Estruturado em seis etapas, o trabalho inicia a partir de um resgate histórico, através das fontes bibliográficas e webgráficas, das bibliotecas no Brasil e no mundo, entendendo a sua evolução até o momento atual, analisando tipologias e conformações espaciais. Nessa primeira etapa analisa-se o papel assumido pelas bibliotecas públicas da atualidade, de modo a entender as suas carências e potencialidades, bem como sua função real na sociedade. Em um segundo momento são estudadas obras correlatas para servirem de referência na concepção do projeto. Antes de dar sequencia rumo à análise do terreno, o trabalho ainda visa, nos dois capítulos seguintes, analisar questões técnicas e arquitetônicas para a elaboração do projeto. A pesquisa trata, em seguida, do local onde será inserida a biblioteca, fazendo uma investigação do terreno e seu entorno, tratando de questões sobre o bairro em que será situada e coletando o perfil dos usuários da biblioteca pública através de pesquisa de campo. Finalmente são reunidos os dados e informações relevantes para a definição, no sétimo capítulo, sexta etapa, do programa de necessidades do edifício e elaboração das diretrizes que servirão de base para a concepção do projeto da biblioteca pública.Abstract : This research aims to investigate and collect public library related data, gathering information to serve as a support for the development of the building design, drawing conclusions and reflections clarifying the guidelines as a basis for the design of the final project. Structured in six stages, the work starts from a historical review through bibliographical and webgraphical sources, about libraries in Brazil and the world, understanding its evolution up to the present, analyzing types and spatial conformations. This first step is an analysis of the role played by public libraries today, in order to understand their needs and capabilities as well as its actual function in society. In a second moment related works are studied to serve as a reference in the project design. Before giving sequence toward terrain analysis, further work is to analyze in the next two chapters technical and architectural issues for the project design of the building.The research studies, then, about where the library will be inserted into, making an investigation of the land and its surroundings, addressing issues about the neighborhood in which it is located and collecting the profile of users of the public library through field research. Finally gather data and information relevant to the definition, in the seventh chapter, the sixth stage, of the building'sprogram needs and preparation of guidelines as a basis for the design of the project to the public library

    Abnormalities of the large ribosomal subunit protein, Rpl35a, in Diamond-Blackfan anemia

    Diamond-Blackfan anemia (DBA) is an inherited bone marrow failure syndrome characterized by anemia, congenital abnormalities, and cancer predisposition. Small ribosomal subunit genes RPS19, RPS24, and RPS17 are mutated in approximately one-third of patients. We used a candidate gene strategy combining high-resolution genomic mapping and gene expression microarray in the analysis of 2 DBA patients with chromosome 3q deletions to identify RPL35A as a potential DBA gene. Sequence analysis of a cohort of DBA probands confirmed involvement RPL35A in DBA. shRNA inhibition shows that Rpl35a is essential for maturation of 28S and 5.8S rRNAs, 60S subunit biogenesis, normal proliferation, and cell survival. Analysis of pre-rRNA processing in primary DBA lymphoblastoid cell lines demonstrated similar alterations of large ribosomal subunit rRNA in both RPL35A-mutated and some RPL35A wild-type patients, suggesting additional large ribosomal subunit gene defects are likely present in some cases of DBA. These data demonstrate that alterations of large ribosomal subunit proteins cause DBA and support the hypothesis that DBA is primarily the result of altered ribosomal function. The results also establish that haploinsufficiency of large ribosomal subunit proteins contributes to bone marrow failure and potentially cancer predisposition
