    Evaluación de la resistencia genética de híbridos de maíz al virus del mosaico de la caña de azúcar (SCMV)

    La resistencia genética es la manera más eficiente de controlar las enfermedades ya que no tiene un costo adicional para el agricultor y no contamina el ambiente. Un cultivo de maíz sano es la regla, la excepción es la enfermedad. El primer paso en todo programa de mejoramiento genético para incorporar resistencia al cultivo es identificar materiales resistentes entre las variedades tradicionales y comerciales, poblaciones mejoradas de alta diversidad genética (pooles), colecciones núcleo y colecciones para estudios genéticos. Para esto es necesario realizar evaluaciones en condiciones controladas donde se inocula el patógeno y se le brindan las condiciones para que la planta se enferme. En caso de enfermedades virales, las técnicas de inoculación incluyen el frotamiento, punción vascular y la utilización de los vectores naturales que transmiten el virus. Sugarcane mosaic virus (SCMV) está entre los principales virus que afectan al cultivo de maíz en los valles altos y trópicos de Latinoamérica y alrededor del mundo. En la Estación Experimental Litoral Sur del INIAP se inocularon 32 híbridos comerciales y experimentales de maíz utilizando la técnica del frotamiento en plántulas, con un aislamiento de SCMV colectado en la Estación Experimental Portoviejo del INIAP. Dos semanas después de la inoculación se evaluó la incidencia de los síntomas de la enfermedad en las hojas nuevas. El experimento tuvo tres réplicas biológicas en un diseño  completo al azar, con 20 plantas por tratamiento. No existieron híbridos resistentes, lo que indica la necesidad de incorporar genes de resistencia a SCMV en las poblaciones de mejoramiento genético de maíces tropicales de grano amarillo duro de las empresas públicas y privadas

    Patterns of Research Effort and Extinction Risk of Marine Mammals in the Philippines

    Global marine mammal research is disproportionately lacking compared to terrestrial mammal research and is strongly biased toward populations in Europe, North America, New Zealand, and Australia. With high extinction risks facing marine mammals in the tropics, we sought to identify potential drivers of research effort and extinction risk evaluations for marine mammals in the Philippines as a model for tropical island nations with limited resources and research capacity. Using a bibliographic approach, we compiled all materials on marine mammal research in the Philippines from 1991 to 2020, which we categorized into eight thematic areas of research focus. We reviewed all materials based on their research focus to assess the current scientific knowledge of local marine mammal populations. Using a simple metric to calculate research effort allocation, we found that all marine mammal species in the Philippines receive inadequate research attention. Using generalized linear models, we analyzed the relationship of potential factors that drive research effort. The model with the lowest Akaike Information Criterion value suggests that frequency of marine mammal stranding incidents may influence an increase in research effort on marine mammals by providing access to biological specimens that would normally be difficult to obtain. Strandings are unfortunate events with often unclear causes, but they provide an opportunity to collect data from behaviorally cryptic animals in areas where financial constraints often hamper scientific progress. We also determined that a national Red List evaluation was predicted by increased research effort. Maximizing local research using all materials from strandings and building research capacity may be an alternative to expensive field-based methods to increase knowledge on local marine mammal populations

    Ultraviolet through Infrared Spectral Energy Distributions from 1000 SDSS Galaxies: Dust Attenuation

    The meaningful comparison of models of galaxy evolution to observations is critically dependent on the accurate treatment of dust attenuation. To investigate dust absorption and emission in galaxies we have assembled a sample of ~1000 galaxies with ultraviolet (UV) through infrared (IR) photometry from GALEX, SDSS, and Spitzer and optical spectroscopy from SDSS. The ratio of IR to UV emission (IRX) is used to constrain the dust attenuation in galaxies. We use the 4000A break as a robust and useful, although coarse, indicator of star formation history (SFH). We examine the relationship between IRX and the UV spectral slope (a common attenuation indicator at high-redshift) and find little dependence of the scatter on 4000A break strength. We construct average UV through far-IR spectral energy distributions (SEDs) for different ranges of IRX, 4000A break strength, and stellar mass (M_*) to show the variation of the entire SED with these parameters. When binned simultaneously by IRX, 4000A break strength, and M_* these SEDs allow us to determine a low resolution average attenuation curve for different ranges of M_*. The attenuation curves thus derived are consistent with a lambda^{-0.7} attenuation law, and we find no significant variations with M_*. Finally, we show the relationship between IRX and the global stellar mass surface density and gas-phase-metallicity. Among star forming galaxies we find a strong correlation between IRX and stellar mass surface density, even at constant metallicity, a result that is closely linked to the well-known correlation between IRX and star-formation rate.Comment: 12 pages, 8 figures, 2 tables, appearing in the Dec 2007 GALEX special issue of ApJ Supp (29 papers

    Cytokinin Determines Thiol-Mediated Arsenic Tolerance and Accumulation

    The presence of arsenic in soil and water is a constant threat to plant growth in many regions of the world. Phytohormones act in the integration of growth control and stress response, but their role in plant responses to arsenic remains to be elucidated. Here, we show that arsenate [As(V)], the most prevalent arsenic chemical species in nature, causes severe depletion of endogenous cytokinins (CKs) in the model plant Arabidopsis (Arabidopsis thaliana). We found that CK signaling mutants and transgenic plants with reduced endogenous CK levels showed an As(V)-tolerant phenotype. Our data indicate that in CK-depleted plants exposed to As(V), transcript levels of As(V)/phosphate-transporters were similar or even higher than in wild-type plants. In contrast, CK depletion provoked the coordinated activation of As(V) tolerance mechanisms, leading to the accumulation of thiol compounds such as phytochelatins and glutathione, which are essential for arsenic sequestration. Transgenic CK-deficient Arabidopsis and tobacco lines show a marked increase in arsenic accumulation. Our findings indicate that CK is an important regulatory factor in plant adaptation to arsenic stress

    Chemical and Photometric Evolution of Extended Ultraviolet Disks: Optical Spectroscopy of M83 (NGC5236) and NGC4625

    We present the results from the analysis of optical spectra of 31 Halpha-selected regions in the extended UV (XUV) disks of M83 (NGC5236) and NGC4625 recently discovered by GALEX. The spectra were obtained using IMACS at Las Campanas Observatory 6.5m Magellan I telescope and COSMIC at the Palomar 200-inch telescope, respectively for M83 and NGC4625. The line ratios measured indicate nebular oxygen abundances (derived from the R23 parameter) of the order of Zsun/5-Zsun/10. For most emission-line regions analyzed the line fluxes and ratios measured are best reproduced by models of photoionization by single stars with masses in the range 20-40 Msun and oxygen abundances comparable to those derived from the R23 parameter. We find indications for a relatively high N/O abundance ratio in the XUV disk of M83. Although the metallicities derived imply that these are not the first stars formed in the XUV disks, such a level of enrichment could be reached in young spiral disks only 1 Gyr after these first stars would have formed.     Mortality According to CD4 Count at Start of Combination Antiretroviral Therapy Among HIV-infected Patients Followed for up to 15 Years After Start of Treatment: Collaborative Cohort Study.

    BACKGROUND: CD4 count at start of combination antiretroviral therapy (ART) is strongly associated with short-term survival, but its association with longer-term survival is less well characterized. METHODS: We estimated mortality rates (MRs) by time since start of ART (<0.5, 0.5-0.9, 1-2.9, 3-4.9, 5-9.9, and ≥10 years) among patients from 18 European and North American cohorts who started ART during 1996-2001. Piecewise exponential models stratified by cohort were used to estimate crude and adjusted (for sex, age, transmission risk, period of starting ART [1996-1997, 1998-1999, 2000-2001], and AIDS and human immunodeficiency virus type 1 RNA at baseline) mortality rate ratios (MRRs) by CD4 count at start of ART (0-49, 50-99, 100-199, 200-349, 350-499, ≥500 cells/µL) overall and separately according to time since start of ART. RESULTS: A total of 6344 of 37 496 patients died during 359 219 years of follow-up. The MR per 1000 person-years was 32.8 (95% confidence interval [CI], 30.2-35.5) during the first 6 months, declining to 16.0 (95% CI, 15.4-16.8) during 5-9.9 years and 14.2 (95% CI, 13.3-15.1) after 10 years' duration of ART. During the first year of ART, there was a strong inverse association of CD4 count at start of ART with mortality. This diminished over the next 4 years. The adjusted MRR per CD4 group was 0.97 (95% CI, .94-1.00; P = .054) and 1.02 (95% CI, .98-1.07; P = .32) among patients followed for 5-9.9 and ≥10 years, respectively. CONCLUSIONS: After surviving 5 years of ART, the mortality of patients who started ART with low baseline CD4 count converged with mortality of patients with intermediate and high baseline CD4 counts

    Tracking cell proliferation using a nanotechnology based approach

    yesTo develop an efficient nanotechnology fluorescence-based method to track cell proliferation to avoid the limitations of current cell-labeling dyes. Material & methods: Synthesis, PEGylation, bifunctionalization and labeling with a fluorophore (Cy5) of 200 nm polystyrene nanoparticles (NPs) were performed. These NPs were characterized and assessed for in vitro long-term monitoring of cell proliferation. Results: The optimization and validation of this method to track long-term cell proliferation assays have been achieved with high reproducibility, without cell cycle disruption. This method has been successfully applied in several adherent and suspension cells including hard-to-transfect cells and isolated human primary lymphocytes. Conclusion: A novel approach to track efficiently cellular proliferation by flow cytometry using fluorescence labeled NPs has been successfully developed

    Complications of Peripherally Inserted Central Venous Catheters: A Retrospective Cohort Study

    Background and Aim The use of venous catheters is a widespread practice, especially in oncological and oncohematological units. The objective of this study was to evaluate the complications associated with peripherally inserted central catheters (PICCs) in a cohort of patients. Materials and Methods In this retrospective cohort study, we included all patient carrying PICCs (n = 603) inserted at our institute between October 2010 and December 2013. The main variables collected were medical diagnosis, catheter care, location, duration of catheterization, reasons for catheter removal, complications, and nursing care. Complications were classified as infection, thrombosis, phlebitis, migration, edema, and/or ecchymosis. Results All patients were treated according to the same “nursing care” protocol. The incidence rate of complications was two cases per 1000 days of catheter duration. The most relevant complications were infection and thrombosis, both with an incidence of 0.17 cases per 1000 days of the total catheterization period. The total average duration of catheterization was 170 days [SD 6.06]. Additionally to “end of treatment” (48.42%) and “exitus”, (22.53%) the most frequent cause of removal was migration (displacement towards the exterior) of the catheter (5.80%). Conclusions PICCs are safe devices that allow the administration of long-term treatment and preserve the integrity of the venous system of the patient. Proper care of the catheter is very important to improve the quality life of patients with oncologic and hematologic conditions. Therefore, correct training of professionals and patients as well as following the latest scientific recommendations are particularly relevant
