    Mitochondrial cytochrome oxidase I in tetranychid mites : a comparison between molecular phylogeny and changes of morphological and life history traits

    Spider mites, #Tetranychidae, represent one of the most cosmopolitan and economically important groups of terrestrial arthropods ; however, many aspects of their evolutionary relationships remain uncertain. We sequenced part of the mitochondrial cytochrome oxidase subunit I (COI) gene in 20 species of phytophagous mites belonging to nine genera and two families (#Tetranychidae and #Tenuipalpidae),includingseveralagriculturalpests.Asreportedininsects,thesequenceswereextremelyrichinA+T(75), including several agricultural pests. As reported in insects, the sequences were extremely rich in A+T (75% on average), especially in the third codon position (95%). However, one of the genera we studied had a significantly lower A+T content (69% on average, 78% in the third codon position), showing that base composition can change substantially over short periods of time. Most interspecific differences were transversions and their number increased steadily with the number of non-synonymous differences, while the number of transitions remained constant. The phylogeny based on COI sequences was inferred using the maximum likelihood method. The results are compatible as a whole with the traditional classification based on morphological characters, but call for some minor taxonomic revisions. Some morphological characters and life history traits (mode of reproduction, adaptation to the host plant) were also analysed within this phylogenetic framework. At the family level, one can see a trend towards thelytoky becoming rarer compared to the general mode of reproduction of the group, arrhenotoky. There is also an evolutionary tendency towards a more complex mode of life, with the production of silk webs and correlated changes of the locomotion apparatus. However, in the #Tetranychidae there seems to have been convergent evolution of these morphological characters together with independent development of a common adaptation to this mode of life in different genera.     Etude de la résistance au méthyl-parathion, au méthidathion et au méthomyl chez une population de #Tetranychus urticae$ Koch originaire de Grèce. A la CL 50 le taux de résistance est hautement variable avec le méthidathion (5 à 63 fois) et avec le méthomyl (6 à 34 fois). Les courbes de mortalité avec le méthyl-parathion présentent un plateau net à une mortalité d'environ 20% et le taux de résistance est d'environ 50 fois à CL 50, dans tous les échantillons. Par utilisation de la focalisation isoélectrique sur des membranes d'acétate de cellulose, les études électrophorétiques de 5 loci codant les estérases (Est-1 et Est-2), l'isomérase gluco-phosphate (Gpi), l'enzyme malique (Me) et la phosphoglucomutase (Pgm), indiquent les différences génétiques importantes parmi les échantillons.     Mitochondrial COI sequences in mites : evidence for variations in base composition

    Studies of mitochondrial DNA sequences in a variety of animals have shown important differences between phyla, including differences in the genetic codes used, and varying constraints on base composition. In that respect, little is known of mites, an important and diversified group. We sequenced a portion (340 nt) of the cytochrome oxidase subunit I (COI) encoding gene in twenty species of phytophagous mites belonging to nine genera of the two families #Tetranychidae and #Tenuipalpidae. The mitochondrial genetic code used in mites appeared to be the same as in insects. As is generally also the case in insects, the mite sequences were very rich in A + T (75% on average), especially at the third codon position (94%). However, important variations of base composition were observed among mite species, one of them showing as little as 69% A + T. Variations of base composition occur mostly through synonymous transitions, and do not have detectable effects on polypeptide evolution in this group. (Résumé d'auteur

    Muscle and liver transcriptome characterization and genetic marker discovery in the farmed meagre, Argyrosomus regius

    Meagre (Argyrosomus regius), a teleost fish of the family Sciaenidae, is part of a group of marine fish species considered new for Mediterranean aquaculture representing the larger fish cultured in the region. Meagre aquaculture started ~ 25 years ago in West Mediterranean, and the supply of juveniles has been dominated by few hatcheries. This fact has raised concerns on possible inbreeding, urging the need for genetic information on the species and for an assessment of the polymorphisms found in the genome. To that end we characterized the muscle and liver transcriptome of a pool of meagre individuals, from different families and phenotypic size, to obtain a backbone that can support future studies regarding physiology, immunology and genetics of the species. The assembled transcripts were assigned to a wide range of biological processes including growth, reproduction, metabolism, development, stress and behavior. Then, to infer its genetic diversity and provide a catalogue of markers for future use, we scanned the reconstructed transcripts for polymorphic genetic markers. Our search revealed a total of 42,933 high quality SNP and 20,581 STR markers. We found a relatively low rate of polymorphism in the transcriptome that may indicate that inbreeding has taken place.     Transforming growth factor-β-inducible early response gene 1 is a novel substrate for atypical protein kinase Cs

    The protein kinase C (PKC) family of serine/threonine kinases consists of ten different isoforms grouped into three subfamilies, denoted classical, novel and atypical PKCs (aPKCs). The aPKCs, PKCι/λ and PKCζ serve important roles during development and in processes subverted in cancer such as cell and tissue polarity, cell proliferation, differentiation and apoptosis. In an effort to identify novel interaction partners for aPKCs, we performed a yeast two-hybrid screen with the regulatory domain of PKCι/λ as bait and identified the Krüppel-like factors family protein TIEG1 as a putative interaction partner for PKCι/λ. We confirmed the interaction of both aPKCs with TIEG1 in vitro and in cells, and found that both aPKCs phosphorylate the DNA-binding domain of TIEG1 on two critical residues. Interestingly, the aPKC-mediated phosphorylation of TIEG1 affected its DNA-binding activity, subnuclear localization and transactivation potential

    Glutamine Acts as a Neuroprotectant against DNA Damage, Beta-Amyloid and H2O2-Induced Stress

    Glutamine is the most abundant free amino acid in the human blood stream and is ‘conditionally essential’ to cells. Its intracellular levels are regulated both by the uptake of extracellular glutamine via specific transport systems and by its intracellular synthesis by glutamine synthetase (GS). Adding to the regulatory complexity, when extracellular glutamine is reduced GS protein levels rise. Unfortunately, this excess GS can be maladaptive. GS overexpression is neurotoxic especially if the cells are in a low-glutamine medium. Similarly, in low glutamine, the levels of multiple stress response proteins are reduced rendering cells hypersensitive to H2O2, zinc salts and DNA damage. These altered responses may have particular relevance to neurodegenerative diseases of aging. GS activity and glutamine levels are lower in the Alzheimer's disease (AD) brain, and a fraction of AD hippocampal neurons have dramatically increased GS levels compared with control subjects. We validated the importance of these observations by showing that raising glutamine levels in the medium protects cultured neuronal cells against the amyloid peptide, Aβ. Further, a 10-day course of dietary glutamine supplementation reduced inflammation-induced neuronal cell cycle activation, tau phosphorylation and ATM-activation in two different mouse models of familial AD while raising the levels of two synaptic proteins, VAMP2 and synaptophysin. Together, our observations suggest that healthy neuronal cells require both intracellular and extracellular glutamine, and that the neuroprotective effects of glutamine supplementation may prove beneficial in the treatment of AD

    Genomic analysis of Sparus aurata reveals the evolutionary dynamics of sex-biased genes in a sequential hermaphrodite fish

    Sexual dimorphism is a fascinating subject in evolutionary biology and mostly results from sex-biased expression of genes, which have been shown to evolve faster in gonochoristic species. We report here genome and sex-specific transcriptome sequencing of Sparus aurata, a sequential hermaphrodite fish. Evolutionary comparative analysis reveals that sex-biased genes in S. aurata are similar in number and function, but evolved following strikingly divergent patterns compared with gonochoristic species, showing overall slower rates because of stronger functional constraints. Fast evolution is observed only for highly ovary-biased genes due to female-specific patterns of selection that are related to the peculiar reproduction mode of S. aurata, first maturing as male, then as female.
