    A low perfusion rate microreactor for continuous monitoring of enzyme characteristics: application to glucose oxidase

    This report describes a versatile and robust microreactor for bioactive proteins physically immobilized on a polyether sulfone filter. The potential of the reactor is illustrated with glucose oxidase immobilized on a filter with a cut-off value of 30 kDa. A flow-injection system was used to deliver the reactants and the device was linked on-line to an electrochemical detector. The microreactor was used for on-line preparation of apoglucose oxidase in strong acid and its subsequent reactivation with flavin adenine dinucleotide. In addition we describe a miniaturized version of the microreactor used to assess several characteristics of femtomole to attomole amounts of glucose oxidase. A low negative potential over the electrodes was used when ferrocene was the mediator in combination with horseradish peroxidase, ensuring the absence of oxidation of electro-active compounds in biological fluids. A low backpressure at very low flow rates is an advantage, which increases the sensitivity. A variety of further applications of the microreactor are suggested

    Genetic screening of Fabry patients with EcoTILLING and HRM technology

    <p>Abstract</p> <p>Background</p> <p>Anderson-Fabry disease (FD) is caused by a deficit of the α-galactosidase A enzyme which leads to the accumulation of complex sphingolipids, especially globotriaosylceramide (Gb3), in all the cells of the body, causing the onset of a multi-systemic disease with poor prognosis in adulthood. In this article, we describe two alternative methods for screening the <it>GLA </it>gene which codes for the α-galactosidase A enzyme in subjects with probable FD in order to test analysis strategies which include or rely on initial pre-screening.</p> <p>Findings</p> <p>We analyzed 740 samples using EcoTILLING, comparing two mismatch-specific<ul/>endonucleases, CEL I and ENDO-1, while conducting a parallel screening of the same samples using HRM (High Resolution Melting). Afterwards, all samples were subjected to direct sequencing. Overall, we identified 12 different genetic variations: -10C>T, -12G>A, -30G>A, IVS2-76_80del5, D165H, C172Y, IVS4+16A>G, IVS4 +68 A>G, c.718_719delAA, D313Y, IVS6-22C>T, G395A. This was consistent with the high genetic heterogeneity found in FD patients and carriers. All of the mutations were detected by HRM, whereas 17% of the mutations were not found by EcoTILLING. The results obtained by EcoTILLING comparing the CEL I and ENDO-1 endonucleases were perfectly overlapping.</p> <p>Conclusion</p> <p>On the basis of its simplicity, flexibility, repeatability, and sensitivity, we believe that<ul/>HRM analysis of the <it>GLA </it>gene is a reliable presequencing screening tool. This method can be applied to any genomic feature to identify known and unknown genetic alterations, and it is ideal for conducting screening and population studies.</p

    Miniaturization in Biocatalysis

    The use of biocatalysts for the production of both consumer goods and building blocks for chemical synthesis is consistently gaining relevance. A significant contribution for recent advances towards further implementation of enzymes and whole cells is related to the developments in miniature reactor technology and insights into flow behavior. Due to the high level of parallelization and reduced requirements of chemicals, intensive screening of biocatalysts and process variables has become more feasible and reproducibility of the bioconversion processes has been substantially improved. The present work aims to provide an overview of the applications of miniaturized reactors in bioconversion processes, considering multi-well plates and microfluidic devices, update information on the engineering characterization of the hardware used, and present perspective developments in this area of research

    Bakalářská práce se zaměřuje na problematiku řízení zásob v podniku Crystalite Bohemia, s.r.o. Tento podnik se zabývá strojovou výrobou skla pro domácnost a nápojového skla. První část práce obecně vymezuje pojem zásoby a jejich role v podniku, dále seznamuje s řízením zásob a jejich metodami. Druhá část se zaměřuje na charakteristiku společnosti a především na analýzu řízení zásob v Crystalite Bohemia, s.r.o. Následně na provedené analýzy navazují návrhy na zlepšení současné situace v podniku. Cílem práce je analyzovat strukturu zásob pomocí metody ABC a následně minimalizovat vázanost kapitálu v zásobách.Bachelor thesis concentrates on the problems of control stock in Crystalite Bohemia s.r.o. enterprise. This enterprise deals with machine production of glass for household and drinking glasses. The first part of the thesis generally defines the term stock and its role in the enterprise as well as it acquaints with the stock management and its methods. The second part concentrates on the characteristics of the company and especially the analysis stock management in Crystalite Bohemia, s.r.o. Subsequently, the analysis continued with proposals to improve the current situation in the company.     The diploma thesis named "Do we need more information? Using of multiple media at the same time" examines the phenomen of simultaneous media usage in young audience aged 18-35 years. This thesis is based on the hypotheses of international surveys that demonstrate a new trend in consumption of media by which people use multiple media simultaneously. The aim of the thesis is to provide an insight into this issue. The first part defines basic concepts such as the media, media audience and related theories. Furthermore, international studies and their conclusions are presented, serving as a starting point for my own research. The second part presents the research methodology. The thesis makes use of a combination of a quantitative and qualitative approach, the so-called mixed research. The methodological section briefly describes the selected method - survey and an semi-structured interview. The next two sections deal with the implementation of the research itself. The sample consisted of 103 young people for the quantitative part of the survey, and 15 young people for the qualitative analysis section. The conclusion summarizes the research results and approaches each thematic area, including the possible hypotheses

    Diplomová práce se zabývá hodnocením konkurenceschopnosti společnosti Lékárny HERBA s.r.o. Tato společnost působí na trhu s lékárenskou péčí v lékárnách základního typu přes 20 let. První část práce vymezuje základní pojmy spojené s konkurenceschopností, jako konkurence, konkurenční výhoda či konkurenční strategie. Dále popisuje vybrané metody hodnocení konkurenceschopnosti společnosti. Druhá část se zaměřuje na charakteristiku společnosti a především na hodnocení konkurenceschopnosti společnosti Lékárny HERBA s.r.o.
