Modulation of ecdysal cyst and toxin dynamics of two Alexandrium (Dinophyceae) species under small-scale turbulence

Some dinoflagellate species have shown different physiological responses to certain turbulent conditions. Here we investigate how two levels of turbulent kinetic energy dissipation rates (epsilon = 0.4 and 27 cm(2) s(-3)) affect the PSP toxins and ecdysal cyst dynamics of two bloom forming species, Alexandrium minutum and A. catenella. The most striking responses were observed at the high epsilon generated by an orbital shaker. In the cultures of the two species shaken for more than 4 days, the cellular GTX(1+4) toxin contents were significantly lower than in the still control cultures. In A. minutum this trend was also observed in the C(1+2) toxin content. For the two species, inhibition of ecdysal cyst production occurred during the period of exposure of the cultures to stirring (4 or more days) at any time during their growth curve. Recovery of cyst abundances was always observed when turbulence stopped. When shaking persisted for more than 4 days, the net growth rate significantly decreased in A. minutum (from 0.25 +/- 0.01 day(-1) to 0.19 +/- 0.02 day(-1)) and the final cell numbers were lower (ca. 55.4%) than in the still control cultures. In A. catenella, the net growth rate was not markedly modified by turbulence although under long exposure to shaking, the cultures entered earlier in the stationary phase and the final cell numbers were significantly lower (ca. 23%) than in the control flasks. The described responses were not observed in the experiments performed at the low turbulence intensities with an orbital grid system, where the population development was favoured. In those conditions, cells appeared to escape from the zone of the influence of the grids and concentrated in calmer thin layers either at the top or at the bottom of the containers. This ecophysiological study provides new evidences about the sensitivity to high levels of small-scale turbulence by two life cycle related processes, toxin production and encystment, in dinoflagellates. This can contribute to the understanding of the dynamics of those organisms in nature

Formal Total Synthesis of the Algal Toxin (−)-Polycavernoside A

    No full text
A concise and largely catalysis-based approach to the potent algal toxin polycavernoside A (1) is described that intercepts a late-stage intermediate of a previous total synthesis; from there on, this challenging target can be reached in a small number of steps. Key to success was a sequence of a molybdenum-catalyzed ring-closing alkyne metathesis (RCAM) reaction to forge the macrocyclic frame, followed by a gold-catalyzed and strictly regioselective transannular hydroalkoxylation of the resulting cycloalkyne that allows the intricate oxygenation pattern of the macrolactone ring of 1 to be properly set. The required cyclization precursor 5 was assembled by the arguably most advanced fragment coupling process based on an Evans–Tishchenko redox esterification known to date, which was optimized to the extent that the precious coupling partners could be used in an almost equimolar ratio (6/7 1:1.3). The preparation of these building blocks features, inter alia, the power of the Sc(OTf)3-catalyzed Leighton crotylation as well as the superb selectivities of alkene cross metathesis, asymmetric keto-ester hydrogenation, and the Jacobsen epoxidation/epoxide resolution technologies

The work in the workshop is surely one of the most revealing moments during the astistic process, and in this case sculptural. The materials, the tools, the machines ... they all speak for themselves. It is our duty as artists to recognize this merit and take advantage of it for the benefit of art. But ... what exactly is this intrusion allowed to the machines? All of them work to make our creations possible. Beneath their dust-and-rust housings hides all their magic. Nuts, washers, toothed wheels, pulleys ... and in many occasions pieces specially built for the occasion. And of course, all of them placed meticulously, respecting the order, pressure or distance to work in unison, as if it were a single piece. The work intends for the viewer to discover both the pieces and their operation within a system of gears. The purpose is to show the logic and genius of these procedures using the machines of my father, a blacksmith by profession, who has transmitted to me the value and beauty of all these mechanisms

Cell cycle and cell mortality of Alexandrium minutum (Dinophyceae) under small-scale turbulence conditions

Decreased net population growth rates and cellular abundances have been observed in dinoflagellate species exposed to small-scale turbulence. Here, we investigated whether these effects were caused by alterations in the cell cycle and/or by cell mortality and, in turn, whether these two mechanisms depended on the duration of exposure to turbulence. The study was conducted on the toxic dinoflagellate Alexandrium minutum Halim, with the same experimental design and setup used in previous studies to allow direct comparison among results. A combination of microscopy and Coulter Counter measurements allowed us to detect cell mortality, based on the biovolume of broken cells and thecae. The turbulence applied during the exponential growth phase caused an immediate transitory arrest in the G2/M phase, but significant mortality did not occur. This finding suggests that high intensities of small-scale turbulence can alter the cell division, likely affecting the correct chromosome segregation during the dinomitosis. When shaking persisted for >4 d, mortality signals and presence of anomalously swollen cells appeared, hinting at the activation of mechanisms that induce programmed cell death. Our study suggests that the sensitivity of dinoflagellates to turbulence may drive these organisms to find the most favorable (calm) conditions to complete their division cycle.Postprin

This project is an exercise built from a critical positioning against the new creative practices from the culture industry. Defending art as an experience; sensitivity and compromise as a driving force. Our current society gives priority to rational knowledge in opposition to sensitive knowledge. The lack of prominence and analysis of this last one has considerable consequences in artistic practices, which is the field this proposal is focusing on. We understand culture as a connecting link in society, in which culture industry has a relevant role as a generator. Putting effort in working the quality of the incentives produced by it requires constant awarness and judgement. To find out how this affects us it¿s a need for being able to build a context according to our interests. This is substancial for creative workers, then we feed ourselves mainly from the emotional. In `Aesthetic Theory¿, Adorno writes: `Artworks come out from the empirical world, building a new one with own essence and in opposition to the first one¿. It¿s a need to base the praxis in sensitive experience to make effective a plastical content coming out from a personal and critical vision of reality.Llaveria Caselles, A. (2018). Barcelona: Espacio Literario, Espacio Urbano en la Novela Catalana Femenina del Siglo XX

This dissertation analyses, through a rhetorical framework and a literary approach, texts written in Catalan and Castilian by four Catalan female writers (Dolors Monserdà, Maria Aurèlia Capmany, Esther Tusquets, Monserrat Roig ), whose works cover from 1900 to the 1980. Utilizing this urban feminine literature, it discusses the historical-geographical vision about the changes in Catalan society during the twentieth century with its consequences for the urban space, especially the space occupied by women. It is also established that Barcelona’s recovery and literary vindication by women has been done through the written text, as literary affirmation and as a matter of conscience in which the city could not be summed up as a backdrop, but rather as an active part of a literary creation, active in the double sense, as a socio-historical space in the novel and as characteristic of their works. The primary purpose of this dissertation is to demonstrate that the use of the city as a setting for the novels determines and characterizes those female writers’ texts. Consequently, these writings are literary material relevant and essential to the understanding of the Barcelonian women’s space. However their use of space is not arbitrary, on the contrary it corresponds to a social order established by the patriarchy where the relation of women to the world is embodied in the intentional and socially restricted space and movements of their bodies. The theoretical perspectives of this study are based on Montserrat Roig’s feminist urban space theories. Her theory advocates the right to individuality, denouncing the patriarchal and hierarchical social system present in gendered space from the outside male world to the domestic feminized space. I also turn to the writings of Maria Aurèlia Capmany, who addresses cultural aspects of women’s roles revealing a purposive controlled patriarchal society according to a historical-geographical analysis. This study of texts permits a new reading of the Catalan capital and demonstrates that Catalan women writers have consciously willed to give birth to a new history of the city: the history of women as protagonist citizens, producers, reproducers, and consumers of the space represented by the Catalan capita
