    The Unexpected Aftermath of Operation 'Iraqi Freedom'

    As Operation Iraqi Freedom has come to a close, the US has declared victory in achieving its objectives. Given the rapid pace of this campaign, most analysis focused on the course of events, with little academic and often inaccurate assessment of the war and its aftermath in Iraq. Such assessments are crucial when addressing the question of whether Iraq will survive as a viable state, as well as what form it will take. Faulty forecasts about Iraq's future, combined with failing to understand its internal political dynamics could have disastrous effects on both the Iraqi people and the American and UK forces stationed there

    Gabapentin for the hemodynamic response to intubation: systematic review and meta-analysis

    Purpose Endotracheal intubation is the gold standard for securing the airway before surgery. Nevertheless, this procedure can produce an activation of the sympathetic nervous system and result in a hemodynamic response which, in high-risk patients, may lead to cardiovascular instability and myocardial ischemia. The aim of this review was to evaluate whether gabapentin can attenuate this response and whether such an attenuation could translate into reduced myocardial ischemia and mortality. Source We searched MEDLINE®, EMBASE™, CINAHL, AMED, and unpublished clinical trial databases for randomized-controlled trials that compared gabapentin with control, fentanyl, clonidine, or beta blockers for attenuating the hemodynamic response to intubation. Primary outcomes were mortality, myocardial infarction, and myocardial ischemia. Secondary outcomes were hemodynamic changes following intubation. Principal findings We included 29 randomized trials with only two studies at low risk of bias. No data were provided for the primary outcomes and no studies included high-risk patients. The use of gabapentin resulted in attenuation in the rise in mean arterial blood pressure [mean difference (MD), −12 mmHg; 95% confidence interval (CI), −17 to −8] and heart rate (MD, −8 beats·min−1; 95% CI, −11 to −5) one minute after intubation. Gabapentin also reduced the risk of hypertension or tachycardia requiring treatment (risk ratio, 0.15; 95% CI, 0.05 to 0.48). Data were limited on adverse hemodynamic events such as bradycardia and hypotension. Conclusion It remains unknown whether gabapentin improves clinically relevant outcomes such as death and myocardial infarction since studies failed to report on these. Nevertheless, gabapentin attenuated increases in heart rate and blood pressure following intubation when compared with the control group. Even so, the studies included in this review were at potential risk of bias. Moreover, they did not include high-risk patients or report adverse hemodynamic outcomes. Future studies are required to address these limitations

    Meneco, a Topology-Based Gap-Filling Tool Applicable to Degraded Genome-Wide Metabolic Networks

    International audienceIncreasing amounts of sequence data are becoming available for a wide range of non-model organisms. Investigating and modelling the metabolic behaviour of those organisms is highly relevant to understand their biology and ecology. As sequences are often incomplete and poorly annotated, draft networks of their metabolism largely suffer from incompleteness. Appropriate gap-filling methods to identify and add missing reactions are therefore required to address this issue. However, current tools rely on phenotypic or taxonomic information, or are very sensitive to the stoichiometric balance of metabolic reactions, especially concerning the co-factors. This type of information is often not available or at least prone to errors for newly-explored organisms. Here we introduce Meneco, a tool dedicated to the topological gap-filling of genome-scale draft metabolic networks. Meneco reformulates gap-filling as a qualitative combinatorial optimization problem, omitting constraints raised by the stoichiometry of a metabolic network considered in other methods, and solves this problem using Answer Set Programming. Run on several artificial test sets gathering 10,800 degraded Escherichia coli networks Meneco was able to efficiently identify essential reactions missing in networks at high degradation rates, outperforming the stoichiometry-based tools in scalability. To demonstrate the utility of Meneco we applied it to two case studies. Its application to recent metabolic networks reconstructed for the brown algal model Ectocarpus siliculosus and an associated bacterium Candidatus Phaeomarinobacter ectocarpi revealed several candidate metabolic pathways for algal-bacterial interactions. Then Meneco was used to reconstruct, from transcriptomic and metabolomic data, the first metabolic network for the microalga Euglena mutabilis. These two case studies show that Meneco is a versatile tool to complete draft genome-scale metabolic networks produced from heterogeneous data, and to suggest relevant reactions that explain the metabolic capacity of a biological system

    Histone Variants and Their Post-Translational Modifications in Primary Human Fat Cells

    Epigenetic changes related to human disease cannot be fully addressed by studies of cells from cultures or from other mammals. We isolated human fat cells from subcutaneous abdominal fat tissue of female subjects and extracted histones from either purified nuclei or intact cells. Direct acid extraction of whole adipocytes was more efficient, yielding about 100 µg of protein with histone content of 60% –70% from 10 mL of fat cells. Differential proteolysis of the protein extracts by trypsin or ArgC-protease followed by nanoLC/MS/MS with alternating CID/ETD peptide sequencing identified 19 histone variants. Four variants were found at the protein level for the first time; particularly HIST2H4B was identified besides the only H4 isoform earlier known to be expressed in humans. Three of the found H2A potentially organize small nucleosomes in transcriptionally active chromatin, while two H2AFY variants inactivate X chromosome in female cells. HIST1H2BA and three of the identified H1 variants had earlier been described only as oocyte or testis specific histones. H2AFX and H2AFY revealed differential and variable N-terminal processing. Out of 78 histone modifications by acetylation/trimethylation, methylation, dimethylation, phosphorylation and ubiquitination, identified from six subjects, 68 were found for the first time. Only 23 of these modifications were detected in two or more subjects, while all the others were individual specific. The direct acid extraction of adipocytes allows for personal epigenetic analyses of human fat tissue, for profiling of histone modifications related to obesity, diabetes and metabolic syndrome, as well as for selection of individual medical treatments

    Microstructural and mechanical characterisation of laser-welded high-carbon and stainless steel

    The final publication is available at Springer via http://dx.doi.org/10.1007/s00170-015-7111-5Laser welding is becoming an important joining technique for welding of stainless steel to carbon steel and is extensively used across various sectors, including aerospace, transportation, power plants, electronics and other industries. However, welding of stainless steel to high-carbon steel is still at its early stage, predominantly due to the formation of hard brittle phases, which undermine the mechanical strength of the joint. This study reports a scientific investigation on controlling the brittle phase formation during laser dissimilar welding of high-carbon steel to stainless steel. Attempts have been made to tailor the microstructure and phase composition of the fusion zone through influencing the alloying composition and the cooling rate. Results show that the heat-affected zone (HAZ) within the high-carbon steel has significantly higher hardness than the weld area, which severely undermines the weld quality. To reduce the hardness of the HAZ, a new heat treatment strategy was proposed and evaluated using a finite element analysis-based numerical simulation model. A series of experiments has been performed to verify the developed thermo-metallurgical finite element analysis (FEA) model, and a qualitative agreement of predicted martensitic phase distribution is shown to exist

    Does Environmental Enrichment Reduce Stress? An Integrated Measure of Corticosterone from Feathers Provides a Novel Perspective

    Enrichment is widely used as tool for managing fearfulness, undesirable behaviors, and stress in captive animals, and for studying exploration and personality. Inconsistencies in previous studies of physiological and behavioral responses to enrichment led us to hypothesize that enrichment and its removal are stressful environmental changes to which the hormone corticosterone and fearfulness, activity, and exploration behaviors ought to be sensitive. We conducted two experiments with a captive population of wild-caught Clark's nutcrackers (Nucifraga columbiana) to assess responses to short- (10-d) and long-term (3-mo) enrichment, their removal, and the influence of novelty, within the same animal. Variation in an integrated measure of corticosterone from feathers, combined with video recordings of behaviors, suggests that how individuals perceive enrichment and its removal depends on the duration of exposure. Short- and long-term enrichment elicited different physiological responses, with the former acting as a stressor and birds exhibiting acclimation to the latter. Non-novel enrichment evoked the strongest corticosterone responses of all the treatments, suggesting that the second exposure to the same objects acted as a physiological cue, and that acclimation was overridden by negative past experience. Birds showed weak behavioral responses that were not related to corticosterone. By demonstrating that an integrated measure of glucocorticoid physiology varies significantly with changes to enrichment in the absence of agonistic interactions, our study sheds light on potential mechanisms driving physiological and behavioral responses to environmental change

    Fragmented realities: The ‘sectarianisation’of space among Iraqi Shias in London

    How do the spaces we inhabit shape our lived experiences? And how do those lived experiences in turn come to shape and influence our political subjectivity? Such questions are rendered all the more important in studies of migrant or diaspora populations who, by definition, conduct their daily lives in spaces and places that were initially alien to them. The way in which migrants interact with the spaces around them can tell us much about the social, political, and religious engagements they invest in, as well as the very real way in which they experience their local milieu. Through a detailed study of Iraqi Shiis living in London, specifically in the north-western borough of Brent, this article will seek to trace the ways in which religious institutions have carved up the physical and social landscape of north-west London in ways that have enduring effect on the communities with which they engage. The increasing diversification of different religious establishments, I argue, has led to a fragmentation of the city-as-lived, in which the vast majority of practising Iraqi Shiis engage with only small isolated pockets of the urban environment on a daily basis. Moreover, the growing number of specifically Shia schools, charities, mosques, community centres and other such institutions has resulted in what I call a ‘sectarianisation’ of space in Brent, in which individuals hailing from different branches of Islam inhabit different spaces within the city despite often living within metres of each other. Drawing on a mixture of interviews, participant observation, and mapping techniques, I bring together theory and practice in order to sketch out the ways migrant lives can come to be localised in certain spaces, and what that can ultimately mean in terms of their political subjectivity and engagement
