    Report on the Fourth Excavation Season (2011) of the Madâ'in Sâlih Archaeological Project

    This volume is the report on the results of the fourth excavation season of the Saudi-French Archaeological Project at Madâ'in Sâlih, ancient Hegra in the Nabataean kingdom, in north-west Saudi Arabia (MAEE, SCTA, CNRS, Univ Paris 1, IFPO). Apart from the results obtained in the different excavation areas (both in the residential area and in tomb IGN 117), the reader will find a study on the cairns/tumuli of the site (W. Abu-Azizeh) as well as intermediary reports on the geophysical detection (Chr. Benech), the fauna (J. Studer) and the pottery (C.     The hallmarks of Alzheimer's disease (AD) are characterized by cognitive decline and behavioral changes. The most prominent brain region affected by the progression of AD is the hippocampal formation. The pathogenesis involves a successive loss of hippocampal neurons accompanied by a decline in learning and memory consolidation mainly attributed to an accumulation of senile plaques. The amyloid precursor protein (APP) has been identified as precursor of Aβ-peptides, the main constituents of senile plaques. Until now, little is known about the physiological function of APP within the central nervous system. The allocation of APP to the proteome of the highly dynamic presynaptic active zone (PAZ) highlights APP as a yet unknown player in neuronal communication and signaling. In this study, we analyze the impact of APP deletion on the hippocampal PAZ proteome. The native hippocampal PAZ derived from APP mouse mutants (APP-KOs and NexCreAPP/APLP2-cDKOs) was isolated by subcellular fractionation and immunopurification. Subsequently, an isobaric labeling was performed using TMT6 for protein identification and quantification by high-resolution mass spectrometry. We combine bioinformatics tools and biochemical approaches to address the proteomics dataset and to understand the role of individual proteins. The impact of APP deletion on the hippocampal PAZ proteome was visualized by creating protein-protein interaction (PPI) networks that incorporated APP into the synaptic vesicle cycle, cytoskeletal organization, and calcium-homeostasis. The combination of subcellular fractionation, immunopurification, proteomic analysis, and bioinformatics allowed us to identify APP as structural and functional regulator in a context-sensitive manner within the hippocampal active zone network

    The creative voice: free indirect speech in the cinema of Rohmer and Bresson

    This article examines a particular use of the voice in cinema which conveys character reflexivity and generates expressive ambiguity in a film's narrative point of view. Drawing on Pasolini's notion of the free indirect style and Deleuze's elaboration of this into his concept of the free indirect speech-act, two different creative uses of speech are analysed: one in Rohmer's Claire's Knee (1970) and the other in Bresson's Diary of a Country Priest (1951). The essay finds that when characters narrate their thoughts, feelings, actions or experiences in a reflexive manner (that is, to themselves, even if also to another), and this activity becomes for a time the primary event of the film, these characters step into the role of storyteller. They become both subject and object of their utterances which take on the style of the free indirect. With this, the voice enters into a new relationship with the image – directing, counterpointing, or disconnecting from it – changing the function of voice and image while seeming momentarily to supplant the film's narration. The symbolic nature of language gives words the power to create images and meaning that cannot be captured or conveyed by the camera alone. Privileging speech as a creative force not only dimensionalises character but, on a formal level, creates a role for voice that allows it to exist not as an appendage to the image but for its own sake. In order to suggest practical uses for these theoretical considerations, Bresson's and Rohmer's reflections on the role of the voice are also discussed

    Parkinsonian gait in elderly people: Significance of the threshold value of two and more falls per year.

    Objectives: Parkinsonism in the elderly presents a major risk factor for recurrent falls (2 and more falls per year), which is associated with increased morbidity. The main objective was to investigate explanatory variables relating to the risk of being recurrent fallers (RF) in persons with parkinsonian gait.Methods: Seventy-nine among 172 eligible persons were enrolled in this prospective study, the findings of which were analyzed at 12 months. Motor and non-motor features, as well as follow-up interviews to identify falls, loss of ability to walk, fluctuating cognition, traumatic falls, all-cause hospitalizations and deaths were collated and results compared between non RF (zero and one fall per year) and RF. Bayesian model averaging was used to predict the probability of patients being RF from their medical history as well as from cognitive assessment, gait velocity, vision and posture.Results: N=79, 0.58 men, 50% had Parkinson's disease, 14% other neurodegenerative parkinsonian syndrome, 23% vascular parkinsonism and 13% Lewy body disease, 58% were RF. Median age 81.2 years and median MMSE 25/30. A history of falls and of hallucinations, median odds ratio respectively 9.06 (CI 2.34-38.22), 4.21 (CI 1.04-18.67) were associated with the highest odds ratios along with fluctuating cognition and abnormal posture. Two or more falls a year was a relevant threshold to distinguish a population with a high risk of comorbidity.Conclusion: The whole history of falls, hallucinations and fluctuating cognition can be considered predictive of recurrent falls in elderly people with parkinsonian gait and provide a tracking tool for patient management

    Sex-Biased Transcriptome of Schistosoma mansoni: Host-Parasite Interaction, Genetic Determinants and Epigenetic Regulators Are Associated with Sexual Differentiation

    Background Among more than 20,000 species of hermaphroditic trematodes, Schistosomatidae are unusual since they have evolved gonochorism. In schistosomes, sex is determined by a female heterogametic system, but phenotypic sexual dimorphism appears only after infection of the vertebrate definitive host. The completion of gonad maturation occurs even later, after pairing. To date, the molecular mechanisms that trigger the sexual differentiation in these species remain unknown, and in vivo studies on the developing schistosomulum stages are lacking. To study the molecular basis of sex determination and sexual differentiation in schistosomes, we investigated the whole transcriptome of the human parasite Schistosoma mansoni in a stage-and sex-comparative manner. Methodology/Principal Findings We performed a RNA-seq on males and females for five developmental stages: cercariae larvae, three in vivo schistosomulum stages and adults. We detected 7,168 genes differentially expressed between sexes in at least one of the developmental stages, and 4,065 of them were functionally annotated. Transcriptome data were completed with H3K27me3 histone modification analysis using ChIP-Seq before (in cercariae) and after (in adults) the phenotypic sexual dimorphism appearance. In this paper we present (i) candidate determinants of the sexual differentiation, (ii) sex-biased players of the interaction with the vertebrate host, and (iii) different dynamic of the H3K27me3 histone mark between sexes as an illustration of sex-biased epigenetic landscapes. Conclusions/Significance Our work presents evidence that sexual differentiation in S. mansoni is accompanied by distinct male and female transcriptional landscapes of known players of the host-parasite crosstalk, genetic determinants and epigenetic regulators. Our results suggest that such combination could lead to the optimized sexual dimorphism of this parasitic species. As S. mansoni is pathogenic for humans, this study represents a promising source of therapeutic targets, providing not only data on the parasite development in interaction with its vertebrate host, but also new insights on its reproductive function
