Digital and Territorial Trails System for Developing Sustainable Tourism and Enhancing Cultural Heritage in Rural Areas: The Case of San Giovanni Lipioni, Italy
Depopulation has become a significant issue for local culture and built heritage conservation of many European rural areas. In San Giovanni Lipioni, a province of Chieti (Italy), this phenomenon has increased to the point that, nowadays, there are only 150 inhabitants and no significant economic activities. In this regard, the present paper aims to describe the crucial role of nature-oriented tourism in an economic, social, and revitalization strategy; how digital tools can be used to map and create a territorial trail system between municipalities; and, finally, outline the operations necessary for reactivation. The proposed methodology consists of a first digital survey phase using GPS receivers and outdoor navigation apps. The second phase would create a web platform with a system of virtual itineraries between villages, named âThe Golden Leaves Pathsâ. After that, the last phase concerns the creation of analysis factsheets to guide the maintenance of paths and the design of iconic signage with artistic illustrations based on the oak leaves leitmotif to be installed along the paths. A local social promotion association will employ the outcomes, technical drawings, and strategies to reactivate paths as an attractive element for nature-oriented tourism and create a digital platform to foster the villageâs territorial and cultural heritage
Passive Sampling of Organic Contaminants as a Novel Approach to Monitor Seawater Quality in Aquarium Ocean Tanks
The determination of trace pollutants in seawater is challenging, and sampling is a crucial step in the entire analytical process. Passive samplers combine in situ sampling and preconcentration, thus limiting the tedious treatment steps of the conventional sampling methods. Their use to monitor water quality in confined marine environment could bring several advantages. In this work, the presence of organic contaminants at trace and ultra-trace levels was assessed in the Genoa Aquarium supply-and-treated water using Polar Organic Integrative Samplers (POCIS). Both untargeted gas chromatography-mass spectrometry and targeted liquid chromatography-tandem mass spectrometry were employed. The untargeted approach showed the presence of hydrocarbons, diphenyl sulfone and 2,4-di-tert-butyl-phenol. Only hydrocarbons were detected in all the samples. Nineteen emerging contaminants, belonging to different classes (pharmaceuticals, UV-filters, hormones and perfluorinated compounds), were selected for the target analysis. Thirteen analytes were detected, mainly in supply water, even though the majority of them were below the quantitation limit. It is worthy to note that two of the detected UV-filters had never been reported in seawater using the POCIS samplers. The comparison of the analytes detected in supply and treated water indicated a good performance of the Aquarium water treatment system in the abatement of seawater contaminants

Is pain sensitivity altered in people with Alzheimer's disease? A systematic review and meta-analysis of experimental pain research
Background: Clinical studies suggest people with Alzheimer's disease (AD) have altered pain sensitivity. Experimental pain research is equivocal.
Objective: Conduct a meta-analysis to investigate if people with AD have altered pain sensitivity compared to healthy controls (HCs).
Methods: Three authors searched electronic databases from inception till November 2015 for experimental pain studies in AD vs. HCs. Outcome measures were pain threshold, tolerance, pain ratings, heart rate response to nox- ious stimuli and the Facial Action Coding System (FACS). Random effect meta-analysis calculating Hedges' g ± 95% confidence intervals (CI) was conducted.
Results: Thirteen studies were identified, including 256 people with AD (74.6 (±5.6) years, 59% females with a mean mini mental state examination (MMSE) score of 19.2) and 260 HCs. Meta-analysis demonstrated no signif- icant difference in pain threshold (g = 0.025, 95% CI â0.315-0.363, p = 0.88, n AD = 135, n HCs = 157), pain tolerance (g = â0.363, 95% CI â2.035-1.309, p = 0.67, n AD = 41, n HCs = 53) or pain intensity ratings (g = 0.03, p = 0.89, n AD = 138, n HCs = 135). Heart rate response to pain was less pronounced in AD but not significant (g = â0.746, p = 0.11). People with AD (n = 90) had significantly higher FACS scores versus HCs (n = 109) (g = 0.442, p = 0.03) indicating increased pain. Meta-regression demonstrated that an increasing percentage of AD female participants moderated pain threshold (p = 0.02) whilst MMSE scores did not (p = 0.19).
Conclusion: People with AD have a greater sensitivity to pain when validated observer ratings of facial expres- sions are used. Verbal response to painful stimuli, even under experimental conditions, may mean pain is not identified in people with AD. Clinically useful observer rated pain tools may be the most appropriate way to as- sess pain in AD
Tailoring Optical Complex Field with Spiral Blade Plasmonic Vortex Lens
Optical complex fields have attracted increasing interests because of the novel effects and phenomena arising from the spatially inhomogeneous state of polarizations and optical singularities of the light beam. In this work, we propose a spiral blade plasmonic vortex lens (SBPVL) that offers unique opportunities to manipulate these novel fields. The strong interaction between the SBPVL and the optical complex fields enable the synthesis of highly tunable plasmonic vortex. Through theoretical derivations and numerical simulations we demonstrated that the characteristics of the plasmonic vortex are determined by the angular momentum (AM) of the light, and the geometrical topological charge of the SBPVL, which is govern by the nonlinear superposition of the pitch and the number of blade element. In addition, it is also shown that by adjusting the geometric parameters, SBPVL can be utilized to focus and manipulate optical complex field with fractional AM. This miniature plasmonic device may find potential applications in optical trapping, optical data storage and many other related fields
Punctual and kinetic MRD analysis from the Fondazione Italiana Linfomi MCL0208 phase III trial in mantle cell lymphoma
Minimal residual disease (MRD) analysis is a known predictive tool in mantle cell lymphoma (MCL). We describe MRD results from the Fondazione Italiana Linfomi phase III MCL0208 prospective clinical trial assessing lenalidomide maintenance vs observation after autologous transplantation (ASCT), in the first prospective comprehensive analysis of different techniques, molecular markers, and tissues (peripheral blood, PB, and bone marrow, BM), taken at well-defined timepoints. Among the 300 patients enrolled, a molecular marker was identified in 250 (83%), allowing us to analyze 234 patients and 4351 analytical findings from 10 timepoints. ASCT induced high rates of molecular remission (91% in PB and 83% in BM, by quantitative real-time PCR [RQ-PCR]). Nevertheless, the number of patients with persistent clinical and molecular remission decreased over time in both arms (up to 30% after 36 months). MRD predicted early progression and long-term outcome, particularly from 6 months after ASCT (6-month TTP HR 3.83, p<0.001). In single-timepoint analysis, BM outperformed PB, and RQ-PCR was more reliable, while nested PCR appeared applicable to a larger number of patients (234 vs 176). To improve MRD performance we developed a time-varying kinetic model, based on regularly updated MRD results and the Mantle Cell Lymphoma International Prognostic Index, showing an area under the ROC curve (AUROC) of up to 0.87 using BM. Most notably, PB reached an AUROC of up to 0.81: with kinetic analysis it was comparable to BM in performance. MRD is a powerful predictor over the entire natural history of MCL and suitable for models with continuous adaptation of patient risk. Glucose transporter type 10âlacking in arterial tortuosity syndromeâfacilitates dehydroascorbic acid transport
Loss-of-function mutations in the gene encoding GLUT10 are responsible for arterial tortuosity syndrome (ATS), a rare connective tissue disorder. In this study GLUT10-mediated dehydroascorbic acid (DAA) transport was investigated, supposing its involvement in the pathomechanism. GLUT10 protein produced by in vitro translation and incorporated into liposomes efficiently transported DAA. Silencing of GLUT10 decreased DAA transport in immortalized human fibroblasts whose plasma membrane was selectively permeabilized. Similarly, the transport of DAA through endomembranes was markedly reduced in fibroblasts from ATS patients. Re-expression of GLUT10 in patientsâ fibroblasts restored DAA transport activity. The present results demonstrate that GLUT10 is a DAA transporter and DAA transport is diminished in the endomembranes of fibroblasts from ATS patients
GLUT10-Lacking in Arterial Tortuosity Syndrome-Is Localized to the Endoplasmic Reticulum of Human Fibroblasts.
GLUT10 belongs to a family of transporters that catalyze the uptake of sugars/polyols by facilitated diffusion. Loss-of-function mutations in the SLC2A10 gene encoding GLUT10 are responsible for arterial tortuosity syndrome (ATS). Since subcellular distribution of the transporter is dubious, we aimed to clarify the localization of GLUT10. In silico GLUT10 localization prediction suggested its presence in the endoplasmic reticulum (ER). Immunoblotting showed the presence of GLUT10 protein in the microsomal, but not in mitochondrial fractions of human fibroblasts and liver tissue. An even cytosolic distribution with an intense perinuclear decoration of GLUT10 was demonstrated by immunofluorescence in human fibroblasts, whilst mitochondrial markers revealed a fully different decoration pattern. GLUT10 decoration was fully absent in fibroblasts from three ATS patients. Expression of exogenous, tagged GLUT10 in fibroblasts from an ATS patient revealed a strict co-localization with the ER marker protein disulfide isomerase (PDI).
