Nanobody mediated inhibition of attachment of F18 fimbriae expressing Escherichia coli
Post-weaning diarrhea and edema disease caused by F18 fimbriated E. coli are important diseases in newly weaned piglets and lead to severe production losses in farming industry. Protective treatments against these infections have thus far limited efficacy. In this study we generated nanobodies directed against the lectin domain of the F18 fimbrial adhesin FedF and showed in an in vitro adherence assay that four unique nanobodies inhibit the attachment of F18 fimbriated E. coli bacteria to piglet enterocytes. Crystallization of the FedF lectin domain with the most potent inhibitory nanobodies revealed their mechanism of action. These either competed with the binding of the blood group antigen receptor on the FedF surface or induced a conformational change in which the CDR3 region of the nanobody displaces the D ''-E loop adjacent to the binding site. This D ''-E loop was previously shown to be required for the interaction between F18 fimbriated bacteria and blood group antigen receptors in a membrane context. This work demonstrates the feasibility of inhibiting the attachment of fimbriated pathogens by employing nanobodies directed against the adhesin domain
Streamwise vortices destabilize swimming bluegill sunfish (Lepomis macrochirus)
In their natural environment, fish must swim stably through unsteady flows and vortices, including vertical vortices, typically shed by posts in a flow, horizontal cross-flow vortices, often produced by a step or a waterfall in a stream, and streamwise vortices, where the axis of rotation is aligned with the direction of the flow. Streamwise vortices are commonly shed by bluff bodies in streams and by ships\u27 propellers and axial turbines, but we know little about their effects on fish. Here, we describe how bluegill sunfish use more energy and are destabilized more often in flow with strong streamwise vorticity. The vortices were created inside a sealed flow tank by an array of four turbines with similar diameter to the experimental fish. We measured oxygen consumption for seven sunfish swimming at 1.5 body lengths (BL) s−1 with the turbines rotating at 2 Hz and with the turbines off (control). Simultaneously, we filmed the fish ventrally and recorded the fraction of time spent maneuvering side-to-side and accelerating forward. Separately, we also recorded lateral and ventral video for a combination of swimming speeds (0.5, 1.5 and 2.5 BL s−1) and turbine speeds (0, 1, 2 and 3 Hz), immediately after turning the turbines on and 10 min later to test for accommodation. Bluegill sunfish are negatively affected by streamwise vorticity. Spills (loss of heading), maneuvers and accelerations were more frequent when the turbines were on than in the control treatment. These unsteady behaviors, particularly acceleration, correlated with an increase in oxygen consumption in the vortex flow. Bluegill sunfish are generally fast to recover from roll perturbations and do so by moving their pectoral fins. The frequency of spills decreased after the turbines had run for 10 min, but was still markedly higher than in the control, showing that fish partially adapt to streamwise vorticity, but not completely. Coping with streamwise vorticity may be an important energetic cost for stream fishes or migratory fishes

Renal safety of lithium in HIV-infected patients established on tenofovir disoproxil fumarate containing antiretroviral therapy: analysis from a randomized placebo-controlled trial
BACKGROUND: The prevalence of bipolar disorder in HIV-infected patients is higher than the general population. Lithium is the most effective mood stabiliser, while tenofovir disoproxil fumarate (TDF) is frequently used as part of combination antiretroviral therapy (ART). Both TDF and lithium are associated with renal tubular toxicity, which could be additive, or a pharmacokinetic interaction may occur at renal transporters with a decrease in TDF elimination. OBJECTIVE: We report on the change in estimated glomerular filtration rate (eGFR) using the modification of diet in renal disease formula in participants who received ART including TDF and were enrolled in a 24Â week randomised trial of lithium versus placebo in patients with HIV-associated neurocognitive impairment. METHODS: We included HIV-infected adults with cognitive impairment established on ART for at least 6Â months with a suppressed viral load attending public sector ART clinics in Cape Town, South Africa. We excluded participants with an eGFRÂ <60Â mL/min and treated with medications predisposing to lithium toxicity. We reviewed participants weekly for the first month for adverse events followed by 4 weekly visits for renal function assessment, adverse event monitoring and adherence. Lithium dose was titrated to achieve the maintenance target plasma concentration of between 0.6 and 1.0Â mmol/L. Sham lithium concentrations were generated for participants receiving placebo. RESULTS: We included 23 participants allocated to the lithium arm and 30 participants allocated to the placebo arm. Baseline characteristics were not statistically different with a mean age of 37.7 and 40.8Â years, a median time on ART of 33 and 40Â months and an eGFR of 139.3 and 131.0Â mL/min in the lithium and placebo arms respectively. There was no statistical significant difference in the reduction in eGFR or increase in potassium between the two arms during the 24Â weeks. CONCLUSIONS: We found that 24-week treatment of HIV-infected patients with lithium and TDF did not result in increased nephrotoxicity. Psychiatric emergency services and their relationships with mental health network in Brazil
OBJETIVO: Revisão de dados da literatura relativos ao papel e à inserção de serviços de emergências psiquiátricas em redes de saúde mental. MÉTODO: Foi realizada uma busca em banco de dados (PubMed e SciELO) de artigos empÃricos e revisões sobre serviços de emergências psiquiátricas e rede de serviços de saúde mental. RESULTADOS: Serviços de emergências psiquiátricas constituem unidade central para o funcionamento adequado de redes de saúde mental, tanto pelo manejo de situações de emergências, como pela regulação da rede em que se insere. Os serviços de emergências psiquiátricas relacionam-se com todos os serviços hospitalares e extra-hospitalares, possibilitando a organização do fluxo das internações e evitando sobrecarga da rede de saúde mental. As funções dos serviços de emergências psiquiátricas são amplas e extrapolam o simples encaminhamento para internação integral, pois estabilização clÃnica e suporte psicossocial podem ser alcançados em serviços de emergências psiquiátricas bem estruturados. No Brasil, estas funções foram ampliadas após a Reforma da Assistência à Saúde Mental e a sobrecarga das redes de saúde mental provocadas pelas dificuldades e limitações dos serviços hospitalares e extra-hospitalares. CONCLUSÃO: Serviços de emergências psiquiátricas devem ser valorizados e ampliados, principalmente aqueles localizados em hospitais gerais. Recomenda-se que o investimento em emergências psiquiátricas seja prioridade das polÃticas de saúde pública brasileiras para o aprimoramento da atenção na saúde mentalOBJECTIVE: To review the literature concerning the role and the inclusion of emergency psychiatric services in mental health networks. METHOD: We performed a search in online databases (PubMed and SciELO) of empirical articles and reviews about emergency psychiatric services and networks of mental health services. RESULTS: Emergency psychiatric services are a core unit for a proper functioning of mental health networks, by both the management of emergencies itself, and the regulation of the network where it belongs. The emergency psychiatric services relate to all inpatient and outpatient services, allowing the organization of the flow of admissions and avoiding the overload of the network of mental health.The functions of emergency psychiatric services are broad and go beyond the simple referral for hospitalization, since clinical stabilization and psychosocial support can be reached in well structured emergency psychiatric services. In Brazil, these functions were expanded after the mental health reform and the burden of mental health network caused by difficulties and limitations of inpatient and outpatient services. CONCLUSION: Emergency psychiatric services should be recovered and expanded; especially those located in general hospitals. Intersectional climate justice : A conceptual pathway for bridging adaptation planning, transformative action, and social equity

However, the compounding and overlapping vulnerabilities of historically marginalized residents are commonly tackled in a fragmented manner by conventional adaptation approaches, even when justice is presented as an overarching goal of these plans. In response, we propose an intersectional pivot in climate adaptation research and practice to analyze the interconnected forms of social-environmental injustices that drive vulnerabilities in cities, paving the way for more concrete and integrated strategies of just urban adaptation and transformation. This paper brings together narrative and analytical review methodologies to inform a new conceptual framework that highlights the need to (1) tackle underlying reinforcers of racial and gender inequalities; (2) redress drivers of differential vulnerabilities; (3) take politics and ethics of care seriously; (4) adopt place-based and place-making approaches; and (5) promote cross-identity forms of activism and community resilience building. We illustrate the framework with examples of ongoing projects in Barcelona, Spain, which is an early adopter of intersectional thinking and justice-driven principles in climate action. Although many initiatives are in a pilot phase and do not all exclusively focus on climate adaptation, experiences from Barcelona do provide illustrative directionality for innovative and integrated approaches that can address multiple and intersecting social-environmental inequities

A análise de óleos lubrificantes e lÃquidos de arrefecimento é um excelente termômetro para indicar as condições protetoras dos fluidos e consequentemente o nivel de desgaste de peças e componentes: como transmissões, motores diesel, sistemas hidráulicos e outros.
Sabemos da grande importancia da lubrificação na manutenção, pois ao colocarmos uma camada de um lubrificante entre duas superfÃcies de um determinado equipamento, impediremos o contato entre estas. Retirando esse contato, evitaremos durante o movimento, cisalhamento e arranhamento com o desprendimento de calor e desgaste dessas superfÃcies. Definiremos então como lubrificante, as substâncias que interpostas entre duas superfÃcies em deslocamento relativo, diminuem a resistência ao movimento. A função dos lubrificantes é evitar o contato metálico, reduzir o atrito, e conseqüentemente o desgaste, refrigerar, etc. Os principais fatores que exercem influência na lubrificação são:
velocidade, temperatura e pressão.
O objetivo de um plano de lubrificação é promover uso correto dos lubrificantes reduzindo a quantidade de manutenções corretivas, diminuindo a reposição de peças e aumentando a produtividade. Ao implantar um sistema de planejamento deste porte e controle de lubrificação e treinamento de pessoal responsável por esta área é possÃvel reduzir o desgaste excessivo de componentes mecânicos e falhas devido ao emprego de lubrificantes não recomendados, bem como por lubrificação inadequada e aplicação correta das técnicas de lubrificação. O emprego racional de lubrificantes elimina o desperdÃcio e prolonga a vida útil dos lubrificantes em uso e das máquinas. Este controle é feito por meio do acompanhamento das cargas por programa de análise.
Pensando neste cenário foi desenvolvido uma planilha (figura 1) inteligente programado em VBA que é capaz de determinar rotas otimizadas, quantidade de lubrificante a ser aplicado e técnicas de lubrificação eficiente a partir de um levantamento de dados de máquinas como rolamentos, temperaturação de operação, classificação, rotação e etc.
Tudo isso visando minimo de perdas possÃveis de consumo de lubrificantes, paradas de equipamentos postergando a vida útil do mesmo, além de promover um ganho de qualidade para o lubrificador.Eje: Lubricación y tribologÃa.Facultad de IngenierÃ
