    Harmonic Analysis and Tidal Prediction by the Method of Least Squares: A User\u27s Manual

    A common objective in the treatment of measured tides is the mathematical separation by harmonic analysis of certain periodic components which can be used to simulate the astronomical tide. Traditionally, astronomical or predicted tides have been represented by the sum of several such components, each one due to a particular aspect of the tide-producing forces. Once determined, the same set of parameters governing each periodic component may be used to predict the tide during any desired time period at a given location. These parameters are known as the tidal constants for that location. Although a number of different methods have been developed for determining tidal constants, both before and since the advent of electronic computing techniques, one rarely finds that any of these are intended for practical application by nonspecialists. This is particularly true in the case of the scientist or technician who has a collection of tidal records and would like to utilize them to accurately predict tides by some convenient means. The certified thesis is available in the Institute Archives and Special Collections.Page 160 blank.Includes bibliographical references (p. 155-159).This thesis examines the phenomenon of vacated churches and analyzes the major issues underlying their adaptive reuse in order to help promulgate an awareness of the range of successful strategies and solutions that are available to stakeholders who are interested in seeing a former church building preserved through its conversion to a new use. The reuse potential of a church is affected by the building typology and its structural condition, the stakeholders involved, the process used, the regulatory context, finance and site issues. Reuse potential for a church can also be informed by researching and documenting examples of previously successful reuse strategies. These issues are examined closely in this thesis, as is a case study of a successful public-private church redevelopment project in Brookline, MA. The results of the research and analysis are used to form a series of findings and recommendations regarding the reuse of churches. The recommendations include combining funding sources, engaging the congregation and the public, retaining the appropriate kind of developer for the end goal, undertaking public-private redevelopment processes and the importance of identifying interim solutions. The recommendations are tested for credibility by applying them to a real situation in Plymouth, MA while working with a team of developers to asses the reuse potential of a historic church for a downsizing congregation.by Christopher John Kiley.M.C.P.and S.M.in Real Estate Developmen

    Reconstructing metabolic pathways of hydrocarbon-degrading bacteria from the Deepwater Horizon oil spill

    The Deepwater Horizon blowout in the Gulf of Mexico in 2010, one of the largest marine oil spills1, changed bacterial communities in the water column and sediment as they responded to complex hydrocarbon mixtures2-4. Shifts in community composition have been correlated to the microbial degradation and use of hydrocarbons2,5,6, but the full genetic potential and taxon-specific metabolisms of bacterial hydrocarbon degraders remain unresolved. Here, we have reconstructed draft genomes of marine bacteria enriched from sea surface and deep plume waters of the spill that assimilate alkane and polycyclic aromatic hydrocarbons during stable-isotope probing experiments, and we identify genes of hydrocarbon degradation pathways. Alkane degradation genes were ubiquitous in the assembled genomes. Marinobacter was enriched with n-hexadecane, and uncultured Alpha- and Gammaproteobacteria populations were enriched in the polycyclic-aromatic-hydrocarbon-degrading communities and contained a broad gene set for degrading phenanthrene and naphthalene. The repertoire of polycyclic aromatic hydrocarbon use varied among different bacterial taxa and the combined capabilities of the microbial community exceeded those of its individual components, indicating that the degradation of complex hydrocarbon mixtures requires the non-redundant capabilities of a complex oil-degrading community

    Marburg virus survivor immune responses are Th1 skewed with limited neutralizing antibody responses.

    Until recently, immune responses in filovirus survivors remained poorly understood. Early studies revealed IgM and IgG responses to infection with various filoviruses, but recent outbreaks have greatly expanded our understanding of filovirus immune responses. Immune responses in survivors of Ebola virus (EBOV) and Sudan virus (SUDV) infections have provided the most insight, with T cell responses as well as detailed antibody responses having been characterized. Immune responses to Marburg virus (MARV), however, remain almost entirely uncharacterized. We report that immune responses in MARV survivors share characteristics with EBOV and SUDV infections but have some distinct differences. MARV survivors developed multivariate CD4+ T cell responses but limited CD8+ T cell responses, more in keeping with SUDV survivors than EBOV survivors. In stark contrast to SUDV survivors, rare neutralizing antibody responses in MARV survivors diminished rapidly after the outbreak. These results warrant serious consideration for any vaccine or therapeutic that seeks to be broadly protective, as different filoviruses may require different immune responses to achieve immunity

    Comprehensive Molecular Characterization of Pheochromocytoma and Paraganglioma

    SummaryWe report a comprehensive molecular characterization of pheochromocytomas and paragangliomas (PCCs/PGLs), a rare tumor type. Multi-platform integration revealed that PCCs/PGLs are driven by diverse alterations affecting multiple genes and pathways. Pathogenic germline mutations occurred in eight PCC/PGL susceptibility genes. We identified CSDE1 as a somatically mutated driver gene, complementing four known drivers (HRAS, RET, EPAS1, and NF1). We also discovered fusion genes in PCCs/PGLs, involving MAML3, BRAF, NGFR, and NF1. Integrated analysis classified PCCs/PGLs into four molecularly defined groups: a kinase signaling subtype, a pseudohypoxia subtype, a Wnt-altered subtype, driven by MAML3 and CSDE1, and a cortical admixture subtype. Correlates of metastatic PCCs/PGLs included the MAML3 fusion gene. This integrated molecular characterization provides a comprehensive foundation for developing PCC/PGL precision medicine

    Global Impact of the COVID-19 Pandemic on Cerebral Venous Thrombosis and Mortality

    Background and purpose: Recent studies suggested an increased incidence of cerebral venous thrombosis (CVT) during the coronavirus disease 2019 (COVID-19) pandemic. We evaluated the volume of CVT hospitalization and in-hospital mortality during the 1st year of the COVID-19 pandemic compared to the preceding year. Methods: We conducted a cross-sectional retrospective study of 171 stroke centers from 49 countries. We recorded COVID-19 admission volumes, CVT hospitalization, and CVT in-hospital mortality from January 1, 2019, to May 31, 2021. CVT diagnoses were identified by International Classification of Disease-10 (ICD-10) codes or stroke databases. We additionally sought to compare the same metrics in the first 5 months of 2021 compared to the corresponding months in 2019 and 2020 (ClinicalTrials.gov Identifier: NCT04934020). Results: There were 2,313 CVT admissions across the 1-year pre-pandemic (2019) and pandemic year (2020); no differences in CVT volume or CVT mortality were observed. During the first 5 months of 2021, there was an increase in CVT volumes compared to 2019 (27.5%; 95% confidence interval [CI], 24.2 to 32.0; P<0.0001) and 2020 (41.4%; 95% CI, 37.0 to 46.0; P<0.0001). A COVID-19 diagnosis was present in 7.6% (132/1,738) of CVT hospitalizations. CVT was present in 0.04% (103/292,080) of COVID-19 hospitalizations. During the first pandemic year, CVT mortality was higher in patients who were COVID positive compared to COVID negative patients (8/53 [15.0%] vs. 41/910 [4.5%], P=0.004). There was an increase in CVT mortality during the first 5 months of pandemic years 2020 and 2021 compared to the first 5 months of the pre-pandemic year 2019 (2019 vs. 2020: 2.26% vs. 4.74%, P=0.05; 2019 vs. 2021: 2.26% vs. 4.99%, P=0.03). In the first 5 months of 2021, there were 26 cases of vaccine-induced immune thrombotic thrombocytopenia (VITT), resulting in six deaths. Conclusions: During the 1st year of the COVID-19 pandemic, CVT hospitalization volume and CVT in-hospital mortality did not change compared to the prior year. COVID-19 diagnosis was associated with higher CVT in-hospital mortality. During the first 5 months of 2021, there was an increase in CVT hospitalization volume and increase in CVT-related mortality, partially attributable to VITT
