    Force measurements with optical tweezers inside living cells

    The force exerted by optical tweezers can be measured by tracking the momentum changes of the trapping beam, a method which is more general and powerful than traditional calibration techniques as it is based on first principles, but which has not been brought to its full potential yet, probably due to practical difficulties when combined with high-NA optical traps, such as the necessity to capture a large fraction of the scattered light. We show that it is possible to measure forces on arbitrary biological objects inside cells without an in situ calibration, using this approach. The instrument can be calibrated by measuring three scaling parameters that are exclusively determined by the design of the system, thus obtaining a conversion factor from volts to piconewtons that is theoretically independent of the physical properties of the sample and its environment. We prove that this factor keeps valid inside cells as it shows good agreement with other calibration methods developed in recent years for viscoelastic media. Finally, we apply the method to measuring the stall forces of kinesin and dynein in living A549 cells.Publisher PD

    Coronal X-Ray Emission from the Stellar Companions to Transiently Accreting Black Holes

    Observations of soft X-ray transients (SXTs) in quiescence have found that the binaries harboring black holes are fainter than those that contain a neutron star. Narayan and collaborators postulated that the faint X-ray emission from black hole binaries was powered by an advection dominated accretion flow (ADAF). We explore an alternative explanation for the quiescent X-ray emission from the black hole systems: coronal emission from the rapidly rotating optical companion. This is commonly observed and well studied in other tidally locked binaries, such as the RS CVns. We show that two of the three X-ray detected black hole binaries (A0620-00 and GRO J1655-40) exhibit X-ray fluxes entirely consistent with coronal emission. The X-ray spectra of these objects should be best fit with thermal Raymond-Smith models rich in lines when coronal emission predominates. One black hole system (V404 Cyg) is too X-ray bright to be explained as coronal emission. The quiescent X-ray emission from the neutron star binaries is far too bright for coronal emission. It might be that all SXT's have variable accretion rates in quiescence and that the basal quiescent X-ray flux is set by either coronal emission from the companion or -- when present -- by thermal emission from the neutron star. We also show that the lithium abundances in the black hole systems are comparable to those in the RS CVns, reducing the need for production mechanisms that involve the compact object.Comment: ApJ, accepted (v541; Oct 1, 2000); Changes to figures and tables, minor modifications to text.     Changes in plasma fatty acid composition are associated with improvements in obesity and related metabolic disorders: A therapeutic approach to overweight adolescents

    Background & aims: In recent years, obesity has reached alarming levels among children and adolescents. The study of plasma fatty acid (FA) composition, as a reflection of diet, and its associations with other parameters, that are closely linked to obesity and the cardiometabolic profile, may be useful for setting nutritional goals for obesity treatment and prevention. This study explored the relationship between plasma FA levels and body fat and cardiometabolic risk markers, in overweight adolescents. Methods: A multidisciplinary weight loss program was followed by 127 overweight and obese adolescents aged 12-17 years old. Plasma FA composition, anthropometric indicators of adiposity and biochemical parameters were analyzed at baseline, two months (the end of the intensive intervention phase) and six months (the end of the extensive phase). Results: While saturated fatty acid (SFA) and n-6 polyunsaturated fatty acid (PUFA) levels decreased significantly during the intervention, monounsaturated fatty acid (MUFA) and n-3 PUFA showed the opposite trend. The decrease in SFA C14:0 was associated with a reduction in total and LDL cholesterol, apolipoprotein B and insulin. The increase in MUFAs, especially C18:1n-9, was related to a reduction in weight, fat mass, fat mass index and glucose. Regarding PUFAs, changes in the n-3 series were not associated with any of the parameters studied, whereas the reduction in n-6 PUFA5 was directly related to weight, fat mass, total and HDL cholesterol, apolipoprotein Al, glucose and insulin, and inversely associated with diastolic blood pressure. The adolescents with greater weight loss presented significant changes in MUFAs, n-6 PUFA5 and C14:0. Conclusions: Modifications in plasma FA composition were associated with adiposity reduction and cardiometabolic profile improvement in an anti-obesity program aimed at adolescents. The changes observed in FA composition were related to the success of the treatment, since the individuals most affected by these variations were those who presented the greatest weight loss

    The First Extrasolar Planet Discovered with a New Generation High Throughput Doppler Instrument

    We report the detection of the first extrasolar planet, ET-1 (HD 102195b), using the Exoplanet Tracker (ET), a new generation Doppler instrument. The planet orbits HD 102195, a young star with solar metallicity that may be part of the local association. The planet imparts radial velocity variability to the star with a semiamplitude of 63.4±2.063.4\pm2.0 m s−1^{-1} and a period of 4.11 days.     GJ 900: A new hierarchical system with low-mass components

    Speckle interferometric observations made with the 6 m telescope of the Special Astrophysical Observatory of the Russian Academy of Sciences in 2000 revealed the triple nature of the nearby (πHip=51.80±1.74\pi_{Hip}=51.80\pm1.74 mas) low-mass young (≈200\approx200 Myr) star GJ 900. The configuration of the triple system allowed it to be dynamically unstable. Differential photometry performed from 2000 through 2004 yielded II- and KK-band absolute magnitudes and spectral types for the components to be IAI_{A}=6.66±\pm0.08, IBI_{B}=9.15±\pm0.11, ICI_{C}=10.08±\pm0.26, KAK_{A}=4.84±\pm0.08, KBK_{B}=6.76±\pm0.20, KCK_{C}=7.39±\pm0.31, SpASp_{A}≈\approxK5--K7, SpBSp_{B}≈\approxM3--M4, SpCSp_{C}≈\approxM5--M6. The ``mass--luminosity'' relation is used to estimate the individual masses of the components: MA\mathcal{M}_{A}≈0.64M⊙\approx0.64\mathcal{M}_{\odot}, MB\mathcal{M}_{B}≈0.21M⊙\approx0.21\mathcal{M}_{\odot}, MC\mathcal{M}_{C}≈0.13M⊙\approx0.13\mathcal{M}_{\odot}. From the observations of the components relative motion in the period 2000--2006, we conclude that GJ 900 is a hierarchical triple star with the possible orbital periods PA−BC_{A-BC}≈\approx80 yrs and PBC_{BC}≈\approx20 yrs. An analysis of the 2MASS images of the region around GJ 900 leads us to suggest that the system can include other very-low-mass components.Comment: 7 pages, 5 figure

    Use of Colorimetric Culture Methods for Detection of Mycobacterium Tuberculosis Complex Isolates from Sputum Samples in Resource-Limited Settings

    Despite recent advances, tuberculosis (TB) diagnosis remains imperfect in resource-limited settings due to its complexity and costs, poor sensitivity of available tests, or long times to reporting. We present a report on the use of colorimetric methods, based on the detection of mycobacterial growth using colorimetric indicators, for the detection of Mycobacterium tuberculosis in sputum specimens. We evaluated the nitrate reductase assay (NRA), a modified NRA using para-nitrobenzoic acid (PNB) (NRAp), and the resazurin tube assay using PNB (RETAp) to differentiate tuberculous and nontuberculous mycobacteria. The performances were assessed at days 18 and 28 using mycobacterium growth indicator tube (MGIT) and Löwenstein-Jensen (LJ) medium culture methods as the reference standards. We enrolled 690 adults with suspected pulmonary tuberculosis from a regional referral hospital in Uganda between March 2010 and June 2011. At day 18, the sensitivities and specificities were 84.6% and 90.0% for the NRA, 84.1% and 92.6% for the NRAp, and 71.2% and 99.3% for the RETAp, respectively. At day 28, the sensitivity of the RETAp increased to 82.6%. Among smear-negative patients with suspected TB, sensitivities at day 28 were 64.7% for the NRA, 61.3% for the NRAp, and 50% for the RETAp. Contamination rates were found to be 5.4% for the NRA and 6.7% for the RETAp, compared with 22.1% for LJ medium culture and 20.4% for MGIT culture. The median times to positivity were 10, 7, and 25 days for colorimetric methods, MGIT culture, and LJ medium culture,respectively. Whereas the low specificity of the NRA/NRAp precludes it from being used for TB diagnosis, the RETAp might provide an alternative to LJ medium culture to decrease the time to culture results in resource-poor settings

    SN 1999ga: a low-luminosity linear type II supernova?

    Type II-linear supernovae are thought to arise from progenitors that have lost most of their H envelope by the time of the explosion, and they are poorly understood because they are only occasionally discovered. It is possible that they are intrinsically rare, but selection effects due to their rapid luminosity evolution may also play an important role in limiting the number of detections. In this context, the discovery of a subluminous type II-linear event is even more interesting. We investigate the physical properties and characterise the explosion site of the type II SN 1999ga, which exploded in the nearby spiral galaxy NGC 2442. Spectroscopic and photometric observations of SN 1999ga allow us to constrain the energetics of the explosion and to estimate the mass of the ejected material, shedding light on the nature of the progenitor star in the final stages of its life. The study of the environment in the vicinity of the explosion site provides information on a possible relation between these unusual supernovae and the properties of the galaxies hosting them. Despite the lack of early-time observations, we provide reasonable evidence that SN 1999ga was probably a type II-linear supernova that ejected a few solar masses of material, with a very small amount of radioactive elements of the order of 0.01 solar masses.Comment: 11 pages, 9 figures.
