    The use of annual killifish in the biocontrol of the aquatic stages of mosquitoes in temporary bodies of fresh water; a potential new tool in vector control

    <p>Abstract</p> <p>Background</p> <p>Mosquitoes that breed in temporary pools in remote areas that dry up seasonally are especially difficult to control through chemical or biological means. The annual killifish has been suggested as a means of eradicating the aquatic stages of mosquitoes in transient pools because they can maintain permanent populations in such habitats by undergoing suspended animation or diapause during the embryonic stages to survive periodic drought. However, very little is known about the predatory activity of annual killifish and their usefulness in mosquito control.</p> <p>Results</p> <p>The annual killifish, <it>Nothobranchius guentheri</it>, native to Tanzania, was used in this investigation. Food preference was tested under laboratory conditions by feeding juvenile killifish with 2<sup>nd </sup>instar mosquito larvae of <it>Culex quinquefasciatus </it>in the presence of alternative food sources, such as rotifers and chironomid larvae. Semi-field tests were conducted by introduction of hibernating killifish embryos and juvenile fish to artificial ponds in an outdoor open environment that allowed natural oviposition of <it>Cx. quinquefasciatus</it>. Food preference studies show that <it>N. guentheri </it>preferred to prey on mosquito larvae than either chironomid or rotifers. When hibernating killifish embryos were added to ponds simultaneously with the addition of freshwater, the embryos hatched and fed on mosquito larval population resulting in complete elimination of the immature stages. The introduction of juvenile fish to ponds with high density of mosquito larvae resulted in total eradication of the mosquito population due to predation by fish. Complete biocontrol of the mosquito larval population was achieved in the presence of 3 fish per m<sup>2 </sup>of pond surface area.</p> <p>Conclusions</p> <p>The annual killifish provides yet another tool that may be employed in the eradication diseases carried by mosquitoes through vector control, particularly in temporary bodies of freshwater. The fish can be conveniently transported in the absence of water in the form of hibernating embryos. Once introduced either as embryos or juveniles in ponds, the annual killifish can effectively reduce the larval population because of its aggressive predatory activity.</p

    Discovering novel systemic biomarkers in photos of the external eye

    External eye photos were recently shown to reveal signs of diabetic retinal disease and elevated HbA1c. In this paper, we evaluate if external eye photos contain information about additional systemic medical conditions. We developed a deep learning system (DLS) that takes external eye photos as input and predicts multiple systemic parameters, such as those related to the liver (albumin, AST); kidney (eGFR estimated using the race-free 2021 CKD-EPI creatinine equation, the urine ACR); bone & mineral (calcium); thyroid (TSH); and blood count (Hgb, WBC, platelets). Development leveraged 151,237 images from 49,015 patients with diabetes undergoing diabetic eye screening in 11 sites across Los Angeles county, CA. Evaluation focused on 9 pre-specified systemic parameters and leveraged 3 validation sets (A, B, C) spanning 28,869 patients with and without diabetes undergoing eye screening in 3 independent sites in Los Angeles County, CA, and the greater Atlanta area, GA. We compared against baseline models incorporating available clinicodemographic variables (e.g. age, sex, race/ethnicity, years with diabetes). Relative to the baseline, the DLS achieved statistically significant superior performance at detecting AST>36, calcium=300, and WBC<4 on validation set A (a patient population similar to the development sets), where the AUC of DLS exceeded that of the baseline by 5.2-19.4%. On validation sets B and C, with substantial patient population differences compared to the development sets, the DLS outperformed the baseline for ACR>=300 and Hgb<11 by 7.3-13.2%. Our findings provide further evidence that external eye photos contain important biomarkers of systemic health spanning multiple organ systems. Further work is needed to investigate whether and how these biomarkers can be translated into clinical impact

    Protective Efficacy of Menthol Propylene Glycol Carbonate Compared to N, N-diethyl-Methylbenzamide Against Mosquito Bites in Northern Tanzania.

    The reduction of malaria parasite transmission by preventing human-vector contact is critical in lowering disease transmission and its outcomes. This underscores the need for effective and long lasting arthropod/insect repellents. Despite the reduction in malaria transmission and outcomes in Tanzania, personal protection against mosquito bites is still not well investigated. This study sought to determine the efficacy of menthol propylene glycol carbonate (MR08), Ocimum suave as compared to the gold standard repellent N, N-diethyl-methylbenzamide (DEET), either as a single dose or in combination (blend), both in the laboratory and in the field against Anopheles gambiae s.l and Culex quinquefasciatus. In the laboratory evaluations, repellents were applied on one arm while the other arm of the same individual was treated with a base cream. Each arm was separately exposed in cages with unfed female mosquitoes. Repellents were evaluated either as a single dose or as a blend. Efficacy of each repellent was determined by the number of mosquitoes that landed and fed on treated arms as compared to the control or among them. In the field, evaluations were performed by human landing catches at hourly intervals from 18:00  hr to 01:00  hr. A total of 2,442 mosquitoes were collected during field evaluations, of which 2,376 (97.30%) were An. gambiae s.l while 66 (2.70%) were Cx. quinquefaciatus. MR08 and DEET had comparatively similar protective efficacy ranging from 92% to 100 for both single compound and blends. These findings indicate that MR08 has a similar protective efficacy as DEET for personal protection outside bed nets when used singly and in blends. Because of the personal protection provided by MR08, DEET and blends as topical applicants in laboratory and field situations, these findings suggest that, these repellents could be used efficiently in the community to complement existing tools. Overall, Cx. quinquefasciatus were significantly prevented from blood feeding compared to An. gambiae s.l. The incorporation of these topical repellents for protection against insect bites can be of additional value in the absence or presence of IRS and ITNs coverage. However, a combination of both the physical (bed nets) and the repellent should be used in an integrated manner for maximum protection, especially before going to bed. Additional research is needed to develop repellents with longer duration of protection

    Is there a role for the quantification of RRM1 and ERCC1 expression in pancreatic ductal adenocarcinoma?

    <p>Abstract</p> <p>Background</p> <p>RRM1 and ERCC1 overexpression has been extensively investigated as potential predictive markers of tumor sensitivity to conventional chemotherapy agents, most thoroughly in lung cancer. However, data in pancreatic cancer are scarce.</p> <p>Methods</p> <p>We investigated the mRNA and protein expression of ERCC1 and RRM1 by RT-PCR and immunohistochemistry (IHC) in formalin-fixed, paraffin-embedded pancreatic ductal carcinoma (PDA) tissues. The primary outcome investigated was the association between RRM1 and ERCC1 expression and overall survival (OS) or disease-free survival (DFS).</p> <p>Results</p> <p>A total of 94 patients with resected PDA were included in this study. Most of them (87%) received gemcitabine based chemotherapy. Data for OS analysis was available in all cases but only 68% had enough information to estimate DFS. IHC analysis revealed information for 99% (93/94) and 100% of the cases for RRM1 and ERCC1 expression respectively. However, PCR data interpretation was possible in only 49 (52%) and 79 (84%) cases respectively. There was no significant association between high or low expression of either RRM1 or ERCC1, detected by IHC and OS (14.4 vs. 19.9 months; <it>P </it>= 0.5 and 17.1 vs. 19.9; <it>P </it>= 0.83 respectively) or PCR and OS (48.0 vs. 24.1 months; <it>P </it>= 0.21 and 22.0 vs. 16.0 months; <it>P </it>= 0.39 respectively). Similar results were obtained for DFS.</p> <p>Conclusions</p> <p>RRM1 and ERCC1 expression does not seem to have a clear predictive or prognostic value in pancreatic cancer. Our data raise some questions regarding the real clinical and practical significance of analyzing these molecules as predictors of outcomes.</p

    Genotypic diversity and phenotypic spectrum of infantile liver failure syndrome type 1 due to variants inLARS1

    Purpose: Biallelic variants in LARS1, coding for the cytosolic leucyl-tRNA synthetase, cause infantile liver failure syndrome 1 (ILFS1). Since its description in 2012, there has been no systematic analysis of the clinical spectrum and genetic findings. Methods: Individuals with biallelic variants in LARS1 were included through an international, multicenter collaboration including novel and previously published patients. Clinical variables were analyzed and functional studies were performed in patient-derived fibroblasts. Results: Twenty-five individuals from 15 families were ascertained including 12 novel patients with eight previously unreported variants. The most prominent clinical findings are recurrent elevation of liver transaminases up to liver failure and encephalopathic episodes, both triggered by febrile illness. Magnetic resonance image (MRI) changes during an encephalopathic episode can be consistent with metabolic stroke. Furthermore, growth retardation, microcytic anemia, neurodevelopmental delay, muscular hypotonia, and infection-related seizures are prevalent. Aminoacylation activity is significantly decreased in all patient cells studied upon temperature elevation in vitro. Conclusion: ILFS1 is characterized by recurrent elevation of liver transaminases up to liver failure in conjunction with abnormalities of growth, blood, nervous system, and musculature. Encephalopathic episodes with seizures can occur independently from liver crises and may present with metabolic stroke

    A dense mini-Neptune orbiting the bright young star HD 18599

    Very little is known about the young planet population because the detection of small planets orbiting young stars is obscured by the effects of stellar activity and fast rotation which mask planets within radial velocity and transit data sets. The few planets that have been discovered in young clusters generally orbit stars too faint for any detailed follow-up analysis. Here we present the characterization of a new mini-Neptune planet orbiting the bright (V=9) and nearby K2 dwarf star, HD 18599. The planet candidate was originally detected in TESS light curves from Sectors 2, 3, 29, and 30, with an orbital period of 4.138 days. We then used HARPS and FEROS radial velocities, to find the companion mass to be 25.5±4.6 M⊕. When we combine this with the measured radius from TESS of 2.70±0.05 R⊕, we find a high planetary density of 7.1±1.4 g cm-3. The planet exists on the edge of the Neptune Desert and is the first young planet (300 Myr) of its type to inhabit this region. Structure models argue for a bulk composition to consist of 23 per cent H2O and 77 per cent Rock and Iron. Future follow-up with large ground- and space-based telescopes can enable us to begin to understand in detail the characteristics of young Neptunes in the galaxy

    Another Shipment of Six Short-Period Giant Planets from TESS

    We present the discovery and characterization of six short-period, transiting giant planets from NASA's Transiting Exoplanet Survey Satellite (TESS) -- TOI-1811 (TIC 376524552), TOI-2025 (TIC 394050135), TOI-2145 (TIC 88992642), TOI-2152 (TIC 395393265), TOI-2154 (TIC 428787891), & TOI-2497 (TIC 97568467). All six planets orbit bright host stars (8.9 <G< 11.8, 7.7 <K< 10.1). Using a combination of time-series photometric and spectroscopic follow-up observations from the TESS Follow-up Observing Program (TFOP) Working Group, we have determined that the planets are Jovian-sized (RP_{P} = 1.00-1.45 RJ_{J}), have masses ranging from 0.92 to 5.35 MJ_{J}, and orbit F, G, and K stars (4753 << Teff_{eff} << 7360 K). We detect a significant orbital eccentricity for the three longest-period systems in our sample: TOI-2025 b (P = 8.872 days, ee = 0.220±0.0530.220\pm0.053), TOI-2145 b (P = 10.261 days, ee = 0.1820.049+0.0390.182^{+0.039}_{-0.049}), and TOI-2497 b (P = 10.656 days, ee = 0.1960.053+0.0590.196^{+0.059}_{-0.053}). TOI-2145 b and TOI-2497 b both orbit subgiant host stars (3.8 << log\log g <<4.0), but these planets show no sign of inflation despite very high levels of irradiation. The lack of inflation may be explained by the high mass of the planets; 5.350.35+0.325.35^{+0.32}_{-0.35} MJ_{\rm J} (TOI-2145 b) and 5.21±0.525.21\pm0.52 MJ_{\rm J} (TOI-2497 b). These six new discoveries contribute to the larger community effort to use {\it TESS} to create a magnitude-complete, self-consistent sample of giant planets with well-determined parameters for future detailed studies.Comment: 20 Pages, 6 Figures, 8 Tables, Accepted by MNRA

    CANDELS: The Cosmic Assembly Near-infrared Deep Extragalactic Legacy Survey

    The Cosmic Assembly Near-infrared Deep Extragalactic Legacy Survey (CANDELS) is designed to document the first third of galactic evolution, over the approximate redshift (z) range 8--1.5. It will image >250,000 distant galaxies using three separate cameras on the Hubble Space Telescope, from the mid-ultraviolet to the near-infrared, and will find and measure Type Ia supernovae at z>1.5 to test their accuracy as standardizable candles for cosmology. Five premier multi-wavelength sky regions are selected, each with extensive ancillary data. The use of five widely separated fields mitigates cosmic variance and yields statistically robust and complete samples of galaxies down to a stellar mass of 10^9 M_\odot to z \approx 2, reaching the knee of the ultraviolet luminosity function (UVLF) of galaxies to z \approx 8. The survey covers approximately 800 arcmin^2 and is divided into two parts. The CANDELS/Deep survey (5\sigma\ point-source limit H=27.7 mag) covers \sim 125 arcmin^2 within GOODS-N and GOODS-S. The CANDELS/Wide survey includes GOODS and three additional fields (EGS, COSMOS, and UDS) and covers the full area to a 5\sigma\ point-source limit of H \gtrsim 27.0 mag. Together with the Hubble Ultra Deep Fields, the strategy creates a three-tiered "wedding cake" approach that has proven efficient for extragalactic surveys. Data from the survey are nonproprietary and are useful for a wide variety of science investigations. In this paper, we describe the basic motivations for the survey, the CANDELS team science goals and the resulting observational requirements, the field selection and geometry, and the observing design.
