    Introduction: Streetlife - the Shifting Sociologies of the Street

    The street has long been a key laboratory for studies of social life, from the roots of urban sociology in the ethnographies of the Chicago School to the diverse range of contemporary studies which consider the performative, affective and non-representational nature of street etiquette and encounter. For all this, the street remains only loosely defined in many studies, and sometimes disappears from view entirely, with social action often privileged over material and environmental context. This Special Issue is intended as a spur to take the street more seriously in contemporary sociology, and explores the importance of the street as a site, scale and field for sociological research. Recognising that the street is both contradictory and complex, in the Introduction to this Issue we draw out emerging themes in the shifting sociologies of the street by highlighting the specific contribution inter-disciplinary work can make to our understanding of streets as distinctive but contested social spaces

    The same neuron may play different functional roles in the neural circuits to which it belongs. For example, neurons in the Tritonia pedal ganglia may participate in variable phases of the swim motor rhythms [1]. While such neuronal functional variability is likely to play a major role the delivery of the functionality of neural systems, it is difficult to study it in most nervous systems. We work on the pyloric rhythm network of the crustacean stomatogastric ganglion (STG) [2]. Typically network models of the STG treat neurons of the same functional type as a single model neuron (e.g. PD neurons), assuming the same conductance parameters for these neurons and implying their synchronous firing [3, 4]. However, simultaneous recording of PD neurons shows differences between the timings of spikes of these neurons. This may indicate functional variability of these neurons. Here we modelled separately the two PD neurons of the STG in a multi-neuron model of the pyloric network. Our neuron models comply with known correlations between conductance parameters of ionic currents. Our results reproduce the experimental finding of increasing spike time distance between spikes originating from the two model PD neurons during their synchronised burst phase. The PD neuron with the larger calcium conductance generates its spikes before the other PD neuron. Larger potassium conductance values in the follower neuron imply longer delays between spikes, see Fig. 17.Neuromodulators change the conductance parameters of neurons and maintain the ratios of these parameters [5]. Our results show that such changes may shift the individual contribution of two PD neurons to the PD-phase of the pyloric rhythm altering their functionality within this rhythm. Our work paves the way towards an accessible experimental and computational framework for the analysis of the mechanisms and impact of functional variability of neurons within the neural circuits to which they belong

    COVID-19 symptoms at hospital admission vary with age and sex: results from the ISARIC prospective multinational observational study

    Background: The ISARIC prospective multinational observational study is the largest cohort of hospitalized patients with COVID-19. We present relationships of age, sex, and nationality to presenting symptoms. Methods: International, prospective observational study of 60 109 hospitalized symptomatic patients with laboratory-confirmed COVID-19 recruited from 43 countries between 30 January and 3 August 2020. Logistic regression was performed to evaluate relationships of age and sex to published COVID-19 case definitions and the most commonly reported symptoms. Results: ‘Typical’ symptoms of fever (69%), cough (68%) and shortness of breath (66%) were the most commonly reported. 92% of patients experienced at least one of these. Prevalence of typical symptoms was greatest in 30- to 60-year-olds (respectively 80, 79, 69%; at least one 95%). They were reported less frequently in children (≀ 18 years: 69, 48, 23; 85%), older adults (≄ 70 years: 61, 62, 65; 90%), and women (66, 66, 64; 90%; vs. men 71, 70, 67; 93%, each P < 0.001). The most common atypical presentations under 60 years of age were nausea and vomiting and abdominal pain, and over 60 years was confusion. Regression models showed significant differences in symptoms with sex, age and country. Interpretation: This international collaboration has allowed us to report reliable symptom data from the largest cohort of patients admitted to hospital with COVID-19. Adults over 60 and children admitted to hospital with COVID-19 are less likely to present with typical symptoms. Nausea and vomiting are common atypical presentations under 30 years. Confusion is a frequent atypical presentation of COVID-19 in adults over 60 years. Women are less likely to experience typical symptoms than men

    Gold nanoparticle probe-based gene expression analysis with unamplified total human RNA

    Microarray-based gene expression analysis plays a pivotal role in modern biology and is poised to enter the field of molecular diagnostics. Current microarray-based gene expression systems typically require enzymatic conversion of mRNA into labeled cDNA or cRNA. Conversion to cRNA involves a target amplification step that overcomes the low sensitivity associated with commonly used fluorescent detection methods. Herein, we present a novel enzyme-free, microarray-based gene expression system that uses unamplified total human RNA sample as the target nucleic acid. The detection of microarray-bound RNA molecules is accomplished by targeting the poly-A tail with an oligo-dT(20) modified gold nanoparticle probe, signal amplification by autometallography, and subsequent measurement of nanoparticle-mediated light scattering. The high sensitivity afforded by the nanoparticle probes allows differential gene expression from as little as 0.5 ÎŒg unamplified total human RNA in a 2 h hybridization without the need for elaborate sample labeling steps

    Areas of importance for seabirds tracked from French southern territories, and recommendations for conservation

    Seabirds are increasingly threatened worldwide, with population declines for many species that are faster than in any other group of birds. Here the Important Bird Area (IBA) criteria recommended by BirdLife International were applied to a large tracking dataset collected from a range of seabirds, to identify areas of importance at an ocean basin scale. Key areas were identified using tracks obtained from both the breeding and non-breeding periods of 10 species that have different habitat requirements. These species range in their IUCN threat status from Least Concern to Critically Endangered. An evaluation of spatial overlap between the key areas for these species and the jurisdiction of Regional Fisheries Management Organisations (RFMOs), national Exclusive Economic Zones (EEZs) and other stakeholder bodies highlighted the major importance of the French EEZs (around Crozet, Kerguelen and Amsterdam Islands) for seabird conservation. The majority of the candidate marine IBAs that were identified were located in the High Seas, where Marine Protected Areas cannot easily be designated under existing international agreements, except in the Commission for the Conservation of Antarctic Marine Living Resources Convention Area. In the short term, it seems that only fisheries regulations (through international agreements) can bring about efficient protection for seabirds in the High Seas. The BirdLife IBA approach, although sensitive to heterogeneity in the data (species selected, inclusion of different life stages, years etc.), proved valuable for selecting important areas corresponding to large-scale oceanographic structures that are considered to be key foraging habitats for many species

    Common architecture of nuclear receptor heterodimers on DNA direct repeat elements with different spacings

    Nuclear hormone receptors (NHRs) control numerous physiological processes through the regulation of gene expression. The present study provides a structural basis for understanding the role of DNA in the spatial organization of NHR heterodimers in complexes with coactivators such as Med1 and SRC-1. We have used SAXS, SANS and FRET to determine the solution structures of three heterodimer NHR complexes (RXR-RAR, PPAR-RXR and RXR-VDR) coupled with the NHR interacting domains of coactivators bound to their cognate direct repeat elements. The structures show an extended asymmetric shape and point to the important role played by the hinge domains in establishing and maintaining the integrity of the structures. The results reveal two additional features: the conserved position of the ligand-binding domains at the 5' ends of the target DNAs and the binding of only one coactivator molecule per heterodimer, to RXR's partner
