    Characterization of Cre recombinase activity for in vivo targeting of adipocyte precursor cells.

    The increased incidence of obesity and metabolic disease underscores the importance of elucidating the biology of adipose tissue development. The recent discovery of cell surface markers for prospective identification of adipose precursor cells (APCs) in vivo will greatly facilitate these studies, yet tools for specifically targeting these cells in vivo have not been identified. Here, we survey three transgenic mouse lines, Fabp4-Cre, PdgfRα-Cre, and Prx1-Cre, precisely assessing Cre-mediated recombination in adipose stromal populations and mature tissues. Our data provide key insights into the utility of these tools to modulate gene expression in adipose tissues. In particular, Fabp4-Cre is not effective to target APCs, nor is its activity restricted to these cells. PdgfRα-Cre directs recombination in the vast majority of APCs, but also targets other populations. In contrast, adipose expression of Prx1-Cre is chiefly limited to subcutaneous inguinal APCs, which will be valuable for dissection of APC functions among adipose depots

    Biomarkers: a strategic tool in the assessment of environmental quality of coastal waters

    Ecosystems are under the pressure of complex mixtures of contaminants whose effects are not always simple to assess. Biomarkers, acting as early warning signals of the presence of potentially toxic xenobiotics, are useful tools for assessing either exposure to, or the effects of these compounds providing information about the toxicant bioavailability. In fact, it has been argued that a full understanding of ecotoxicological processes must consider an integrated multi-level approach, in which molecular impact is related with higher-order biological consequences at the individual, population and community levels. Monitoring programs should make use of this tool to link contaminants and ecological responses fulfilling strategies like those launched by OSPAR (Commissions of Oslo and Paris) Convention on the protection of the marine environment of the North-East Atlantic and the International Council for the Exploration of the Sea (ICES). An overview of the work done in the past few years using biomarkers as in situ tools for pollution assessment in Portuguese coastal waters is presented as a contribution to the set up of a biomonitoring program for the Portuguese coastal zone. Considering the data set available the biomonitoring proposal should include the analysis of biomarkers and effects at individual levels. The aim of the program will include a spatial and temporal characterization of the biomarkers acetyl-cholinesterase, metallothioneins, DNA damage, adenylate energy charge and scope-for-growth levels. The investigation of the spatial variation of biomarkers is crucial to define sites for long term monitoring, which will be integrated with a chemical monitoring program.     Characterization of triacylglycerols in Madeira laurel oil by HPLC-atmospheric pressure chemical ionization-MS

    Madeira laurel oil was fractionated by liquid ex traction combined with TLC, and TAGs were analyzed by HPLC coupled with atmospheric pressure chemical ionization-MS (APCI-MS). Eluted molecular species compositions of the eluted TAG in the complex natural mixture were determined by GC identification of FAME and by LC-atmospheric pressure chemi cal ionization (APCI)-MS analysis of the lipid. The APCI-MS spectra of most TAG exhibited [M + H]+ and [M − RCOO]+ ions, which defined the M.W. and the molecular association of fatty acyl residues, respectively. Despite the relatively high de gree of saturation, with a saturated/unsaturated ratio of 0.70, no totally saturated TAG nor mixed asymmetric TAG with two sat urated FA (SSM or SSU, where S is saturated, M is monounsatu rated, and U is unsaturated) were found. This type of molecular structure provides a possible explanation for the relatively low m.p. (12–15°C) and also the high oxidative resistance observed.info:eu-repo/semantics/publishedVersio

    Reproductive biology of Megabalanus azoricus (Pilsbry), the Azorean Barnacle

    Copyright © 2007, Taylor & Francis.The reproductive biology of Megabalanus azoricus (Pilsbry), the commercially exploited barnacle in the Azores, was studied in an attempt to provide the scientific basis for the sustainable management of this heavily exploited regional marine resource. Both the Ospar Commission and WWF have expressed concern for this species, considering it at risk and in urgent need for scientific study. Barnacles were collected every month from shallow water (.3 m depth) around São Miguel Island from October 2004 to September 2005. Individuals were measured and gonads processed for histology and analysed with a stereological method. M. azoricus has a hermaphroditic reproductive system with separate gonads and it was possible to describe the various stages of gametic maturation in both. Data on fertility were also obtained by egg counts and calculation of the Gonadosomatic index (GSI). Throughout the year, some specimens of M. azoricus can be found that rare mature, but two reproductive peaks were observed, one in January and a smaller one in July. Esta comunicação traz à tona questões que envolvem o currículo nos cursos de licenciaturas para a formação de professores no Instituto Federal de Educação, Ciência e Tecnologia do Rio Grande do Norte, IFRN, Brasil. A partir do conjunto de conhecimentos contemplados na disciplina de Didática, a qual compõe o núcleo pedagógico do referido currículo, tratamos aqui de um recorte epistemológico que evidencia os conhecimentos da Didática, na perspectiva crítica, como norteadores de uma construção reflexiva da identidade profissional docente. Nosso objetivo geral é discutir o potencial curricular da Didática em um currículo praticado na formação inicial para a docência e suas interfaces com os construtos identitários. As referencias teóricas do estudo balizam-se em produções do campo da Didática crítica, com ênfase em Freire (2006), Candau (2002), Rios (2010). Os conhecimentos e princípios da didática sinalizam uma formação e uma identidade profissional pautadas na reflexão do fazer pedagógico, nos processos de conscientização sobre a dimensão social e ética contidas no ser professor, bem como valorizam o fazer técnico-instrumental, o saber científico e o saber ético.     Decision support system for accessing costs and risks of connected and autonomous vehicles as mobility service in urban contexts

    A predicted increase of connected autonomous vehicles (CAVs) in our roads paved the way for new opportunities and challenges towards the promotion of sustainable mobility. However, the impacts of CAVs on the road environment and its implications are widely dependent on technological choices and public policy [1,2]. Therefore, this research (PhD Workplan) intends to assess whether CAVs could be effective mobility solutions for improving social, economic and environmental efficiency [4]. This question will be addressed by developing a decision support tool driven by comprehensive data analysis and modelling processes. The outputs achieved will integrate a tool that will support transport system’s planning and the implementation of urban strategies to introduce CAVs [3,5]. Thus, the research’s main focus encompasses the development of a model-driven decision support system (DSS) that allows assessing the costs and risks of implementing CAVs in urban context [3,4]. Three specific research objectives are assumed: I) Predicting impacts of CAVs operation in urban contexts, by analyzing cost-efficiency, transportation demand and mobility patterns considering market penetration scenarios of CAVs in Portugal; II) Conceiving a hybrid transport planning tool to assess possible restrictions to CAVs on different types of links through field data testing and simulating scenarios using a microscopic traffic model. Data will support the development of a macroscopic model for a full network assessment performance; III) Developing a multidimensional decision tool directed to a wide range of stakeholders, both from public or private sectors, to compute the benefits, costs, constraints and risks of implementing CAVs on urban mobility systems. Preliminary results from different urban arterials indicate that CAVs can have negative or negligible impacts in some urban contexts. However, if the impact on the traffic flow’s energy performance is considered into the internal car following algorithm, global energy savings of up to 12% can be achieved.publishe

    The 50 years experience of adrenocorticotropic hormone (ACTH) use in West Syndrome: literatura review and UNIFESP protocol

    INTRODUÇÃO: A eficácia do ACTH no tratamento da Síndrome de West (SW) é extensivamente debatida na literatura, o mesmo ocorrendo em relação às doses, efeitos colaterais e protocolos de utilização. OBJETIVO: Revisar, analisar criticamente e discutir os achados das publicações mais recentes e relevantes relacionadas ao uso do ACTH no tratamento da SW. METODOLOGIA: Pesquisou-se as bases de dados MEDLINE e PUBMED, utilizando-se as palavras-chave ACTH e WEST SYNDROME, período de 1997 a 2007. RESULTADOS: Foram obtidos 96 títulos no MEDLINE e 158 no PUBMED, e dentre eles foram selecionados 22 publicações. Outros trabalhos anteriores ao período avaliado mas que apresentavam relevância especial no contexto desta revisão também foram incluídos. CONCLUSÕES: Há evidências de que o ACTH provavelmente apresente eficácia maior no controle imediato dos espasmos e da hipsarritmia. Entretanto, não há evidências concretas desta superioridade no longo prazo (controle de crises a longo prazo e interrrupção ou prevenção de comprometimento do desenvolvimento neuropsicomotor).INTRODUCTION: It is extensively discussed in the literature the efficacy of ACTH in West Syndrome (WS) as well, dosage, best protocols and side effects. OBJECTIVE: Analyze and discuss the most recent and relevant publication related to ACTH in patients with WS. METHODOLOGY: Literature publication was selected from MEDLINE and PUBMED between 1997 to 2007 using the following key words: "ACTH" and " West Syndrome". RESULTS: 96 titles were referral at medline and 158 at pubmed. 22 were selected and previous studies with special relevance were also included. CONCLUSIONS: There are evidences that ACTH probably has efficacy for immediately control of spasms and hypsarrhythmia.     Phase diagram and critical properties within an effective model of QCD: the Nambu-Jona-Lasinio model coupled to the Polyakov loop

    We investigate the phase diagram of the so-called Polyakov--Nambu--Jona-Lasinio model at finite temperature and non-zero chemical potential with three quark flavors. Chiral and deconfinement phase transitions are discussed and the relevant order-like parameters are analyzed. The results are compared with simple thermodynamic expectations and lattice data. We present the phase diagram in the (T, μB)T,\,\mu_B) plane, paying special attention to the critical end point: as the strength of the flavor-mixing interaction becomes weaker, the critical end point moves to low temperatures and can even disappear.Comment: 46 pages, 11 figures, 3 table
