    Cannabis use during pregnancy and its relationship with fetal developmental outcomes and psychiatric disorders. A systematic review

    This study analyze factors associated to cannabis use in pregnant women, its perceived availability, its risk perception and the relationship between prenatal exposure to cannabis and developmental and mental disorders. We present a review of the literature on cannabis use among pregnant women. The objective is to analyze factors associated to cannabis use during pregnancy and assess the potential effects of prenatal exposure to cannabis on the development of the fetus and the mental health of those exposed. Systematic review of studies on the maternal use of cannabis and the relationship between early exposure and the development of psychiatric disorders in the PubMed database until July 2018 in English and Spanish with the following keywords: Marijuana, Cannabinoids, Mental disorders, Pregnancy, Prenatal Cannabis Exposure, Risk factors. The use of cannabis among pregnant women is frequent but it has not been extensively researched. Prenatal exposure to cannabis may be associated with affective symptoms and ADHD. Mental healthcare professionals who treat women during their fertile life need to be able to explain the relationship between prenatal exposure to cannabis and the presence of developmental and mental disorders

    Pharmacologic concentrations of linezolid modify oxidative phosphorylation function and adipocyte secretome

    The oxidative phosphorylation system is important for adipocyte differentiation. Therefore, xenobiotics inhibitors of the oxidative phosphorylation system could affect adipocyte differentiation and adipokine secretion. As adipokines impact the overall health status, these xenobiotics may have wide effects on human health. Some of these xenobiotics are widely used therapeutic drugs, such as ribosomal antibiotics. Because of its similarity to the bacterial one, mitochondrial translation system is an off-target for these compounds. To study the influence of the ribosomal antibiotic linezolid on adipokine production, we analyzed its effects on adipocyte secretome. Linezolid, at therapeutic concentrations, modifies the levels of apolipoprotein E and several adipokines and proteins related with the extracellular matrix. This antibiotic also alters the global methylation status of human adipose tissue-derived stem cells and, therefore, its effects are not limited to the exposure period. Besides their consequences on other tissues, xenobiotics acting on the adipocyte oxidative phosphorylation system alter apolipoprotein E and adipokine production, secondarily contributing to their systemic effects

    La restauración de la villa romana de La Ontavia (Terrinches, Ciudad Real)

    En el mes de julio de 2010 se iniciaron las labores de conservación y restauración en el yacimiento romano de La Ontavia (Terrinches, Ciudad Real). Durante cuatro meses se han simultaneado estos trabajos con la excavación arqueológica, en la que han colaborado once alumnos de la Escuela Superior de Conservación y Restauración de Bienes Culturales de la Comunidad de Madrid, bajo la dirección del equipo técnico, realizando prácticas para completar su formación. La intervención llevada a cabo ha estado enfocada a minimizar el efecto de los agentes de deterioro a los que se halla expuesto el yacimiento, que se encuentra al aire libre. El presente artículo tiene como finalidad la difusión de los trabajos de conservación y restauración realizados durante esta campaña, prestando especial atención a la metodología y a los materiales utilizadosIn July 2010 conservation and restoration work began at the Roman site of La Ontavia (Terrinches, Ciudad Real). For four months, the work was carried out alongside the archaeological excavation, involving eleven students from the Madrid School of Cultural Asset Conservation and Restoration, doing work experience under the guidance of a technical team to complete their training. The intervention focused on minimising the effect of the deterioration factors that this open-air site has been exposed to.     Transcriptomic and genetic studies identify NFAT5 as a candidate gene for cocaine dependence

    Cocaine reward and reinforcing effects are mediated mainly by dopaminergic neurotransmission. In this study, we aimed at evaluating gene expression changes induced by acute cocaine exposure on SH-SY5Y-differentiated cells, which have been widely used as a dopaminergic neuronal model. Expression changes and a concomitant increase in neuronal activity were observed after a 5 μ cocaine exposure, whereas no changes in gene expression or in neuronal activity took place at 1 μ cocaine. Changes in gene expression were identified in a total of 756 genes, mainly related to regulation of transcription and gene expression, cell cycle, adhesion and cell projection, as well as mitogen-activeated protein kinase (MAPK), CREB, neurotrophin and neuregulin signaling pathways. Some genes displaying altered expression were subsequently targeted with predicted functional single-nucleotide polymorphisms (SNPs) in a case-control association study in a sample of 806 cocaine-dependent patients and 817 controls. This study highlighted associations between cocaine dependence and five SNPs predicted to alter microRNA binding at the 3'-untranslated region of the NFAT5 gene. The association of SNP rs1437134 with cocaine dependence survived the Bonferroni correction for multiple testing. A functional effect was confirmed for this variant by a luciferase reporter assay, with lower expression observed for the rs1437134G allele, which was more pronounced in the presence of hsa-miR-509. However, brain volumes in regions of relevance to addiction, as assessed with magnetic resonance imaging, did not correlate with NFAT5 variation. These results suggest that the NFAT5 gene, which is upregulated a few hours after cocaine exposure, may be involved in the genetic predisposition to cocaine dependence

    Personal Construct Therapy vs Cognitive Behavioral Therapy in the Treatment of Depression in Women with Fibromyalgia: Study Protocol for a Multicenter Randomized Controlled Trial

    Background: Fibromyalgia (FM) is a debilitating syndrome, more prevalent in women, which is aggravated by the presence of depressive symptoms. In the last decade, cognitive behavioral therapy (CBT) has demonstrated to reduce such depressive symptoms and pain in these patients, but there are still a considerable number of them who do not respond to interventions. The complexity of the disorder requires the consideration of the unique psychological characteristics of each patient to attain good outcomes. One approach that could accomplish this goal might be personal construct therapy (PCT), an idiographic approach that considers identity features and interpersonal meanings as their main target of intervention. Then, the aim of the study is to test the efficacy of PCT as compared to a well-established treatment in the reduction of depressive symptoms in women with fibromyalgia. Methods and Analysis: This is a multicenter randomized controlled trial. In each condition participants will attend up to eighteen 1-hr weekly therapy sessions and up to three 1-hr booster sessions during the following 3- 5 months after the end of treatment. The depression subscale of the Hospital Anxiety and Depression Scale (HADS-D) will be the primary outcome measure and it will be assessed at baseline, at the end of therapy, and at 6-month follow-up. Other secondary measures will be applied following the same schedule. Participants will be 18- to 70-years-old women with a diagnosis of FM, presenting depressive symptoms evinced by scores above seven in depression items of the HADS-D. Intention-to-treat and complete case analyses will be performed for the main statistical tests. Linear mixed models will be used to analyze and to compare the treatment effects of both conditions

    Six-year follow-up of slaughterhouse surveillance (2008-2013): the Catalan Slaughterhouse Support Network (SESC)

    Meat inspection has the ultimate objective of declaring the meat and offal obtained from carcasses of slaughtered animals fit or unfit for human consumption. This safeguards the health of consumers by ensuring that the food coming from these establishments poses no risk to public health. Concomitantly, it contributes to animal disease surveillance. The Catalan Public Health Protection Agency (Generalitat de Catalunya) identified the need to provide its meat inspectors with a support structure to improve diagnostic capacity: the Slaughterhouse Support Network (SESC). The main goal of the SESC was to offer continuing education to meat inspectors to improve the diagnostic capacity for lesions observed in slaughterhouses. With this aim, a web-based application was designed that allowed meat inspectors to submit their inquiries, images of the lesions, and samples for laboratory analysis. This commentary reviews the cases from the first 6 years of SESC operation (2008–2013). The program not only provides continuing education to inspectors but also contributes to the collection of useful information on animal health and welfare. Therefore, SESC complements animal disease surveillance programs, such as those for tuberculosis, bovine cysticercosis, and porcine trichinellosis, and is a powerful tool for early detection of emerging animal diseases and zoonoses

    Landscape-scale drivers of glacial ecosystem change in the montane forests of the eastern Andean flank, Ecuador

    Understanding the impact of landscape-scale disturbance events during the last glacial period is vital in accu- rately reconstructing the ecosystem dynamics of montane environments. Here, a sedimentary succession from the tropical montane cloud forest of the eastern Andean flank of Ecuador provides evidence of the role of non- climate drivers of vegetation change (volcanic events, fire regime and herbivory) during the late-Pleistocene. Multiproxy analysis (pollen, non-pollen palynomorphs, charcoal, geochemistry and carbon content) of the se- diments, radiocarbon dated to ca. 45–42 ka, provide a snap shot of the depositional environment, vegetation community and non-climate drivers of ecosystem dynamics. The geomorphology of the Vinillos study area, along with the organic‐carbon content, and aquatic remains suggest deposition took place near a valley floor in a swamp or shallow water environment. The pollen assemblage initially composed primarily of herbaceous types (Poaceae-Asteraceae-Solanaceae) is replaced by assemblages characterised by Andean forest taxa, (first Melastomataceae-Weinmannia-Ilex, and later, Alnus-Hedyosmum-Myrica). The pollen assemblages have no modern analogues in the tropical montane cloud forest of Ecuador. High micro-charcoal and rare macro-charcoal abundances co-occur with volcanic tephra deposits suggesting transportation from extra-local regions and that volcanic eruptions were an important source of ignition in the wider glacial landscape. The presence of the coprophilous fungi Sporormiella reveals the occurrence of herbivores in the glacial montane forest landscape. Pollen analysis indicates a stable regional vegetation community, with changes in vegetation population co- varying with large volcanic tephra deposits suggesting that the structure of glacial vegetation at Vinillos was driven by volcanic activity

    Synthesis of Janus compounds for the recognition of G-U mismatched nucleobase pairs

    The design and synthesis of two Janus-type heterocycles with the capacity to simultaneously recognize guanine and uracyl in G-U mismatched pairs through complementary hydrogen bond pairing is described. Both compounds were conveniently functionalized with a carboxylic function and efficiently attached to a tripeptide sequence by using solid-phase methodologies. Ligands based on the derivatization of such Janus compounds with a small aminoglycoside, neamine, and its guanidinylated analogue have been synthesized, and their interaction with Tau RNA has been investigated by using several biophysical techniques, including UV-monitored melting curves, fluorescence titration experiments, and 1H NMR. The overall results indicated that Janus-neamine/guanidinoneamine showed some preference for the +3 mutated RNA sequence associated with the development of some tauopathies, although preliminary NMR studies have not confirmed binding to G-U pairs. Moreover, a good correlation has been found between the RNA binding affinity of such Janus-containing ligands and their ability to stabilize this secondary structure upon complexation
