    Tämän opinnäytteen tarkoituksena on selvittää BisnesAkatemia-yhteisön kehittämiskohteita erilaisten tiedonkeruumenetelmien ja teorioiden kautta. Pääroolissa on organisaation psykologista pääomaa etsivä kyselytutkimus, joka pyrkii muodostamaan tiimiyrittäjien keskuudesta yksittäisiä ryhmiä, jotka näkevät yhteisön arvot, rakenteet, käytänteet sekä toimisen siellä omalla uniikilla tavallaan. Kyselytutkimuksen taustatukena on teemahaastatteluita. Tärkein teoreettinen viitekehys on oppivat yhteisöt ja organisaatiopsykologia. Työ painottaa yksilön merkitystä koko yhteisön kannalta ja keskittyy löytämään ratkaisut yksilön kautta, yksilöä ajatellen. Opinnäytettä ohjaa halu kehittää toimintaa ja toimia kehittämistutkimuksena, joka tähtää muutokseen konkreettisin keinoin. Tutkimusosio tehdään noudattaen tieteellisiä opinnäytteen standardeja. Tulokset analysoidaan liittäen ne suurempiin kokonaisuuksiin ja BisnesAkatemia-yhteisön antamiin signaaleihin vapaasti ja rohkeasti. Kehittämistutkimus antoi suoria viestejä siitä, että yhteisön kehittämisessä on vielä paljon tehtävää. Erityisesti esille nousi yhteisten arvojen, ilmapiirin ja vision puute. Kehittämistutkimus antaa lopuksi toiminnan kehittämiseen suoria konkreettisia suosituksia, jotka ovat perusteltavissa eri teorioiden ja käytäntöjen pohjalta. Monet suositukset on sovellettavissa välittömästi toimintaan.The purpose of the thesis is to study BusinessAcademy community development areas through a variety of data collection methods and theories. The main role is in psychological capital survey that seeks to form a student teams, who see the community, values, and struc-tures in different ways and work there in their own way. The survey is supported by theme inter-views. The most important theoretical framework includes learning communities and organiza-tional psychology. The study emphasizes the importance of the individual to community as a whole and focuses on finding solutions based on the individual. This thesis is driven by the desire to develop and as a development research it aims to change things through concrete means. Research section is done in accordance to the scientific thesis standards. The results are analyzed, attributing them to larger entities and to the signals given by the BusinessAcademy-community. Development Research gave direct messages that there is still a lot to be done in community development. Particular problems were the lack of shared values, atmosphere and vision. As a whole, the development research provides concrete development recommendations, which are justified on the basis of different theories and practices. Many of the recommenda-tions are directly applicable to BusinessAcademy

    Analysis of long branch extraction and long branch shortening.

Research has shown that parsimony is particularly vulnerable to inferring the wrong tree in Felsenstein topologies. The long branch extraction method is a procedure to detect a data set suffering from this problem so that Maximum Likelihood could be used instead of Maximum Parsimony. RESULTS: The long branch extraction method has been well cited and used by many authors in their analysis but no strong validation has been performed as to its accuracy. We performed such an analysis by an extensive search of the branch length search space under two topologies of six taxa, a Felsenstein-like topology and Farris-like topology. We also examine a long branch shortening method. CONCLUSIONS: The long branch extraction method seems to mask the majority of the search space rendering it ineffective as a detection method of LBA. A proposed alternative, the long branch shortening method, is also ineffective in predicting long branch attraction for all tree topologies

    Phylogenetic search through partial tree mixing.

    BACKGROUND: Recent advances in sequencing technology have created large data sets upon which phylogenetic inference can be performed. Current research is limited by the prohibitive time necessary to perform tree search on a reasonable number of individuals. This research develops new phylogenetic algorithms that can operate on tens of thousands of species in a reasonable amount of time through several innovative search techniques. RESULTS: When compared to popular phylogenetic search algorithms, better trees are found much more quickly for large data sets. These algorithms are incorporated in the PSODA application available at http://dna.cs.byu.edu/psoda CONCLUSIONS: The use of Partial Tree Mixing in a partition based tree space allows the algorithm to quickly converge on near optimal tree regions. These regions can then be searched in a methodical way to determine the overall optimal phylogenetic solution

    Inferring gene regulatory networks from asynchronous microarray data with AIRnet

    Background Modern approaches to treating genetic disorders, cancers and even epidemics rely on a detailed understanding of the underlying gene signaling network. Previous work has used time series microarray data to infer gene signaling networks given a large number of accurate time series samples. Microarray data available for many biological experiments is limited to a small number of arrays with little or no time series guarantees. When several samples are averaged to examine differences in mean value between a diseased and normal state, information from individual samples that could indicate a gene relationship can be lost. Results Asynchronous Inference of Regulatory Networks (AIRnet) provides gene signaling network inference using more practical assumptions about the microarray data. By learning correlation patterns for the changes in microarray values from all pairs of samples, accurate network reconstructions can be performed with data that is normally available in microarray experiments. Conclusions By focussing on the changes between microarray samples, instead of absolute values, increased information can be gleaned from expression data

    Thermofield Dynamics and Casimir Effect for Fermions

    A generalization of the Bogoliubov transformation is developed to describe a space compactified fermionic field. The method is the fermionic counterpart of the formalism introduced earlier for bosons (J. C. da Silva, A. Matos Neto, F. C. Khanna and A. E. Santana, Phys. Rev. A 66 (2002) 052101), and is based on the thermofield dynamics approach. We analyse the energy-momentum tensor for the Casimir effect of a free massless fermion field in a dd-dimensional box at finite temperature. As a particular case the Casimir energy and pressure for the field confined in a 3-dimensional parallelepiped box are calculated. It is found that the attractive or repulsive nature of the Casimir pressure on opposite faces changes depending on the relative magnitude of the edges. We also determine the temperature at which the Casimir pressure in a cubic boc changes sign and estimate its value when the edge of the cybe is of the order of confining lengths for baryons.Comment: 21 pages, 3 figures, to appear in Annals of Physic

    Basis for enhanced barrier function of pigmented skin

    Humans with darkly-pigmented skin display superior permeability barrier function in comparison to humans with lightly-pigmented skin. The reduced pH of the stratum corneum (SC) of darkly-pigmented skin could account for enhanced function, because acidifying lightly-pigmented human SC resets barrier function to darkly-pigmented levels. In SKH1 (non-pigmented) vs. SKH2/J (pigmented) hairless mice, we evaluated how a pigment-dependent reduction in pH could influence epidermal barrier function. Permeability barrier homeostasis is enhanced in SKH2/J vs. SKH1 mice, correlating with a reduced pH in the lower SC that co-localizes with the extrusion of melanin granules. Darkly-pigmented human epidermis also shows substantial melanin extrusion in the outer epidermis. Both acute barrier disruption and topical basic pH challenges accelerate re-acidification of SKH2/J (but not SKH1) SC, while inducing melanin extrusion. SKH2/J mice also display enhanced expression of the SC acidifying enzyme, secretory phospholipase A2f (sPLA2f). Enhanced barrier function of SKH2/J mice could be attributed to enhanced activity of two acidic pH-dependent, ceramide-generating enzymes, β-glucocerebrosidase and acidic sphingomyelinase, leading to accelerated maturation of SC lamellar bilayers. Finally, organotypic cultures of darkly-pigmented-bearing human keratinocytes display enhanced barrier function in comparison to lightly-pigmented cultures. Together, these results suggest that the superior barrier function of pigmented epidermis can be largely attributed to the pH-lowering impact of melanin persistence/extrusion and enhanced sPLA2f expression

    On the use of cartographic projections in visualizing phylo-genetic tree space

    Phylogenetic analysis is becoming an increasingly important tool for biological research. Applications include epidemiological studies, drug development, and evolutionary analysis. Phylogenetic search is a known NP-Hard problem. The size of the data sets which can be analyzed is limited by the exponential growth in the number of trees that must be considered as the problem size increases. A better understanding of the problem space could lead to better methods, which in turn could lead to the feasible analysis of more data sets. We present a definition of phylogenetic tree space and a visualization of this space that shows significant exploitable structure. This structure can be used to develop search methods capable of handling much larger data sets

    Pharmacological Stimulation of Edar Signaling in the Adult Enhances Sebaceous Gland Size and Function

    Impaired ectodysplasin A (EDA) receptor (EDAR) signaling affects ectodermally derived structures including teeth, hair follicles, and cutaneous glands. The X-linked hypohidrotic ectodermal dysplasia (XLHED), resulting from EDA deficiency, can be rescued with lifelong benefits in animal models by stimulation of ectodermal appendage development with EDAR agonists. Treatments initiated later in the developmental period restore progressively fewer of the affected structures. It is unknown whether EDAR stimulation in adults with XLHED might have beneficial effects. In adult Eda mutant mice treated for several weeks with agonist anti-EDAR antibodies, we find that sebaceous gland size and function can be restored to wild-type levels. This effect is maintained upon chronic treatment but reverses slowly upon cessation of treatment. Sebaceous glands in all skin regions respond to treatment, although to varying degrees, and this is accompanied in both Eda mutant and wild-type mice by sebum secretion to levels higher than those observed in untreated controls. Edar is expressed at the periphery of the glands, suggesting a direct homeostatic effect of Edar stimulation on the sebaceous gland. Sebaceous gland size and sebum production may serve as biomarkers for EDAR stimulation, and EDAR agonists may improve skin dryness and eczema frequently observed in XLHED
