    Evaluación del efecto sinergico del inhibidor de la "kinesin spindle protein" filanesib (arry-520) en combinación con fármacos inmunomoduladores en modelos preclínicos de mieloma múltiple

    [ES] En el presente trabajo de se ha evaluado preclínicamente la eficacia y el mecanismo de acción de un nuevo fármaco (filanesib, ARRY-520) en combinación con agentes inmunomoduladores junto con dexametasona en mielloma múltiple, una enfermedad considerada a día de hoy incurable. Filanesib es un fármaco que ha mostrado eficacia en pacientes altamente tratados. Los resultados de nuestro trabajo han conseguido identificar una combinación con este fármaco aún más eficaz en MM y han sentado las bases para el ensayo clínico de la misma en pacientes en recaida o refractarios a tratamientos, este ensayo clínico denominado POMDEFIL se está realizando en el contexto del grupo español de mieloma múltiple (GEM). Así mismo estos datos se han publicado en una revista europea recientemente y favorecen la aplicación de una medicina personalizada, identificando al grupo de pacientes que se puede beneficiar del tratamiento (el grupo de peor pronóstico, con mielomas muy proliferativos)

    The activation of the Sox2 RR2 pluripotency transcriptional reporter in human breast cancer cell lines is dynamic and labels cells with higher tumorigenic potential

    This is an open-access article distributed under the terms of the Creative Commons Attribution License (CC BY).-- et al.The striking similarity displayed at the mechanistic level between tumorigenesis and the generation of induced pluripotent stem cells and the fact that genes and pathways relevant for embryonic development are reactivated during tumor progression highlights the link between pluripotency and cancer. Based on these observations, we tested whether it is possible to use a pluripotency-associated transcriptional reporter, whose activation is driven by the SRR2 enhancer from the Sox2 gene promoter (named S4+ reporter), to isolate cancer stem cells (CSCs) from breast cancer cell lines. The S4+ pluripotency transcriptional reporter allows the isolation of cells with enhanced tumorigenic potential and its activation was switched on and offin the cell lines studied, reflecting a plastic cellular process. Microarray analysis comparing the populations in which the reporter construct is active versus inactive showed that positive cells expressed higher mRNA levels of cytokines (IL-8, IL-6, TNF) and genes (such as ATF3, SNAI2, and KLF6) previously related with the CSC phenotype in breast cancer.This work is supported by grants from Obra Social Kutxa, Gobierno Vasco (Saiotek program and Consejería de Educación PI2010-25), and Instituto de Salud Carlos III Acción Estratégica en Salud (PI2010-01035).     Percepción y satisfacción sobre la insulinoterapia en pacientes con diabetes mellitus tipo 2

    Introducción: Existen múltiples barreras que pueden afectar la adherencia al tratamiento con insulina en pacientes con diabetes mellitus tipo 2, entre ellas, aquellas intrínsecas al tipo de insulina, las asociadas a los conocimientos y abordaje clínico del médico, y las barreras biopsicosociales del paciente, como sus propias experiencias con el uso, los mitos existentes hacia la insulinoterapia, la percepción personal y cultural, entre otras cosas. Es indispensable abordar todas las esferas que puedan afectar la adherencia, control y satisfacción hacia el tratamiento, así como la calidad de vida que tienen los pacientes que utilizan esta terapia o que la vayan a requerir. Objetivo: Conocer la percepción y satisfacción hacia la insulinoterapia que tienen los pacientes con diabetes mellitus tipo 2 que utilizan insulina actualmente o estén utilizando antidiabéticos orales en su esquema terapéutico, así como su efecto sobre la satisfacción. Material y métodos: Se trata de un estudio transversal, prospectivo, observacional, y comparativo. Se invitaron a pacientes mayores de 18 años con diagnóstico de diabetes mellitus tipo 2 en manejo con insulina o antidiabéticos orales, que acudieron a nuestro Departamento de Medicina Familiar durante el periodo de julio a noviembre de 2020. Se obtuvieron características sociodemográficas y comorbilidades de la población. Para medir la percepción, se utilizó la escala de percepción del tratamiento con insulina (ITAS), instrumento validado en población mexicana con un alfa de Cronbach de 0.85, conformado por 16 ítems de percepción negativa y 4 ítems de percepción positiva, en escala tipo Likert donde un mayor puntaje significa mayor percepción positiva y negativa, de acuerdo con la dimensión y un mayor puntaje global en la escala representa mayor percepción negativa. Para la medición de la satisfacción con el tratamiento, se utilizó el instrumento DTSQ (The Diabetes Treatment Satisfaction Questionnaire) validado en población española con un alfa de Cronbach de 0.90, constituido por 8 ítems que permiten 7 posibles respuestas, que oscilan entre 0 (muy insatisfecho) y 6 (satisfecho) puntos cada uno. Se realizó un muestro no probabilístico de casos consecutivos. Resultados: El 58% de los pacientes fueron manejados con insulinoterapia para su control glucémico, y el 42% con fármacos antidiabéticos orales. Las principales comorbilidades del paciente con diabetes son hipertensión, obesidad y dislipidemias y la principal discapacidad fue la visual. Los pacientes con manejo de insulinoterapia tienen mayor satisfacción con el tratamiento y mejor percepción de la terapia, comparado con los pacientes con manejo farmacológico oral. Encontramos que los pacientes con insulinoterapia muestran puntajes más altos de satisfacción positiva y menor puntaje de satisfacción negativa. También observamos una asociación inversamente proporcional moderada entre la satisfacción y la percepción negativa. Conclusión: Los pacientes con tratamiento con insulina tienen mayor satisfacción y mejor percepción del tratamiento que los pacientes con antidiabéticos orales

    Eclectic Methodology and Intercultural Mediation in Teaching Initial Spanish to Native Chinese Speakers

    The aim of this study is to know the appraisal of the agents involved in the implementation of a course designed for developing the oral communication effectiveness of Chinese workers. For the course, we advocated an eclectic methodology in which teaching resources of different kinds were applied to optimize language learning considering the specific needs of the learners. We decided the participation of two teachers per group and a linguistic and cultural Chinese mediator and we also took into account the methodology currently followed in the Chinese centers: the traditional approach (grammar-translation) and structural (Yang, 2013). In this observational descriptive study, the course participants (6 teachers, 6 assistants and 59 students) answered a questionnaire about different factors such as methodology, design of materials, resources used, use of the mother tongue, learners reactions to certain tasks, role of teachers and assistants.     Chain‑End Functional di‑Sorbitan Oleate Monomer Obtained from Renewable Resources as Precursors for Bio‑Based Polyurethanes

    Artículo derivado de una tesis de MCMA three-step synthetic route was proposed and tested to obtain a chain-end functional di-sorbitan oleate monomer: First, 1,18-octadec-9-enedioic acid compound was produced by self-metathesis reaction of an oleic acid; then, the 1,18-octadec 9-enoyl dichloride compound was yielded by chlorination of the di-acid with thionyl chloride, and fnally, the 1,18-di-sorbitan oleate monomer was yielded by esterifcation of the dichloride with 1,4-sorbitan. The di-sorbitan oleate monomer was purifed and then characterized by FTIR, 1 H-NMR, DSC and TGA to establish its structure and properties. A bio-based polyurethane (PU) was synthesized by reacting the obtained 1,18-di-sorbitan oleate monomer and MDI. Rheological analysis showed that a curing reaction occurs as a signifcant increase of the storage modulus (G’) and the complex viscosity (η*) at 100 °C. The obtained bio-based PU was characterized by FTIR, TGA and DMA, confrming that 1,18-di-sorbitan oleate is a feasible monomer for synthesizing polyurethanes.Conacyt CB-2015–01-257591

    Antitumoral effect of Ocoxin on acute myeloid leukemia

    This is an open-access article distributed under the terms of the Creative Commons Attribution License.Acute myeloid leukemia (AML) is a heterogeneous hematological malignancy whose incidence is growing in developed countries. In the relapse setting, very limited therapeutic options are available and in most cases only palliative care can be offered to patients. The effect of a composite formulation that contains several antioxidants, Ocoxin Oral solution (OOS), was tested in this condition. When analyzed in vitro, OOS exhibited anti-AML action that was both time and dose dependent. In vivo OOS induced a ralentization of tumor growth that was due to a decrease in cell proliferation. Such effect could, at least partially, be due to an increase in the cell cycle inhibitor p27, although other cell cycle proteins seemed to be altered. Besides, OOS induced an immunomodulatory effect through the induction of IL6. When tested in combination with other therapeutic agents normally used in the treatment of AML patients, OOS demonstrated a higher antiproliferative action, suggesting that it may be used in combination with those standard of care treatments to potentiate their antiproliferative action in the AML clinic.This work was partially supported by Catalysis S.L. as well as the Ministry of Economy and Competitiveness of Spain (BFU2012-39151).     Los programas de formación ocupacional: una alternativa al término de la educación obligatoria

    This paper analyzes the knowledge of pupils finishing the compulsory education about the program of professional qualification. We present some results and conclusions from a study carried out in the Principality of Asturias, with 2,260 pupils, who are in the 4th year of Secondary Compulsory Education, about their academic and professional expectations for the future. The results obtained allow us to conclude that pupils haven’t got enough information about these programs, regardless of where they live. So we believe that this situation would be able to change if both school centres and local agents together develop a proposal of vocational counselling
