    Do intraspecific or interspecific interactions determine responses to predators feeding on a shared size‐structured prey community?

    Summary Coexistence of predators that share the same prey is common. This is still the case in size‐structured predator communities where predators consume prey species of different sizes (interspecific prey responses) or consume different size classes of the same species of prey (intraspecific prey responses). A mechanism has recently been proposed to explain coexistence between predators that differ in size but share the same prey species, emergent facilitation, which is dependent on strong intraspecific responses from one or more prey species. Under emergent facilitation, predators can depend on each other for invasion, persistence or success in a size‐structured prey community. Experimental evidence for intraspecific size‐structured responses in prey populations remains rare, and further questions remain about direct interactions between predators that could prevent or limit any positive effects between predators [e.g. intraguild predation (IGP)]. Here, we provide a community‐wide experiment on emergent facilitation including natural predators. We investigate both the direct interactions between two predators that differ in body size (fish vs. invertebrate predator), and the indirect interaction between them via their shared prey community (zooplankton). Our evidence supports the most likely expectation of interactions between differently sized predators that IGP rates are high, and interspecific interactions in the shared prey community dominate the response to predation (i.e. predator‐mediated competition). The question of whether emergent facilitation occurs frequently in nature requires more empirical and theoretical attention, specifically to address the likelihood that its pre‐conditions may co‐occur with high rates of IGP. </jats:p

    Citizen meets social science : Predicting volunteer involvement in a global freshwater monitoring experiment

    FreshWater Watch is a global citizen science project that seeks to advance the understanding and stewardship of freshwater ecosystems across the globe through analysis of their physical and chemical properties by volunteers. To date, literature concerning citizen science has mainly focused on its potential to generate unprecedented volumes of data. In this paper, we focus instead on the data relating to the volunteer experience and ask key questions about volunteer engagement with the project. For example, we ask what factors influence: a) volunteer data submission following a training event and b) the number of water quality samples volunteers subsequently submit. We used a binomial model to identify the factors that influence the retention of volunteers after training. In addition, we used a generalized linear model (GLM) to examine the factors that affected the number of samples each citizen scientist submitted. In line with other citizen science projects, most people trained did not submit any data, and 1% of participants contributed 47% of the data. We found that the statistically significant factors associated with submission of data after training were: whether training was given on how to upload data, the number of volunteers that attended the training, whether the volunteer was assigned to a research team, the outside temperature, and the average engagement of others in the training group. The statistically significant factors associated with the quantity of data submitted were: the length of time volunteers were active in the project, whether training took place as part of a paid work day, the difficulty of the sampling procedure, how socially involved volunteers were in the project, average sampling group size, and engagement with online learning modules. Based on our results, we suggest that intrinsic motivation may be important for predicting volunteer retention after training and the number of samples collected subsequently. We suggest that, to maximize the contribution of citizen science to our understanding of the world around us, there is an urgent need to better understand the factors that drive volunteer retention and engagement

    EBP-Colombia and the bioeconomy: Genomics in the service of biodiversity conservation and sustainable development

    The 2016 Peace Agreement has increased access to Colombia’s unique ecosystems, which remain understudied and increasingly under threat. The Colombian government has recently announced its National Bioeconomic Strategy (NBS), founded on the sustainable characterization, management, and conservation of the nation's biodiversity as a means to achieve sustainability and peace. Molecular tools will accelerate such endeavors, but capacity remains limited in Colombia. The Earth Biogenome Project's (EBP) objective is to characterize the genomes of all eukaryotic life on Earth through networks of partner institutions focused on sequencing either specific taxa or eukaryotic communities at regional or national scales. Colombia’s immense biodiversity and emerging network of stakeholders have inspired the creation of the national partnership “EBP-Colombia.” Here, we discuss how this Colombian-driven collaboration between government, academia, and the private sector is integrating research with sustainable, environmentally focused strategies to develop Colombia’s postconflict bioeconomy and conserve biological and cultural diversity. EBP-Colombia will accelerate the uptake of technology and promote partnership and exchange of knowledge among Colombian stakeholders and the EBP’s global network of experts; assist with conservation strategies to preserve Colombia’s vast biological wealth; and promote innovative approaches among public and private institutions in sectors such as agriculture, tourism, recycling, and medicine. EBP-Colombia can thus support Colombia’s NBS with the objective of sustainable and inclusive development to address the many social, environmental, and economic challenges, including conflict, inequality, poverty, and low agricultural productivity, and so, offer an alternative model for economic development that similarly placed countries can adopt

    A hypoxia biomarker does not predict benefit from giving chemotherapy with radiotherapy in the BC2001 randomised controlled trial.

    BACKGROUND: BC2001 showed combining chemotherapy (5-FU + mitomycin-C) with radiotherapy improves loco-regional disease-free survival in patients with muscle-invasive bladder cancer (MIBC). We previously showed a 24-gene hypoxia-associated signature predicted benefit from hypoxia-modifying radiosensitisation in BCON and hypothesised that only patients with low hypoxia scores (HSs) would benefit from chemotherapy in BC2001. BC2001 allowed conventional (64Gy/32 fractions) or hypofractionated (55Gy/20 fractions) radiotherapy. An exploratory analysis tested an additional hypothesis that hypofractionation reduces reoxygenation and would be detrimental for patients with hypoxic tumours. METHODS: RNA was extracted from pre-treatment biopsies (298 BC2001 patients), transcriptomic data generated (Affymetrix Clariom-S arrays), HSs calculated (median expression of 24-signature genes) and patients stratified as hypoxia-high or -low (cut-off: cohort median). PRIMARY ENDPOINT: invasive loco-regional control (ILRC); secondary overall survival. FINDINGS: Hypoxia affected overall survival (HR = 1.30; 95% CI 0.99-1.70; p = 0.062): more uncertainty for ILRC (HR = 1.29; 95% CI 0.82-2.03; p = 0.264). Benefit from chemotherapy was similar for patients with high or low HSs, with no interaction between HS and treatment arm. High HS associated with poor ILRC following hypofractionated (n = 90, HR 1.69; 95% CI 0.99-2.89 p = 0.057) but not conventional (n = 207, HR 0.70; 95% CI 0.28-1.80, p = 0.461) radiotherapy. The finding was confirmed in an independent cohort (BCON) where hypoxia associated with a poor prognosis for patients receiving hypofractionated (n = 51; HR 14.2; 95% CI 1.7-119; p = 0.015) but not conventional (n = 24, HR 1.04; 95% CI 0.07-15.5, p = 0.978) radiotherapy. INTERPRETATION: Tumour hypoxia status does not affect benefit from BC2001 chemotherapy. Hypoxia appears to affect fractionation sensitivity.     Task sharing in Zambia: HIV service scale-up compounds the human resource crisis

    BACKGROUND: Considerable attention has been given by policy makers and researchers to the human resources for health crisis in Africa. However, little attention has been paid to quantifying health facility-level trends in health worker numbers, distribution and workload, despite growing demands on health workers due to the availability of new funds for HIV/AIDS control scale-up. This study analyses and reports trends in HIV and non-HIV ambulatory service workloads on clinical staff in urban and rural district level facilities. METHODS: Structured surveys of health facility managers, and health services covering 2005-07 were conducted in three districts of Zambia in 2008 (two urban and one rural), to fill this evidence gap. Intra-facility analyses were conducted, comparing trends in HIV and non-HIV service utilisation with staff trends. RESULTS: Clinical staff (doctors, nurses and nurse-midwives, and clinical officers) numbers and staff population densities fell slightly, with lower ratios of staff to population in the rural district. The ratios of antenatal care and family planning registrants to nurses/nurse-midwives were highest at baseline and increased further at the rural facilities over the three years, while daily outpatient department (OPD) workload in urban facilities fell below that in rural facilities. HIV workload, as measured by numbers of clients receiving antiretroviral treatment (ART) and prevention of mother to child transmission (PMTCT) per facility staff member, was highest in the capital city, but increased rapidly in all three districts. The analysis suggests evidence of task sharing, in that staff designated by managers as ART and PMTCT workers made up a higher proportion of frontline service providers by 2007. CONCLUSIONS: This analysis of workforce patterns across 30 facilities in three districts of Zambia illustrates that the remarkable achievements in scaling-up HIV/AIDS service delivery has been on the back of sustained non-HIV workload levels, increasing HIV workload and stagnant health worker numbers. The findings are based on an analysis of routine data that are available to district and national managers. Mixed methods research is needed, combining quantitative analyses of routine health information with follow-up qualitative interviews, to explore and explain workload changes, and to identify and measure where problems are most acute, so that decision makers can respond appropriately. This study provides quantitative evidence of a human resource crisis in health facilities in Zambia, which may be more acute in rural areas

    Multiple novel prostate cancer susceptibility signals identified by fine-mapping of known risk loci among Europeans

    Genome-wide association studies (GWAS) have identified numerous common prostate cancer (PrCa) susceptibility loci. We have fine-mapped 64 GWAS regions known at the conclusion of the iCOGS study using large-scale genotyping and imputation in 25 723 PrCa cases and 26 274 controls of European ancestry. We detected evidence for multiple independent signals at 16 regions, 12 of which contained additional newly identified significant associations. A single signal comprising a spectrum of correlated variation was observed at 39 regions; 35 of which are now described by a novel more significantly associated lead SNP, while the originally reported variant remained as the lead SNP only in 4 regions. We also confirmed two association signals in Europeans that had been previously reported only in East-Asian GWAS. Based on statistical evidence and linkage disequilibrium (LD) structure, we have curated and narrowed down the list of the most likely candidate causal variants for each region. Functional annotation using data from ENCODE filtered for PrCa cell lines and eQTL analysis demonstrated significant enrichment for overlap with bio-features within this set. By incorporating the novel risk variants identified here alongside the refined data for existing association signals, we estimate that these loci now explain ∼38.9% of the familial relative risk of PrCa, an 8.9% improvement over the previously reported GWAS tag SNPs. This suggests that a significant fraction of the heritability of PrCa may have been hidden during the discovery phase of GWAS, in particular due to the presence of multiple independent signals within the same regio

    Testing the efficacy of river restoration across multiple levels of biological organisation

    As we start to count the cost of unprecedented biodiversity loss in terms of impaired provisional, supporting and cultural ecosystem goods and services, so to are we increasingly implementing measures to halt and reverse these impacts. A major focus of these efforts has been the restoration river habitats, which rank among the most degraded globally. However, while river restoration has become a global enterprise, biomonitoring to test the ecological effectiveness of these activities remains exceptional and even then typically lacking the rigour necessary for detecting and tracking ecological responses across space and time. Here I use standardised and quantitative biomonitoring techniques to measure the ecological response to replicated, experimental, reach-scale large wood installations in heavily modified calcareous system. In Chapters 3-5, I used biannual biomonitoring to investigate the response of a target fish species and the wider fish assemblage to restoration and found strong, rapid responses in the target species’ predators (Chapter 3). In Chapter 4, I build food webs revealing changes to the food web structure that indicate enhanced energy transfer between predators and prey following restoration. In Chapter 5 I investigate the effect of restoration on ecosystem functioning, by comparing leaf-litter breakdown rates and colonisation in restored and unrestored reaches. In Chapter 6, I discuss how engaging river users as citizen scientists could advance the ecological success of their interventions and restoration science. The findings presented here highlight the importance of robust biomonitoring methods for revealing, measuring and characterising the response of ecological communities to habitat interventions. Repairing degraded ecosystems is among the most necessary and exciting challenges we have undertaken as a species, and only by advancing our understanding the factors that influence restoration success and failure will we be able to advance the practice of restoration towards an ever more efficient and ecologically effective management practice.Open Acces
