    Development of a device compatible with universal testing machine to perform hole expansion and Erichsen cupping tests

    The material characterization regarding sheet metal formability is usually assessed by the forming limit curve (FLC). The FLC requires specialized and expensive equipment, several samples with different geometries, and can be a very time-consuming procedure for data treatment. Alternatively, the hole expansion test (HET) and the Erichsen Cupping Test (ECT) can be used for routine evaluations of sheet metal mechanical behavior. These formability tests require fewer quantities of material and easy specimen preparation with a fast analysis of results. The HET and ECT procedures provide a proper evaluation of sheet material stretch-flangeability, formability, strength, and ductility. In this work, we developed a low-cost mechanical device capable of performing the HET and ECT tests using a universal testing machine. The equipment is designed to meet the test parameters set by ISO 16630 (HET) and ISO 20482 (ECT) standards. In order to verify its functionality, tests were carried out with the dual-phase steels DP600 and DP780. The corresponding values determined for the hole expansion ratio and the Erichsen index provided reliable results in terms of the accuracy and repeatability of the proposed testing device.publishe

    True absolute determination of photoluminescence quantum yields by coupling multiwavelength thermal lens and photoluminescence spectroscopies

Their determination, by means of methodologies employing reference standard materials, innevitably leads to large uncertainties. In response to this, herein we report for the first time an innovative and elegant methodology, whereby the use of neat solvent/reference material required by thermal lens approaches is eliminated by coupling it to photoluminescence spectroscopy, allowing for the discrimination between materials with similar photoluminescence quantum yields. To achieve that, both radiative and non-radiative transitions are simultaneously measured by means of a photoluminescence spectrometer coupled to a multiwavelength thermal lens spectroscopy setup in a mode-mismatched dual-beam configuration, respectively. The absorption factor independent ratio of the thermal lens and photoluminescence signals can then be used to determine the fluorescence quantum yield both accurately and precisely. We validated our reported method by means of rhodamine 6G and further applied in three novel structurally related diketopyrrolopyrrole based materials to, contrary to results obtained by other methods, unveil significant differences in their photoluminescence quantum yields.Peer reviewe

    Institutional factors governing the deployment of remote experiments: lessons from the rexnet project

    Remote labs offer many unique advantages to students as they provide opportunities to access experiments and learning scenarios that would be otherwise unavailable. At the same time, however, these opportunities introduce real challenges to the institutions hosting the remote labs. This paper draws on the experiences of the REXNET project consortium to expose a number of these issues as a means of furthering the debate on the value of remote labs and the best practices in deploying them. The paper presents a brief outline of the various types of remote lab scenarios that might be deployed. It then describes the key human and technological actors that have an interest in or are intrinsic to a remote lab instance, with a description of the role of each actor and their interest. Some relationships between these various actors are then discussed with some factors that might influence those relationships. Finally some general issues are briefly described

    Association between Chronotype and Nutritional, Clinical and Sociobehavioral Characteristics of Adults Assisted by a Public Health Care System in Brazil

    Chronotype (CT) has been associated with predisposition to chronic noncommunicable diseases (CNCDs), such as diabetes mellitus and obesity. However, the effects of CT on individuals assisted by public health systems (PHSs) in middle-up economies are still poorly explored. The objective of this study was to evaluate the relationship between CT and clinical, sociobehavioral and nutritional aspects in adults assisted by a PHS in Brazil. This is a population-based cross-sectional study. The sample consisted of 380 individuals, selected through probabilistic sampling by clusters, in all health units in a city of approximately 100 thousand inhabitants. Data collection was performed during home visits, by means of general and nutritional interviews, anthropometric measurements and the Morningness–Eveningness Questionnaire (MEQ). Statistical analysis comprised chi-square test and principal component analysis (CPA) followed by Fisher’s discriminant analysis to determine aspects associated with each CT (morning, evening or intermediate). With the aim of explaining the variation in the CT scores, the consumption of micronutrients (corrected to the total energy intake) and other individual and sociodemographic variables were used as explanatory factors in the adjustment of a linear regression model. The morning group was characterized by older men, with less than eight years of schooling, with low body mass index (BMI) and with low intake of omega-6, omega-3, sodium, zinc, thiamine, pyridoxine and niacin. The evening group, on the other hand, was composed of younger individuals, with a high consumption of these same nutrients, with high BMI and a higher frequency of heart diseases (p < 0.05). It was concluded that most morning CT individuals were elderly thin males with lower consumption of omega-6 and -3, sodium, zinc, thiamine, pyridoxine and niacin, whereas evening individuals were younger, had higher BMI and had higher consumption of the studied micronutrients. The identification of circadian and behavioral risk groups can help to provide preventive and multidisciplinary health promotion measures

    Fibronectin rescues estrogen receptor α from lysosomal degradation in breast cancer cells

    Estrogen receptor α (ERα) is expressed in tissues as diverse as brains and mammary glands. In breast cancer, ERα is a key regulator of tumor progression. Therefore, understanding what activates ERα is critical for cancer treatment in particular and cell biology in general. Using biochemical approaches and superresolution microscopy, we show that estrogen drives membrane ERα into endosomes in breast cancer cells and that its fate is determined by the presence of fibronectin (FN) in the extracellular matrix; it is trafficked to lysosomes in the absence of FN and avoids the lysosomal compartment in its presence. In this context, FN prolongs ERα half-life and strengthens its transcriptional activity. We show that ERα is associated with β1-integrin at the membrane, and this integrin follows the same endocytosis and subcellular trafficking pathway triggered by estrogen. Moreover, ERα+ vesicles are present within human breast tissues, and colocalization with β1-integrin is detected primarily in tumors.     Most of the computational biology analysis is made comparing genomic features. The nucleotide and amino acid sequence alignments are frequently used in gene function identification and genome comparison. Despite its widespread use, there are limitations in their analysis capabilities that need to be considered but are often overlooked or unknown by many researchers. This paper presents a gene based whole genome comparison toolkit which can be used not only as an alternative and more robust way to compare a set of whole genomes, but, also, to understand the tradeoff of the use of sequence local alignment in this kind of comparison. A study case was performed considering fifteen whole genomes of the Xanthomonas genus. The results were compared with the 16S rRNA-processing protein RimM phylogeny and some thresholds for the use of sequence alignments in this kind of analysis were discussed.Grande parte das analises realizadas na biologia computacional ? feita comparando caracter?sticas gen?micas. Apesar de seu uso generalizado, h? limita??es em suas capacidades de analise que precisam ser consideradas, mas s?o frequentemente negligenciadas ou desconhecidas por muitos pesquisadores. Este artigo apresenta um conjunto de ferramentas de compara??o de genomas completos baseado em genes que pode ser usado n?o somente como uma maneira alternativa e mais robusta de comparar um conjunto de genomas completos, mas tamb?m para entender as vantagens e desvantagens do uso do alinhamento local de sequencias neste tipo de compara??o. Um estudo de caso foi realizado considerando quinze genomas completos do g?nero Xanthomonas. Os resultados foram comparados com a filogenia produzida utilizando a prote?na 16S rRNA-processing protein RimM e alguns limiares para o uso de alinhamentos de sequencias neste tipo de analise foram discutidos
