Pulmonary arterial hypertension is a severe life-threatening disease. Up to now there is no medical treatment to permanently stop or even reverse the course of disease. Mainly the small arterial vessels of the lung are affected, while they increase pulmonary resistance and thereby lead to pulmonary hypertension. Intimal fibrosis, thickening of the media, neointima proliferation as well as plexiform lesions are typical pathohistological findings.
The nitric oxide (NO)- soluble guanylate cyclase (sGC)- cyclic guanosine monophosphate (cGMP) signal transduction pathway is impaired in many cardiovascular diseases, including PAH. Riociguat (BAY 63-2521) is a stimulator of sGC that works both in synergy with and independently from NO to increase levels of cGMP. The aim of the study was to investigate the role of NO-sGC-cGMP signaling in a model of severe PAH and to evaluate the effects of sGC stimulation by riociguat and PDE5 inhibition by sildenafil in comparison to a placebo treatment on hemodynamics and vascular remodeling in severe experimental PAH. Severe angioproliferative PAH was induced in rats by combined exposure to the vascular endothelial growth factor receptor antagonist SU5416 and hypoxia at 10% O2. The employed model enabled not only to increase pulmonary pressure, but also to mimic the vascular alteration observed in human patients.
Therapeutic effects have been demonstrated with help of different parameters: invasive hemodynamics, echocardiographic investigations and histological studies. Both compounds show positive influence on hemodynamics, vascular remodeling, right heart hypertrophy, and heart function in comparison with vehicle treatment. Riociguat had greater effects on these parameters than sildenafil.
The outcome of this study contributes to the understanding of the process and the disease itself and paves the way for an optimized medicamentation. Besides sildenafil, which is already approved for PAH therapy (RevatioÂź), riociguat is a promising possibility for a treatment

(DIPF/Orig.
Mitochondrial Haplogroups, Control Region Polymorphisms and Malignant Melanoma: A Study in Middle European Caucasians
Because mitochondria play an essential role in energy metabolism, generation of reactive oxygen species (ROS), and apoptosis, sequence variation in the mitochondrial genome has been postulated to be a contributing factor to the etiology of multifactorial age-related diseases, including cancer. The aim of the present study was to compare the frequencies of mitochondrial DNA (mtDNA) haplogroups as well as control region (CR) polymorphisms of patients with malignant melanoma (nâ=â351) versus those of healthy controls (nâ=â1598) in Middle Europe.Using primer extension analysis and DNA sequencing, we identified all nine major European mitochondrial haplogroups and known CR polymorphisms. The frequencies of the major mitochondrial haplogroups did not differ significantly between patients and control subjects, whereas the frequencies of the one another linked CR polymorphisms A16183C, T16189C, C16192T, C16270T and T195C were significantly higher in patients with melanoma compared to the controls. Regarding clinical characteristics of the patient cohort, none of the nine major European haplogroups was associated with either Breslow thickness or distant metastasis. The CR polymorphisms A302CC-insertion and T310C-insertion were significantly associated with mean Breslow thickness, whereas the CR polymorphism T16519C was associated with metastasis. Our results suggest that mtDNA variations could be involved in melanoma etiology and pathogenesis, although the functional consequence of CR polymorphisms remains to be elucidated
PAK1 modulates a PPARÎł/NF-ÎșB cascade in intestinal inflammation
P21-activated kinases (PAKs) are multifunctional effectors of Rho GTPases with both kinase and scaffolding activity. Here, we investigated the effects of inflammation on PAK1 signaling and its role in colitis-driven carcinogenesis. PAK1 and p-PAK1 (Thr423) were assessed by immunohistochemistry, immunofluorescence, and Western blot. C57BL6/J wildtype mice were treated with a single intraperitoneal TNFα injection. Small intestinal organoids from these mice and from PAK1-KO mice were cultured with TNFα. NF-ÎșB and PPARÎł were analyzed upon PAK1 overexpression and silencing for transcriptional/translational regulation. PAK1 expression and activation was increased on the luminal intestinal epithelial surface in inflammatory bowel disease and colitis-associated cancer. PAK1 was phosphorylated upon treatment with IFNÎł, IL-1ÎČ, and TNFα. In vivo, mice administered with TNFα showed increased p-PAK1 in intestinal villi, which was associated with nuclear p65 and NF-ÎșB activation. p65 nuclear translocation downstream of TNFα was strongly inhibited in PAK1-KO small intestinal organoids. PAK1 overexpression induced a PAK1âp65 interaction as visualized by co-immunoprecipitation, nuclear translocation, and increased NF-ÎșB transactivation, all of which were impeded by kinase-dead PAK1. Moreover, PAK1 overexpression downregulated PPARÎł and mesalamine recovered PPARÎł through PAK1 inhibition. On the other hand PAK1 silencing inhibited NF-ÎșB, which was recovered using BADGE, a PPARÎł antagonist. Altogether these data demonstrate that PAK1 overexpression and activation in inflammation and colitis-associated cancer promote NF-ÎșB activity via suppression of PPARÎł in intestinal epithelial cells
Balancing the benefits and risks of publicâprivate partnerships to address the global double burden of malnutrition
Objective: Transnational food, beverage and restaurant companies, and their corporate foundations, may be potential collaborators to help address complex public health nutrition challenges. While UN system guidelines are available for private-sector engagement, non-governmental organizations (NGO) have limited guidelines to navigate diverse opportunities and challenges presented by partnering with these companies through public–private partnerships (PPP) to address the global double burden of malnutrition.Design: We conducted a search of electronic databases, UN system websites and grey literature to identify resources about partnerships used to address the global double burden of malnutrition. A narrative summary provides a synthesis of the interdisciplinary literature identified. Results: We describe partnership opportunities, benefits and challenges; and tools and approaches to help NGO engage with the private sector to address global public health nutrition challenges. PPP benefits include: raising the visibility of nutrition and health on policy agendas; mobilizing funds and advocating for research; strengthening food-system processes and delivery systems; facilitating technology transfer; and expanding access to medications, vaccines, healthy food and beverage products, and nutrition assistance during humanitarian crises. PPP challenges include: balancing private commercial interests with public health interests; managing conflicts of interest; ensuring that co-branded activities support healthy products and healthy eating environments; complying with ethical codes of conduct; assessing partnership compatibility; and evaluating partnership outcomes. Conclusions: NGO should adopt a systematic and transparent approach using available tools and processes to maximize benefits and minimize risks of partnering with transnational food, beverage and restaurant companies to effectively target the global double burden of malnutrition.<br /

Incorporating translation into sociolinguistic research: translation policy in an international non-governmental organisation
This article explores aspects of translation, multilingualism and language policy in the field of transnational civil society. By focusing on translation policies at Amnesty International, an international non-governmental organisation that performs a key role in global governance, this article seeks to contribute to a globalisation-sensitive sociolinguistics. It argues that combining a sociolinguistic approach, more precisely linguistic ethnography, with translation studies leads to an increased understanding of the language practices under study. Furthermore, the article calls for more interdisciplinary research, stating that there is a space for sociolinguistics and translation studies to contribute to research in international relations and development studies by highlighting the role of multilingualism and challenging the traditionally powerful position of English in transnational civil society
