    Productividad secundaria en los esteros de la barra de arena El Mogote en la Ensenada de La Paz, Baja California Sur, México

    Mangrove ecosystems are critical habitats for many species, and their productivity is important because of their contribution of organic detritus and secondary production. The objective of this study was to identify suitable indicators of secondary productivity of mangrove systems, in order to establish a baseline for measuring changes in the mangrove system due to natural or anthropogenic potential disturbances. The bivalve mollusks chosen as indicators of secondary productivity were the mangrove cockle Anadara tuberculosa and the mangrove oyster Crassostrea palmula. Samples were collected in three estuaries from August 2007 to July 2009. Abundance and secondary production were estimated from biomass increase, calculated from individual growth rates determined using length frequency distributions analysis. The average abundance was 1.27 and 510 ind · m-2 of mangrove cockle and mangrove oyster respectively. It was estimated a growth rate of 4.91 mm · month-1 (3.67 g · month-1 ) for mangrove cockle and 3.80 mm · month-1 (0.18 g · month-1 ) for the mangrove oyster. The mean secondary production of mangrove cockle was 4.51 g · m-2 · month-1 and was higher during spring, while for the mangrove oyster it was 97.9 g · m-2 · month-1 , and the maximum production was recorded during summer. The results show that these indicators of benthos quality are suitable for measuring changes of the mangrove system in the Ensenada of La Paz, B. C. S., Mexico.Los ecosistemas de manglar son un hábitat crítico para numerosas especies y su productividad es importante por su contribución de detritos orgánicos y su producción secundaria. El objetivo del trabajo fue definir indicadores de productividad secundaria para establecer una línea base que permita medir alteraciones en el sistema de manglar por posibles perturbaciones naturales o antropogénicas. Como indicadores se escogieron los moluscos bivalvos pata de mula, Anadara tuberculosa y el ostión de mangle, Crassostrea palmula. Se realizaron muestreos en tres esteros de agosto de 2007 a julio de 2009. Se estimó la abundancia y la producción secundaria con base en el incremento de biomasa. Para la estimación del incremento en peso se usaron las tasas de crecimiento individual obtenidas mediante un análisis de distribuciones de frecuencia de tallas. La abundancia promedio fue de 1.27 y 510 ind · m-2 de la pata de mula y ostión de mangle respectivamente. Se estimó una tasa de crecimiento de 4.91 mm · mes-1 (3.67 g · mes-1 ) para la pata de mula y de 3.80 mm · mes-1 (0.18 g · mes-1 ) para el ostión de mangle. La producción secundaria promedio de pata de mula fue de 4.51 g · m-2 · mes-1 y fue mayor durante la primavera, mientras que para el ostión de mangle fue de 97.9 g · m-2 · mes-1 , registrándose la máxima producción durante el verano. Los resultados obtenidos demuestran que estos indicadores de calidad del bentos son adecuados para medir alteraciones al sistema de manglar de la Ensenada de La Paz, B. C. S., México

    Asociaciones de larvas de peces en relación a cambios ambientales en las Bahías Chamela, Jalisco y Santiago-Manzanillo, Colima (2001-2002)

    Spatial and temporal structure of fish larvae associations and their relationship to the environmental variability were studied at Bahía Chamela, Jalisco, and Santiago-Manzanillo complex, Colima, during 2001-2002. Two spatial aggregations were found, related to the coastal physiography and to the adult habitat. The first was observed at Bahía Chamela, which integrated a combination of species belonging to coastal, demersal and inshore habitats (Bregmaceros bathymaster, Bairdiella sp., Cynoscion sp., Engraulis mordax, and Labrisomidae type 1) and the other in Santiago-Manzanillo composed by species from lagoon and inshore areas (Eucinostomus sp., Abudefduf troschelii, Haemulidae type 2 and 3, and Enneanectes sexmaculatus). Temporal organization was influenced by the environmental shift between two periods (warm and temperate) defined by the regional environmental variability. The warm period was characterized by tropical species (Harengula thrissina, Eucinostomus sp., Auxis sp., and Haemulidae type 3), related to high temperature and rainfall, while temperate period was dominated by species with a tropical-subtropical affinity (B. bathymaster, Bairdiella sp., Cynoscion sp., and E. mordax) and related to lower temperature and elevated nutrients, chlorophyll a and plankton concentrations.Se analizó la influencia de los factores ambientales sobre las asociaciones espacio-temporales de larvas de peces frente a Bahía Chamela, Jalisco y el conjunto de bahías Santiago-Manzanillo, Colima, durante un ciclo anual (2001-2002). Se identificaron dos agrupaciones relacionadas con la fisiografía de la costa y el hábitat de los adultos, una frente a Bahía Chamela integrada por una mezcla de especies de hábitats costero, demersal e intermareal-submareal (Bregmaceros bathymaster, Bairdiella sp., Cynoscion sp., Engraulis mordax y Labrisomidae tipo 1) y otra en Santiago-Manzanillo compuesta por especies de origen lagunar e intermareal-submareal (Eucinostomus sp., Abudefduf troschelii, Haemulidae tipos 2 y 3, así como Enneanectes sexmaculatus). La organización temporal estuvo influenciada por la alternancia entre dos periodos (cálido y templado) definidos por la variabilidad ambiental regional. El periodo cálido estuvo caracterizado por especies principalmente tropicales (Harengula thrissina, Eucinostomus sp., Auxis sp. y Haemulidae tipo 3), relacionadas con una mayor temperatura y precipitación pluvial, en cambio durante el periodo templado dominaron las especies de afinidad tropical-subtropical (B. bathymaster, Bairdiella sp., Cynoscion sp. y E. mordax), relacionadas con una menor temperatura e incrementos en las concentraciones de nutrimentos, clorofila-a y zooplancton

    Impact of uterine manipulator on oncological outcome in endometrial cancer surgery

    Background: There are limited data available to indicate whether oncological outcomes might be influenced by the uterine manipulator, which is used at the time of hysterectomy for minimally invasive surgery in patients with endometrial cancer. The current evidence derives from retrospective studies with limited sample sizes. Without substantial evidence to support its use, surgeons are required to make decisions about its use based only on their personal choice and surgical experience. Objective: To evaluate the use of the uterine manipulator on oncological outcomes after minimally invasive surgery, for apparent early-stage endometrial cancer. Study Design: We performed a retrospective multicentric study to assess the oncological safety of uterine manipulator use in patients with apparent early-stage endometrial cancer, treated with minimally invasive surgery. The type of manipulator, surgical staging, histology, lymphovascular space invasion, International Federation of Gynecology and Obstetrics stage, adjuvant treatment, recurrence, and pattern of recurrence were evaluated. The primary objective was to determine the relapse rate. The secondary objective was to determine recurrence-free survival, overall survival, and the pattern of recurrence. Results: A total of 2661 women from 15 centers were included; 1756 patients underwent hysterectomy with a uterine manipulator and 905 without it. Both groups were balanced with respect to histology, tumor grade, myometrial invasion, International Federation of Gynecology and Obstetrics stage, and adjuvant therapy. The rate of recurrence was 11.69% in the uterine manipulator group and 7.4% in the no-manipulator group (P<.001). The use of the uterine manipulator was associated with a higher risk of recurrence (hazard ratio, 2.31; 95% confidence interval, 1.27–4.20; P=.006). The use of uterine manipulator in uterus-confined endometrial cancer (International Federation of Gynecology and Obstetrics [FIGO] I–II) was associated with lower disease-free survival (hazard ratio, 1.74; 95% confidence interval, 0.57–0.97; P=.027) and higher risk of death (hazard ratio, 1.74; 95% confidence interval, 1.07–2.83; P=.026). No differences were found regarding the pattern of recurrence between both groups (chi-square statistic, 1.74; P=.63). Conclusion: In this study, the use of a uterine manipulator was associated with a worse oncological outcome in patients with uterus-confined endometrial cancer (International Federation of Gynecology and Obstetrics I–II) who underwent minimally invasive surgery. Prospective trials are essential to confirm these results

    Dendritic cell deficiencies persist seven months after SARS-CoV-2 infection

    Severe Acute Respiratory Syndrome Coronavirus (SARS-CoV)-2 infection induces an exacerbated inflammation driven by innate immunity components. Dendritic cells (DCs) play a key role in the defense against viral infections, for instance plasmacytoid DCs (pDCs), have the capacity to produce vast amounts of interferon-alpha (IFN-α). In COVID-19 there is a deficit in DC numbers and IFN-α production, which has been associated with disease severity. In this work, we described that in addition to the DC deficiency, several DC activation and homing markers were altered in acute COVID-19 patients, which were associated with multiple inflammatory markers. Remarkably, previously hospitalized and nonhospitalized patients remained with decreased numbers of CD1c+ myeloid DCs and pDCs seven months after SARS-CoV-2 infection. Moreover, the expression of DC markers such as CD86 and CD4 were only restored in previously nonhospitalized patients, while no restoration of integrin β7 and indoleamine 2,3-dyoxigenase (IDO) levels were observed. These findings contribute to a better understanding of the immunological sequelae of COVID-19

    SARS-CoV-2 viral load in nasopharyngeal swabs is not an independent predictor of unfavorable outcome

    The aim was to assess the ability of nasopharyngeal SARS-CoV-2 viral load at first patient’s hospital evaluation to predict unfavorable outcomes. We conducted a prospective cohort study including 321 adult patients with confirmed COVID-19 through RT-PCR in nasopharyngeal swabs. Quantitative Synthetic SARS-CoV-2 RNA cycle threshold values were used to calculate the viral load in log10 copies/mL. Disease severity at the end of follow up was categorized into mild, moderate, and severe. Primary endpoint was a composite of intensive care unit (ICU) admission and/or death (n = 85, 26.4%). Univariable and multivariable logistic regression analyses were performed. Nasopharyngeal SARS-CoV-2 viral load over the second quartile (≥ 7.35 log10 copies/mL, p = 0.003) and second tertile (≥ 8.27 log10 copies/mL, p = 0.01) were associated to unfavorable outcome in the unadjusted logistic regression analysis. However, in the final multivariable analysis, viral load was not independently associated with an unfavorable outcome. Five predictors were independently associated with increased odds of ICU admission and/or death: age ≥ 70 years, SpO2, neutrophils > 7.5 × 103/µL, lactate dehydrogenase ≥ 300 U/L, and C-reactive protein ≥ 100 mg/L. In summary, nasopharyngeal SARS-CoV-2 viral load on admission is generally high in patients with COVID-19, regardless of illness severity, but it cannot be used as an independent predictor of unfavorable clinical outcome

    Cross-disease Meta-analysis of Genome-wide Association Studies for Systemic Sclerosis and Rheumatoid Arthritis Reveals IRF4 as a New Common Susceptibility Locus

    Objectives: Systemic sclerosis (SSc) and rheumatoid arthritis (RA) are autoimmune diseases that share clinical and immunological characteristics. To date, several shared SSc- RA loci have been identified independently. In this study, we aimed to systematically search for new common SSc-RA loci through an inter-disease meta-GWAS strategy. Methods: We performed a meta-analysis combining GWAS datasets of SSc and RA using a strategy that allowed identification of loci with both same-direction and opposingdirection allelic effects. The top single-nucleotide polymorphisms (SNPs) were followed-up in independent SSc and RA case-control cohorts. This allowed us to increase the sample size to a total of 8,830 SSc patients, 16,870 RA patients and 43,393 controls. Results: The cross-disease meta-analysis of the GWAS datasets identified several loci with nominal association signals (P-value < 5 x 10-6), which also showed evidence of association in the disease-specific GWAS scan. These loci included several genomic regions not previously reported as shared loci, besides risk factors associated with both diseases in previous studies. The follow-up of the putatively new SSc-RA loci identified IRF4 as a shared risk factor for these two diseases (Pcombined = 3.29 x 10-12). In addition, the analysis of the biological relevance of the known SSc-RA shared loci pointed to the type I interferon and the interleukin 12 signaling pathways as the main common etiopathogenic factors. Conclusions: Our study has identified a novel shared locus, IRF4, for SSc and RA and highlighted the usefulness of cross-disease GWAS meta-analysis in the identification of common risk loci

    RICORS2040 : The need for collaborative research in chronic kidney disease

    Chronic kidney disease (CKD) is a silent and poorly known killer. The current concept of CKD is relatively young and uptake by the public, physicians and health authorities is not widespread. Physicians still confuse CKD with chronic kidney insufficiency or failure. For the wider public and health authorities, CKD evokes kidney replacement therapy (KRT). In Spain, the prevalence of KRT is 0.13%. Thus health authorities may consider CKD a non-issue: very few persons eventually need KRT and, for those in whom kidneys fail, the problem is 'solved' by dialysis or kidney transplantation. However, KRT is the tip of the iceberg in the burden of CKD. The main burden of CKD is accelerated ageing and premature death. The cut-off points for kidney function and kidney damage indexes that define CKD also mark an increased risk for all-cause premature death. CKD is the most prevalent risk factor for lethal coronavirus disease 2019 (COVID-19) and the factor that most increases the risk of death in COVID-19, after old age. Men and women undergoing KRT still have an annual mortality that is 10- to 100-fold higher than similar-age peers, and life expectancy is shortened by ~40 years for young persons on dialysis and by 15 years for young persons with a functioning kidney graft. CKD is expected to become the fifth greatest global cause of death by 2040 and the second greatest cause of death in Spain before the end of the century, a time when one in four Spaniards will have CKD. However, by 2022, CKD will become the only top-15 global predicted cause of death that is not supported by a dedicated well-funded Centres for Biomedical Research (CIBER) network structure in Spain. Realizing the underestimation of the CKD burden of disease by health authorities, the Decade of the Kidney initiative for 2020-2030 was launched by the American Association of Kidney Patients and the European Kidney Health Alliance. Leading Spanish kidney researchers grouped in the kidney collaborative research network Red de Investigación Renal have now applied for the Redes de Investigación Cooperativa Orientadas a Resultados en Salud (RICORS) call for collaborative research in Spain with the support of the Spanish Society of Nephrology, Federación Nacional de Asociaciones para la Lucha Contra las Enfermedades del Riñón and ONT: RICORS2040 aims to prevent the dire predictions for the global 2040 burden of CKD from becoming true

    Complement component C4 structural variation and quantitative traits contribute to sex-biased vulnerability in systemic sclerosis

We investigated the association of C4 CN with systemic sclerosis (SSc) risk. Imputed total C4, C4A, C4B, and HERV-K CN were analyzed in 26,633 individuals and validated in an independent cohort. Our results showed that higher C4 CN confers protection to SSc, and deviations from CN parity of C4A and C4B augmented risk. The protection contributed per copy of C4A and C4B differed by sex. Stronger protection was afforded by C4A in men and by C4B in women. C4 CN correlated well with its gene expression and serum protein levels, and less C4 was detected for both in SSc patients. Conditioned analysis suggests that C4 genetics strongly contributes to the SSc association within the major histocompatibility complex locus and highlights classical alleles and amino acid variants of HLA-DRB1 and HLA-DPB1 as C4-independent signals

    New insights into the genetic etiology of Alzheimer's disease and related dementias

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele
