    A Holocene tephra layer within coastal aeolian deposits north of Caleta Olivia (Santa Cruz Province, Argentina)

    In this paper we illustrate the stratigraphy, geochronology, and geochemistry (major, minor, trace elements and Sr-isotopes) of a Holocene tephra layer found within coastal sedimentary deposits north of Caleta Olivia (Santa Cruz Province, Argentina). The stratigraphic succession comprises beach deposits with basal erosive surface resting on the local substrate (“FormaciĂłn Patagonia”) followed by a poorly developed paleosoil. The paleosoil is covered by a lenticular fine grained (Mdφ: 5.2, 0.027 mm), well sorted (σφ: 1.2) volcanic ash layer and aeolian sands. The geochemical composition of shard fragments points to an origin from the Hudson volcano, located in the southern Andes, ca. 400 km to the west. The geochemistry, Sr-isotopes and the radiometric constraints (younger than the age of the underlying marine layer dated at ca. 4,100 a cal BP) further allow correlating this tephra with the so-called H2 eruption (ca. 3,900 a cal BP). This finding is of interest owing to the poor preservation potential of tephra within the Late Holocene sedimentary deposits of the Atlantic coast of Patagonia and represents the first finding of H2 eruption in this area, improving our knowledge of the dispersion of the fine-grained distal deposit of the Hudson volcanic explosive activity, thus allowing a better estimate of the eruptive dynamics and the risks associated with the Hudson volcano

    5-Aza Exposure Improves Reprogramming Process Through Embryoid Body Formation in Human Gingival Stem Cells

    Embryoid bodies (EBs) are three-dimensional aggregates formed by pluripotent stem cells, including embryonic stem cells and induced pluripotent stem cells. They are used as an in vitro model to evaluate early extraembryonic tissue formation and differentiation process. In the adult organisms, cell differentiation is controlled and realized through the epigenetic regulation of gene expression, which consists of various mechanisms including DNA methylation. One demethylating agent is represented by 5-Azacytidine (5-Aza), considered able to induce epigenetic changes through gene derepression. Human gingival mesenchymal stem cells (hGMSCs), an easily accessible stem cells population, migrated from neural crest. They are particularly apt as an in vitro study model in regenerative medicine and in systemic diseases. The ability of 5-Aza treatment to induce hGMSCs toward a dedifferentiation stage and in particular versus EBs formation was investigated. For this purpose hGMSCs were treated for 48 h with 5-Aza (5 ÎŒM). After treatment, hGMSCs are organized as round 3D structures (EBs-hGMSCs). At light and transmission electron microscopy, the cells at the periphery of EBs-hGMSCs appear elongated, while ribbon-shaped cells and smaller cells with irregular shape surrounded by extracellular matrix were present in the center. By RT-PCR, EBs-hGMSCs expressed specific transcription markers related to the three germ layers as MAP-2, PAX-6 (ectoderm), MSX-1, Flk-1 (mesoderm), GATA-4, and GATA-6 (endoderm). Moreover, in EB-hGMSCs the overexpression of DNMT1 and ACH3 other than the down regulation of p21 was detectable. Immunofluorescence staining also showed a positivity for specific etodermal and mesodermal markers. In conclusion, 5-Aza was able to induce the direct conversion of adult hGMSCs into cells of three embryonic lineages: endoderm, ectoderm, and mesoderm, suggesting their possible application in autologous cell therapy for clinical organ repair

    Astrophysics with the Laser Interferometer Space Antenna

    Laser Interferometer Space Antenna (LISA) will be a transformative experiment for gravitational wave astronomy as it will offer unique opportunities to address many key astrophysical questions in a completely novel way. The synergy with ground-based and other space-based instruments in the electromagnetic domain, by enabling multi-messenger observations, will add further to the discovery potential of LISA. The next decade is crucial to prepare the astrophysical community for LISA's first observations. This review outlines the extensive landscape of astrophysical theory, numerical simulations, and astronomical observations that are instrumental for modeling and interpreting the upcoming LISA datastream. To this aim, the current knowledge in three main source classes for LISA is reviewed: ultra-compact stellar-mass binaries, massive black hole binaries, and extreme or intermediate mass ratio inspirals. The relevant astrophysical processes and the established modeling techniques are summarized. Likewise, open issues and gaps in our understanding of these sources are highlighted, along with an indication of how LISA could help make progress in the different areas. New research avenues that LISA itself, or its joint exploitation with studies in the electromagnetic domain, will enable, are also illustrated. Improvements in modeling and analysis approaches, such as the combination of numerical simulations and modern data science techniques, are discussed. This review is intended to be a starting point for using LISA as a new discovery tool for understanding our Universe

    Effects of Probiotic Strains on Disease Activity and Enteric Permeability in Psoriatic Arthritis–A Pilot Open-Label Study

    (1) Background: Psoriatic Arthritis (PsA) is a painful disease of the joints and spine. Recent reports observed distinct enteric dysbiosis in PsA; intake of probiotic strains is considered to ameliorate enteric dysbiosis. If probiotics are effective in PsA is elusive. (2) Methods: In this pilot open-label study we enrolled 10 PsA patients with low to medium disease activity who received probiotics for 12 weeks. Analysis of faecal zonulin, α1-antitrypsin and calprotectin, as well as peripheral immune phenotyping was performed at baseline, after 12 weeks and 12 weeks after termination of probiotic intake. (3) Results: All patients showed increased levels of the enteric permeability marker zonulin which correlated with the frequency of peripheral Th17 cells. Calprotectin, a marker for intestinal inflammation was elevated in 6 out of 10 patients. Probiotic intake resulted in a reduction of disease activity and gut permeability. These effects, however, were not sustained beyond termination of probiotic intake. (4) Conclusions: PsA patients suffer from enhanced enteric permeability and inflammation. Probiotics may ameliorate disease activity in PsA by targeting these alterations

    Probabilistic life cycle costing of existing buildings retrofit interventions towards nZE target: Methodology and application example

    One of the major challenge facing the achievement of nZE standards in existing buildings is the economic issue: the evidence of monetary gains of energy savings facing high investment costs seems still rather limited to the investors’ eyes. In this context, LCC methods have gained much importance in recent years. However, they present a limitation due to the notable simplifications and hypothesis usually made for input parameters that may affect the results. In order to overcome this limit, this work suggests a probabilistic LCC based on uncertainty and sensitivity analysis via Monte Carlo methods and illustrates it through a building case study under several retrofit scenarios sighting the target nZE. The methodology allows investigating the economic effectiveness of alternative measures, giving insight into possible ranges of the economic indicator related to a specific design option. The analysis is focused on a micro-economic dimension and based on the availability and reliability of inputs data and on their proper characterization with Probability Density Functions. Variance-based methods for sensitivity analysis are employed to establish the most influential parameters on output uncertainty. The paper demonstrates the potentials of a probabilistic LCC in providing a more realistic decision support about investments for energy efficient projects

    Smile In<sup>TM</sup> Totems in Radiotherapy: Patients’ Satisfaction with Limited Equipment and COVID-19

    Background: We report a mono-institutional experience regarding patient-perceived quality regarding the Chieti Radiotherapy Department, through RAMSI (Radiotherapy Amica Mia—SmileINTM(SI)—My Friend RadiotherapySI) project, in critical scenarios of limited equipment and COVID-19. Material and methods: Patient-reported experience measures (PREMs) were assessed as follows: Patient-centric welcome perception (PCWP), Comfort, Professional skills and Punctuality. Patients could give anonymous feedback using HappyOrNot technology through four totems located in strategic areas within the center. An internal benchmark was obtained using the feedback received after a preliminary observation period. The SI Experience Index was collected, analyzed and compared. Weekly and monthly reports were generated. Results: From February 2019 to February 2022, 8924 patients accessed the department; 17,464 daily treatments were recorded and 5830 points of feedback were collected: 896, 1267, 1125 and 2542 for PCWP, Comfort, Professional skills and Punctuality, respectively. A LINAC decommissioning period was analyzed, with decreases in the SI-Index score and Smile-IN approved percentage and an improvement after this period. Additionally, the COVID-19 pandemic was analyzed with a mild evaluations decrease for PREM’s Welcome, Comfort and Punctuality (Δ-value: −9%, −3% and −4%, respectively), while Professional skills were always optimal. Conclusion: The RAMSI project was effective for assessing treatment quality perception, allowing for improving clinical procedures with corrective actions. The RAMSI project is ongoing
