    The purpose of this study was to investigate how pupils with social dysfunction were defined and described and how the responsibility of the school concerning the needs of these pupils was apprehended. For this matter, in depth interviews with four headmasters and four teachers at three different schools were carried out. The possibilities to help pupils with social dysfunction were regarded as limited due to limited resources. The school can be described, as a huge refined apparatus of classification where some children very early are eliminated and there does not appear to exist any back up plan for these individuals. In this respect the function of the school is very similar to that of yesterdays school where the elimination process worked just as well, the denotations change but the problems remain the same. The material used in this study is too small to make general conclusions but the results give some indication of the problems of the interaction between the school and its staff and pupils with social dysfunction. We strongly believe that the results warrants further investigation as to in what way the school can improve in helping pupils with social dysfunction

    Metabolic alterations in preneoplastic development revealed by untargeted metabolomic analysis

    Metabolic rewiring is a critical hallmark of tumorigenesis and is essential for the development of cancer. Although many key features of metabolic alteration that are crucial for tumor cell survival, proliferation and progression have been identified, these are obtained from studies with established tumors and cancer cell lines. However, information on the essential metabolic changes that occur during pre-neoplastic cell (PNC) development that enables its progression to full blown tumor is still lacking. Here, we present an untargeted metabolomics analysis of human oncogene HRAS(G12V) induced PNC development, using a transgenic inducible zebrafish larval skin development model. By comparison with normal sibling controls, we identified six metabolic pathways that are significantly altered during PNC development in the skin. Amongst these altered pathways are pyrimidine, purine and amino acid metabolism that are common to the cancer metabolic changes that support rapid cell proliferation and growth. Our data also suggest alterations in post transcriptional modification of RNAs that might play a role in PNC development. Our study provides a proof of principle work flow for identifying metabolic alterations during PNC development driven by an oncogenic mutation. In the future, this approach could be combined with transcriptomic or proteomic approaches to establish the detailed interaction between signaling networks and cellular metabolic pathways that occur at the onset of tumor progression

    GTN-P borehole data management towards global assessment of permafrost temperature change

    In 1999, the International Permafrost Association (IPA) established the Global Terrestrial Network for Permafrost (GTN-P, gtnp.org). The goal of the network is systematic and long-term documentation of the distribution, variability, and trends of permafrost (an Essential Climate Variable, ECV) based on a global network of field measurements. The two current cryospheric indicators are permafrost temperature and active layer thickness, throughout the Earth’s permafrost regions. The network has been mainly operated by scientist and research institutions and programs. GTN-P developed a Data Management System (gtnpdatabase.org) for the collection, processing (including standardisation), and dissemination of permafrost data and metadata. Recent ground temperature and active layer thickness data are being compiled to provide an update to the current permafrost state. GTN-P is part of the Global Climate Observing System (GCOS) Global Terrestrial Observing System (GTOS). GCOS is a joint undertaking of the World Meteorological Organization (WMO), the Intergovernmental Oceanographic Commission (IOC) of the United Nations Educational Scientific and Cultural Organization (UNESCO), the United Nations Environment Programme (UNEP) and the International Council for Science (ICSU). Permafrost temperature measurements, commonly performed with permanently installed multi-thermistor cables in boreholes, enable a good accuracy of 0.1°C. The logger resolution and measurement frequency, however, varies with the type and the depth of the individual borehole. Due to high geomorphological surface and subground dynamics, the relative vertical position of testing probes can change and bias the depth indications of old boreholes in sensitive areas. Most important quality concerns are measurement accuracy, zero annual amplitude depth, data gaps, incomplete time series, and spatial clustering of boreholes. We developed a methodological approach to filter the data by defined quality rules in order to calculate global to regional weighted averages of permafrost temperature anomalies. In this presentation we aim to give an overview on the systematical data pathway from borehole principal investigators over National Correspondents in GTN-P, followed by data processing algorithms in the GTN-P DMS towards quality checked time series data

    Hierarchical patterning modes orchestrate hair follicle morphogenesis

    Two theories address the origin of repeating patterns, such as hair follicles, limb digits, and intestinal villi, during development. The Turing reaction–diffusion system posits that interacting diffusible signals produced by static cells first define a prepattern that then induces cell rearrangements to produce an anatomical structure. The second theory, that of mesenchymal self-organisation, proposes that mobile cells can form periodic patterns of cell aggregates directly, without reference to any prepattern. Early hair follicle development is characterised by the rapid appearance of periodic arrangements of altered gene expression in the epidermis and prominent clustering of the adjacent dermal mesenchymal cells. We assess the contributions and interplay between reaction–diffusion and mesenchymal self-organisation processes in hair follicle patterning, identifying a network of fibroblast growth factor (FGF), wingless-related integration site (WNT), and bone morphogenetic protein (BMP) signalling interactions capable of spontaneously producing a periodic pattern. Using time-lapse imaging, we find that mesenchymal cell condensation at hair follicles is locally directed by an epidermal prepattern. However, imposing this prepattern’s condition of high FGF and low BMP activity across the entire skin reveals a latent dermal capacity to undergo spatially patterned self-organisation in the absence of epithelial direction. This mesenchymal self-organisation relies on restricted transforming growth factor (TGF) β signalling, which serves to drive chemotactic mesenchymal patterning when reaction–diffusion patterning is suppressed, but, in normal conditions, facilitates cell movement to locally prepatterned sources of FGF.     Influential factors of spousal relationship quality in couples living with dementia – A narrative synthesis systematic review

    Aim The aim of this systematic review is to identify factors that influence relationship quality in couples living with dementia. Previous research has shown how maintaining a positive spousal relationship quality is important for quality of life and coping for both the caregiver and the person with dementia. Knowledge of influential factors could contribute to a deeper understanding of the value of a couple-centred clinical practice and research, within the field of dementia. Research design and methods Systematic procedures to database search, screening, data extraction and synthesis were followed. Qualitative, quantitative and mixed methods studies were included. A narrative synthesis was conducted through narrative summaries of included studies, thematic analysis and narrative descriptions of factors influencing relationship quality. Results 39 studies were included in the study: 28 qualitative, 8 quantitative and 3 mixed methods. Through the narrative synthesis, 20 factors were identified. The factors were grouped into two overarching themes: The world of us and The world outside of us, and further to six influencing factor categories: (1) Attitudes and strategies, (2) Behaviour and activities, (3) Emotional connectedness, (4) Activities and experiences outside of the home, (5) Social behaviour and roles, and (6) Belonging and safety. Discussion and conclusion The identified factors influence relationship quality in couples living with dementia on various levels. The findings of this review study should inform clinical, couple-centred dementia care practise and intervention studies, and further research should seek to gain deeper understandings of the individual factors and broader understandings of the correlations between factors

    Enclosing a pen reduced time to response to questionnaire mailings

    OBJECTIVES: To assess the effectiveness of including a pen in postal questionnaires on response rate, necessity of reminders, time to response, and completeness of response to the primary outcome question (POQ).   STUDY DESIGN AND SETTING: A two-arm randomized controlled trial (RCT) embedded within the screening of older women for prevention of fracture trial (SCOOP). Women, aged 70-75 years, were randomized to receive a pen with their questionnaire (n = 3,826) or to receive the questionnaire alone (n = 3,829). The results were combined with another embedded RCT in a meta-analysis.   RESULTS: A response rate of 92.4% was observed in the pen group compared with 91.3% in the control group (odds ratio [OR] = 1.16; 95% confidence interval [CI]: 0.98, 1.37; P = 0.08). There was a difference in reminders required (OR = 0.88; 95% CI: 0.79, 0.98; P = 0.02), time to response (hazard ratio = 1.06; 95% CI: 1.01, 1.11; P = 0.01) and some difference in the completeness of response to the POQ (OR = 1.18; 95% CI: 1.00, 1.39; P = 0.05). The pooled OR from the meta-analysis for response rate was 1.21 (95% CI: 1.05, 1.39; P = 0.01).   CONCLUSION: Inclusion of a pen with postal questionnaires potentially has a positive impact on response rates and the number of reminders required. There may be some reduction in time to response. Studies of different participant groups are needed to test the effectiveness over more diverse populations

    PRL3-DDX21 transcriptional control of endolysosomal genes restricts melanocyte stem cell differentiation

    Melanocytes, replenished throughout life by melanocyte stem cells (MSCs), play a critical role in pigmentation and melanoma. Here, we reveal a function for the metastasis-associated phosphatase of regenerating liver 3 (PRL3) in MSC regeneration. We show that PRL3 binds to the RNA helicase DDX21, thereby restricting productive transcription by RNAPII at master transcription factor (MITF)-regulated endolysosomal vesicle genes. In zebrafish, this mechanism controls premature melanoblast expansion and differentiation from MSCs. In melanoma patients, restricted transcription of this endolysosomal vesicle pathway is a hallmark of PRL3-high melanomas. Our work presents the conceptual advance that PRL3-mediated control of transcriptional elongation is a differentiation checkpoint mechanism for activated MSCs and has clinical relevance for the activity of PRL3 in regenerating tissue and cancer
