    Wie wirken sich Geburtsverhalten und Wurfqualität auf die Aufzuchtleistung aus?

    In dieser Untersuchung wurde der Effekt des Geburtsverhaltens und der Wurfqualität auf die Aufzuchtleistung in 132 Würfen von Deutsche Landrasse Sauen geprüft. Das Geburtsverhalten wurde mit einem Notensystem (sehr gut, gut, mittel, schlecht) bewertet. Die Wurfqualität wurde anhand der Vitalität und Homogenität der Ferkel zur Geburt erfasst. Von der Geburt bis zum Absetzen wurden Daten verschiedener Leistungsmerkmale gesammelt. Unterschiede zwischen Würfen mit verschiedenen Noten wurden mittels ANOVA und Tukey-Test untersucht und der Effekt des Geburtsverhaltens und der Wurfqualität auf die Aufzuchtleistung mittels eines linearen Modells (GLM) analysiert. Der Einfluss der Wurfqualität auf die Anzahl aufgezogener Ferkel, das Wurfgewicht am 35. Lebenstag und die prozentualen Saugferkelverluste war statistisch signifikant. Darüber hinaus zeigten Würfe mit hoher und niedriger Qualität einen signifikanten Unterschied bezüglich der Anzahl lebend und tot geborener Ferkel und des durchschnittlichen Geburtsgewichts der Ferkel

    An improved folate stable isotope dilution assay of unexploited food sources from Brazil

    Brazil has a diverse plant community, including underutilized non-conventional food crops (PANCs), which have the potential to be a rich source of food and contribute to food security. For assessing the folate content in a range of Brazilian PANCs, we extended the validation of an existing stable isotope dilution assay (SIDA) for the stably 13C-labelled 10-formyl-Pte[13C5]Glu (10-CHO-Pte[13C5]Glu). The SIDA method with an enzymatic treatment, purification step, and an LC–MS/MS measurement was validated regarding linearity, precision, LoD/LoQ, and recovery for 10-CHO-PteGlu. After successful validation, the study of some underutilized Brazilian non-conventional fruits and leaves from the São Paulo State University campus revealed them as an important source of folates. It provided the first insights into the folate content of unexploited food sources from Brazil. Pequi had the highest folate content among the fruits studied, with mean values of 333 μg/100 g based on fresh weight (FW). The analysis also shows that different cultivars of fruit or fruits from different growing locations have a high variability in folate content or other nutritional factors. In most fruits, the main vitamer was 5-CH3-H4folate, but jenipapo and taioba showed the highest content of 10-CHO-PteGlu with 28.22 μg/100 g (FW) in jenipapo peel and 75.64 μg/100 g (FW) in the taioba leaves. Thus, this study also provides results on the importance of the folate vitamer 10-CHO-PteGlu contributing to the total folate content

    Virus-Receptor Mediated Transduction of Dendritic Cells by Lentiviruses Enveloped with Glycoproteins Derived from Semliki Forest Virus

    Lentiviruses have recently attracted considerable interest for their potential as a genetic modification tool for dendritic cells (DCs). In this study, we explore the ability of lentiviruses enveloped with alphaviral envelope glycoproteins derived from Semliki Forest virus (SFV) to mediate transduction of DCs. We found that SFV glycoprotein (SFV-G)-pseudotyped lentiviruses use C-type lectins (DC-SIGN and L-SIGN) as attachment factors for transduction of DCs. Importantly, SFV-G pseudotypes appear to have enhanced transduction towards C-type lectin-expressing cells when produced under conditions limiting glycosylation to simple high-mannose, N-linked glycans. These results, in addition to the natural DC tropism of SFV-G, offer evidence to support the use of SFV-G-bearing lentiviruses to genetically modify DCs for the study of DC biology and DC-based immunotherapy

    Search of the Orion spur for continuous gravitational waves using a loosely coherent algorithm on data from LIGO interferometers

    We report results of a wideband search for periodic gravitational waves from isolated neutron stars within the Orion spur towards both the inner and outer regions of our Galaxy. As gravitational waves interact very weakly with matter, the search is unimpeded by dust and concentrations of stars. One search disk (A) is 6.87° in diameter and centered on 20h10m54.71s+33°33′25.29′′, and the other (B) is 7.45° in diameter and centered on 8h35m20.61s-46°49′25.151′′. We explored the frequency range of 50-1500 Hz and frequency derivative from 0 to -5×10-9 Hz/s. A multistage, loosely coherent search program allowed probing more deeply than before in these two regions, while increasing coherence length with every stage. Rigorous follow-up parameters have winnowed the initial coincidence set to only 70 candidates, to be examined manually. None of those 70 candidates proved to be consistent with an isolated gravitational-wave emitter, and 95% confidence level upper limits were placed on continuous-wave strain amplitudes. Near 169 Hz we achieve our lowest 95% C.L. upper limit on the worst-case linearly polarized strain amplitude h0 of 6.3×10-25, while at the high end of our frequency range we achieve a worst-case upper limit of 3.4×10-24 for all polarizations and sky locations. © 2016 American Physical Society

    First low frequency all-sky search for continuous gravitational wave signals

    In this paper we present the results of the first low frequency all-sky search of continuous gravitational wave signals conducted on Virgo VSR2 and VSR4 data. The search covered the full sky, a frequency range between 20 and 128 Hz with a range of spin-down between −1.0×10−10 and +1.5×10−11  Hz/s, and was based on a hierarchical approach. The starting point was a set of short fast Fourier transforms, of length 8192 s, built from the calibrated strain data. Aggressive data cleaning, in both the time and frequency domains, has been done in order to remove, as much as possible, the effect of disturbances of instrumental origin. On each data set a number of candidates has been selected, using the FrequencyHough transform in an incoherent step. Only coincident candidates among VSR2 and VSR4 have been examined in order to strongly reduce the false alarm probability, and the most significant candidates have been selected. The criteria we have used for candidate selection and for the coincidence step greatly reduce the harmful effect of large instrumental artifacts. Selected candidates have been subject to a follow-up by constructing a new set of longer fast Fourier transforms followed by a further incoherent analysis, still based on the FrequencyHough transform. No evidence for continuous gravitational wave signals was found, and therefore we have set a population-based joint VSR2-VSR4 90% confidence level upper limit on the dimensionless gravitational wave strain in the frequency range between 20 and 128 Hz. This is the first all-sky search for continuous gravitational waves conducted, on data of ground-based interferometric detectors, at frequencies below 50 Hz. We set upper limits in the range between about 10−24 and 2×10−23 at most frequencies. Our upper limits on signal strain show an improvement of up to a factor of ∼2 with respect to the results of previous all-sky searches at frequencies below 80 H

    Microplastic in the water cycle : sampling, sample preparation, analyses, occurrence and assessment

    Gedruckt erschienen im Universitätsverlag der TU Berlin, ISBN 978-3-7983-3162-4Das interdisziplinäre Forschungsprojekt MiWa widmete sich grundlegenden Fragestellungen zur Analytik und Wirkung von Mikroplastik-Partikeln im Wasserkreislauf. Es wurden Methoden der Umweltprobennahme, der Probenaufbereitung und verschiedene Detektionsverfahren zur Charakterisierung und Quantifizierung von Mikroplastik intensiv untersucht, miteinander verglichen und weiterentwickelt. Öko- und humantoxikologische Untersuchungen dienten dem Zweck, die potenziell von Mikroplastik ausgehende Gefährdung für die aquatische Umwelt und den Menschen zu analysieren und zu bewerten. Eine Harmonisierung und Standardisierung von Methoden der Probennahme, Probenaufbereitung und Mikroplastik-Detektion sind trotz der erheblichen Fortschritte derzeit nur teilweise möglich. Die ökotoxikologischen Studien zeigen zwar die Aufnahme von Mikroplastik-Partikeln durch einige Organismen, jedoch konnte bisher keine schädigende Wirkung nachgewiesen werden. Dabei wurden für eine Auswahl aquatischer Modellspezies sowohl Szenarien direkter als auch indirekter Exposition innerhalb einer Nahrungskette betrachtet. Interaktionen mit menschlichen Modellzellen wurden bislang nur bei Mikroplastik-Partikeln mit Größen weit unterhalb von 1 µm (also Nanoplastik) beobachtet. Eine umfassende Bewertung ist bislang nicht möglich.The interdisciplinary research project MiWa focused on principle knowledge gaps of analytical detection and effects of microplastic in fresh water cycles. Methods for environmental sampling, sample preparation and different analytical identification and quantification were intensively investigated, compared and further developed. Toxicological studies were conducted to assess potential risks of microplastic particles towards the environment and human health. Harmonization and standardization are still only partially possible despite various improvements. The eco-toxicological experiments confirmed the intake of microplastic particles by different organisms but no hazardous effects could be found. Both direct ingestions and indirect exposition within food webs were tested. Interactions with exemplary human cells were only observed for particle sizes far below 1 µm (thus nanoplastic).
