    Hexavalent Chromium in Waters for Human Consumption and Irrigation in the Guano Canton

    Introduction: Considering the geographical location and industrial activities of the county Guano, which lies on an aquifer that supply water for both consumption and crop irrigation, it is necessary to examine the levels of chromium (Cr) in its watershed and tributaries. Objective: To evaluate the amount of Cr existing in the surface waters of the Guano river basin and aquifer. Methods: Several samples of the river and aquifer were taken and the amount of Cr present was measured. Using Z-test, Cr levels found in the water were compared with those recommended as normal by the World Health Organization (WHO), national standards of the Texto Unificado de Legislación Secundaria de Medio Ambiente (TULSMA) and the Servicio Ecuatoriano de Normalización 1108 ( INEN 1108). Results: Relatively high Cr values were found when compared with those established by the WHO. However, they were normal when compared with TULSMA and INEN 1108. Conclusion: The levels of Cr found in the Guano river and the aquifer were higher than those recommended by the WHO. It is recommended to replicate our findings in order to corroborate the results and to take appropriate measures in order to reduce water contamination. Keywords: chromium, water, Guano, Ecuador. Resumen Introducción: Considerando la ubicación geográfica y actividades industriales del cantón Guano el cual yace sobre un acuífero que sustenta las necesidades de agua tanto para consumo y riego de la población es necesario examinar los niveles de Cromo (Cr) en su vertiente como sus afluentes. Objetivo: Evaluar la cantidad de Cr existente en las aguas superficiales de la cuenca del río Guano y en pequeños ojos de agua provenientes del acuífero. Métodos: Se tomó varias muestras de agua del río y ojos de agua y se midió la cantidad de Cr presente. Por medio de la prueba Z se comparó los niveles de Cr con los recomendados como normales por la Organización Mundial de la Salud (OMS), normas nacionales del Texto Unificado de Legislación Secundaria de Medio Ambiente (TULSMA) y Servicio Ecuatoriano de Normalización 1108 (INEN 1108). Resultados: Se encontró valores relativamente altos de Cr al compararlos con los establecidos por la OMS. Sin embargo, estos se encontraron dentro de rangos normales al compararlos con TULSMA e INEN 1108. Conclusiones: Los niveles de Cr encontrados en el río Guano y ojos de agua del acuífero son superiores a los recomendados por la OMS. Se recomienda replicar los hallazgos para corroborar los resultados y tomar las medidas adecuadas para reducir la contaminación del agua. Palabras clave: cromo, agua, Guano, Ecuador

    Arquitectura genérica de una red en chip de enrutamiento unidireccional en FPGA

    El constante aumento de los componentes que contiene un sistema on-chip ha incrementado la complejidad de comunicación entre los elementos de procesamiento (EPs) del sistema. Un recurso utilizado para disminuir la complejidad es el diseño de enrutamiento de conexiones (cableado), el cual ha sido suficiente para interconectar algunos EPs, dicho diseño se conoce como redes en chip o por sus siglas en inglés NoC (Network on Chip), de manera alternativa, enrutar paquetes permite una mayor escalabilidad de las redes, tener una latencia aceptable y una utilización moderada de área. Sin embargo, las redes en chip (NoC) suelen ser implementadas en tecnologías rígidas y deterministas como los ASIC (Circuito Integrado de Aplicación Específica), limitando la flexibilidad, arquitectura y modularidad que ofrece una NoC de enrutamiento de paquetes. Este trabajo propone una arquitectura de una red en chip de switcheo o enrutamiento unidireccional utilizando un router genérico para topología de mariposa, de enrutamiento de paquetes, implementado en una FPGA de la familia Xilinx. Donde el diseño permite enviar paquetes desde 16 puntos de origen, hacia 16 puntos de destino, así como la flexibilidad de enviar paquetes de diferentes tamaños, divididos enflits.     Improvement in detecting cytomegalovirus drug resistance mutations in solid organ transplant recipients with suspected resistance using next generation sequencing

    Objetives The aim of this study was to identify CMV drug resistance mutations (DRM) in solid organ transplant (SOT) recipients with suspected resistance comparing next-generation sequencing (NGS) with Sanger sequencing and assessing risk factors and the clinical impact of resistance. Methods Using Sanger sequencing as the reference method, we prospectively assessed the ability of NGS to detect CMV DRM in the UL97 and UL54 genes in a nationwide observational study from September 2013 to August 2016. Results Among 44 patients recruited, 14 DRM were detected by Sanger in 12 patients (27%) and 20 DRM were detected by NGS, in 16 (36%). NGS confirmed all the DRM detected by Sanger. The additional six mutations detected by NGS were present in <20% of the sequenced population, being located in the UL97 gene and conferring high-level resistance to ganciclovir. The presence of DRM by NGS was associated with lung transplantation (p = 0.050), the administration of prophylaxis (p = 0.039), a higher mean time between transplantation and suspicion of resistance (p = 0.038) and longer antiviral treatment duration before suspicion (p = 0.024). However, the latter was the only factor independently associated with the presence of DRM by NGS in the multivariate analysis (OR 2.24, 95% CI 1.03 to 4.87). Conclusions NGS showed a higher yield than Sanger sequencing for detecting CMV resistance mutations in SOT recipients.     Productive performance, carcass and meat quality of intact and castrated gilts slaughtered at 106 or 122 kg BW

    A total of 200 (Landrace3Large White dam3Pietrain3Large White sire) gilts of 5063 days of age (23.361.47 kg BW) were used to investigate the effects of castration (intact gilt, IG v. castrated gilt, CG) and slaughter weight (SW; 106 v. 122 kg BW) on productive performance, carcass and meat quality. Four treatments were arranged factorially and five replicates of 10 pigs each per treatment. Half of the gilts were ovariectomized at 58 days of age (8 days after the beginning of the trial at 29.861.64 kg BW), whereas the other half remained intact. The pigs were slaughtered at 106 or 122 kg BW. Meat samples were taken at Musculus longissimus thoracis at the level of the last rib and subcutaneous fat samples were taken at the tail insertion. For the entire experimental period, CG had higher ( P,0.05) BW gain and higher ( P,0.001) backfat and Musculus gluteus medius fat thickness than IG. However, IG had higher ( P,0.05) loin and trimmed primal cut yields than CG. Meat quality was similar for IG and CG but the proportion of linoleic acid in subcutaneous fat was higher ( P,0.001) for IG. Pigs slaughtered at 122 kg BW had higher ( P,0.001) feed intake and poorer feed efficiency than pigs slaughtered at 106 kg BW. An increase in SW improved ( P,0.001) carcass yield but decreased ( P,0.05) trimmed primal cut yield. Meat from pigs slaughtered at the heavier BW was redder (a*; P,0.001) and had more ( P,0.01) intramuscular fat and less thawing ( P,0.05) and cooking ( P,0.10) loss than meat from pigs slaughtered at the lighter BW. In addition, pigs slaughtered at 122 kg BW had less ( P,0.01) linoleic acid content in subcutaneous fat than pigs slaughtered at 106 kg BW. Castration of gilts and slaughtering at heavier BW are useful practices for the production of heavy pigs destined to the dry-cured industry in which a certain amount of fat in the carcass is required. In contrast, when the carcasses are destined to fresh meat production, IG slaughtered at 106 kg BW is a more efficient alternative

    In vitro Induction of Entamoeba histolytica Cyst-like Structures from Trophozoites

    Inhibition of encystment can be conceived as a potentially useful mechanism to block the transmission of Entamoeba histolytica under natural conditions. Unfortunately, amoeba encystment has not been achieved in vitro and drugs inhibiting the formation of cysts are not available. Luminal conditions inducing encystment in vivo are also unknown, but cellular stress such as exposure to reactive oxygen species from immune cells or intestinal microbiota could be involved. A role for certain divalent cations as cofactors of enzymes involved in excystment has also been described. In this study, we show that trophozoite cultures, treated with hydrogen peroxide in the presence of trace amounts of several cations, transform into small-sized spherical and refringent structures that exhibit resistance to different detergents. Ultrastructural analysis under scanning and transmission electron microscopy revealed multinucleated structures (some with four nuclei) with smooth, thick membranes and multiple vacuoles. Staining with calcofluor white, as well as an ELISA binding assay using wheat germ agglutinin, demonstrated the presence of polymers of N-acetylglucosamine (chitin), which is the primary component of the natural cyst walls. Over-expression of glucosamine 6-phosphate isomerase, likely to be the rate-limiting enzyme in the chitin synthesis pathway, was also confirmed by RT-PCR. These results suggest that E. histolytica trophozoites activated encystment pathways when exposed to our treatment

    The intracellular domain of β-dystroglycan mediates the nucleolar stress response by suppressing UBF transcriptional activity

    β-dystroglycan (β-DG) is a key component of multiprotein complexes in the plasma membrane and nuclear envelope. In addition, β-DG undergoes two successive proteolytic cleavages that result in the liberation of its intracellular domain (ICD) into the cytosol and nucleus. However, stimuli-inducing ICD cleavage and the physiological relevance of this proteolytic fragment are largely unknown. In this study we show for the first time that β-DG ICD is targeted to the nucleolus where it interacts with the nuclear proteins B23 and UBF (central factor of Pol I-mediated rRNA gene transcription) and binds to rDNA promoter regions. Interestingly DG silencing results in reduced B23 and UBF levels and aberrant nucleolar morphology. Furthermore, β-DG ICD cleavage is induced by different nucleolar stressors, including oxidative stress, acidosis, and UV irradiation, which implies its participation in the response to nucleolar stress. Consistent with this idea, overexpression of β-DG elicited mislocalization and decreased levels of UBF and suppression of rRNA expression, which in turn provoked altered ribosome profiling and decreased cell growth. Collectively our data reveal that β-DG ICD acts as negative regulator of rDNA transcription by impeding the transcriptional activity of UBF, as a part of the protective mechanism activated in response to nucleolar stress

    Host factor PLAC8 is required for pancreas infection by SARS-CoV-2

    Although mounting evidence demonstrated that pancreas is infected by SARS-CoV-2 the severity and pathophysiology of pancreatic COVID-19 disease are still unclear. Here we investigated the consequences of SARS-CoV-2 infection of the pancreas and the role of Placenta-associated protein-8 (PLAC8). Our data showed pancreatic damage in patients who died from COVID-19. Notably, circulating pancreatic enzymes stratified patients according to COVID-19 severity and outcome. PLAC8 expression was associated with SARS-CoV-2 infection in postmortem analysis of COVID-19 patients and functional assays demonstrated the requirement of PLAC8 in SARS-CoV-2 pancreatic infection. Full SARS-CoV-2 infectious virus revealed a requirement of PLAC8 for efficient viral infection of pancreatic cell lines. Finally, we observed colocalization of PLAC8 and SARS-CoV-2 in the pancreas of deceased patients. In conclusion, our data confirm the human pancreas as a SARS-CoV-2 target and demonstrate the requirement of PLAC8 for SARS-CoV-2 pancreatic infection thereby opening new target opportunities for COVID-19-associated pancreatic pathogenesis.N

    Efficacy of β-lactam/β-lactamase inhibitors to treat extended-spectrum beta-lactamase-producing Enterobacterales bacteremia secondary to urinary tract infection in kidney transplant recipients (INCREMENT-SOT Project)

    REIPI/INCREMENT-SOT Group.[Background] Whether active therapy with β-lactam/β-lactamase inhibitors (BLBLI) is as affective as carbapenems for extended-spectrum β-lactamase-producing Enterobacterales (ESBL-E) bloodstream infection (BSI) secondary to urinary tract infection (UTI) in kidney transplant recipients (KTRs) remains unclear.[Methods] We retrospectively evaluated 306 KTR admitted to 30 centers from January 2014 to October 2016. Therapeutic failure (lack of cure or clinical improvement and/or death from any cause) at days 7 and 30 from ESBL-E BSI onset was the primary and secondary study outcomes, respectively.[Results] Therapeutic failure at days 7 and 30 occurred in 8.2% (25/306) and 13.4% (41/306) of patients. Hospital-acquired BSI (adjusted OR [aOR]: 4.10; 95% confidence interval [CI]: 1.50-11.20) and Pitt score (aOR: 1.47; 95% CI: 1.21-1.77) were independently associated with therapeutic failure at day 7. Age-adjusted Charlson Index (aOR: 1.25; 95% CI: 1.05-1.48), Pitt score (aOR: 1.72; 95% CI: 1.35-2.17), and lymphocyte count ≤500 cells/μL at presentation (aOR: 3.16; 95% CI: 1.42-7.06) predicted therapeutic failure at day 30. Carbapenem monotherapy (68.6%, primarily meropenem) was the most frequent active therapy, followed by BLBLI monotherapy (10.8%, mostly piperacillin-tazobactam). Propensity score (PS)-adjusted models revealed no significant impact of the choice of active therapy (carbapenem-containing vs any other regimen, BLBLI- vs carbapenem-based monotherapy) within the first 72 hours on any of the study outcomes.[Conclusions] Our data suggest that active therapy based on BLBLI may be as effective as carbapenem-containing regimens for ESBL-E BSI secondary to UTI in the specific population of KTR.
