Should the poultry red mite Dermanyssus gallinae be of wider concern for veterinary and medical science?
The poultry red mite Dermanyssus gallinae is best known as a threat to the laying-hen industry; adversely affecting production and hen health and welfare throughout the globe, both directly and through its role as a disease vector. Nevertheless, D. gallinae is being increasingly implemented in dermatological complaints in non-avian hosts, suggesting that its significance may extend beyond poultry. The main objective of the current work was to review the potential of D. gallinae as a wider veterinary and medical threat. Results demonstrated that, as an avian mite, D. gallinae is unsurprisingly an occasional pest of pet birds. However, research also supports that these mites will feed from a range of other animals including: cats, dogs, rodents, rabbits, horses and man. We conclude that although reported cases of D. gallinae infesting mammals are relatively rare, when coupled with the reported genetic plasticity of this species and evidence of permanent infestations on non-avian hosts, potential for host-expansion may exist. The impact of, and mechanisms and risk factors for such expansion are discussed, and suggestions for further work made. Given the potential severity of any level of host-expansion in D. gallinae, we conclude that further research should be urgently conducted to confirm the full extent of the threat posed by D. gallinae to (non-avian) veterinary and medical sectors
Biomorphic Engineering of Multifunctional Polylactide Stomatocytes toward Therapeutic Nano-Red Blood Cells
Morphologically discrete nanoarchitectures, which mimic the structural complexity of biological systems, are an increasingly popular design paradigm in the development of new nanomedical technologies. Herein, engineered polymeric stomatocytes are presented as a structural and functional mimic of red blood cells (RBCs) with multifunctional therapeutic features. Stomatocytes, comprising biodegradable poly(ethylene glycol) block-poly(D,L-lactide), possess an oblate-like morphology reminiscent of RBCs. This unique dual-compartmentalized structure is augmented via encapsulation of multifunctional cargo (oxygen-binding hemoglobin and the photosensitizer chlorin e6). Furthermore, stomatocytes are decorated with a cell membrane isolated from erythrocytes to ensure that the surface characteristics matched those of RBCs. In vivo biodistribution data reveal that both the uncoated and coated nano-RBCs have long circulation times in mice, with the membrane-coated ones outperforming the uncoated stomatoctyes. The capacity of nano-RBCs to transport oxygen and create oxygen radicals upon exposure to light is effectively explored toward photodynamic therapy, using 2D and 3D tumor models; addressing the challenge presented by cancer-induced hypoxia. The morphological and functional control demonstrated by this synthetic nanosystem, coupled with indications of therapeutic efficacy, constitutes a highly promising platform for future clinical application.</p
Effect of dimerized melittin on gastric cancer cells and antibacterial activity
Melittin is the peptide toxin found in bee venom and is effective against cancer cells. To enhance its activity, a branched dimeric form of melittin was designed. The monomeric form of the peptide was more cytotoxic against gastric cancer cells at low concentrations (1-5 μM) than the dimer form, while the cytotoxic effect was comparable at higher concentrations (10 μM). Confocal microscopy showed that both the monomer and dimer forms of melittin with fluorescent label at the C terminus penetrated the cytoplasm and localized at the cell nucleus and disrupted the cell membrane. The results indicated that both peptides localized in the nucleus and no significant difference in penetration was observed between monomer and dimer of melittin. Although the C and N termini are important for melittin activity, using C terminus for dimerization of the peptide resulted in similar activity for the monomer and dimer against bacteria and gastric cancer cells
Manipulating thyroid status alters endoplasmic reticulum calcium homeostasis in rat cerebellum

hormones regulate the efficiency and expression of sarco-endoplasmic reticulum
calcium ATPases in cardiac and skeletal muscle. However, little is known about
the relationship between thyroid hormones and calcium (Ca2+)
homeostasis in the brain. It is hypothesized that manipulating rat thyroid
hormone levels would induce significant brain Ca2+ adaptations
consistent with clinical findings. Adult male Sprague-Dawley rats were assigned
to one of three treatment groups for 28 days: control, hypothyroid
(6-n-propyl-2-thiouracil (PTU), an inhibitor of thyroxine (T4) synthesis), and
hyperthyroid (T4). Throughout, rats were given weekly behavioral tests. Ca2+
accumulation decreased in the cerebellum in both hyper- and hypothyroid
animals. This was specific to different ER pools of calcium with regional
heterogeneity in the response to thyroid hormone manipulation. Behavioral tasks
demonstrated sensitivity to thyroid manipulation, and corresponded to
alterations in calcium homeostasis. Ca2+ accumulation heterogeneity
in chronic hyper- and hypothyroid animals potentially explains clinical
manifestations of altered thyroid status
