    Microbial use of low molecular weight DOM in filtered and unfiltered freshwater:Role of ultra-small microorganisms and implications for water quality monitoring

    Dissolved organic matter (DOM) plays a central role in regulating productivity and nutrient cycling in freshwaters. It is therefore vital that we can representatively sample and preserve DOM in freshwaters for subsequent analysis. Here we investigated the effect of filtration, temperature (5 and 25 °C) and acidification (HCl) on the persistence of low molecular weight (MW) dissolved organic carbon (DOC), nitrogen (DON) and orthophosphate in oligotrophic and eutrophic freshwater environments. Our results showed the rapid loss of isotopically-labelled glucose and amino acids from both filtered (0.22 and 0.45 Όm) and unfiltered waters. We ascribe this substrate depletion in filtered samples to the activity of ultra-small (< 0.45 Όm) microorganisms (bacteria and archaea) present in the water. As expected, the rate of C, N and P loss was much greater at higher temperatures and was repressed by the addition of HCl. Based on our results and an evaluation of the protocols used in recently published studies, we conclude that current techniques used to sample water for low MW DOM characterisation are frequently inadequate and lack proper validation. In contrast to the high degree of analytical precision and rigorous statistical analysis of most studies, we argue that insufficient consideration is still given to the presence of ultra-small microorganisms and potential changes that can occur in the low MW fraction of DOM prior to analysis

    Conflation between self-report and neurocognitive assessments of cognitive flexibility: a critical review of the Jingle Fallacy

    Cognitive flexibility is a widely studied construct and is considered an important treatment target for several psychological disorders. The convergence of several independent fields of research has led to assumptions about the assessment of cognitive flexibility – assumptions that are not empirically supported and often conflate different notions of flexibility. This critical review discusses how the conflation of self-report and neurocognitive assessments has seemingly arisen from literature on eating disorders. We describe how seminal early observations of “inflexible” personality characteristics, communication competence research, and investigations of frontal lobe function after injury led to two methods of assessing “cognitive flexibility”. We discuss the impact that conflation of self-report and neurocognitive assessments has had on the field, and we provide recommendations for assessing cognitive flexibility in both research and clinical settings.Caitlin A.     Blocking endothelial apoptosis revascularises the retina in a model of ischemic retinopathy

    Aberrant, neovascular retinal blood vessel growth is a vision-threatening complication in ischemic retinal diseases. It is driven by retinal hypoxia frequently caused by capillary nonperfusion and endothelial cell (EC) loss. We investigated the role of EC apoptosis in this process using a mouse model of ischemic retinopathy, in which vessel closure and EC apoptosis cause capillary regression and retinal ischemia followed by neovascularization. Protecting ECs from apoptosis in this model did not prevent capillary closure or retinal ischemia. Nonetheless, it prevented the clearance of ECs from closed capillaries, delaying vessel regression and allowing ECs to persist in clusters throughout the ischemic zone. In response to hypoxia, these preserved ECs underwent a vessel sprouting response and rapidly reassembled into a functional vascular network. This alleviated retinal hypoxia, preventing subsequent pathogenic neovascularization. Vessel reassembly was not limited by VEGFA neutralization, suggesting it was not dependent on the excess VEGFA produced by the ischemic retina. Neutralization of ANG2 did not prevent vessel reassembly, but did impair subsequent angiogenic expansion of the reassembled vessels. Blockade of EC apoptosis may promote ischemic tissue revascularization by preserving ECs within ischemic tissue that retain the capacity to reassemble a functional network and rapidly restore blood supply

    This study examines the dynamics of the development of Japan's notorious ‘lost decade’ from 1990--2003. This economic downturn marked the end of four decades of strong economic growth and is still affecting the Japanese economy today. While previous studies have focused on government policies to explain the nation's slow response to this crisis, the attitudes of the Japanese business community merit more attention. For example, the leaders of Keidanren, the powerful representative of big business in Japan, defined a set of economic challenges facing Japan in the late 1980s and neoliberal solutions that blinded them to the significance of the economic bubble that developed at that time and its subsequent collapse.     Suspended timber ground floors: measured heat loss compared with models

    There are approximately 6.6 million dwellings in the UK built before 1919, predominantly constructed with suspended timber ground floors whose thermal performance has not been extensively investigated. The results are presented from an in-situ heat-flow measuring campaign conducted at 27 locations on a suspended timber ground floor, and the estimated whole-floor U-value compared with modelled results. Findings highlight a significant variability in heat flow, with increased heat loss near the external perimeter. In-situ measured-point U-values ranged from 0.54 ± 0.09 Wm−2 K−1, when away from the external wall perimeter, to nearly four times as high (2.04 ± 0.21 Wm−2 K−1) when near the perimeter. The results highlight the fact that observing only a few measurements is likely to bias any attempts to derive a whole-floor U-value, which was estimated to be 1.04 ± 0.12 Wm−2 K−1 and nearly twice that derived from current models. This raises questions about the validity of using such models in housing stock models to inform retrofit decision-making and space-heating-reduction interventions. If this disparity between models and measurements exists in the wider stock, a reappraisal of the performance of suspended timber ground floors and heat-loss-reduction potential through this element will be required to support the UK’s carbon-emission-reduction targets

    Tight-binding parameters for charge transfer along DNA

    We systematically examine all the tight-binding parameters pertinent to charge transfer along DNA. The π\pi molecular structure of the four DNA bases (adenine, thymine, cytosine, and guanine) is investigated by using the linear combination of atomic orbitals method with a recently introduced parametrization. The HOMO and LUMO wavefunctions and energies of DNA bases are discussed and then used for calculating the corresponding wavefunctions of the two B-DNA base-pairs (adenine-thymine and guanine-cytosine). The obtained HOMO and LUMO energies of the bases are in good agreement with available experimental values. Our results are then used for estimating the complete set of charge transfer parameters between neighboring bases and also between successive base-pairs, considering all possible combinations between them, for both electrons and holes. The calculated microscopic quantities can be used in mesoscopic theoretical models of electron or hole transfer along the DNA double helix, as they provide the necessary parameters for a tight-binding phenomenological description based on the π\pi molecular overlap. We find that usually the hopping parameters for holes are higher in magnitude compared to the ones for electrons, which probably indicates that hole transport along DNA is more favorable than electron transport.     Anthropogenic Space Weather

    Anthropogenic effects on the space environment started in the late 19th century and reached their peak in the 1960s when high-altitude nuclear explosions were carried out by the USA and the Soviet Union. These explosions created artificial radiation belts near Earth that resulted in major damages to several satellites. Another, unexpected impact of the high-altitude nuclear tests was the electromagnetic pulse (EMP) that can have devastating effects over a large geographic area (as large as the continental United States). Other anthropogenic impacts on the space environment include chemical release ex- periments, high-frequency wave heating of the ionosphere and the interaction of VLF waves with the radiation belts. This paper reviews the fundamental physical process behind these phenomena and discusses the observations of their impacts.Comment: 71 pages, 35 figure

    Heat-flow variability of suspended timber ground floors: Implications for in-situ heat-flux measuring

    Reducing space heating energy demand supports the UK’s legislated carbon emission reduction targets and requires the effective characterisation of the UK’s existing housing stock to facilitate retrofitting decision-making. Approximately 6.6 million UK dwellings pre-date 1919 and are predominantly of suspended timber ground floor construction, the thermal performance of which has not been extensively investigated. This paper examines suspended timber ground floor heat-flow by presenting high resolution in-situ heat-flux measurements undertaken in a case study house at 15 point locations on the floor. The results highlight significant variability in observed heat-flow: point U-values range from 0.56 ± 0.05 to 1.18 ± 0.11 Wm−2 K−1. This highlights that observing only a few measurements is unlikely to be representative of the whole floor heat-flow and the extrapolation from such point values to whole floor U-value estimates could lead to its over- or under- estimation. Floor U-value models appear to underestimate the actual measured floor U-value in this case study. This paper highlights the care with which in-situ heat-flux measuring must be undertaken to enable comparison with models, literature and between studies and the findings support the unique, high-resolution in-situ monitoring methodology used in this study for further research in this area

Location: Two hundred and eighty-three c. 1 ha forest inventory plots from across Amazonia. Methods: We evaluated PD as the total phylogenetic branch length across species in each plot (PDss), the mean pairwise phylogenetic distance between species (MPD), the mean nearest taxon distance (MNTD) and their equivalents standardized for species richness (ses.PDss, ses.MPD, ses.MNTD). We compared PD of tree communities growing (1) on substrates of varying geological age; and (2) in environments with varying ecophysiological barriers to growth and survival. Results: PDss is strongly positively correlated with species richness (SR), whereas MNTD has a negative correlation. Communities on geologically young- and intermediate-aged substrates (western and central Amazonia respectively) have the highest SR, and therefore the highest PDss and the lowest MNTD. We find that the youngest and oldest substrates (the latter on the Brazilian and Guiana Shields) have the highest ses.PDss and ses.MNTD. MPD and ses.MPD are strongly correlated with how evenly taxa are distributed among the three principal angiosperm clades and are both highest in western Amazonia. Meanwhile, seasonally dry tropical forest (SDTF) and forests on white sands have low PD, as evaluated by any metric. Main conclusions: High ses.PDss and ses.MNTD reflect greater lineage diversity in communities. We suggest that high ses.PDss and ses.MNTD in western Amazonia results from its favourable, easy-to-colonize environment, whereas high values in the Brazilian and Guianan Shields may be due to accumulation of lineages over a longer period of time. White-sand forests and SDTF are dominated by close relatives from fewer lineages, perhaps reflecting ecophysiological barriers that are difficult to surmount evolutionarily.
