ExCyto PCR Amplification
ExCyto PCR cells provide a novel and cost effective means to amplify DNA transformed into competent bacterial cells. ExCyto PCR uses host E. coli with a chromosomally integrated gene encoding a thermostable DNA polymerase to accomplish robust, hot-start PCR amplification of cloned sequences without addition of exogenous enzyme.Because the thermostable DNA polymerase is stably integrated into the bacterial chromosome, ExCyto cells can be transformed with a single plasmid or complex library, and then the expressed thermostable DNA polymerase can be used for PCR amplification. We demonstrate that ExCyto cells can be used to amplify DNA from different templates, plasmids with different copy numbers, and master mixes left on ice for up to two hours. Further, PCR amplification with ExCyto cells is comparable to amplification using commercial DNA polymerases. The ability to transform a bacterial strain and use the endogenously expressed protein for PCR has not previously been demonstrated.ExCyto PCR reduces pipetting and greatly increases throughput for screening EST, genomic, BAC, cDNA, or SNP libraries. This technique is also more economical than traditional PCR and thus broadly useful to scientists who utilize analysis of cloned DNAs in their research
To what extent is behaviour a problem in English schools?:Exploring the scale and prevalence of deficits in classroom climate
The working atmosphere in the classroom is an important variable in the process of education in schools, with several studies suggesting that classroom climate is an important influence on pupil attainment. There are wide differences in the extent to which classroom climate is considered to be a problem in English schools. Some ‘official’ reports suggest that behaviour in schools is ‘satisfactory or better’ in the vast majority of schools; other sources have pointed to behaviour being a serious and widespread problem. The paper details four studies conducted over the past decade which aimed to explore these disparities. The aim of the research was to gain a more accurate insight into the extent to which deficits in classroom climate limit educational attainment and equality of educational opportunity in English schools. The findings question the suggestion that behaviour is satisfactory or better in 99.7% of English schools and the concluding section suggests ways in which deficits in classroom climate might be addressed. Although the study is limited to classrooms in England, OECD studies suggest that deficits in the working atmosphere in classrooms occur in many countries. The study therefore has potential relevance for education systems in other countries
The Representative XMM-Newton Cluster Structure Survey (REXCESS) of an X-ray Luminosity Selected Galaxy Cluster Sample
The largest uncertainty for cosmological studies using clusters of galaxies
is introduced by our limited knowledge of the statistics of galaxy cluster
structure, and of the scaling relations between observables and cluster mass.
To improve on this situation we have started an XMM-Newton Large Programme for
the in-depth study of a representative sample of 33 galaxy clusters, selected
in the redshift range z=0.055 to 0.183 from the REFLEX Cluster Survey, having
X-ray luminosities above 0.4 X 10^44 h_70^-2 erg s^-1 in the 0.1 - 2.4 keV
band. This paper introduces the sample, compiles properties of the clusters,
and provides detailed information on the sample selection function. We describe
the selection of a nearby galaxy cluster sample that makes optimal use of the
XMM-Newton field-of-view, and provides nearly homogeneous X-ray luminosity
coverage for the full range from poor clusters to the most massive objects in
the Universe. For the clusters in the sample, X-ray fluxes are derived and
compared to the previously obtained fluxes from the ROSAT All-Sky Survey. We
find that the fluxes and the flux errors have been reliably determined in the
ROSAT All-Sky Survey analysis used for the REFLEX Survey. We use the sample
selection function documented in detail in this paper to determine the X-ray
luminosity function, and compare it with the luminosity function of the entire
REFLEX sample. We also discuss morphological peculiarities of some of the
sample members. The sample and some of the background data given in this
introductory paper will be important for the application of these data in the

The Hubble Space Telescope Cluster Supernova Survey: V. Improving the Dark Energy Constraints Above z>1 and Building an Early-Type-Hosted Supernova Sample
We present ACS, NICMOS, and Keck AO-assisted photometry of 20 Type Ia
supernovae SNe Ia from the HST Cluster Supernova Survey. The SNe Ia were
discovered over the redshift interval 0.623 < z < 1.415. Fourteen of these SNe
Ia pass our strict selection cuts and are used in combination with the world's
sample of SNe Ia to derive the best current constraints on dark energy. Ten of
our new SNe Ia are beyond redshift , thereby nearly doubling the
statistical weight of HST-discovered SNe Ia beyond this redshift. Our detailed
analysis corrects for the recently identified correlation between SN Ia
luminosity and host galaxy mass and corrects the NICMOS zeropoint at the count
rates appropriate for very distant SNe Ia. Adding these supernovae improves the
best combined constraint on the dark energy density \rho_{DE}(z) at redshifts
1.0 < z < 1.6 by 18% (including systematic errors). For a LambdaCDM universe,
we find \Omega_\Lambda = 0.724 +0.015/-0.016 (68% CL including systematic
errors). For a flat wCDM model, we measure a constant dark energy
equation-of-state parameter w = -0.985 +0.071/-0.077 (68% CL). Curvature is
constrained to ~0.7% in the owCDM model and to ~2% in a model in which dark
energy is allowed to vary with parameters w_0 and w_a. Tightening further the
constraints on the time evolution of dark energy will require several
improvements, including high-quality multi-passband photometry of a sample of
several dozen z>1 SNe Ia. We describe how such a sample could be efficiently

SPT-CL J0546-5345: A Massive z > 1 Galaxy Cluster Selected Via the Sunyaev-Zel'dovich Effect with the South Pole Telescope
We report the spectroscopic confirmation of SPT-CL J0546-5345 at = 1.067.
To date this is the most distant cluster to be spectroscopically confirmed from
the 2008 South Pole Telescope (SPT) catalog, and indeed the first z > 1 cluster
discovered by the Sunyaev-Zel'dovich Effect (SZE). We identify 21 secure
spectroscopic members within 0.9 Mpc of the SPT cluster position, 18 of which
are quiescent, early-type galaxies. From these quiescent galaxies we obtain a
velocity dispersion of 1179^{+232}_{-167} km/s, ranking SPT-CL J0546-5345 as
the most dynamically massive cluster yet discovered at z > 1. Assuming that
SPT-CL J0546-5345 is virialized, this implies a dynamical mass of M_200 =
1.0^{+0.6}_{-0.4} x 10^{15} Msun, in agreement with the X-ray and SZE mass
measurements. Combining masses from several independent measures leads to a
best-estimate mass of M_200 = (7.95 +/- 0.92) x 10^{14} Msun. The spectroscopic
confirmation of SPT-CL J0546-5345, discovered in the wide-angle, mass-selected
SPT cluster survey, marks the onset of the high redshift SZE-selected galaxy

SPT-CL J0205-5829: A z = 1.32 Evolved Massive Galaxy Cluster in the South Pole Telescope Sunyaev-Zel'dovich Effect Survey
The galaxy cluster SPT-CL J0205-5829 currently has the highest
spectroscopically-confirmed redshift, z=1.322, in the South Pole Telescope
Sunyaev-Zel'dovich (SPT-SZ) survey. XMM-Newton observations measure a
core-excluded temperature of Tx=8.7keV producing a mass estimate that is
consistent with the Sunyaev-Zel'dovich derived mass. The combined SZ and X-ray
mass estimate of M500=(4.9+/-0.8)e14 h_{70}^{-1} Msun makes it the most massive
known SZ-selected galaxy cluster at z>1.2 and the second most massive at z>1.
Using optical and infrared observations, we find that the brightest galaxies in
SPT-CL J0205-5829 are already well evolved by the time the universe was <5 Gyr
old, with stellar population ages >3 Gyr, and low rates of star formation
(<0.5Msun/yr). We find that, despite the high redshift and mass, the existence
of SPT-CL J0205-5829 is not surprising given a flat LambdaCDM cosmology with
Gaussian initial perturbations. The a priori chance of finding a cluster of
similar rarity (or rarer) in a survey the size of the 2500 deg^2 SPT-SZ survey

Predictors of reading literacy for first and second language learners
In this study an attempt was made to construct a multi-factor model predicting the development of reading literacy in the upper grades of primary school in the Netherlands for subgroups of 729 first language (L1) learners and 93 second language (L2) learners. Following a longitudinal design, it was explored to what extent the variation in reading literacy development in L1 and L2 from grade 4 to grade 6 can be explained from children’s word decoding, language, mathematics and nonverbal reasoning skills, reading motivation and self confidence as well as their home reading resources. The results showed that L1 and L2 learners differed in reading literacy skills, language, mathematics, and reasoning skills. Structural equation modelling showed that the reading literacy development in both L1 and L2 learners could be explained from decoding, language, mathematics and reasoning skills, as well as their motivation and self-confidence. A striking difference was the fact that home reading resources had an impact on reading literacy in L1 learners but not in L2 learners
Catalog of Radio Galaxies with z>0.3. I:Construction of the Sample
The procedure of the construction of a sample of distant () radio
galaxies using NED, SDSS, and CATS databases for further application in
statistical tests is described. The sample is assumed to be cleaned from
objects with quasar properties. Primary statistical analysis of the list is
performed and the regression dependence of the spectral index on redshift is

Diagnostic Accuracy of the Leishmania OligoC-TesT and NASBA-Oligochromatography for Diagnosis of Leishmaniasis in Sudan
The leishmaniases are a group of vector-borne diseases caused by protozoan parasites of the genus Leishmania. The parasites are transmitted by phlebotomine sand flies and can cause, depending on the infecting species, three clinical manifestations of leishmaniasis: visceral leishmaniasis (VL), post kala-azar dermal leishmaniasis (PKDL) and cutaneous leishmaniasis (CL) including the mucocutaneous form. VL, PKDL as well as CL are endemic in several parts of Sudan, and VL especially represents a major health problem in this country. Molecular tests such as the polymerase chain reaction (PCR) or nucleic acid sequence based assay (NASBA) are powerful techniques for accurate detection of the parasite in clinical specimens, but broad use is hampered by their complexity and lack of standardisation. Recently, the Leishmania OligoC-TesT and NASBA-Oligochromatography were developed as simplified and standardised PCR and NASBA formats.
