'He said he was going to kill me'
This article uses a case study design to explore attempted intimate femicide in metropolitan SĂŁo Paulo, Brazil. We conducted 30 in-depth interviews with adult women on community and personal relationship experiences, health-care services, and national legislation about violence against women. Through a thorough transcript review, we identified two participants, Maria* and Raquel*, whose intimate partners attempted to kill them. We used a modified grounded theory approach to code the entire sample, and further analysed these transcripts to identify missed opportunities for intervention in both the prevention of- and responses to intimate partner violence (IPV), and the antecedents of attempted intimate femicide.
Both women had normalized experiences of violence, and experienced psychological abuse prior to the attempted intimate femicide. Social and familial networks were sources of shame for Raquel and support for Maria. Neither woman expressed confidence in law enforcementâs ability to protect women and girls from IPV.
These cases illustrate the need for multi-tiered interventions to prevent femicide in Brazil, which boasts one of the highest global female homicide rates. Although anti-femicide laws exist, better mechanisms are needed to integrate health, legal and social services for IPV and femicide prevention. In addition, community- and interpersonal- level interventions that counteract the Brazilian âculture of violenceâ and machismo may provide support for at-risk women and girls.

Antigen-specific clonal expansion and cytolytic effector function of CD8+ T lymphocytes depend on the transcription factor Bcl11b
CD8+ T lymphocytes mediate the immune response to viruses, intracellular bacteria, protozoan parasites, and tumors. We provide evidence that the transcription factor Bcl11b/Ctip2 controls hallmark features of CD8+ T cell immunity, specifically antigen (Ag)-dependent clonal expansion and cytolytic activity. The reduced clonal expansion in the absence of Bcl11b was caused by altered proliferation during the expansion phase, with survival remaining unaffected. Two genes with critical roles in TCR signaling were deregulated in Bcl11b-deficient CD8+ T cells, CD8 coreceptor and PlcÎł1, both of which may contribute to the impaired responsiveness. Bcl11b was found to bind the E8I, E8IV, and E8V, but not E8II or E8III, enhancers. Thus, Bcl11b is one of the transcription factors implicated in the maintenance of optimal CD8 coreceptor expression in peripheral CD8+ T cells through association with specific enhancers. Short-lived Klrg1hiCD127lo effector CD8+ T cells were formed during the course of infection in the absence of Bcl11b, albeit in smaller numbers, and their Ag-specific cytolytic activity on a per-cell basis was altered, which was associated with reduced granzyme B and perforin
The 3D Topography of Mitotic Chromosomes
A long-standing conundrum is how mitotic chromosomes can compact, as required for clean separation to daughter cells, while maintaining close parallel alignment of sister chromatids. Pursuit of this question, by high resolution 3D fluorescence imaging of living and fixed mammalian cells, has led to three discoveries. First, we show that the structural axes of separated sister chromatids are linked by evenly spaced âmini-axisâ bridges. Second, when chromosomes first emerge as discrete units, at prophase, they are organized as co-oriented sister linear loop arrays emanating from a conjoined axis. We show that this same basic organization persists throughout mitosis, without helical coiling. Third, from prophase onward, chromosomes are deformed into sequential arrays of half-helical segments of alternating handedness (perversions), accompanied by correlated kinks. These arrays fluctuate dynamically over \textless15Â s timescales. Together these discoveries redefine the foundation for thinking about the evolution of mitotic chromosomes as they prepare for anaphase segregation

Hostile, aggressive family conflict trajectories during the transition to adulthood: Associations with adolescent Big Five and emerging adulthood adjustment problems
The goal of this longitudinal study was to investigate the relation of adolescents' Big Five factor (BFF) personality to the development of different patterns of mother-adolescent hostile, aggressive conflict (MHAC) from late adolescence to young adulthood. Furthermore, we examined the prediction of Antisocial Problems (AP) and Depressive Problems (DP) in emerging adulthood from BFF and MHAC trajectories. 385 adolescents participated in this study (age 15-16 at Time 1 and 21-22 at Time 4). Using latent growth curve analysis, Low stable (69.1%), Medium Increasing, (23.3%), and High decreasing (7.6%) trajectories were distinguished. Low adolescents' emotional stability was directly related to AP and indirectly related to DP, throughout the mediation of both Medium Increasing and High Decreasing trajectories. Agreeableness was directly negatively related to DP and indirectly throughout the mediation of High Decreasing trajectory. Low Conscientiousness was indirectly related to DP, throughout the mediation of Medium Increasing trajectory. 385 adolescents participated in this study (age 15-16 at Time 1 and 21-22 at Time 4). Arl2-GTP and Arl3-GTP regulate a GDI-like transport system for farnesylated cargo
Lipidated Rho and Rab GTP-binding proteins are transported between membranes in complex with solubilizing factors called 'guanine nucleotide dissociation inhibitors' (GDIs). Unloading from GDIs using GDI displacement factors (GDFs) has been proposed but remains mechanistically elusive. PDEÎŽ is a putative solubilizing factor for several prenylated Ras-subfamily proteins. Here we report the structure of fully modified farnesylated Rheb-GDP in complex with PDEÎŽ. The structure explains the nucleotide-independent binding of Rheb to PDEÎŽ and the relaxed specificity of PDEÎŽ. We demonstrate that the G proteins Arl2 and Arl3 act in a GTP-dependent manner as allosteric release factors for farnesylated cargo. We thus describe a new transport system for farnesylated G proteins involving a GDI-like molecule and an unequivocal GDF. Considering the importance of PDEÎŽ for proper Ras and Rheb signaling, this study is instrumental in developing a new target for anticancer therapy
