    Low incidence of severe bacterial infections in hospitalised patients with COVID-19 : A population-based registry study

    Background Bacterial infections complicating COVID-19 are rare but present a challenging clinical entity. The aim of this study was to evaluate the incidence, aetiology and outcome of severe laboratory-verified bacterial infections in hospitalised patients with COVID-19. Methods All laboratory-confirmed patients with COVID-19 admitted to specialised healthcare hospitals in the Capital Province of Finland during the first wave of COVID-19 between 27 February and 21 June 2020 were retrospectively studied. We gathered the blood and respiratory tract culture reports of these patients and analysed their association with 90-day case-fatality using multivariable regression analysis. Results A severe bacterial infection was diagnosed in 40/585 (6.8%) patients with COVID-19. The range of bacteria was diverse, and the most common bacterial findings in respiratory samples were gram-negative, and in blood cultures gram-positive bacteria. Patients with severe bacterial infection had longer hospital stay (mean 31; SD 20 days) compared to patients without (mean 9; SD 9 days; p < 0.001). Case-fatality was higher with bacterial infection (15% vs 11%), but the difference was not statistically significant (OR 1.38 CI95% 0.56-3.41). Conclusions Severe bacterial infection complicating COVID-19 was a rare occurrence in our cohort. Our results are in line with the current understanding that antibiotic treatment for hospitalised COVID-19 patients should only be reserved for situations where a bacterial infection is strongly suspected. The ever-evolving landscape of the pandemic and recent advances in immunomodulatory treatment of COVID-19 patients underline the need for continuous vigilance concerning the possibility and frequency of nosocomial bacterial infections.Peer reviewe

    Nordic Homicide in Deep Time draws a unique, detailed picture of developments in human violence and presents new findings on homicide in Northern Europe in two eras – the 17th century and early 21st century. The book provides answers to questions, such as where and when did homicide typically occur, who were the victims and the offenders, and what were the circumstances of their conflicts? Additionally, it offers an empirically grounded view on how state consolidation and changing routines of everyday life transformed the patterns of criminal homicide in the Nordics. This publication is also a methodological experiment. When developing a new approach for extending homicide research into the deep past, the authors created a new instrument, the Historical Homicide Monitor. This tool combines wide explanatory scope, measurement standardization, and articulated theory expression. By retroactively expanding research data to the pre-statistical era, the method enables long-duration comparison of different periods and areas.     Nordic Homicide in Deep Time draws a unique and detailed picture of developments in human interpersonal violence and presents new findings on rates, patterns, and long-term changes in lethal violence in the Nordics. Conducted by an interdisciplinary team of criminologists and historians, the book analyses homicide and lethal violence in northern Europe in two eras – the 17th century and early 21st century. Similar and continuous societal structures, cultural patterns, and legal cultures allow for long-term and comparative homicide research in the Nordic context. Reflecting human universals and stable motives, such as revenge, jealousy, honour, and material conflicts, homicide as a form of human behaviour enables long-duration comparison. By describing the rates and patterns of homicide during these two eras, the authors unveil continuity and change in human violence. Where and when did homicide typically take place? Who were the victims and the offenders, what where the circumstances of their conflicts? Was intimate partner homicide more prevalent in the early modern period than in present times? How long a time elapsed from violence to death? Were homicides often committed in the context of other crime? The book offers answers to these questions among others, comparing regions and eras. We gain a unique and empirically grounded view on how state consolidation and changing routines of everyday life transformed the patterns of criminal homicide in Nordic society. The path to pacification was anything but easy, punctuated by shorter crises of social turmoil, and high violence. The book is also a methodological experiment that seeks to assess the feasibility of long-duration standardized homicide analysis and to better understand the logic of homicide variation across space and over time. In developing a new approach for extending homicide research into the deep past, the authors have created the Historical Homicide Monitor. The new instrument combines wide explanatory scope, measurement standardization, and articulated theory expression. By retroactively expanding research data to the pre-statistical era, the method enables long-duration comparison of different periods and areas.     How consistent are the transcriptome changes associated with cold acclimation in two species of the Drosophila virilis group?

(NE/I528634/1).For many organisms the ability to cold acclimate with the onset of seasonal cold has major implications for their fitness. In insects, where this ability is widespread, the physiological changes associated with increased cold tolerance have been well studied. Despite this, little work has been done to trace changes in gene expression during cold acclimation that lead to an increase in cold tolerance. We used an RNA-Seq approach to investigate this in two species of the Drosophila virilis group. We found that the majority of genes that are differentially expressed during cold acclimation differ between the two species. Despite this, the biological processes associated with the differentially expressed genes were broadly similar in the two species. These included: metabolism, cell membrane composition, and circadian rhythms, which are largely consistent with previous work on cold acclimation/cold tolerance. In addition, we also found evidence of the involvement of the rhodopsin pathway in cold acclimation, a pathway that has been recently linked to thermotaxis. Interestingly, we found no evidence of differential expression of stress genes implying that long-term cold acclimation and short-term stress response may have a different physiological basis.PostprintPeer reviewe

    Nordic homicide report : Homicide in Denmark, Finland, Iceland, Norway and Sweden, 2007–2016

    This report compares the trends and patterns of lethal violence in from 2007 to 2016, in five Nordic countries: Denmark, Finland, Iceland, Norway and Sweden. The report is a product of the “Nordic Homicide from Past to Present” research project, funded by the Scandinavian Research Council for criminology. The main findings include: * Based on homicide mortality rates, the Nordic countries form currently three groups: compared to the rates in Denmark and Sweden, the homicide mortality rate is about 30 per cent higher in Finland and 30 per cent lower in Norway and Iceland. However, by global standards, all the countries have extremely low rates of homicide mortality. * In Denmark, Iceland, Norway, and Sweden, the hotspots of lethal violence are the metropolitan areas of the largest cities; in Finland, the rural areas and small towns in the eastern and northern provinces. In this respect, the situation in Scandinavia and Iceland resembles that in Western Europe, in Finland again, it resembles that in some of the eastern European countries. * In all five countries, non-working working-age male substance abusers are hugely over-represented among homicide offenders and victims. * In the three Scandinavian countries, immigrants make up to 25 to 40 per cent of homicide offenders, while in Iceland and Finland their proportion is about 10 per cent. While this difference reflects the sizes of the immigrant populations, differential risks are also involved. In all Nordic countries, the homicide offending rates of immigrants are higher than those of native residents, but this difference is substantially larger in Scandinavia than in Iceland or Finland. * The role of alcohol and drinking situations in lethal violence is central in Finland, Iceland and Sweden, but only moderate in Denmark and insignificant in Norway. This is reflected in the temporal distribution of homicide incidents; and to the lower percentage of crimes in near relations and the higher percentage of male victims in Finland, Iceland and Sweden. * Concerning firearm homicides, Sweden is currently a clear outlier in the region with every fourth homicide being perpetrated by firearms. The firearm homicide rate in Sweden is the highest of all the Nordic countries. The situation has deteriorated fast in the last few years. Firearm homicides are concentrated in the metropolitan areas of Stockholm, Gothenburg and Malmö and to a large extent are linked to gang violence in a few residential districts. * Nordic homicide clearance rates are among the highest in the world; offenders are caught and sentenced almost without exception. The official control policies are effective and arguably contribute to very low homicide rates in the region when compared with the global situation

    DNA methylation regulates expression of VEGF-R2 (KDR) and VEGF-R3 (FLT4)

    Abstract Background Vascular Endothelial Growth Factors (VEGFs) and their receptors (VEGF-Rs) are important regulators for angiogenesis and lymphangiogenesis. VEGFs and VEGF-Rs are not only expressed on endothelial cells but also on various subtypes of solid tumors and leukemias contributing to the growth of the malignant cells. This study was performed to examine whether VEGF-R2 (KDR) and VEGF-R3 (FLT4) are regulated by DNA methylation. Methods Real-time (RT) PCR analysis was performed to quantify KDR and FLT4 expression in some ninety leukemia/lymphoma cell lines, human umbilical vein endothelial cells (HUVECs) and dermal microvascular endothelial cells (HDMECs). Western blot analyses and flow cytometric analyses confirmed results at the protein level. After bisulfite conversion of DNA we determined the methylation status of KDR and FLT4 by DNA sequencing and by methylation specific PCR (MSP). Western blot analyses were performed to examine the effect of VEGF-C on p42/44 MAPK activation. Results Expression of KDR and FLT4 was observed in cell lines from various leukemic entities, but not in lymphoma cell lines: 16% (10/62) of the leukemia cell lines expressed KDR, 42% (27/65) were FLT4 positive. None of thirty cell lines representing six lymphoma subtypes showed more than marginal expression of KDR or FLT4. Western blot analyses confirmed KDR and FLT4 protein expression in HDMECs, HUVECs and in cell lines with high VEGF-R mRNA levels. Mature VEGF-C induced p42/44 MAPK activation in the KDR- /FLT4+ cell line OCI-AML1 verifying the model character of this cell line for VEGF-C signal transduction studies. Bisulfite sequencing and MSP revealed that GpG islands in the promoter regions of KDR and FLT4 were unmethylated in HUVECs, HDMECs and KDR + and FLT4 + cell lines, whereas methylated cell lines did not express these genes. In hypermethylated cell lines, KDR and FLT4 were re-inducible by treatment with the DNA demethylating agent 5-Aza-2'deoxycytidine, confirming epigenetic regulation of both genes. Conclusions Our data show that VEGF-Rs KDR and FLT4 are silenced by DNA methylation. However, if the promoters are unmethylated, other factors (e.g. transactivation factors) determine the extent of KDR and FLT4 expression

    Filicide in Austria and Finland - A register-based study on all filicide cases in Austria and Finland 1995-2005

    <p>Abstract</p> <p>Background</p> <p>Filicide is the tragic crime of murdering one's own child. Previous research has found that the offending parents are commonly depressed and that suicide is often associated as an actual act or an intention. Yet, filicide is an underreported crime and previous studies have been strained with methodological problems. No comprehensive international studies on filicide have been presented in the literature until now.</p> <p>Methods</p> <p>This was a descriptive, comprehensive, register-based study of all filicides in Austria and Finland during 1995-2005. Filicide-suicide cases were also included.</p> <p>Results</p> <p>Most of the perpetrators were the biological mothers; in Austria 72%, in Finland 52%. Suicide followed filicide either as an attempt or a fulfilled act in 32% and 54% of the cases in Austria and Finland, respectively. Psychotic mood disorders were diagnosed for 10% of the living perpetrators in Austria, and 12% in Finland. Non-psychotic depression was diagnosed in 9% of surviving perpetrators in Austria, 35% in Finland.</p> <p>Conclusion</p> <p>The data from the two countries demonstrated that filicide is such a multifaceted and rare phenomenon that national data from individual countries seldom offer sufficient scope for its thorough study. Further analyses are needed to produce a complete picture of filicide.</p

    Development of the SIOPE DIPG network, registry and imaging repository : a collaborative effort to optimize research into a rare and lethal disease

    Diffuse intrinsic pontine glioma (DIPG) is a rare and deadly childhood malignancy. After 40 years of mostly single-center, often non-randomized trials with variable patient inclusions, there has been no improvement in survival. It is therefore time for international collaboration in DIPG research, to provide new hope for children, parents and medical professionals fighting DIPG. In a first step towards collaboration, in 2011, a network of biologists and clinicians working in the field of DIPG was established within the European Society for Paediatric Oncology (SIOPE) Brain Tumour Group: the SIOPE DIPG Network. By bringing together biomedical professionals and parents as patient representatives, several collaborative DIPG-related projects have been realized. With help from experts in the fields of information technology, and legal advisors, an international, web-based comprehensive database was developed, The SIOPE DIPG Registry and Imaging Repository, to centrally collect data of DIPG patients. As for April 2016, clinical data as well as MR-scans of 694 patients have been entered into the SIOPE DIPG Registry/Imaging Repository. The median progression free survival is 6.0 months (95% Confidence Interval (CI) 5.6-6.4 months) and the median overall survival is 11.0 months (95% CI 10.5-11.5 months). At two and five years post-diagnosis, 10 and 2% of patients are alive, respectively. The establishment of the SIOPE DIPG Network and SIOPE DIPG Registry means a paradigm shift towards collaborative research into DIPG. This is seen as an essential first step towards understanding the disease, improving care and (ultimately) cure for children with DIPG.Peer reviewe
