    Facies characteristics and diversity in carbonate eolianites

    Carbonate eolian dunes can form huge sand bodies along the coasts but are seldom described in the pre-Quaternary record. The study of more than 600 thin-sections collected in present-day, Holocene and Pleistocene dunes from Sardinia, Crete, Cyprus, Tunisia, Morocco, Australia and Baja California confirms that these deposits can be easily misinterpreted as shallow marine at core or thin-section scale. The classical eolian criteria (fine-grained and well-sorted sands) are exceptional in carbonate dunes because the diversity of shapes and densities of carbonate particles lowers the critical shear velocity of the sediment thus blurring the sedimentary structures. Wind carbonate deposits are mainly heterogeneous in size and often coarse-grained. The paucity of eolianites in the pre-Quaternary record could be due to misinterpretation of these deposits. The recognition should be based on converging sedimentological and stratigraphic elements at core scale, and diagenetic (vadose diagenesis, pedogenetic imprints) and petrographical (grain verticalization, scarcity of micritic envelopes, broken and/or reworked foraminifera) clues in thin-section.     Symptom-based stratification of patients with primary Sjögren's syndrome: multi-dimensional characterisation of international observational cohorts and reanalyses of randomised clinical trials

    Background Heterogeneity is a major obstacle to developing effective treatments for patients with primary Sjögren's syndrome. We aimed to develop a robust method for stratification, exploiting heterogeneity in patient-reported symptoms, and to relate these differences to pathobiology and therapeutic response. Methods We did hierarchical cluster analysis using five common symptoms associated with primary Sjögren's syndrome (pain, fatigue, dryness, anxiety, and depression), followed by multinomial logistic regression to identify subgroups in the UK Primary Sjögren's Syndrome Registry (UKPSSR). We assessed clinical and biological differences between these subgroups, including transcriptional differences in peripheral blood. Patients from two independent validation cohorts in Norway and France were used to confirm patient stratification. Data from two phase 3 clinical trials were similarly stratified to assess the differences between subgroups in treatment response to hydroxychloroquine and rituximab. Findings In the UKPSSR cohort (n=608), we identified four subgroups: Low symptom burden (LSB), high symptom burden (HSB), dryness dominant with fatigue (DDF), and pain dominant with fatigue (PDF). Significant differences in peripheral blood lymphocyte counts, anti-SSA and anti-SSB antibody positivity, as well as serum IgG, κ-free light chain, β2-microglobulin, and CXCL13 concentrations were observed between these subgroups, along with differentially expressed transcriptomic modules in peripheral blood. Similar findings were observed in the independent validation cohorts (n=396). Reanalysis of trial data stratifying patients into these subgroups suggested a treatment effect with hydroxychloroquine in the HSB subgroup and with rituximab in the DDF subgroup compared with placebo. Interpretation Stratification on the basis of patient-reported symptoms of patients with primary Sjögren's syndrome revealed distinct pathobiological endotypes with distinct responses to immunomodulatory treatments. Our data have important implications for clinical management, trial design, and therapeutic development. Similar stratification approaches might be useful for patients with other chronic immune-mediated diseases.     Value of ultrasonography as a marker of early response to abatacept in patients with rheumatoid arthritis and an inadequate response to methotrexate: results from the APPRAISE study

    Objectives: To study the responsiveness of a combined power Doppler and greyscale ultrasound (PDUS) score for assessing synovitis in biologic-naïve patients with rheumatoid arthritis (RA) starting abatacept plus methotrexate (MTX). Methods: In this open-label, multicentre, single-arm study, patients with RA (MTX inadequate responders) received intravenous abatacept (∼10 mg/kg) plus MTX for 24 weeks. A composite PDUS synovitis score, developed by the Outcome Measures in Rheumatology–European League Against Rheumatism (OMERACT–EULAR)-Ultrasound Task Force, was used to evaluate individual joints. The maximal score of each joint was added into a Global OMERACT–EULAR Synovitis Score (GLOESS) for bilateral metacarpophalangeal joints (MCPs) 2–5 (primary objective). The value of GLOESS containing other joint sets was explored, along with clinical efficacy. Results: Eighty-nine patients completed the 24-week treatment period. The earliest PDUS sign of improvement in synovitis was at week 1 (mean change in GLOESS (MCPs 2–5): −0.7 (95% CIs −1.2 to −0.1)), with continuous improvement to week 24. Early improvement was observed in the component scores (power Doppler signal at week 1, synovial hyperplasia at week 2, joint effusion at week 4). Comparable changes were observed for 22 paired joints and minimal joint subsets. Mean Disease Activity Score 28 (C reactive protein) was significantly reduced from weeks 1 to 24, reaching clinical meaningful improvement (change ≥1.2) at week 8. Conclusions: In this first international prospective study, the composite PDUS score is responsive to abatacept. GLOESS demonstrated the rapid onset of action of abatacept, regardless of the number of joints examined. Ultrasound is an objective tool to monitor patients with RA under treatment.     Les sédiments éoliens carbonatés représentent des aquifères et des réservoirs d'hydrocarbures potentiels. Ces dépôts, fréquents le long des côtes semi-arides actuelles, se retrouvent très fréquemment dans les terrasses marines quaternaires mais n'ont été que très rarement signalés dans des séries sédimentaires plus anciennes au point que de nombreux auteurs considèrent que ces accumulations ne se développent que durant les périodes glaciaires. L'examen pétrographique d'éolianites actuelles et holocènes dans l'est de la Méditerranée et en Australie indique que la rareté de ces faciès dans les séries pré-quaternaires provient de la difficulté qu'il y a à les distinguer des dépôts marins. L'utilisation des techniques d'analyse d'images a permis de mettre en évidence un critère pétrotextural diagnostique. Ce critère pourrait être utilisé pour réinterpréter le milieu de dépôt de certains réservoirs pétroliers et constituer un nouvel indicateur paléoclimatique

    At a time when new communication technologies seem to promise a revival of participatory democracy, analysing the operation mode of a ten-year-old local television channel allows the assessment of a medium that was originally presented as a new tool for local democracy. Beyond that specific case, the survey brings out the determining factors which may lead a local channel to become a kind of tranquilizing factor producing a discourse that erases the conflicts of interests that shape local politics while actually contributing very little to a full-scale "public debate".Au moment où l'apparition de nouvelles technologies de communication semble promettre un renouveau de la démocratie participative, l'analyse - après 10 années d'existence - du fonctionnement d'une chaîne de télévision locale présentée à l'époque de son installation comme un « nouveau média » permet de tirer un bilan provisoire.     Recognition of ancient carbonate wind deposits: lessons from a modern analogue, Chrissi Island, Crete

    Carbonate aeolian deposits are common along arid to semiarid, wind-exposed, present-day coastlines bordered by productive carbonate ramps. Lithified carbonate dunes (aeolianites) have been described around the world in marine terraces of Quaternary age, but these deposits have seldom been identified in the Pre-Quaternary record. Several authors have suggested that this scarcity reflects that these deposits form and are preserved only during icehouse periods characterized by high-amplitude sea-level changes. Others [e.g. McKee and Ward Carbonate Depositional Environments (1983), AAPG Memoirs, Vol. 33, pp. 131–170] suggest that the scarcity of aeolianites in the Pre-Quaternary record could reflect the ‘great difficulty in recognising wind blown carbonate deposits and in differentiating between them [aeolianites] and other carbonate sands of nearshore environments'. It has been considered that carbonate shoreface/foreshore deposits are very difficult to discriminate petrographically from backshore deposits. This petrographic study of recent sediments from the shoreface to backshore along the northern coast of Chrissi Island, Crete, confirms that carbonate aeolian sands can be very easily misinterpreted as shoreface deposits. Textural examination of thin sections by image analysis techniques indicates, however, that grain orientation patterns differ between facies. Shoreface deposits exhibit a unimodal distribution of grain orientation (flat rose diagram), whereas backshore deposits show a tendency towards a bimodal distribution with a significant proportion of vertical grains. This observation has been confirmed in Pleistocene aeolianites from Tunisia and Western Australia. Grain verticality thus seems to be a reliable criterion for discriminating wind-lain carbonate bodies from shoreface deposits. Vertical grains in aeolian carbonate deposits could reflect gravity effects (e.g. reorientation of grains because of meteoric water percolation and air pull-up). Laboratory experiments conducted on carbonate sands under the action of percolating waters confirm this hypothesis. This reorganization process is preferentially developed in recently deposited and loosely packed sands resulting from grainfall and/or grainflow. In addition, this suggests that the presence of vertical grain orientation might be an indicator of the frequency and intensity of rainfalls during deposition

    Identifying the role of historical anthropogenic activities on urban soils: geochemical impact and city scale mapping

    International audienceRecently, European cities have faced several changes including deindustrialization and population increase. To limit urban sprawl, urban densification is preferred. It conducts to (re)develop available areas such as brownfields. Although these areas can be attractive for housing due to their location (in proximity to the city centre or to a riverside), their soils and subsoils are often contaminated. They are therefore potentially harmful for human health and the environment, and potentially costly to remediate. Currently, in case of contamination suspicion, depth geochemical characterization of urban soil and subsoil are carried out at site scale. Nevertheless, large redevelopment project occur at quarter to city scale. It appears therefore useful to acquire the preliminary knowledge on the structure and quality of soil and subsoils, as well as on the potential sources of contamination at quarter to city scale. In the frame of the Ile de Nantes (France) redevelopment project, we considered more particularly anthropogenic deposits and former industrial activities as main sources of contamination linked to human activities. To face the low traceability of the use of anthropogenic deposits and the lack of synthesis of former industrial activities, we carried out a historical study, synthetizing the information spread in numerous archive documents to spatialize the extent of the deposits and of the former activities. In addition we developed a typology of made grounds according to their contamination potential to build a 3D geological model with a geochemical coherence. In this frame, we valorized existing borehole descriptions coming mainly from pollution diagnosis and geotechnical studies. We also developed a methodology to define urban baseline compatibility levels using the existing analytical data at depth from pollution diagnosis. These data were previously gathered in a local geodatabase towards with borehole descriptions (more than 2000 borehole descriptions, more than 1800 analyzed samples, almost 100 000 analyzed parameters). The potential quality of soil and subsoil was spatialized in 2D and 3D on the basis of anthropogenic deposits structure and typology as well as of the potential sources of contamination linked to former industrial activities. Volumes were also calculated to help the developer anticipating the management of excavated materials. Comparison with effective soil and subsoil quality (existing chemical data) shows fairly good anticipation of contamination problems, confirming the interest of spatializing the historical anthropogenic activities to anticipate the quality of urban soil and subsoil and guide city scale mapping. Urban geochemical compatibility levels will be used operationally to enhance the reuse of excavated materials. A better knowledge of soils and subsoils at depth is very useful to optimize urban redevelopment projects, anticipating contamination problems, and managing excavated materials (e.g. local reuse possibilities, disposal costs etc.). The potential economic, environmental and social consequences render it essential for urban sustainable development. 3D geochemical characterization of soil and subsoil for urban (re)development is an ambitious task.
