Margaret Chase Smith Essay: High School Student Essay Winners
Maine has benefited from the public service of many well-respected and influential national leaders over the last two centuries. One of them, Senator Margaret Chase Smith, offered her reflections on leadership at a time when the United States faced a struggle for civil rights at home and the tensions of the Cold War abroad. With the country currently confronting challenges such as the threat of terrorism, ongoing tensions in the Middle East, and the taint of corporate scandals, the Margaret Chase Smith Library annual essay contest invited Maine high school seniors to reflect on the qualities leaders will need to possess in order to be more effective in the twenty-first century. Transmitochondrial embryonic stem cells containing pathogenic mtDNA mutations are compromised in neuronal differentiation
Objectives:â Defects of the mitochondrial genome (mtDNA) cause a series of rare, mainly neurological disorders. In addition, they have been implicated in more common forms of movement disorders, dementia and the ageing process. In order to try to model neuronal dysfunction associated with mitochondrial disease, we have attempted to establish a series of transmitochondrial mouse embryonic stem cells harbouring pathogenic mtDNA mutations.
Materials and methods:âTransmitochondrial embryonic stem cell cybrids were generated by fusion of cytoplasts carrying a variety of mtDNA mutations, into embryonic stem cells that had been pretreated with rhodamine 6G, to prevent transmission of endogenous mtDNA. Cybrids were differentiated into neurons and assessed for efficiency of differentiation and electrophysiological function.
Results:â Neuronal differentiation could occur, as indicated by expression of neuronal markers. Differentiation was impaired in embryonic stem cells carrying mtDNA mutations that caused severe biochemical deficiency. Electrophysiological tests showed evidence of synaptic activity in differentiated neurons carrying non-pathogenic mtDNA mutations or in those that caused a mild defect of respiratory activity. Again, however, neurons carrying mtDNA mutations that resulted in severe biochemical deficiency had marked reduction in post-synaptic events.
Conclusions:â Differentiated neurons carrying severely pathogenic mtDNA defects can provide a useful model for understanding how such mutations can cause neuronal dysfunction
Evolution of surface gravity waves over a submarine canyon
The effects of a submarine canyon on the propagation of ocean surface waves
are examined with a three-dimensional coupled-mode model for wave propagation
over steep topography. Whereas the classical geometrical optics approximation
predicts an abrupt transition from complete transmission at small incidence
angles to no transmission at large angles, the full model predicts a more
gradual transition with partial reflection/transmission that is sensitive to
the canyon geometry and controlled by evanescent modes for small incidence
angles and relatively short waves. Model results for large incidence angles are
compared with data from directional wave buoys deployed around the rim and over
Scripps Canyon, near San Diego, California, during the Nearshore Canyon
Experiment (NCEX). Wave heights are observed to decay across the canyon by
about a factor 5 over a distance shorter than a wavelength. Yet, a spectral
refraction model predicts an even larger reduction by about a factor 10,
because low frequency components cannot cross the canyon in the geometrical
optics approximation. The coupled-mode model yields accurate results over and
behind the canyon. These results show that although most of the wave energy is
refractively trapped on the offshore rim of the canyon, a small fraction of the
wave energy 'tunnels' across the canyon. Simplifications of the model that
reduce it to the standard and modified mild slope equations also yield good
results, indicating that evanescent modes and high order bottom slope effects
are of minor importance for the energy transformation of waves propagating
across depth contours at large oblique angles
Study Design of a Cluster-Randomized Controlled Trial to Evaluate a Large-scale Distribution of Cook Stoves and Water Filters in Western Province, Rwanda
Background: In Rwanda, pneumonia and diarrhea are the first and second leading causes of death, respectively, among children under five. Household air pollution (HAP) resultant from cooking indoors with biomass fuels on traditional stoves is a significant risk factor for pneumonia, while consumption of contaminated drinking water is a primary cause of diarrheal disease. To date, there have been no largescale effectiveness trials of programmatic efforts to provide either improved cookstoves or household water filters at scale in a low-income country. In this paper we describe the design of a clusterrandomized trial to evaluate the impact of a national-level program to distribute and promote the use of improved cookstoves and advanced water filters to the poorest quarter of households in Rwanda.
Methods/Design: We randomly allocated 72 sectors (administratively defined units) in Western Province to the intervention, with the remaining 24 sectors in the province serving as controls. In the intervention sectors, roughly 100,000 households received improved cookstoves and household water filters through a government-sponsored program targeting the poorest quarter of households nationally. The primary outcome measures are the incidence of acute respiratory infection (ARI) and diarrhea among children under five years of age. Over a one-year surveillance period, all cases of acute respiratory infection (ARI) and diarrhea identified by health workers in the study area will be extracted from records maintained at health facilities and by community health workers (CHW). In addition, we are conducting intensive, longitudinal data collection among a random sample of households in the study area for in-depth assessment of coverage, use, environmental exposures, and additional health measures.
Discussion: Although previous research has examined the impact of providing household water treatment and improved cookstoves on child health, there have been no studies of national-level programs to deliver these interventions at scale in a developing country. The results of this study, the first RCT of a large-scale programmatic cookstove or household water filter intervention, will inform global efforts to reduce childhood morbidity and mortality from diarrheal disease and pneumonia
Use of Serologic Responses against Enteropathogens to Assess the Impact of a Point-of-Use Water Filter: A Randomized Controlled Trial in Western Province, Rwanda.
Diarrhea is a leading contributor to childhood morbidity and mortality in sub-Saharan Africa. Given the challenge of blinding most water, sanitation, and hygiene (WASH) interventions, diarrheal disease outcome measures in WASH intervention trials are subject to potential bias and misclassification. Using the platform of a cluster-randomized controlled trial of a household-based drinking water filter in western province, Rwanda, we assessed the impact of the drinking water filter on enteric seroconversion in young children as a health outcome and examined the association between serologic responses and caregiver-reported diarrhea. Among the 2,179 children enrolled in the trial, 189 children 6-12 months of age were enrolled in a nested serology study. These children had their blood drawn at baseline and 6-12 months after the intervention was distributed. Multiplex serologic assays for Giardia, Cryptosporidium, Entamoeba histolytica, norovirus, Campylobacter, enterotoxigenic Escherichia coli and Vibrio cholerae were performed. Despite imperfect uptake, receipt of the water filter was associated with a significant decrease in seroprevalence of IgG directed against Cryptosporidium parvum Cp17 and Cp23 (relative risk [RR]: 0.62, 95% confidence interval [CI]: 0.44-0.89). Serologic responses were positively associated with reported diarrhea in the previous 7 days for both Giardia intestinalis (RR: 1.94, 95% CI: 1.04-3.63) and C. parvum (RR: 2.21, 95% CI: 1.09-4.50). Serologic responses for all antigens generally increased in the follow-up round, rising sharply after 12 months of age. The water filter is associated with reduced serologic responses against C. parvum, a proxy for exposure and infection; therefore, serologic responses against protozoa may be a suitable health outcome measure for WASH trials among children with diarrhea

Use, microbiological effectiveness and health impact of a household water filter intervention in rural Rwanda-A matched cohort study.
Unsafe drinking water is a substantial health risk contributing to child diarrhoea. We investigated impacts of a program that provided a water filter to households in rural Rwandan villages. We assessed drinking water quality and reported diarrhoea 12-24 months after intervention delivery among 269 households in the poorest tertile with a child under 5 from 9 intervention villages and 9 matched control villages. We also documented filter coverage and use. In Round 1 (12-18 months after delivery), 97.4% of intervention households reported receiving the filter, 84.5% were working, and 86.0% of working filters contained water. Sensors confirmed half of households with working filters filled them at least once every other day on average. Coverage and usage was similar in Round 2 (19-24 months after delivery). The odds of detecting faecal indicator bacteria in drinking water were 78% lower in the intervention arm than the control arm (odds ratio (OR) 0.22, 95% credible interval (CrI) 0.10-0.39, p<0.001). The intervention arm also had 50% lower odds of reported diarrhoea among children <5 than the control arm (OR=0.50, 95% CrI 0.23-0.90, p=0.03). The protective effect of the filter is also suggested by reduced odds of reported diarrhoea-related visits to community health workers or clinics, although these did not reach statistical significance
Brain Cells in the Avian âPrefrontal Cortexâ Code for Features of Slot-Machine-Like Gambling
Slot machines are the most common and addictive form of gambling. In the current study, we recorded from single neurons in the âprefrontal cortexâ of pigeons while they played a slot-machine-like task. We identified four categories of neurons that coded for different aspects of our slot-machine-like task. Reward-Proximity neurons showed a linear increase in activity as the opportunity for a reward drew near. I-Won neurons fired only when the fourth stimulus of a winning (four-of-a-kind) combination was displayed. I-Lost neurons changed their firing rate at the presentation of the first nonidentical stimulus, that is, when it was apparent that no reward was forthcoming. Finally, Near-Miss neurons also changed their activity the moment it was recognized that a reward was no longer available, but more importantly, the activity level was related to whether the trial contained one, two, or three identical stimuli prior to the display of the nonidentical stimulus. These findings not only add to recent neurophysiological research employing simulated gambling paradigms, but also add to research addressing the functional correspondence between the avian NCL and primate PFC
Water wave propagation and scattering over topographical bottoms
Here I present a general formulation of water wave propagation and scattering
over topographical bottoms. A simple equation is found and is compared with
existing theories. As an application, the theory is extended to the case of
water waves in a column with many cylindrical steps
Practical use of variational principles for modeling water waves
This paper describes a method for deriving approximate equations for
irrotational water waves. The method is based on a 'relaxed' variational
principle, i.e., on a Lagrangian involving as many variables as possible. This
formulation is particularly suitable for the construction of approximate water
wave models, since it allows more freedom while preserving the variational
structure. The advantages of this relaxed formulation are illustrated with
various examples in shallow and deep waters, as well as arbitrary depths. Using
subordinate constraints (e.g., irrotationality or free surface impermeability)
in various combinations, several model equations are derived, some being
well-known, other being new. The models obtained are studied analytically and
