Development of cyclodextrin-hydrogel polymeric systems in scCO2 for drug delivery

Thesis for the Degree of Master of Science in Bioorganic Universidade Nova de Lisboa, Faculdade de Ciências e TecnologiaThis work describes the studies on the development of new cyclodextrin-hydrogel systems in supercritical carbon dioxide (scCO2) with potential application in drug delivery. Three β-cyclodextrin (CDs) derivatives were synthesized: 6-monoacryloyl-β-CD, 2-monoacryloyl-β-CD and 6-monoacryloly-heptakis-(2,3-di-O-benzyl)-β-CD. Their structures were assigned by nuclear magnetic resonance (NMR), infrared (IR) and mass spectrometry (MS) using the technique of matrix-assisted laser desorption/ionisation-time-of-flight (MALDI-TOF). These functionalized β-CDs were co-polymerized in scCO2 and the resulting co-polymers were characterized by high resolution magnetic angle spinning (HR-MAS) NMR. Swelling tests were performed showing that the presence of CD decreases the swelling capacity of the corresponding co-polymers. The β-CD co-polymers were impregnated with a model drug, metronidazole, using a batch supercritical fluid impregnation process. Experiments in vitro were realized in order to evaluate the performance of the cyclodextrin-hydrogel system as drug release device at different pHs, 2.2 and 7.4. The co-polymer with 2.5 % of 2-monoacryloyl-β-CD was the one that impregnated more drug and showed more interesting results, since at pH 2.2 the release is more controlled. The effect of the percentage of β-CD in the co-polymers performance was also investigated. The co-polymer with more percentage of 2-monoacryloyl-β-CD (8.8%) showed a more controlled drug release at pH 7.4. The results with 2.5 % of β-CD indicate that the co-polymer would be more suitable for oral administration, whereas with 8.8 % would be suitable for parenteral administration

Gene expression regulation by upstream open reading frames in rare diseases

Upstream open reading frames (uORFs) constitute a class of cis-acting elements that regulate translation initiation. Mutations or polymorphisms that alter, create or disrupt a uORF have been widely associated with several human disorders, including rare diseases. In this mini-review, we intend to highlight the mechanisms associated with the uORF-mediated translational regulation and describe recent examples of their deregulation in the etiology of human rare diseases. Additionally, we discuss new insights arising from ribosome profiling studies and reporter assays regarding uORF features and their intrinsic role in translational regulation. Translation of ABCE1 Is Tightly Regulated by Upstream Open Reading Frames in Human Colorectal Cells

This article belongs to the Special Issue mRNA Metabolism in Health and Disease.ATP-binding cassette subfamily E member 1 (ABCE1) belongs to the ABC protein family of transporters; however, it does not behave as a drug transporter. Instead, ABCE1 actively participates in different stages of translation and is also associated with oncogenic functions. Ribosome profiling analysis in colorectal cancer cells has revealed a high ribosome occupancy in the human ABCE1 mRNA 5'-leader sequence, indicating the presence of translatable upstream open reading frames (uORFs). These cis-acting translational regulatory elements usually act as repressors of translation of the main coding sequence. In the present study, we dissect the regulatory function of the five AUG and five non-AUG uORFs identified in the human ABCE1 mRNA 5'-leader sequence. We show that the expression of the main coding sequence is tightly regulated by the ABCE1 AUG uORFs in colorectal cells. Our results are consistent with a model wherein uORF1 is efficiently translated, behaving as a barrier to downstream uORF translation. The few ribosomes that can bypass uORF1 (and/or uORF2) must probably initiate at the inhibitory uORF3 or uORF5 that efficiently repress translation of the main ORF. This inhibitory property is slightly overcome in conditions of endoplasmic reticulum stress. In addition, we observed that these potent translation-inhibitory AUG uORFs function equally in cancer and in non-tumorigenic colorectal cells, which is consistent with a lack of oncogenic function. Análise de acontecimentos que resultaram em perdas superiores ao VAR estimado para empresas do setor do retalho e das telecomunicações em Portugal

Mestrado em Econometria Aplicada e PrevisãoO objetivo desta dissertação é quantificar o Value at Risk, calculado a partir de diferentes modelos econométricos GARCH e do modelo Risk Metrics, utilizando previsões da volatilidade a 1 passo, para as empresas Jerónimo Martins, SONAE, NOS e Pharol, presentes no Índice da Bolsa de Valores de Lisboa, de forma a verificar quando é que esta métrica foi ultrapassada pelas desvalorizações dos preços das ações das cotadas procurando destacar, neste trabalho, o "porquê" dos retornos serem de tal modo negativos que até ultrapassaram o VaR. A análise foca-se no período entre dezembro de 2012 e junho de 2017 e procura apresentar eventos que possam ter provocado tais perdas.The purpose of this dissertation is to quantify the Value at Risk, calculated from different GARCH econometric models and the Risk Metrics model, using one-step volatility forecasts, for the companies Jerónimo Martins, SONAE, NOS and Pharol, quoted in the Lisbon Stock Exchange Index, to verify when this metric has been exceeded by the devaluation of the listed company's shares prices. A satisfação com o trabalho e a liderança nos bombeiros de Penafiel

Porto, Instituto Politécnico de Viana do CasteloA satisfação com o trabalho e a liderança demonstram-se fundamentais numa organização pela tradução de uma maior produtividade, influenciando a qualidade de vida do colaborador e da própria entidade empregadora, a criação de uma visão estratégica e o alcance dos objetivos. A presente investigação pretende avaliar a satisfação profissional e a liderança nos bombeiros de Penafiel. Para tal foi desenvolvido um estudo quantitativo, transversal, descritivo e correlacional. A amostra é constituída por 55 bombeiros que exercem funções na Associação Humanitária de Bombeiros Voluntários de Penafiel, obedecendo aos critérios de inclusão e à participação na amostra. A recolha de dados foi efetuada através da aplicação de um questionário constituído por três partes: caracterização sociodemográfica, profissional e de saúde da amostra, avaliação dos níveis de satisfação e avaliação da liderança na organização. Os resultados obtidos demostram que os inquiridos deste estudo apresentam um nível médio de satisfação global (média=3,850; desvio padrão=0,7500) no entanto, numa análise dimensional para a “Motivação pessoal” e a “Satisfação com a liderança de topo”, foram encontrados scores altos com média de 4,116 e desvio padrão de 0,9727 e 4,028, com desvio padrão de 0,8243, respetivamente. Esta análise permite-nos ainda constatar que o “líder democrático” foi o mais escolhido pelos colaboradores, para caracterizar o estilo de liderança presente. No que respeita à comparação entre as médias da variável satisfação profissional e a liderança, verificamos que estas não são estatisticamente significativas, uma vez que p value é superior a 0,05. Numa análise dimensional entre a satisfação e a liderança, as diferenças são significativas apenas na dimensão “Satisfação com a liderança de topo”, onde p value é inferior a 0,05. Relativamente às variáveis de caraterização, apenas se verificaram diferenças entre as médias da satisfação, em relação ao sexo e à satisfação com o rendimento, apresentando melhores pontuações os participantes do sexo feminino e aqueles que dizem estar satisfeitos com o rendimento.Satisfaction with labor and leadership prove to be fundamental in an organization by translating into greater productivity, influencing the quality of life of the employee and the employer itself, the creation of a strategic vision and the achievement of objectives. The present investigation intends to evaluate the professional satisfaction and the leadership in the firefighters of Penafiel. To this end, a quantitative, cross-sectional, descriptive and correlational study was developed. The sample consists of 55 firefighters who work in the Humanitarian Association of Volunteer Firefighters of Penafiel, complying with the inclusion criteria and participation in the sample. The data collection was carried out through the application of a questionnaire, consisting of three parts: sociodemographic, professional and health characterization of the sample, assessment of satisfaction levels and assessment of leadership in the organization. The results obtained show that the respondents of this study have an average level of overall satisfaction (average= 3,850; standard deviation= 0,7500) however, in a dimensional analysis for "Personal motivation" and "Satisfaction with top leadership", high scores were found with an average of 4,116 and a standard deviation of 0,9727, and 4,028 and a standard deviation of 0,8243, respectively. This analysis also allows us to verify that the “democratic leader” was the most selected by the employees, to characterize the present leadership style. Regarding the comparison between the means of the variables, professional satisfaction and leadership, we found that these are not statistically significant, since p is greater than 0.05. In a dimensional analysis between satisfaction and leadership, the differences are significant only in the dimension “Satisfaction with top leadership”, where p value is less than 0.05. Regarding the characterization variables, there were only differences between the means of satisfaction, in relation to gender and satisfaction with the performance, with better scores for female participants and those who say they are satisfied with their performance

Modelos para estimar taxas de retenção de clientes : aplicação a uma carteira de seguro automóvel

A comparação entre condições tarifárias de diferentes seguradoras é hoje mais frequente, com efeito nas taxas de retenção de clientes e respetivos contratos de seguro. A importância que é dada a este tema é cada vez maior e a construção de ferramentas para estimar as referidas taxas permite tomar medidas para a retenção de negócio rentável e o agravamento dos prémios de contratos menos rentáveis. Este trabalho teve como objetivo estimar a probabilidade de retenção à data de vencimento de uma apólice de seguro, numa carteira do ramo automóvel. Verificado o problema de desequilíbrio entre as classes da variável resposta, a escolha das metodologias a usar baseou-se essencialmente na procura de aumentar a exatidão do modelo final e contornar esse problema.With an increasingly easy accessibility to information, there is a growing concern about customer retention rates. Insurers are giving more importance on having accurate tools to monitor the policies renewal process, making them allowed to keep with the profitable business and increase premiums on the less profitable one. The objective of this study was to estimate the probability of renewing a policy in a motor insurance portfolio. Atenção, memória e estruturação visuo-perceptiva em crianças e adolescentes com Perturbação de Hiperactividade com Défice de Atenção (PHDA)

A Perturbação de Hiperatividade com Défice de Atenção (PHDA) é a perturbação neuro-comportamental mais frequente na infância e é caracterizada por um padrão persistente de falta de atenção e/ou impulsividade-hiperatividade. Estas dificuldades comportamentais podem resultar em défices cognitivos. Os diversos estudos publicados sobre esta temática não apresentam consenso quanto à natureza, magnitude e especificidade das funções cognitivas alteradas nesta perturbação. Deste modo, o objetivo principal neste estudo é o de identificar a existência de um perfil neuropsicológico nas crianças com PHDA dos 7 aos 15 anos. Realizou-se uma avaliação neuropsicológica nos domínios da atenção, da memória de trabalho, da estruturação visuo-perceptiva e memória visual, e do rendimento intelectual. Neste estudo, comparou-se um grupo de PHDA com um grupo de controlo, para identificar os défices no grupo de PHDA. Os resultados deste estudo mostraram que as crianças com PHDA têm maior dificuldade em provas de atenção, memória de trabalho, estruturação visuo-perceptiva e memória visual. Em relação às crianças de desenvolvimento normativo, as crianças com diagnóstico de PHDA obtiveram piores resultados em todas as provas.Attention Défices Hyperactivity Disorder (ADHD) is a neurobehavioral disorder, common in childhood, characterized by a persistent pattern of inattention and/or hyperactivity-impulsivity. These behaviors may result in cognitive difficulties. There is no consensus in the literature about the nature, magnitude and specificity of the cognitive difficulties caused by this disturbance, thus the main objective of this study is to identify the existence of a neuropsychological profile in ADHD children from 7 to 15 years through a neuropsychological assessment of the areas of attention, working memory, test of visual-perceptual ability and visual memory. In this study, we compared a group of ADHD with a control group to identify the déficess of ADHD group. The results of this study suggest that children with ADHD have difficulty in tests of attention, working memory, visual-perceptual ability and visual memory

A educação das relações raciais no currículo: as Licenciaturas em Geografia em Santa Catarina

A Geografia como campo de conhecimento contribui para o empoderamento dos sujeitos para a construção de sua identidade e de seu posicionamento e atuação no mundo. Desse modo, a discussão sobre as relações raciais é essencial para a percepção da necessidade de mudanças estruturais na sociedade brasileira e para a compreensão da segregação urbana, do direito à terra e das dinâmicas sociais e econômicas da população. Considerando esses aspectos, o artigo analisa os currículos de cursos de Licenciatura em Geografia de cinco Instituições de Ensino Superior localizadas no Estado de Santa Catarina, procurando perceber de que maneira se apresentam as questões raciais nesses cursos. Considerando que as licenciaturas têm como objetivo a formação de professores da educação básica, realizamos também uma análise dos textos legais que normatizam a formação de professores no Brasil, bem como da Geografia e os que interpretam e complementam a Lei n. 10.639/03 no que se refere a sua aplicação, buscando observar a adequação dos cursos de Geografia. A partir dos dados analisados, percebeu-se que, embora a legislação atual e suas normativas curriculares tenham assimilado no corpo de seus textos as questões raciais como estruturantes e princípios educacionais, a presença de conhecimentos e a discussão sobre estas ainda são tímidas nesses cursos, que estão, em sua maioria, cumprindo o mínimo exigido pela lei
