    How fMRI Can Inform Cognitive Theories

    How can functional magnetic resonance imaging (fMRI) advance cognitive theory? Some have argued that fMRI can do little beyond localizing brain regions that carry out certain cognitive functions (and may not even be able to do that). However, in this article, we argue that fMRI can inform theories of cognition by helping to answer at least four distinct kinds of questions. Which mental functions are performed in brain regions specialized for just that function (and which are performed in more general-purpose brain machinery)? When fMRI markers of a particular Mental Process X are found, is Mental Process X engaged when people perform Task Y? How distinct are the representations of different stimulus classes? Do specific pairs of tasks engage common or distinct processing mechanisms? Thus, fMRI data can be used to address theoretical debates that have nothing to do with where in the brain a particular process is carried out

    Background: Acquired Immuno-Deficiency Syndrome (AIDS) is the most severe manifestation of Human Immunodeficiency Virus (HIV) infection. AIDS was first reported in the world in 1981 by the Centers for Disease Control and Prevention. Statewide surveillance for AIDS was begun in 1982. Because the cause of AIDS was unknown at that time, the surveillance case definition included many opportunistic infections and tumors. Persons with AIDS were noted to have abnormalities in their immune system that left them susceptible to certain infections. As more information became available, the AIDS surveillance case definition was modified. In 1984, HIV was found to be the cause of AIDS. HIV infects a specific cell of the immune system, the T-lymphocyte, and kills the cell. Very often, HIV infection is without symptoms, and people do not know they are infected. However, they carry the virus in their blood and other body fluids and can infect other persons exposed to these fluids. Persons with unrecognized and untreated HIV infection may not have symptoms for years. The average time from untreated HIV infection to AIDS is 8 to 10 years. Many drugs are now available to treat HIV infection. The usual regimen is a combination of drugs that are taken daily. The goal of treatment is to reduce the amount of virus in the blood to “undetectable” levels by laboratory methods, and to maintain a level of T-lymphocytes that keeps the immune system function intact. When a person with HIV infection stays on an effective treatment regimen they may never reach the AIDS stage. Therefore, AIDS surveillance will not be a true indicator of the burden of HIV disease in our communities. In 1992, Nevada initiated mandatory reporting of HIV infection by name. The purpose was to find persons with early HIV infection and ensure that they were educated about their disease and referred to appropriate treatment. Therefore, Nevada has a surveillance system both for HIV infection and for AIDS. Not all states have HIV surveillance; therefore AIDS cases are used for comparison of relative rates of cases between states. AIDS cases in this report are cases where the person who has HIV has developed the disease called AIDS. HIV cases in this report are cases where the person has the virus called HIV, but has not yet developed the disease called AIDS. Once an HIV case becomes an AIDS case, the AIDS and HIV surveillance system is updated to reflect that occurrence. This report provides the results of both cases and gives definitions for both

    Early temperamental traits in an octopus (Octopus bimaculoides).

    During their 3rd week of life, 73 Octopus bimaculoides were observed to test whether discrete behaviors could be grouped reliably to reflect dimensions of temperament. Frequencies of behaviors during Week 3 were subjected to principal-components analysis (PCA), resulting in 4 components (active engagement, arousal/readiness, aggression, and avoidance/disinterest) that explain 53% of the variance. Levels of temperamental traits were then evaluated for 37 octopuses using composite scores at 3 time points across the first 9 weeks of life. Profile analysis revealed significant change for the testing group as a whole in trait expression levels from Week 3 to Week 6. Results also suggest a significant effect of relatedness on developing temperamental profiles of octopuses. Discussion focuses on how results apply to the life history of 0. bimaculoides and what temperament can reveal about adaptive individuality in a protostome

    Statewide Molecular Epidemiology of Mycobacterium tuberculosis Transmission in a Moderate- to Low-Incidence State: Are Contact Investigations Enough?

    To assess the circumstances of recent transmission of tuberculosis (TB) (progression to active disease <2 years after infection), we obtained DNA fingerprints for 1,172 (99%) of 1,179 Mycobacterium tuberculosis isolates collected from Maryland TB patients from 1996 to 2000. We also reviewed medical records and interviewed patients with genetically matching M. tuberculosis strains to identify epidemiologic links (cluster investigation). Traditional settings for transmission were defined as households or close relatives and friends; all other settings were considered nontraditional. Of 436 clustered patients, 114 had recently acquired TB. Cluster investigations were significantly more likely than contact investigations to identify patients who recently acquired TB in nontraditional settings (33/42 vs. 23/72, respectively; p<0.001). Transmission from a foreign-born person to a U.S.-born person was rare and occurred mainly in public settings. The time from symptom onset to diagnosis was twice as long for transmitters as for nontransmitters (16.8 vs. 8.5 weeks, respectively; p<0.01). Molecular epidemiologic studies showed that eliminating diagnostic delays can prevent TB transmission in nontraditional settings, which elude contact investigations

    Genetic Variants of Ehrlichia phagocytophila1, Rhode Island and Connecticut

    Primers were used to amplify a 561-bp region of the 16S rRNA gene of Ehrlichia phagocytophila from Ixodes scapularis ticks and small mammals collected in Rhode Island and Connecticut. DNA sequences for all 50 E. phagocytophila-positive samples collected from 1996 through 1998 in southwestern Connecticut were identical to the sequence reported for E. phagocytophila DNA from confirmed human cases. In contrast, the sequences from 92 of 123 E. phagocytophila-positive Rhode Island samples collected from 1996 through 1999 included several variants differing by 1-2 nucleotides from that in the agent infecting humans. While 11.9% of 67 E. phagocytophila-positive ticks collected during 1997 in Rhode Island harbored ehrlichiae with sequences identical to that of the human agent, 79.1% had a variant sequence not previously described. The low incidence of human ehrlichiosis in Rhode Island may in part result from interference by these variant ehrlichiae with maintenance and transmission of the true agent of human disease

    Polygenic resilience scores capture protective genetic effects for Alzheimer’s disease

    Polygenic risk scores (PRSs) can boost risk prediction in late-onset Alzheimer’s disease (LOAD) beyond apolipoprotein E (APOE) but have not been leveraged to identify genetic resilience factors. Here, we sought to identify resilience-conferring common genetic variants in (1) unaffected individuals having high PRSs for LOAD, and (2) unaffected APOE-ε4 carriers also having high PRSs for LOAD. We used genome-wide association study (GWAS) to contrast “resilient” unaffected individuals at the highest genetic risk for LOAD with LOAD cases at comparable risk. From GWAS results, we constructed polygenic resilience scores to aggregate the addictive contributions of risk-orthogonal common variants that promote resilience to LOAD. Replication of resilience scores was undertaken in eight independent studies. We successfully replicated two polygenic resilience scores that reduce genetic risk penetrance for LOAD. We also showed that polygenic resilience scores positively correlate with polygenic risk scores in unaffected individuals, perhaps aiding in staving off disease. Our findings align with the hypothesis that a combination of risk-independent common variants mediates resilience to LOAD by moderating genetic disease risk

    Distinctive Gut Microbiota of Honey Bees Assessed Using Deep Sampling from Individual Worker Bees

    Surveys of 16S rDNA sequences from the honey bee, Apis mellifera, have revealed the presence of eight distinctive bacterial phylotypes in intestinal tracts of adult worker bees. Because previous studies have been limited to relatively few sequences from samples pooled from multiple hosts, the extent of variation in this microbiota among individuals within and between colonies and locations has been unclear. We surveyed the gut microbiota of 40 individual workers from two sites, Arizona and Maryland USA, sampling four colonies per site. Universal primers were used to amplify regions of 16S ribosomal RNA genes, and amplicons were sequenced using 454 pyrotag methods, enabling analysis of about 330,000 bacterial reads. Over 99% of these sequences belonged to clusters for which the first blastn hits in GenBank were members of the known bee phylotypes. Four phylotypes, one within Gammaproteobacteria (corresponding to “Candidatus Gilliamella apicola”) one within Betaproteobacteria (“Candidatus Snodgrassella alvi”), and two within Lactobacillus, were present in every bee, though their frequencies varied. The same typical bacterial phylotypes were present in all colonies and at both sites. Community profiles differed significantly among colonies and between sites, mostly due to the presence in some Arizona colonies of two species of Enterobacteriaceae not retrieved previously from bees. Analysis of Sanger sequences of rRNA of the Snodgrassella and Gilliamella phylotypes revealed that single bees contain numerous distinct strains of each phylotype. Strains showed some differentiation between localities, especially for the Snodgrassella phylotype

    Immunity against Ixodes scapularis Salivary Proteins Expressed within 24 Hours of Attachment Thwarts Tick Feeding and Impairs Borrelia Transmission

    In North America, the black-legged tick, Ixodes scapularis, an obligate haematophagus arthropod, is a vector of several human pathogens including Borrelia burgdorferi, the Lyme disease agent. In this report, we show that the tick salivary gland transcriptome and proteome is dynamic and changes during the process of engorgement. We demonstrate, using a guinea pig model of I. scapularis feeding and B. burgdorferi transmission, that immunity directed against salivary proteins expressed in the first 24 h of tick attachment — and not later — is sufficient to evoke all the hallmarks of acquired tick-immunity, to thwart tick feeding and also to impair Borrelia transmission. Defining this subset of proteins will promote a mechanistic understanding of novel I. scapularis proteins critical for the initiation of tick feeding and for Borrelia transmission
