    Integration of Peripheral and Glandular Regulation of Triiodothyronine Production by Thyrotropin in Untreated and Thyroxine-Treated Subjects

    Objective: We evaluated the roles of central and peripheral T3 regulation. Design and Methods: In a prospective study involving 1796 patients the equilibria between FT3 and TSH were compared in untreated and L-T4-treated patients with varying functional states, residual thyroid secretory capacities and magnitudes of TSH stimulation. Results: T3 concentrations were stable over wide variations in TSH levels (from 0.2 to 7 mU/l) and endogenous T4 production in untreated patients, but unbalanced in L-T4-treated athyreotic patients where T3 correlated with exogenous T4 supply. T3 stability was related to TSH-stimulated deiodinase activity by clinical observation, as predicted by theoretical modelling. Deiodinase activity in treated patients was reduced due to both diminished responsiveness to TSH and lack of thyroidal capacity. Deiodinase activity was increased in high thyroid volume, compared to lower volumes in euthyroid patients (< 5 ml, p<0.001). While deiodinase differed between euthyroid and subclinically hypothyroid patients in high volume, 26.7 nmol/s (23.6, 29.2), n=214 vs 28.9 nmol/s (26.7, 31.5), n=20, p=0.02, it was equivalent between the two functional groups in low volume, 23.3 nmol/s (21.3, 26.1), n=117 vs 24.6 nmol/s (22.2, 27.5), n=38, p=0.22. Conclusions: The findings suggest that the thyroid gland and peripheral tissues are integrated in the physiological process of T3 homeostasis in humans via a feed-forward TSH motif, which coordinates peripheral and central regulatory mechanisms. Regulatory and capacity deficiencies collectively impair T3 homeostasis in L-T4-treated patients

Dále se zabývá návrhem a realizací elektronického ovládání cévního odporu. Ventil automaticky nastavuje hodnoty odporu na základě nastavených parametrů. Z celého systému dostáváme zpětnou vazbu na základě měření tlaku. Výstup je zdigitalizován. Na konci je vyhodnocení dosažených výsledků.This bachelor thesis deal with Dynamic Control of Systemic Vascular Resistance of Vascular Model. It focuses on the issue of systemic vascular resistance. It also deals with the design and implementation of electronic control of vascular resistance. The valve automatically sets the resistance values based on the set parameters. We receive feedback from the entire system based on pressure measurements. The output is digitized.     La crítica ha alabado la última novela de Rafael Chirbes, En la orilla (2013), como la “gran novela de la crisis”. Se han destacado de ella su collage de imágenes y su coro de voces, con los que el autor consigue retratar la crisis económica y ética de España. En el presente análisis se hace un acercamiento diferente a la novela: por el olfato. Para ello, se discute cómo el olor influye en la construcción moral del ser humano y se presenta brevemente un estudio en el que se representa un país a través de su olor, para posteriormente identificar imágenes olfativas en el texto que, junto con las imágenes visuales y las voces, crean una sinergia de potenciación que sumerge al lector en el universo de la novela. Dado que la misma es un reflejo de los valores morales actuales de España, se analizan cuáles son los olores que representan al país y por tanto son característicos de la presente situación. Se concluye que el olor de los valores morales que rezuma España actualmente es el de la podredumbre, del dinero, del oportunismo, del engaño, del egoísmo y del silencio, olores que a su vez se pueden encontrar reflejados en las metáforas olfativas de la prensa nacional

    The alternative sigma factor SigB of Corynebacterium glutamicum modulates global gene expression during transition from exponential growth to stationary phase

    BACKGROUND: Corynebacterium glutamicum is a gram-positive soil bacterium widely used for the industrial production of amino acids. There is great interest in the examination of the molecular mechanism of transcription control. One of these control mechanisms are sigma factors. C. glutamicum ATCC 13032 has seven putative sigma factor-encoding genes, including sigA and sigB. The sigA gene encodes the essential primary sigma factor of C. glutamicum and is responsible for promoter recognition of house-keeping genes. The sigB gene codes for the non-essential sigma factor SigB that has a proposed role in stress reponse. RESULTS: The sigB gene expression was highest at transition between exponential growth and stationary phase, when the amount of sigA mRNA was already decreasing. Genome-wide transcription profiles of the wild-type and the sigB mutant were recorded by comparative DNA microarray hybridizations. The data indicated that the mRNA levels of 111 genes are significantly changed in the sigB-proficient strain during the transition phase, whereas the expression profile of the sigB-deficient strain showed only minor changes (26 genes). The genes that are higher expressed during transition phase only in the sigB-proficient strain mainly belong to the functional categories amino acid metabolism, carbon metabolism, stress defense, membrane processes, and phosphorus metabolism. The transcription start points of six of these genes were determined and the deduced promoter sequences turned out to be indistinguishable from that of the consensus promoter recognized by SigA. Real-time reverse transcription PCR assays revealed that the expression profiles of these genes during growth were similar to that of the sigB gene itself. In the sigB mutant, however, the transcription profiles resembled that of the sigA gene encoding the house-keeping sigma factor. CONCLUSION: During transition phase, the sigB gene showed an enhanced expression, while simultaneously the sigA mRNA decreased in abundance. This might cause a replacement of SigA by SigB at the RNA polymerase core enzyme and in turn results in increased expression of genes relevant for the transition and the stationary phase, either to cope with nutrient limitation or with the accompanying oxidative stress. The increased expression of genes encoding anti-oxidative or protection functions also prepares the cell for upcoming limitations and environmental stresses

Large energy efficiency defined by a heat to data ratio (HBR) of only 240 fJ/bit @ 50 Gb/s is observed. Tuning the cavity photon lifetime is demonstrated to lead to an increase of the data rate in concert with a reduction of the HDR. The large linearity of our L-I-characteristics will allow easily higher order modulation rates.     Encapsulation of ascorbic acid promotes the reduction of Maillard reaction products in UHT milk

    The presence of amino groups and carbonyls renders fortified milk with ascorbic acid particularly susceptible to the reduction of available lysine and to the formation of Maillard reaction products (MRPs), as Nε-(Carboxyethyl)-L-lysine (CEL), Nε-(Carboxymethyl)-L-lysine (CML), Amadori products (APs) and off-flavors. A novel approach was proposed to control the Maillard reaction (MR) in fortified milk: ascorbic acid was encapsulated in a lipid coating and the effects were tested after a lab scale UHT treatment. Encapsulation promoted a delayed release of ascorbic acid and a reduction in the formation of MRPs. Total lysine increased up to 45% in milk with encapsulated ascorbic acid, while reductions in CML, CEL and furosine ranged from 10% to 53% compared with control samples. The effects were also investigated towards the formation of amide-AGEs (advanced glycation end products) by high resolution mass spectrometry (HRMS) revealing that several mechanisms coincide with the MR in the presence of ascorbic acid (AA)
