    XMCD studies of thin Co films on BaTiO3_3

    Different layer thicknesses of Cobalt ranging from 2.6 {\AA} (1.5 ML) up to 55 {\AA} (30.5 ML) deposited on ferroelectric BaTiO3_3 have been studied regarding their magnetic behavior. The layers have been characterized using XMCD spectroscopy at remanent magnetization. After careful data analysis the magnetic moments of the Cobalt could be determined using the sum rule formalism. There is a sudden and abrupt onset in magnetism starting at thicknesses of 9 {\AA} (5 ML) of Cobalt for measurements at 120 K and of 10 {\AA} (5.5 ML) if measured at room temperature. Initial island growth and subsequent coalescence of Co on BaTiO3_3 is suggested to explain the sudden onset.     Impaired endotoxin-induced interleukin-1β secretion, not total production, of mononuclear cells from ESRD patients. Lipopolysaccharide (LPS)-induced interleukin-1 beta (IL-1β) and tumor necrosis factor alpha (TNFα) production and secretion from peripheral blood mononuclear cells (PBMC) were determined in a longitudinal study with repeated measurements in PBMC from patients with chronic uremia not on hemodialysis (N = 8), end-stage renal disease (ESRD) patients (N = 8), and healthy controls (N = 7). ESRD patients were studied while using low-flux Cuprophan dialyzers and again using high-flux AN 69 dialyzers. Total (cell-associated plus secreted) LPS-induced IL-1β production was enhanced in uremic patients, but similar to controls in ESRD patients on Cuprophan. In contrast, LPS-induced IL-1β secretion (secreted amounts in % of total production) was similar to controls in uremic patients, but significantly reduced in ESRD patients on Cuprophan (P < 0.01). During AN 69 hemodialysis, LPS-induced total IL-1β production remained unchanged but IL-1β secretion increased significantly (P < 0.05) compared to Cuprophan dialysis. Increased IL-1β secretion coincided with a suppression in PGE2 synthesis (P < 0.02). Similarly, blockade of endogenous PGE2 by indomethacin increased LPS-induced IL-1β secretion (P < 0.01) but did not enhance total IL-1β production in PBMC from controls and patients on Cuprophan hemodialysis. Neither total production nor secretion of TNFα was different comparing the three study groups. We conclude that LPS-induced IL-1β secretion, but not total production, is impaired in PBMC from ESRD patients on long-term Cuprophan hemodialysis. This functional change in the PBMC response is specific for IL-1β, not due to uremia per se but hemodialysis-dependent and reversible. Hemodialysis with AN 69 suppresses endogenous PGE2 synthesis in PBMC which is associated with increased LPS-induced IL-1β secretion in the presence of unchanged total IL-1β production. We speculate that PGE2 could inactivate the IL-1β converting enzyme which is essential for processing and secretion of mature IL-1β

    Background: The bacterial GlgE pathway is the third known route to glycogen and is the only one present in mycobacteria. It contributes to the virulence of Mycobacterium tuberculosis. The involvement of GlgE in glycogen biosynthesis was discovered twenty years ago when the phenotype of a temperature-sensitive Mycobacterium smegmatis mutation was rescued by the glgE gene. The evidence at the time suggested glgE coded for a glucanase responsible for the hydrolysis of glycogen, in stark contrast with recent evidence showing GlgE to be a polymerase responsible for its biosynthesis. Methods: We reconstructed and examined the temperature-sensitive mutant and characterised the mutated GlgE enzyme. Results: The mutant strain accumulated the substrate for GlgE, α-maltose-1-phosphate, at the non-permissive temperature. The glycogen assay used in the original study was shown to give a false positive result with α-maltose-1-phosphate. The accumulation of α-maltose-1-phosphate was due to the lowering of the kcat of GlgE as well as a loss of stability 42 °C. The reported rescue of the phenotype by GarA could potentially involve an interaction with GlgE, but none was detected. Conclusions: We have been able to reconcile apparently contradictory observations and shed light on the basis for the phenotype of the temperature-sensitive mutation. General significance: This study highlights how the lowering of flux through the GlgE pathway can slow the growth mycobacteria

    Predicting plankton net community production in the Atlantic Ocean

    We present, test and implement two contrasting models to predict euphotic zone net community production (NCP), which are based on 14C primary production (PO14CP) to NCP relationships over two latitudinal (ca. 30°S–45°N) transects traversing highly productive and oligotrophic provinces of the Atlantic Ocean (NADR, CNRY, BENG, NAST-E, ETRA and SATL, Longhurst et al., 1995 [An estimation of global primary production in the ocean from satellite radiometer data. Journal of Plankton Research 17, 1245–1271]). The two models include similar ranges of PO14CP and community structure, but differ in the relative influence of allochthonous organic matter in the oligotrophic provinces. Both models were used to predict NCP from PO14CP measurements obtained during 11 local and three seasonal studies in the Atlantic, Pacific and Indian Oceans, and from satellite-derived estimates of PO14CP. Comparison of these NCP predictions with concurrent in situ measurements and geochemical estimates of NCP showed that geographic and annual patterns of NCP can only be predicted when the relative trophic importance of local vs. distant processes is similar in both modeled and predicted ecosystems. The system-dependent ability of our models to predict NCP seasonality suggests that trophic-level dynamics are stronger than differences in hydrodynamic regime, taxonomic composition and phytoplankton growth. The regional differences in the predictive power of both models confirm the existence of biogeographic differences in the scale of trophic dynamics, which impede the use of a single generalized equation to estimate global marine plankton NCP. This paper shows the potential of a systematic empirical approach to predict plankton NCP from local and satellite-derived P estimates

    Structure and cation distribution of (Mn0.5Zn0.5)Fe2O4 thin films on SrTiO3(001)

    A comprehensive study on growth of ferrimagnetic manganese zinc ferrite (Mn0.5Zn0.5Fe2O4) films on single crystalline strontium titanate(001) (SrTiO3) substrates was carried out. Under the optimized conditions, a thin film with a layer thickness of 200 nm was deposited, and the structural properties were investigated. Contrary to data published in literature, no buffer layer was necessary to achieve epitaxial growth of a poorly lattice-matched layer. This was confirmed for Mn0.5Zn0.5Fe2O4(001) on SrTiO3(001) by x-ray diffraction and the adjoined phi scans, which also revealed a lattice compression of 1.2% of the manganese zinc ferrite film in the out-of-plane direction. Using x-ray photoelectron spectroscopy, the near surface stoichiometry of the film could be shown to agree with the intended one within the uncertainty of the method. X-ray absorption spectroscopy showed an electronic structure close to that published for bulk samples. Additional x-ray magnetic circular dichroism investigations were performed to answer detailed structural questions by a comparison of experimental data with the calculated ones. The calculations took into account ion sites (tetrahedral vs. octahedral coordination) as well as the charge of Fe ions (Fe2+ vs. Fe3+). Contrary to the expectation for a perfect normal spinel that only Fe3Ăľ ions are present in octahedral sites, hints regarding the presence of additional Fe2+ in octahedral sites as well as Fe3+ ions in tetrahedral sites have been obtained. Altogether, the layer could be shown to be mostly in a normal spinel configuration
