    Antifungal drug susceptibility profile of Pichia anomala isolates from patients presenting with nosocomial fungemia

    In vitro susceptibility of 58 isolates of Pichia anomala to five antifungal drugs using two broth microdilution methods (CLSI and EUCAST) was analyzed. Low susceptibility to itraconazole was observed.     Exploring the dynamics of Programmed Death-Ligand 1 in canine lymphoma: unraveling mRNA amount, surface membrane expression and plasmatic levels

    IntroductionProgrammed Death-Ligand 1 is a well-known immune checkpoint molecule. Recent studies evaluated its expression in different canine cancer types through different laboratory techniques. The present study aims to evaluate the surface membrane protein expression (mPD-L1) by means of flow cytometry (FC) in different canine lymphoma immunophenotypes. Furthermore, in a subset of cases, mRNA and plasmatic soluble protein (sPD-L1) have been assessed in the same patient, and correlations among results from the three analyses investigated.MethodsSamples obtained for diagnostic purpose from untreated dogs with a confirmed lymphoma immunophenotype were included: surface protein was assessed via FC and quantified with median fluorescence index ratio (MFI ratio), gene expression was evaluated by real time quantitative polymerase chain reaction (RT-qPCR) and plasmatic concentration of soluble protein (sPD-L1) measured with ELISA. Statistical analyses were performed to investigate any difference among FC immunophenotypes, updated Kiel cytological classes, and in the presence of blood infiltration.ResultsConsidering FC, most B-cell lymphomas (BCL) were positive, with higher MFI ratios than other subtypes (81%, median MFI ratio among positive samples = 1.50, IQR 1.21–2.03, range 1.01–3.47). Aggressive T-cell lymphomas had a lower percentage of positive samples (56%) and showed low expression (median MFI ratio in positive samples = 1.14, IQR 1.07–1.32, range 1.02–2.19), while T-zone lymphomas (TZL) were frequently positive (80%) but with low expression (median MFI ratio in positive samples = 1.19, IQR 1.03–1.46, range 1.02–6.03). Cellular transcript and sPD-L1 were detected in all samples, without differences among immunophenotypes. No correlation between results from different techniques was detected, but sPD-L1 resulted significantly increased in FC-negative lymphomas (p = 0.023).DiscussionPD-L1 molecule is involved in canine lymphoma pathogenesis, with differences among immunophenotypes detected by FC. Specifically, BCL have the highest expression and aggressive T-cell lymphomas the lowest, whereas TZL need further investigations

    Paracoccidioidomicose e infecção pelo virus da imunodeficiência humana

    We present two cases of paracoccidioidorrvycosis, one occurring in an AIDS patient and the other in an HIV infected man. This is the first report of such association. The first patient, which was already followed for HIV infection (group IV-A) presented with high fever and hepatosplenomegaly. Plain X-ray, ultrasound and CT-scan of the abdomen showed solid nodules in the spleen, some of them with calcification. Both the direct smear and the culture of a bone marrow aspiration revealed Paracoccidioides brasiliensis. The patient died of acute disseminated Paracoccidioidomycosis. The second patient, a man anti-HIV seropositive presented with a mass on the right lower abdomen and inguinal region. A biopsy of the mass showed the association of Hodgkin's disease of the mixed cellularity type and paracoccidioidomycosis. With the expanding AIDS epidemic we believe this report emphasizes the need to consider Paracoccidioidomycosis in HIV infected persons in countries where this mycosis is endemic. We also suggest the inclusion of Paracoccidioidomycosis as a potential opportunistic infection in these areas.São apresentados dois casos de paracoccidioidomicose, um em paciente com a síndrome da imunodeficiência adquirida e o outro em paciente com infecção pelo HIV. Trata-se dos primeiros relatos em que esta associação é descrita na literatura. No primeiro, a micose se evidenciou durante o acompanhamento de paciente com AIDS, que passou a apresentar hépato-esplenomegalia e febre elevada. A ecografia, radiografia simples e tomografia computadorizada do abdômen, demonstraram nódulos sólidos, alguns calcificados, no parênquima esplénico. A punção aspirativa da medula óssea confirmou o diagnóstico; o conjunto dos achados caracterizou a forma aguda disseminada da paracoccidioidomicose, a qual levou o paciente ao óbito. No segundo relato, em paciente com infecção pelo HIV, a propósito de investigação de tumoração na região inguinal e fossa ilíaca à direita, constatou-se a associação de doença de Hodgkin, tipo celularidade mista e paracoccidioidomicose. Avalia-se a importância destes relatos frente a expansão da infecção pelo HIV e estima-se que mais casos venham a ser relatados em pacientes com AIDS, procedentes de áreas endêmicas desta micose. Propõe-se a inclusão da paracoccidioidomicose como infecção oportunística potencial em pacientes HIV positivos nestas áreas

    A alfaxalona é um esteroide neuroativo derivado da progesterona com propriedades anestésicas, possuindo poucos efeitos adversos e alterações cardiorrespiratórias mínimas. Ela pode ser usada em cães e gatos de todas as faixas etárias incluindo cadelas gestantes, proporcionando uma alternativa para a indução anestésica. O mecanismo de ação está associado à sua modulação alostérica positiva do receptor ácido gama-aminobutírico tipo A (GABA). Visto que o cenário atual da pandemia do novo coronavírus (COVID-19) induziu à escassez de fármacos anestésicos como o propofol, e dessa forma afetando a Medicina Veterinária, existe a necessidade de introduzir alternativas anestésicas viáveis para indução anestésica em cães e gatos. Visto que que a alfaxalona possa ser um substituto do propofol em procedimentos anestésica e pouco usado no Brasil, objetiva-se com o presente trabalho realizar uma revisão bibliográfica atualizada sobre o uso da alfaxalona como alternativa para indução anestésica em cães e gatos. Concluiu-se que a alfaxalona, similar ao propofol, pode ser usada de forma segura na indução anestésica em cães e gatos, diante do COVID-19, e induz alterações cardiorrespiratórias mínimas

    As abraçadeiras autoestáticas de náilon (AAN) são dispositivos produzidos pelo mesmo material do fio de náilon cirúrgico – a poliamida 6.6, e tem sido estudados como alternativa em procedimentos cirúrgicos na área da Medicina Humana e Veterinária. Visto que as AAN são produzidas pelo mesmo material do fio de náilon cirúrgico, e possuem um sistema autrotravante que garante maior eficácia na realização de hemostasia vascular e na fixação de fragmentos ósseos, o objetivou-se a realizar uma atualização bibliográfica sobre o uso das AAN (poliamida 6.6) como alternativa nas ligaduras vasculares e cerclagem óssea em cães e gatos, permitindo, também, a desmitificação sobre o seu uso na Medicina Veterinária. As AAN de poliamida 6.6 podem ser usadas como método alternativo na ligadura vascular de vasos de médio e grande calibre e na cerclagem óssea em cães e gatos visto que garantiram a segurança mecânica e biológica, porém existe a necessidade de se realizar estudos in vivo da reação tecidual das abraçadeiras a longo prazo. É imprescindível que os cirurgiões que queiram utilizar as AAN sejam submetidos à uma capacitação, permitindo um aprofundamento da técnica cirúrgica

    Macro-scale (biomes) differences in neotropical stream processes and community structure

    The definition of conservation strategies and ecological assessment schemes requires understanding ecosystem patterns over multiple spatial scales. This study aimed to determine if macro-scale structural and functional (processes) patterns associated with stream ecosystems differed among three neotropical biomes (Cerrado, Amazon, Atlantic Forest). We compared the aquatic communities (benthic invertebrates and hyphomycetes) and processes (decomposition rates, primary production and biofilms growth and aquatic hyphomycetes reproduction rates-sporulation) of Cerrado stream sites (neotropical savannah) against those of stream sites in the connecting biomes of the Atlantic Forest and Amazon (rainforests). We expected that, contrary to the biome dependency hypothesis the community structure and processes rates of streams at the biome-scale would not differ significantly, because those ecosystems are strongly influenced by their dense riparian forests, which have a transitional character among the three biomes. Fifty-three stream sites were selected covering a wide range of geographic locations (Table 1), from near the Equator (2° S) in the Amazon, to intermediate latitudes in the Cerrado (12-19° S), and latitudes closer to the tropic of Capricorn in the Atlantic Forest (19º-25° S). We found that: 1) at the abiotic level, the aquatic ecosystems of the three biomes differed, which was mostly explained by large-scale factors such as temperature, precipitation and altitude; 2) functional and structural variables did not behave similarly among biomes: decomposition and sporulation rates showed larger differences among biomes than invertebrate and aquatic hyphomycete assemblages structure; 3) invertebrate assemblages structure differed between the rainforests and Cerrado but not between rainforests (Amazon and Atlantic Forest) whereas aquatic hyphomycetes were similar among all biomes; 4) biofilm growth and algae concentration in biofilms of artificial substrates were highly variable within biomes and not significantly different between biomes. Overall, aquatic ecosystem processes and community structure differed across biomes, being influenced by climatic variables, but the variation is not as pronounced as that described for terrestrial systems. Considering the potential use of these functional and structural indicators in national-wide ecological assessments, our results indicate the need to define different reference values for different biomes, depending on the variable used. The approach followed in this study allowed an integrative analysis and comparison of the stream ecosystems across three tropical biomes, being the first study of this kind. Future studies should try to confirm the patterns evidenced here with more sites from other areas of the three biomes, and especially from the Amazon, which was the least represented biome in our investigation. © 201
