    Physical and hydraulic properties of porous concrete

    [Abstract:] The work presented includes a review of the state of art of porous concrete. Its purpose is to evaluate the potential use of porous concrete in constructions where the level of surface runoff justifies it. A review of the literature presented here has been necessary where parameters of special consideration have been defined in the dosage of permeable mixtures. The study includes the definition of porous concrete in terms of its main components: cement, coarse aggregate, water, additives, and sand, in little or no quantity, to cause the generation of an effective content of interconnected voids that allow rapid storm drainage. Given the reports of variables of high incidence in the mechanical behavior of porous concrete (resistance/permeability relationship), an investigation is warranted to synthesize the effects of the variables in the preparation of the mixture: water–cement ratio, granulometry, and morphology of the aggregates, compaction pressure, and curing techniques, among others. Likewise, the protocols for the characterization of porous concrete and additional aspects relevant to support the experimental phase are exposed, constituting a reference or anchor point for developing technologies associated with the manufacture of this material and the possibilities of its implementation in constructions

    Mathematical models of cytotoxic effects in endpoint tumor cell line assays: Critical assessment of the application of a single parametric value as a standard criterion to quantify the dose-response effects and new unexplored proposal formats

    The development of convenient tools for describing and quantifying the effects of standard and novel therapeutic agents is essential for the research community, to perform more precise evaluations. Although mathematical models and quantification criteria have been exchanged in the last decade between different fields of study, there are relevant methodologies that lack proper mathematical descriptions and standard criteria to quantify their responses. Therefore, part of the relevant information that can be drawn from the experimental results obtained and the quantification of its statistical reliability are lost. Despite its relevance, there is not a standard form for the in vitro endpoint tumor cell lines' assays (TCLA) that enables the evaluation of the cytotoxic dose-response effects of anti-tumor drugs. The analysis of all the specific problems associated with the diverse nature of the available TCLA used is unfeasible. However, since most TCLA share the main objectives and similar operative requirements, we have chosen the sulforhodamine B (SRB) colorimetric assay for cytotoxicity screening of tumor cell lines as an experimental case study. In this work, the common biological and practical non-linear dose-response mathematical models are tested against experimental data and, following several statistical analyses, the model based on the Weibull distribution was confirmed as the convenient approximation to test the cytotoxic effectiveness of anti-tumor compounds. Then, the advantages and disadvantages of all the different parametric criteria derived from the model, which enable the quantification of the dose-response drug-effects, are extensively discussed. Therefore, model and standard criteria for easily performing the comparisons between different compounds are established. The advantages include a simple application, provision of parametric estimations that characterize the response as standard criteria, economization of experimental effort and enabling rigorous comparisons among the effects of different compounds and experimental approaches.     Generation of recycled aggregates and technical requirements for some applications

    In the present framework of sustainable construction, management of construction and demolition (C and amp;D) waste or debris is stressed and its correct implementation requires a previous estimation of the precise quantities and detailed composition of C and amp;D waste.This allows us to make an estimate of the production of recycled aggregate and to analyze the impact of its use in relation to the use of natural aggregate. Research of existing bibliography, the results obtained from both experimental testing and previous experience at construction works where C and amp;D debris is used, enabled us to develop technical specifications for each application of the recycled aggregates. This includes recommendations for use, quality control specifications, and the maximum percentage of recycled aggregate that ought to substitute natural aggregate in concrete mixes in order to get admissible performances

    Logopenic Aphasia due to a Strategic Stroke: New Evidence from a Single Case

    Among primary progressive aphasias (PPAs), logopenic variant PPA (lv-PPA) is usually related to Alzheimer's disease. Although it has been widely clinically and pathologically evaluated, the topography in LPA is still controversial. We report a patient presenting with a logopenic syndrome due to a strategic lesion located in the superior and middle temporal gyrus and compare our findings with those of a PiB-PET positive lv-PPA patient matched by age, gender, and education. We consider that our study provides new anatomical clues to better understand the cognitive mechanisms underlying the logopenic syndrome

    Characterization of in vitro antioxidant, antitumor and anti-inflammatory properties of plant species from Rosaceae family

    Since ancient times, humans have employed different plant species to prepare traditional remedies and treat diseases, due to the health-promoting effects associated with these plants. Among them, several species belonging to the Rosaceae family, namely Agrimonia eupatoria L., Filipendula ulmaria (L.) Maxim. and Rosa canina L., which have been used in different preparations, like infusions, decoctions or tinctures, to treat diverse illnesses and diseases [1]. Nowadays, the search for natural derived ingredients has prompted the study of these traditional plants. The present work aimed to evaluate the biological properties of the above-mentioned Rosaceae species, in order to promote their applications in different sectors. Antioxidant activity was evaluated by different in vitro assays: 2,2-diphenylpycrilhydrazyl (DPPH) radical scavenging assay, thiobarbituric acid reactive substances (TBARS) assay for inhibition of lipid peroxidation, OxHLIA assay for inhibition of oxidative hemolysis, and reducing power determination. Antitumor activity was tested against four tumor cell lines: MCF-7, CaCo, AGS and NCI-H460. Finally, anti-inflammatory activity was assessed by the inhibition of inflammation on lipopolysaccharide-induced RAW264.7 murine macrophages. Considering the results, all plant extracts showed antioxidant effects in the tested assays, especially F. ulmaria. All tested extracts also displayed relevant antitumor effects against the four tested tumor cell lines, mostly F. ulmaria, which exhibited the lowest growth inhibition 50 values (GI50) ranging from 34 to 86 μg/mL of extract, presenting a noteworthy cytotoxic effect. Finally, R. canina showed significant anti-inflammatory effects, with effective concentration 50 values (EC50) of 20 μg/mL of extract.     ¿Qué ocurre a los 24 meses del inicio del tratamiento en una muestra de personas con dependencia de la cocaína?

    La evolución de las personas que están en tratamiento por consumo de cocaína en Centros de Drogodependencias públicos, es un tema relevante debido al notable incremento de la demanda de estos tratamientos en los últimos años. El objetivo del presente estudio es analizar la evolución de personas con dependencia de la cocaína que han recibido tratamiento en Centros públicos de Drogodependencias de Galicia, tras dos años de haberse realizado una evaluación inicial. La muestra el presente estudio es de 38 sujetos que formaban parte de una muestra inicial más amplia (N = 115), y las variables que se analizan son: el consumo de sustancias, el ajuste social con la escala del O.T.I., problemas con la justicia y el malestar psicológico percibido por el sujeto evaluado con las subescalas e índices del SCL-90 R. Los resultados obtenidos son: a los dos años, el 78.9% de la muestra localizada no ha consumido cocaína en el mes previo a la evaluación, lo cual representa un 26% de la muestra inicial, y una disminución del malestar percibido por el sujeto. Estos resultados indican una evolución positiva de la muestra de personas en tratamiento por dependencia de la cocaína a los dos años que ha sido analizada en el presente estudioThe evolution of people in treatment for cocaine dependence at public Drug Abuse Centers is a relevant topic in view of the notable increase in demand for such treatment in recent years. The aim of the present study was to analyze the evolution of people with cocaine dependence receiving treatment at public Drug Abuse Centers in Galicia (NW Spain), two years after the initial assessment for inclusion on the treatment program. The sample for the present study was 38 participants who formed part of a larger initial sample (N = 115). The variables analyzed were: substance use, social adjustment on the O.T.I. scale, problems with the legal system and perceived psychological distress as assessed with the scales and indices of the SCL-90 R. The results indicate that, after two years of treatment, 78.9% of those in the sample located (26% of the initial sample) had not consumed cocaine in the month prior to assessment, and that there was a reduction in psychological distress. These results indicate a positive evolution after two years of the sample of people in treatment for cocaine dependence assessed in this studyS

    Cognitive Composites Domain Scores Related to Neuroimaging Biomarkers within Probable-Amnestic Mild Cognitive Impairment-Storage Subtype

    The probable-amnestic (Pr-a) mild cognitive impairment (MCI)-storage subtype is a phenotype with 8.5 times more risk of conversion to dementia, mainly Alzheimer's disease (AD), than the possible non-amnestic (Pss-na) MCI. The aim of this study was to find the optimized cognitive composites (CCs) domain scores most related to neuroimaging biomarkers within Pr-aMCI-storage subtype patients. The Fundació ACE (ACE) study with 20 Pr-aMCI-storage subtype subjects (MCI) were analyzed. All subjects underwent a neuropsychological assessment, a structural MRI, FDG-PET, and PIB-PET. The adjusted hippocampal volume (aHV) on MRI, the standard uptake value ratio (SUVR) on FDG-PET and PIB-PET SUVR measures were analyzed. The construction of the CCs domain scores, and the aHV on MRI and FDG-PET SUVR measures, were replicated in the parental AB255 study database (n = 133 MCI). Partial correlations adjusted by age, gender, and education were calculated with the associated p -value among every CC domain score and the neuroimaging biomarkers. The results were replicated in the "MCI due to AD" with memory storage impairments from ADNI. Delayed Recall CC domain score was significantly correlated with PIB-PET SUVR (β= -0.61, p = 0.003) in the ACE study and also with aHV on MRI (β= 0.27, p = 0.01) and FDG-PET SUVR (β= 0.27, p = 0.01) in the AB255 study. After a median survival time of 20.6 months, 85% from the ACE MCI converted to AD. The replication of our results in the ADNI dataset also confirmed our findings. Delayed Recall is the CC domain score best correlated with neuroimaging biomarkers associated with prodromal AD diagnosis

    Genome-wide association analysis of dementia and its clinical endophenotypes reveal novel loci associated with Alzheimer's disease and three causality networks : The GR@ACE project

    Introduction: Large variability among Alzheimer's disease (AD) cases might impact genetic discoveries and complicate dissection of underlying biological pathways. Methods: Genome Research at Fundacio ACE (GR@ACE) is a genome-wide study of dementia and its clinical endophenotypes, defined based on AD's clinical certainty and vascular burden. We assessed the impact of known AD loci across endophenotypes to generate loci categories. We incorporated gene coexpression data and conducted pathway analysis per category. Finally, to evaluate the effect of heterogeneity in genetic studies, GR@ACE series were meta-analyzed with additional genome-wide association study data sets. Results: We classified known AD loci into three categories, which might reflect the disease clinical heterogeneity. Vascular processes were only detected as a causal mechanism in probable AD. The meta-analysis strategy revealed the ANKRD31-rs4704171 and NDUFAF6-rs10098778 and confirmed SCIMP-rs7225151 and CD33-rs3865444. Discussion: The regulation of vasculature is a prominent causal component of probable AD. GR@ACE meta-analysis revealed novel AD genetic signals, strongly driven by the presence of clinical heterogeneity in the AD series

    Genomic Characterization of Host Factors Related to SARS-CoV-2 Infection in People with Dementia and Control Populations: The GR@ACE/DEGESCO Study

    Emerging studies have suggested several chromosomal regions as potential host genetic factors involved in the susceptibility to SARS-CoV-2 infection and disease outcome. We nested a COVID-19 genome-wide association study using the GR@ACE/DEGESCO study, searching for susceptibility factors associated with COVID-19 disease. To this end, we compared 221 COVID-19 confirmed cases with 17,035 individuals in whom the COVID-19 disease status was unknown. Then, we performed a meta-analysis with the publicly available data from the COVID-19 Host Genetics Initiative. Because the APOE locus has been suggested as a potential modifier of COVID-19 disease, we added sensitivity analyses stratifying by dementia status or by disease severity. We confirmed the existence of the 3p21.31 region (LZTFL1, SLC6A20) implicated in the susceptibility to SARS-CoV-2 infection and TYK2 gene might be involved in COVID-19 severity. Nevertheless, no statistically significant association was observed in the COVID-19 fatal outcome or in the stratified analyses (dementia-only and non-dementia strata) for the APOE locus not supporting its involvement in SARS-CoV-2 pathobiology or COVID-19 prognosis

We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer's disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer's disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer's disease
