta iniciativas institucionales dirigidas a identificar,
prevenir y tratar la IC en los pacientes con ERC de manera multidisciplinar, prevaleciendo
las actuaciones de las especialidades individuales. Los autores de este artículo de revisión
respaldan la necesidad de crear equipos multidisciplinares cardiorrenales, en los que participen nefrólogos y enfermeras renales, que gestionen colaborativamente las intervenciones
clínicas apropiadas en los entornos de pacientes con ERC e IC hospitalizados y ambulatorios.
Es necesario y urgente que se elaboren guías y modelos de práctica clínica sobre la ERC con IC
por parte de las sociedades profesionales de cardiología y nefrología, así como financiación
para la investigación concertada entre ambas especialidades sobre la necesidad de futuros
tratamientos para la IC en pacientes con ERC. La implementación de programas educativos
cardiorrenales a todos los niveles en cardiología y nefrología ayudará a formar a los futuros
especialistas y enfermeras para que tengan la capacidad de diagnosticar, tratar y prevenir

Propagating Quantum Microwaves: Towards Applications in Communication and Sensing
The field of propagating quantum microwaves has started to receive
considerable attention in the past few years. Motivated at first by the lack of
an efficient microwave-to-optical platform that could solve the issue of secure
communication between remote superconducting chips, current efforts are
starting to reach other areas, from quantum communications to sensing. Here, we
attempt at giving a state-of-the-art view of the two, pointing at some of the
technical and theoretical challenges we need to address, and while providing
some novel ideas and directions for future research. Hence, the goal of this
paper is to provide a bigger picture, and -- we hope -- to inspire new ideas in
quantum communications and sensing: from open-air microwave quantum key
distribution to direct detection of dark matter, we expect that the recent
efforts and results in quantum microwaves will soon attract a wider audience,
not only in the academic community, but also in an industrial environment
Cosmic CARNage II: the evolution of the galaxy stellar mass function in observations and galaxy formation models
We present a comparison of the observed evolving galaxy stellar mass functions with the predictions of eight semi-analytic models and one halo occupation distribution model. While most models are able to fit the data at low redshift, some of them struggle to simultaneously fit observations at high redshift. We separate the galaxies into 'passive' and 'star-forming' classes and find that several of the models produce too many low-mass star-forming galaxies at high redshift compared to observations, in some cases by nearly a factor of 10 in the redshift range 2.5 < z < 3.0. We also find important differences in the implied mass of the dark matter haloes the galaxies inhabit, by comparing with halo masses inferred from observations. Galaxies at high redshift in the models are in lower mass haloes than suggested by observations, and the star formation efficiency in low-mass haloes is higher than observed. We conclude that many of the models require a physical prescription that acts to dissociate the growth of low-mass galaxies from the growth of their dark matter haloes at high redshift.Facultad de Ciencias Astronómicas y GeofísicasInstituto de Astrofísica de La Plat
