    Open ICT tools project

    The paper will introduce a project titled the ‘Open ICT Tools’ which aims to explore and trial out ICT tools to facilitate a global collaborative and secured engagement with external business and community partners. The challenge is to facilitate a communication and multimedia data exchange between Northumbria University and participating external educational and business organisations without compromising the security of either Northumbria University IT infrastructure or that of the partner organisations. This is one of eight projects funded by the JISC infoNet from across the country under its Trialling of Online Collaborative Tools for Business and Community Engagement programme. The Open ICT Tools project is directly connected with the Global Studio, an innovative model of research informed teaching and learning. The Global Studio is a cross-institutional collaboration between Northumbria University and international universities based in the USA, Australia, UK and Korea as well as industry partners such as Intel, Motorola and Inverness Medical. The Global Studio was initiated by the School of Design three years ago and since then it included nearly 300 students from six international universities such as Hong-ik in Korea and RMIT in Australia. The aim of the Global Studio is to equip design students with skills for working in globally networked organisations particularly development of skills in intercultural communication and collaboration. To achieve this aim, students from the participating universities work together on industry led projects. However, attempting to use existing university technical infrastructure has been problematic. A particular problem relates to software licence agreements, which limit use of certain software and/or the virtual learning environment to a particular institution’s students. Attempts to use open source software has not been straightforward, for example using Skype for synchronous cross – institutional communication has often failed as students cannot log into a university’s technical infrastructure, begin a Skype based dialogue with students in another institution and simultaneously view a PowerPoint presentation. Online file sharing has often failed, primarily due to the file size. Therefore the aim of the Global Studio project is to build on the excellent ICT infrastructure at Northumbria and to identify and trial a diverse range of collaborative Information Communication Technology (ICT) tools that: (a) could support engagement between the university and its external collaborative business and community partners and (b) be embedded with the current university IT infrastructure. This paper explores how various ICTs are being trialled in the Global Studio to facilitate information and data exchange between students, teachers and industry partners and how this enables/constrains collaboration. The paper will explore the technologies that have been chosen as well as the rationale underpinning their choice

    Carrying capacity of traditional farming in South East England: A case study

    Traditional farming in South East (SE) England is presented as a highly-evolved form of sustainable farming. The carrying capacity of traditional farming on a 2.75 ha family smallholding in SE England is assessed from production data recorded over a period of 8 years. The key elements of the farming system were mixed farming (livestock, dairy, arable and horticultural), self-sufficiency in terms of inputs and organic principles. Ten types of food were produced with the aim to comprise all the elements of a balanced diet. The holding and farming system are described and an analysis of the food produced is presented, in terms of weight and energy content, for the years 2010 to 2017. An average carrying capacity of 0.64 people ha-1 was demonstrated on the basis of food energy content alone. Carrying capacity increased to 1.09 people ha-1 when production was re-proportioned to align with the UK Government’s currently recommended balanced diet. The latter figure is similar to carrying capacity estimates, derived from national statistics, for the UK’s total farmland in the middle part of the 20th Century but significantly lower than theoretical predictions of national carrying capacity

    [EN] The aim of this work was to assess the relation between carbohydrate levels and flower and fruit production, as well as the role of carbohydrates on CO(2) fixation activity, by analysis of leaves, twigs and roots from the alternate bearing 'Salustiana' cultivar of sweet orange (Citrus sinensis [L.] Osbeck). A heavy crop load (on year) did not affect photosynthesis activity when compared to non-fruiting trees (off year). Fruiting trees accumulated most of the fixed carbon in mature fruits, whilst no accumulation was observed in roots before harvest. Non-fruiting trees transported part of the fixed carbon to the roots and mobilize it for growth processes and, at the end of the season (December), store it as reserves. Reserve carbohydrates accumulation in leaves started by early December for both tree types, showing the same levels in on and off trees until spring bud sprouting. A heavy flowering after an off year caused the rapid mobilization of the stored reserves, which were exhausted at full bloom. We found no evidence on carbon fixation regulation by either fruit demand or carbohydrate levels in leaves.     Peri-conceptual and mid-pregnancy drinking:a cross-sectional assessment in two Scottish health board areas using a 7-day Retrospective Diary

    Aims: The aim of this study was to evaluate the use of a 7-day Retrospective Diary to assess peri-conceptual and mid-pregnancy alcohol consumption. Background: Alcohol consumption among women has increased significantly and is of international concern. Heavy episodic (‘binge’) drinking is commonplace and is associated with unintended pregnancy. Pre-pregnancy drinking is strongly associated with continued drinking in pregnancy. Routine antenatal assessment of alcohol history and current drinking is variable; potentially harmful peri-conceptual drinking may be missed if a woman reports low or no drinking during pregnancy. Design: Cross-sectional study (n=510) in two Scottish health board areas. Methods: Face-to-face Retrospective Diary administration from February to June 2015 assessing alcohol consumption in peri-conceptual and mid-pregnancy periods. Women were recruited at the mid-pregnancy ultrasound clinic. Results: Of 510 women, 470 (92·0%) drank alcohol before their pregnancy; 187 (39·9%) drank every week. Retrospective assessment of peri-conceptual consumption identified heavy episodic drinking (more than six units on one occasion) in 52·2% (n=266); 19·6% (n=100) reported drinking more than 14 units per week, mostly at the weekend; ‘mixing’ of drinks was associated with significantly higher consumption. While consumption tailed off following pregnancy recognition, 5·5% (n=28) still exceeded the recommended daily two-unit limit in pregnancy. Multivariable logistic regression identified that women who ‘binged’ peri-conceptually were 3·2 times more likely to do this. Conclusion: Statistically significant peri-conceptual consumption levels suggest a substantial proportion of alcohol-exposed pregnancies before pregnancy recognition. Not taking a detailed alcohol history, including patterns of consumption, will result in under-detection of alcohol-exposed pregnancies. The Retrospective Diary offers practitioners a detailed way of enquiring about alcohol history for this population

    Adaptor SKAP-55 Binds p21ras Activating Exchange Factor RasGRP1 and Negatively Regulates the p21ras-ERK Pathway in T-Cells

    While the adaptor SKAP-55 mediates LFA-1 adhesion on T-cells, it is not known whether the adaptor regulates other aspects of signaling. SKAP-55 could potentially act as a node to coordinate the modulation of adhesion with downstream signaling. In this regard, the GTPase p21ras and the extracellular signal-regulated kinase (ERK) pathway play central roles in T-cell function. In this study, we report that SKAP-55 has opposing effects on adhesion and the activation of the p21ras -ERK pathway in T-cells. SKAP-55 deficient primary T-cells showed a defect in LFA-1 adhesion concurrent with the hyper-activation of the ERK pathway relative to wild-type cells. RNAi knock down (KD) of SKAP-55 in T-cell lines also showed an increase in p21ras activation, while over-expression of SKAP-55 inhibited activation of ERK and its transcriptional target ELK. Three observations implicated the p21ras activating exchange factor RasGRP1 in the process. Firstly, SKAP-55 bound to RasGRP1 via its C-terminus, while secondly, the loss of binding abrogated SKAP-55 inhibition of ERK and ELK activation. Thirdly, SKAP-55−/− primary T-cells showed an increased presence of RasGRP1 in the trans-Golgi network (TGN) following TCR activation, the site where p21ras becomes activated. Our findings indicate that SKAP-55 has a dual role in regulating p21ras-ERK pathway via RasGRP1, as a possible mechanism to restrict activation during T-cell adhesion

    Encoding optical control in LCK kinase to quantitatively investigate its activity in live cells.

    LCK is a tyrosine kinase that is essential for initiating T-cell antigen receptor (TCR) signaling. A complete understanding of LCK function is constrained by a paucity of methods to quantitatively study its function within live cells. To address this limitation, we generated LCK*, in which a key active-site lysine is replaced by a photocaged equivalent, using genetic code expansion. This strategy enabled fine temporal and spatial control over kinase activity, thus allowing us to quantify phosphorylation kinetics in situ using biochemical and imaging approaches. We find that autophosphorylation of the LCK active-site loop is indispensable for its catalytic activity and that LCK can stimulate its own activation by adopting a more open conformation, which can be modulated by point mutations. We then show that CD4 and CD8, T-cell coreceptors, can enhance LCK activity, thereby helping to explain their effect in physiological TCR signaling.
